Description

b. その結晶表面にアンカー物質の層を適用する工程と、
c. その結晶表面上に、化学吸着されたアンカー層を形成する工程と、を含む。
a. シリケート、ジルコン酸塩またはチタン酸塩の溶液を圧電材料の結晶表面に適用し、酸と反応させて、シリカ、ジルコニア、またはチタニアのそれぞれの中間層を形成する工程と、
b. アンカー物質の層を中間層に適用して、その結晶表面上にアンカー層を形成する工程と、を含む、プロセス。
a. 直接圧電結晶の表面に結合したアンカー物質の層を含む圧電圧結晶を含み、ここにおいて該層内のアンカー物質は、捕捉試薬にも結合し、該捕捉試薬は、アンカー物質の特異的結合パートナーを含むかまたは構成し、かつ生物学的流体中に存在する分析物を特異的に識別し、および、

上記のバイオセンサー構成要素を、捕捉試薬を含む組成物に接触させる工程を含み、ここにおいてこの捕捉試薬は、アンカー物質の特異的結合パートナーを含むかまたは構成し、かつ分析物を特異的に識別し、かつ
その捕捉試薬を、アンカー物質と結合させ、捕捉試薬の層を形成する工程を含み、かつ
その結合した捕捉試薬層を生物学的流体サンプルと接触させる工程と、圧電性の表面に弾性波を発生させる工程と、その捕捉試薬層に結合している分析物の結果として生じる波の振幅、位相または周波数の何らかの変化を測定する工程と、を含む。
次に示す用語は、それらに帰する意味を以下のように有するものとする。
「アンカー物質」は、(i)圧電結晶(「直接」結合について)または、その上の中間コーティング、および(ii)「捕捉試薬」(以下に定義)の両方に結合するコーティング材料を意味する。この用語は、ビオチンを結合できる能力によって機能的に定義されるタンパク質群の一員であるアビジンを含み、これは、それぞれの特異的結合パートナー(例えば、アビジン、ストレプトアビジン、ニュートラアビジン)として機能し、また、特異的結合パートナーを有するオリゴヌクレオチドおよびポリヌクレオチドならびにタンパク質であり、この結合パートナーは、捕捉試薬を改質し、これによって、捕捉試薬を、アンカーコーティングされた圧電材料に結合させる。また、自然に生じる炭水化物結合レクチンを含んでおり、これは、例えば、抗体および抗体フラグメント(例えば、Fcフラグメント)などの炭水化物基と結合する。一般的に、捕捉試薬をアンカーとして使用することは好ましくない。その理由は、捕捉試薬が構造の変化やさらには部分的な変性まで起こす危険性があり、これにより検査の精度に影響を及ぼし得るからである。オリゴヌクレオチドおよびポリヌクレオチドは、イオン性または双極性の部位を介して、直接に、または、適用されている中間銀コーティングを介して(例えばイオン交換法により)、圧電材料に結合することができる。その特異的結合パートナーは相補的なヌクレオチド分子であり、これらは、捕捉試薬を改質するために使用することができる。
本発明は、弾性波センサーの製造に利用できる、全てのタイプの弾性波と全てのタイプ圧電材料二適応できる新しいバイオコーティングのプロセスについて記載している。一実施形態は、バルクモードにおけるそれらの使用を含んでおり、ここにおいて、IDTは、バイオフィルムが堆積された場所の反対側の圧電材料表面上に配置されている。適切な弾性波は、全てのバルクモード、弾性波プレートモードおよび横波型弾性表面波プレートモードを含むことができる。別の実施形態では、関連するバイオコーティングを、表面波および横波型弾性表面波モードの弾性波導波路として利用することが可能であり、これにより圧電面の反対側にIDTをバイオコーティングする必要がなくなる。
このコーティングは、時間単位ではなく数秒または数分単位で実行される、簡単で迅速なコーティング化学反応を含み、このコーティングは、規模拡張可能で、連続的なインライン方式を使用して製造され、最小限のオペレータの介入で簡単に自動化され、不良品の数が少なく、そして少量の生成有害廃棄物を発生する。このコーティング方法は、中間層材料なしに、アンカー物質を圧電表面上に直接堆積する。
一度コーティングされると、安定したアンカー層は、必要に応じて、ホルムアルデヒド蒸気のような可逆的な架橋剤で架橋させることによって、さらに安定化される。ただし、グリオキサールまたはグルタルアルデヒドのような他の相同二官能性架橋剤も利用可能であるが、架橋は、リン酸緩衝液(10%)に最長24時間露出させるか、または標準的な組織化学分野で行われるように、メタノール、イソプロパノールまたはアセトンで短時間溶媒固定することによって、達成される。過剰な架橋は、供給業者の指示に記載されている推奨時間および濃度に従うことによって回避され、これによって、アビジン上の2つまたは3つの残りの露出したビオチン結合部位は影響を受けない。所望により、その結合部位は、以前の可逆的に結合されたデスチオビオチン(Sigma)への露出による崩壊から保護することができ、これは次に、ビオチンまたはビオチン化捕捉試薬によって容易に置換されるこれはまた、ストレプトアビジンのような特定のアンカー物質のための最大70oCの温度で数分間熱固定を行ってもよい。ストレプトアビジンは、最高80 oCで溶液中で安定であることがわかっている(Bang’s Labsによる)。
直接結晶表面をコーティングすることに加えて、本明細書に記載された方法は、結晶表面をコーティングするための、従来技術のプレコーティング(限定されないが)を含む他の材料上に、アビジンをコーティングするのに使用できる。より広い適応も可能である。例えば、直接アビジン被覆の磁気粒子、またはラテックス粒子、あるいは、現在複数のステップで行われる、ラテラルフロー膜などを作ることである。ケイ酸ナトリウム、または二酸化ケイ素は、多くの場合、弾性波導波路として、横波型弾性表面波を保護するために、表面をコーティングするのに使用される。本発明のアビジンの結合方法は、二酸化ケイ素またはガラス表面によく結合し、これによって、導波管面上に重畳することができる。さらに、US 2011/0053139号に教示されるように、そのプロセスは、結合とシリカ導波路層の両方により、酸とシリカNav層に変換される不可逆ガラス質層において、アビジンを捕捉する際に適用される。他の実施形態では、例えば、銀(表面に結合して黒くなる)、銅、金、または、鉄などの結晶表面上に被覆された金属イオンは、本明細書に開示されるプロセスのための間接的なコーティングのための基質を提供することができる。銀または金の塩も結合して、表面結合チオールおよびジスルフィドのタンパク質(IgG、およびアビジンなど)を形成している。
いくつかの実施形態では、結晶表面上に結合したアビジンは、目的の分析物を結合するための活性化を必要とする。活性化には、抗体などのビオチン化結合剤が含まれており、その目的の分析物抗原に特異的である。その抗体または他の薬剤は、アビジン被覆チップに固定する前に、ビオチン化される。抗体は、アビジン基質に固定された後、またはそれより前に、分析物抗原に結合することができる。分析物ビオチン化抗体複合体が、センサーの外側に形成され、次いで複合体がセンサーと接触させることができ、それによって、抗体上のビオチンがアビジン被覆チップに結合する。2つの方法のどちらが好ましいかは、分析物とそのサンプル処理のプロセスに依存している。両方の方法は、本発明の範囲内にある。チップ表面でアビジンに結合している、特定の抗体でコーティングされた表面の分析は、6〜9nmの深さであることが測定された。再度、AFMを用いた際、実際に抗体はアビジン層に結合していることを示した。(図2参照)
本発明によると、特異的に対象の分析物に特異的に結合する捕捉剤を含有する、適切で正しいバイオフィルムコーティングが備わった時に、バイオセンサーは、種々の薬剤および生化学的マーカーを検出するために、容易に大量製造することができる。この統合されたバイオセンサーの使用例としては、ヒトおよび動物の診断が含まれる。分析物とは、感染因子の任意の物質、また、感染因子の中に発見される任意の物質、または、感染因子によって生成され、検出に使用することができる物質であると定義され、これには、オリゴヌクレオチド、核酸、タンパク質、ペプチド、病原体断片、溶解した病原体、および抗体(IgA、IgG、IgM、IgE)、酵素、酵素補因子、酵素阻害剤、毒素、膜受容体、キナーゼ、プロテインA、ポリU、ポリA、ポリリシン、多糖類、およびキレート剤が挙げられるがこれらに限定されない。D抗原-抗体相互作用の検出は、以前に記載されている(米国特許第4236893号、同第4242096号、および同第4314821号、これらは全体が本明細書中に参考として明示的に組み込まれる)。また、細胞全体(原核生物(病原性バクテリアなど)、真核生物細胞(哺乳類の腫瘍細胞を含む))、ウイルス(レトロウイルス、ヘルペス・ウイルス、アデノウイルス、レンチウイルスなどを含む)、真菌、寄生虫および胞子、感染因子の表現型バリエーション(例えば、血清型)の検出における応用は、本発明の範囲内である。
多くの圧電材料は、制限なしに、本発明での使用に適している。また、弾性表面波(SAW)、バルク弾性波(BAW)モードのいずれかにおける弾性表面波の伝搬のために、特に適している多様な結晶方位で、市販されている全てのサイズの研磨されたウェハーが、制限なく含まれる。それは結晶両面に研磨することが好ましいが、単一の研磨された材料はまた、ある用途に適している。変化する結晶方位は全て、本発明の範囲内に包含される。また、適切なものとされるのはランガサイト結晶である。例としては、限定されるものではないが、マグネシウムニオブ酸鉛/チタン酸鉛(PMN-PT)、ジルコン酸鉛、ニオブ酸/チタン酸鉛(PZN-PT)、ニオブ酸リチウム(LiNbO3)、ドーパントを有するニオブ酸リチウム、四ホウ酸リチウム(Li2B4O7)、タンタル酸リチウム、石英である。チタン酸バリウム(BaTiO3)は、室温用途のための、圧電性結晶の非鉛源でもある。
圧電結晶共振器では、弾性波が結晶のバルク内(すなわちバルクモード)または結晶の表面上を移動し、この両方とも、いくつかの形態で発見されている。バルク弾性波(「BAW」)は、液晶媒体を通って移動し、一般的に使用されるBAWデバイスは、厚みすべりモード(TSM)、弾性波プレートモード(APM)、および剪断水平プレートモード(SH-APM)がある。一般的に使用されるSAWデバイスは、剪断水平表面弾性波(SH-SAW)、表面横波(STW)とラブ・ウェーブ(LW)が含まれる。バイオセンサーにおいては、これらの波が減衰されるべきではなく、さもなければ、必要な感度で生体液中の分析物を検出する能力を失う。波型に関係なく、あまり波を減衰させることなく質量変化の結合を維持することは、センサーの開発をする際、重要な問題である。本発明では、すべてのこれらの波モードを考慮しながら、最終的なモード選択をする際、流体と接触した時に波を減衰させることのない生物学的な質量変化に関して、非常に敏感である必要がある。そのセンサーの構成も、この目標を達成するのに重要である。このように、本発明のセンサーの構造とそれぞれに使用される電波の性質は、最適化の対象となっている。
本発明のさらなる実施形態は、様々な波モードと拡張機能を利用している、単一のセンサーや複数のセンサーを含む、唯一の使い捨てカートリッジシステムの製造である。各カートリッジは、最小、あるセンサー、センサーをプセル化するシステム、そして、流体カートリッジ内の流体を処理するためのシステムを最低でも搭載している。センサーの利用は、リーダシステムを使った直接有線接触または無線モードを介してのいずれであってもよい。
これらの方法の全ては、実際に行われている。
a. 固定なしのコーティング:プラズマ発生装置(例えば、Plasma Treat USA、イリノイ州)を用いて、大気圧で約1〜10秒間、LiNbO3の表面を清掃し活性化させる。次に、0.5mg/mlの濃度のアビジンで、PBS中50%グリセロールを含む溶液中のアビジンをインクジェットすることによって、アビジンを適用する。その後、50oCでヒートガンからの温風を用い短時間乾燥させるか、または、約30分間室温で乾燥させて、表面にアビジンを結合させる。これで、バイオセンサーは、現在使用できる状態、またはパッケージングの準備ができた状態となる。
任意選択的に、ニュートラアビジンの堆積は定着工程へと続く。
a. ニュートラアビジン・バイオセンサーのための自動化された直接コーティングモード:
いくつかの実施形態のオンライン製造は、バイオセンサー・ハウジングの下部に固定された圧電結晶上にニュートラアビジンコーティングを堆積させ、次いで、コンベヤーベルトに取り付けることを伴う。プラズマ処理後、センサーは、ニュートラアビジンのインクジェットステーションに移動され、0.5センチメートル平方面積当たりの重量で、0.01〜5.0mg/ml、または0.01〜2.0 mg/ml、または0.01〜1.0 mg/mlのニュートラアビジンを含む流体5〜10μlで被覆される。ヒートガンからの熱(50oC程度)を短時間曝露することによって、流体は蒸発し、ニュートラアビジンは、バイオセンサー上に固定される。ハウジングの上カバーは、その後、バイオセンサーカートリッジの漏れ防止の流体チャンバーを形成するように固定されている。
上記2a同様のインクジェット方式を使用して、室温で、0.01M PBS -0.1% BSA、pH7.4、1〜10%トレハロース、1〜5%グリセロール、0.05%アジ化ナトリウムの非乾燥安定化ゲルマトリックス中の適切な希釈度に希釈したビオチン化抗体をコーティングする。続いて、約1分間のヒートガンからの暖かい空気によって部分的に蒸発させる。バイオセンサー上のゲルコーティングは、表面上に残され、プラスチックパウチの中でその後保管される間に、固定化されたニュートラアビジンとの反応が完了する。
実施例2bの工程が行われるが、代わりに、ビオチン化オリゴヌクレオチドのゲルマトリックス中に希釈される。
本実施例の自動化されたコーティングプロセスは、従来のタイプのセンサー(すなわち、中間コーティングを有するもの)の上、ならびに、中間酸化物コーティングが実施の形態に係る単純な二段階プロセスで適用されているセンサー上でも実施できる。
実験は、女性と男性の両方で性感染症の原因となる偏性細胞内細菌C.trachomatisを用いて実施した。アビジンおよび抗体をコーティングするために使用される手順は、実施例1に記載されている。使用した抗体は、ビオチン化したC. trachomatis(Abcam)の主要な外膜(MOMP)に対するモノクローナル抗体であり、0.25μg/mlで使用された。C.trachomatisはMicrobiaから入手し、1ml当たり105の濃度のPBS/緩衝液に懸濁し、同様の実験では、モノクローナル抗体は、C.trachomatis素体のリポ多糖(Medix)に対して実施された。一定分量の基本小体(EB)、105EBと0.25μg/mlのビオチン化抗EB LPSが、30分間インキュベートされた後、チップに添加された。図6および7は、これらの特定の感染性抗原を用いて行った実験の結果を示す。図6に、ニュートラアビジンに結合した抗体と結合したC. trachomatis素体をもつAMTチップを撮影した蛍光顕微鏡用スライドの黒と白のトレーシングの一連を示している。セクションAはネガティブコントロール(アビジンのみで、抗体を持たないチップ)の図を描いており、現れたエージェントは、セクションBの核酸DAPI、およびセクションCの反-反トラコーマ症抗体FITC標識抗である。この実験によって、チップ上の細胞物質の存在を確認され、抗原は固定化抗体に結合し、結果、あるアビジンアンカーと結合したことを示している。
アビジンと抗体を覆うのに用いられる手順は、例1で記述される。図8は、PBS緩衝液に懸濁した場合に、デング熱ウイルスを用いて行った実験のための周波数の変化を示す。デング熱ウイルスは、デング熱の原因となるあるフラビウイルスであり、毎年約100万人に影響を与えている。これは、都市部の住宅にいる蚊、Aedipus egyptiiによって伝染される。4つの血清型が知られているが、一種の血清型のみが時間内のいずれの点でも感染症を引き起こす。血清型は、数年の間に変更することができる。血清型2は、現在知られている最も一般的な血清型である。デングウイルスのための実験は、0.05%のBSA-PBS中で4μg/mlの2抗体(Feldan)の血清型2に対するモノクローナルエンベロープタンパク質を用いて、室温で30分間インキュベートされた。PBS洗浄によって、過剰の抗体を除去した。デング2型抗原は、Microbixから入手した。ウイルスは、市販のストック(これは急性感染条件下での人間のウイルス血症の程度に似ている)の標準(1X)、PBS/緩衝液(1mlあたり104 または8μg/ml)を濃縮したものに懸濁した。弾性波プレートモードセンサーおよび波発生器は、実施例3と同様とした。抗体調製物の適用および付着して、デングウイルス抗原を塗布した後、500メガヘルツの周波数のシフトが、ベースライン緩衝液(対照)とセンサーとの間に観察された。最後の洗浄にもかかわらず、周波数のシフトは、残っており、抗原が弾性波プレートモードセンサーの周波数の変化に責任があったことを示した。その結果は、時間に対するピーク周波数のプロットである、図8に示されるように、周波数シフトを実証する 。2つのPBS(1×)ベースライン測定は、チップの安定性を、1時間ごとに確認するために行った。デングウイルスは、1時間の間、第3セグメント内のチップに添加した。第4のセグメントは、チップは、余分なPBSで激しく洗浄した後に測定された。周波数シフトは、ウイルスが、チップに結合したことを示す、278.62ヘルツの値を得るために、ベースライン1から最終洗浄を差し引くことにより決定した。
図9は、周波数対時間のグラフであり、チップを抗体に接触される前および後に見られる周波数シフトを示す。この抗体をコーティングするために使用されるプロセスは、実施例1に記載されている。センサーは、実施例3と同じであった。矢印1はアビジンが結合された時にセンサーから受信した周波数を示している。矢印2は、抗体が適用された後のセンサーから受信した周波数を示している抗体は、ビオチン化したC.trachomatisの基本小体に対するポリクローナル抗体であり、1:200希釈で使用された。(Abcam, Cambridge, Massachusetts-AV20387)。100メガヘルツの変化が見られる。そのシフトは、チップへの抗体の添加により生じたものである。
Claims (28)
- 圧電材料と、前記圧電材料の表面に、直接かつ不可逆的に、化学吸着的に単層として結合しているアビジンアンカー物質の層と、を含む弾性波バイオセンサー構成要素であって、前記アビジンアンカー物質が、ビオチン化捕捉試薬と結合する特性を備えている、弾性波バイオセンサー構成要素。
- 前記アビジンアンカー物質が、アビジン、ニュートラアビジン、又はストレプトアビジンを備える、請求項1に記載のバイオセンサー構成要素。
- 前記圧電材料が、ランガナイト結晶、マグネシウムニオブ酸鉛、チタン酸鉛、ジルコン酸ニオブ酸鉛、チタン酸鉛、ニオブ酸リチウム、ドーパントを備えたニオブ酸リチウム、四ホウ酸リチウム、タンタル酸リチウム、石英、チタン酸バリウム、ベルリナイト、オルトリン酸ガリウム、ニオブ酸カリウム、ジルコンチタン酸バリウム、ランタンカルシウムオキシボレート、ランガサイト結晶、ランタンガリウムケイ酸塩、セラミックペロブスカイト構造、ビスマスフェライト、チタン酸ジルコン酸鉛、硫化カドミウム、酸化亜鉛、ガリウム砒素、ビスマス及びゲルマニウム酸化物、窒化アルミニウム、並びにポリフッ化ビニリデンからなる群から選択される、請求項1に記載のバイオセンサー構成要素。
- 前記圧電材料がニオブ酸リチウムである、請求項1に記載のバイオセンサー構成要素。
- ハウジング及び流体工学チャンバーをさらに含み、前記アビジンアンカー層を有する圧電材料の表面が、該チャンバーの壁をなす、請求項1に記載のバイオセンサー構成要素。
- 弾性波発生器及び弾性波受信器をさらに含む、請求項1に記載のバイオセンサー構成要素。
- 前記波動発生器がバルク弾性波又は表面弾性波を発生させる、請求項6に記載のバイオセンサー構成要素。
- 前記バルク弾性波が、厚みすべりモード、弾性波プレートモード、及び水平プレートモードからなる群から選択される、請求項7に記載のバイオセンサー構成要素。
- 前記表面弾性が、剪断水平表面弾性波、表面横断波、及びラブ波からなる群から選択される、請求項7に記載のバイオセンサー構成要素。
- アビジンアンカー物質であって、ビオチン化捕捉試薬と結合する特性を有するアビジンアンカー物質を含むバイオフィルムで圧電材料の表面をコーティングするプロセスであって、該プロセスが:
a.結晶表面の表面エネルギーを増加させるために、前記圧電材料の結晶表面を処理する工程と、
b.前記結晶表面に前記アビジンアンカー物質の層を適用する工程と、
c.前記結晶表面上に、化学吸着されたアビジンアンカー物質の層を形成する工程と、
を含む、プロセス。 - 前記処理工程がプラズマ処理を含む、請求項10に記載のプロセス。
- 前記プラズマ処理が、約5から10秒間の大気圧プラズマジェット流への曝露を含む、請求項11に記載のプロセス。
- 前記圧電材料が、ランガナイト結晶、マグネシウムニオブ酸鉛、チタン酸鉛、ジルコン酸ニオブ酸鉛、チタン酸鉛、ニオブ酸リチウム、ドーパントを備えたニオブ酸リチウム、四ホウ酸リチウム、タンタル酸リチウム、石英、チタン酸バリウム、ベルリナイト、オルトリン酸ガリウム、ニオブ酸カリウム、ジルコンチタン酸バリウム、ランタンカルシウムオキシボレート、ランガサイト結晶、ランタンガリウムケイ酸塩、セラミックペロブスカイト構造、ビスマスフェライト、チタン酸ジルコン酸鉛、硫化カドミウム、酸化亜鉛、ガリウム砒素、ビスマス及びゲルマニウム酸化物、窒化アルミニウム、並びにポリフッ化ビニリデンからなる群から選択される、請求項10に記載のプロセス。
- 前記圧電材料がニオブ酸リチウムである、請求項10に記載のプロセス。
- 前記適用工程が、前記表面層上に前記アビジンアンカー物質を噴霧又は接触転写させて、該表面に薄い均一な液膜を形成する工程を含む、請求項10に記載のプロセス。
- 前記薄い均一な液膜がマイクロドットである、請求項15に記載のプロセス。
- 前記アビジンアンカー物質が、アビジン、ニュートラアビジン、又はストレプトアビジンを備える、請求項10に記載のプロセス。
- 前記適用工程が、さらに、前記アビジンアンカー物質の層を乾燥させる工程を含む、請求項15に記載のプロセス。
- 前記の結合したアビジンアンカー物質の層を、生物学的流体中の分析物を特異的に識別する特性を有するビオチン化捕捉試薬を含む組成物に接触させる工程と、
前記ビオチン化捕捉試薬を前記アビジンアンカー物質に結合させる工程と、
をさらに含む、請求項10に記載のプロセス。 - 弾性波バイオセンサーであって、
a.直接かつ不可逆的に、化学吸着的に単層として圧電材料の表面に結合したアビジンアンカー物質の層を含む、バイオコートされた圧電材料と、
b.弾性波発生器と、

前記層における前記アビジンアンカー物質が、ビオチン化捕捉試薬にも結合し、
前記ビオチン化捕捉試薬が、生物学的流体中に存在する分析物を特異的に識別し、
前記発生器が波を発生し、前記ビオチン化捕捉試薬と分析物との間の反応が、弾性波の特性に検出可能な変化を引き起こす、
弾性波バイオセンサー。 - 前記結晶がニオブ酸リチウムである、請求項20に記載のバイオセンサー。
- 前記分析物が、全細胞、細菌、真核生物細胞、腫瘍細胞、ウイルス、真菌、寄生虫、及び胞子、並びにこれらの任意のものの断片、タンパク質、核酸、及び毒素からなる群から選択される、請求項20に記載のバイオセンサー。

前記圧電性の表面に弾性波を発生させる工程と、
前記ビオチン化捕捉試薬層に結合している分析物の結果として生じる波の振幅、位相又は周波数の任意の変化を測定する工程と、
