Description
の化合物を提供する。
本実施例では、実施例1のPEG試薬1を以下の試薬、即ち、上記CongらのMPEGがCH3O.(CH2CH2O)m-1-CH2CH2-であるPEG試薬2と比較した。
複合体の調製:IFNα-2a(6.5mg、0.845mg/mL)のpH7.4の50mMリン酸ナトリウム緩衝液(150mMのNaClと20mMのEDTAを含有)溶液を調製した。このタンパク質溶液を緩衝液(313μL)で希釈し、次いで1.0mMのDTT水溶液(187.5mL)を添加して最終DTT濃度25mM及び反応物体積7.5mLとした。穏やかに混合した後、反応物を室温にて30分間放置した。還元したタンパク質をPD-10カラムを用いてpH7.4の50mMリン酸ナトリウム(150mMのNaClと20mMのEDTAを含有)へと緩衝液交換した。溶出したタンパク質溶液を遠心分離(3000g、4℃、5分)にかけ、次いで上澄みを280nm(0.532mg/mL)でのUV吸光度測定によって定量化した。このタンパク質溶液を緩衝液で0.10mg/mLに希釈した。PEG1及びPEG2を20mg/mLの割合でpH7.4の50mMリン酸ナトリウム(150mMのNaClと20mMのEDTAを含有)に溶解させた。2つのバイアルそれぞれに還元IFNα-2a(2.5mg、24.8mL)を充填し、一方のバイアルにPEG試薬1(4.9mg、0.245mL)を添加し、他方のバイアルにPEG試薬2(4.9mg、0.245mL)を添加した。反応物を穏やかに混合し、次いで4℃にて18時間放置した。最終反応混合物中の還元タンパク質を5mg/mLの硫酸銅(12.18μL)と50:50(mM)のGSH/GSSG(0.25mL)とを順次添加することによって全て酸化させた。再酸化反応を4℃にて一晩実施した。反応混合物をpH4の100mM酢酸ナトリウムで4倍に希釈し、次いで陽イオン交換クロマトグラフィー(Macrocap(商標)SP)により、0.60〜0.65MのNaClにて溶出する所望の複合体を用いたpH4の100mM酢酸ナトリウム(1.0MのNaCl)の段階的勾配溶出を使用して精製した。
ジスルフィドを還元したpH7.3のIFN-β-1b(9.5mg、0.3mg/mL)に、pH7.3のPEG試薬1(1.7mL、20mg/mL)の溶液を添加した。得られた溶液を22℃にて4時間インキュベートし、直後に粗反応混合物をSDS-PAGEによって分析した。ゲルはInstantBlue(商標)で染色し、IMAGEQUANT(商標)LAS 4010装置を用いて画像化した。結果を図5に示す。図5では、レーンMはNovexタンパク質スタンダードであり、レーン1は開始時のIFN-β-1bであり、レーン2は還元したIFN-β-1bであり、レーン3はPEG試薬1とIFN-β-1bとの反応混合物である。SDS-PAGE分析から、生成物帯域可視レベルが110kDaタンパク質スタンダードと同じIFN-β-1bのPEG化が成功したことがわかる。
Claims (17)
- Yが-NH-CO-を表す、請求項1に記載の化合物。
- pが3である、請求項1又は2に記載の化合物。
- 各Lが独立して-SR、-SO2R、-OSO2R、-N+R3、-N+HR2、-N+H2R、ハロゲン、又は-Oφを表し、式中、Rが水素原子又はアルキル、アリール若しくはアルキル-アリール基を表し、φが少なくとも1つの電子吸引基を含む置換アリール基を表す、請求項1から3のいずれか一項に記載の化合物。
- 各Lが独立してフェニルスルホニル又はトシルを表す、請求項4に記載の化合物。
- 各Xが独立して式CH3O-(CH2CH2O)m-のポリエチレングリコールを表し、式中、mがX中の酸化エチレン単位(-CH 2 CH 2 O-)の数である、請求項6に記載の化合物。
- Yが-NH-CO-を表す、請求項8に記載の化合物。
- pが3である、請求項8又は9に記載の化合物。
- Prが、前記タンパク質若しくはペプチド中のジスルフィド結合に由来する2つの硫黄原子に、又は前記タンパク質若しくはペプチドに付加したポリヒスチジンタグ中に存在する2つのヒスチジン残基に結合した単一のタンパク質又はペプチドを表す、請求項11に記載の化合物。
- PrがIgGのFab断片、INF-α、IFN-β、又はコンセンサスIFNを表す、請求項11又は12に記載の化合物。
- AがCOを表す、請求項8から13のいずれか一項に記載の化合物を調製する方法であって、請求項1から7のいずれか一項に記載の化合物をタンパク質又はペプチドと反応させ、Aが-CO-を表す一般式IIbの化合物を製造する工程を含む、方法。
- 一般式IIbの化合物における基Aである-CO-基を還元して、AがCHOH、CH-NH 2 、CH-NHR、CH-NR 2 、CH-OR、CH-O-C(O)R、CH-NHC(O)R、又はCH-N(C(O)R) 2 基を表し、各Rは水素原子又はアルキル、アリール若しくはアルキル-アリール基を表す、一般式IIbの化合物を形成するさらなる工程を含む、請求項14に記載の方法。
- 請求項1から7のいずれか一項に記載の化合物を含む、医薬組成物。
- 療法に使用される、請求項1から7のいずれか一項に記載の化合物又は請求項16に記載の組成物。
