Description
チロシン--->ドーパ--->ドパキノン--->ドパクロム--->メラニン
a)CB1受容体アンタゴニストを予選択するステップと
b)CB1受容体アンタゴニストと接触させたメラニン生成細胞により生成されるメラニンの量が、このCB1受容体アンタゴニストと接触させていないメラニン生成細胞により生成されるメラニンの量よりも有意に少ないまたは等しいときに、白色化剤および/または抗褐変剤として前記アンタゴニストを最終選択するステップと
を含む方法である。
1.材料および方法
1.1.試験する化合物
アナンダミド(AEA)およびメトアナンダミド(mAEA)が、Sigma Chemical Co.(セントルイス、MO)から供給される。
化合物SR1=N-ピペリジノ-5-(4-クロロフェニル)-1-(2,4-ジクロロフェニル)-4-メチル-3-ピラゾールカルボキサミド(特異なCB1受容体アンタゴニスト)が、Sanofi-Aventis Recherche(フランス、モンペリエ)から供給される。
Promocell(ドイツ、ハイデルベルグ)から供給される、包皮から単離された、健常なヒトの表皮性メラニン生成細胞(NHEM)を、適当な培地M2(Promocell)において、5%CO2の湿潤環境中37℃で24時間培養する。
NHEM細胞中に存在するメラニンの量を、当業者に公知の方法(Xiaoら、Arch Dermatol Res、2007年、299、245〜57頁)で測定する。この方法にしたがって、NHEM細胞をトリプシンで処理し、リン酸緩衝生理食塩水で2回洗浄する。次いで、該試料をMilliQ水200μl中で再懸濁させ、メラニン以外の不透明物質を除去するために、エタノール:エーテル混合物1ml(1:1体積/体積)を加える。15分後、該試料を、600gで5分間、遠心分離機にかける。該ペレットを空気乾燥して1N水酸化ナトリウム200μl中に溶解させる。次いで、該試料を80℃で1時間加熱して冷却する。
所与の結果は、3つの独立した試験方法それぞれを3回実施した、標準偏差による平均値である。それらを平方偏差の分析(ANOVA)により比較した。
2.1.試験した化合物の脱色素効果の実証
これらの結果を鑑みると、化合物SR1は、したがって、皮膚用の白色化剤または抗褐変剤として使用することができる。
脱色素または白色化するクリーム
色素沈着表皮を、健常なヒトのメラニン生成細胞およびケラチン生成細胞を用いて、コラーゲン支持体[BPER(再構築された表皮のためのバイオドレッシング)]において再構築させる。
次いで、メラニンの顕微鏡定量化による色素沈着の定量化を実施して、試験化合物の色素沈着促進活性が例証可能となるようにする。
Claims (7)
- ケラチン物質用の白色化剤および/または抗褐変剤としての、CB1受容体アンタゴニスト5-(4-クロロフェニル)-1-(2,4-ジクロロフェニル)-4-メチル-N-(ピペリジン-1-イル)-1H-ピラゾール-3-カルボキサミドの、皮膚および/もしくは粘膜への局所塗布、または経口投与を含む、非治療的な、化粧品としての使用。
- 皮膚用および/または粘膜用の白色化剤としての、請求項1に記載の使用。
- 皮膚および/または粘膜の色素沈着を低減させる作用剤としての、請求項1または2に記載の使用。
- メラニン形成を抑制する作用剤としての、請求項1から3のいずれか一項に記載の使用。
- 請求項1に規定されるCB1受容体アンタゴニストを生理的に許容される媒質中に含む組成物の、皮膚および/もしくは粘膜への局所塗布、または経口投与を含む、皮膚および/または粘膜を白色化する非治療的な化粧方法。
- 前記CB1受容体アンタゴニストが、組成物の総質量に対して、0.00001質量%から10質量%の間の濃度で存在することを特徴とする、請求項5に記載の方法。
- ケラチン物質用の白色化剤および/または抗褐変剤を選択する方法であって、
a)CB1受容体アンタゴニスト5-(4-クロロフェニル)-1-(2,4-ジクロロフェニル)-4-メチル-N-(ピペリジン-1-イル)-1H-ピラゾール-3-カルボキサミドを予選択するステップと
b)前記CB1受容体アンタゴニストと接触させたメラニン生成細胞により生成されるメラニンの量が、このCB1受容体アンタゴニストと接触させていないメラニン生成細胞により生成されるメラニンの量よりも有意に少ないまたは等しいときに、白色化剤および/または抗褐変剤として前記アンタゴニストを最終選択するステップと
を含む、方法。
