以下に他の定義付けを行わない限りにおいて、本願に使用する全ての技術的および科学的用語は、本発明が属する技術分野の当業者によって一般的に理解されるものと同一の意味を有するものである。
組み合わせBMPのインビトロおよびインビボ機能の特徴を決定するアッセイ法は当該技術分野において周知である(米国特許第4,761,471号;米国特許第4,789,732号;米国特許第4,795,804号;米国特許4,877,864号;米国特許第5,013,649号;米国特許第5,166,058号;米国特許第5,618,924号;米国特許第5,631,142号;米国特許第6,150,328号;米国特許第6,593,109号;Clokie and Urist, Plast. 多様なアッセイ法を使用して、担体への組み換えBMPの結合および担体からの組み換えBMPの放出を測定することができる。例えば、当該技術分野に周知の任意の技術によって組み換えBMPのタンパク質の量を定量化することができる。上記技術としては、ドットブロット法、免疫学的検定法(例えば、酵素免疫測定法(ELISA:enzyme linked immunosorbent assay)、生体インプラントに放射性標識BMPを取り込んだ場合に放出バッファ中に存在する放射能増加の測定法、およびクロマトグラフィー法(例えば、高圧液体クロマトグラフィー法(HPLC:high pressure liquid chromatography)およびイオン交換クロマトグラフィー法)が含まれる。
一態様では、本発明は、本明細書に記載のシステムを収容するためのキットを提供する。一態様では、上記キットは、送達媒体および担体を作製するために必要な要素、並びに必要とされる増殖因子を備える。すなわち、本発明のキットは、送達媒体および担体を作製するために必要な要素、前記送達媒体に関連付けられる少なくとも1つの増殖因子、および前記担体に関連付けられる少なくとも1つの増殖因子を備えることになる。
以下の実施例を用いて本発明を説明する。以下の実施例は、増殖因子(BMPを持続放出で放出する担体中のBMPの付加部分とBMPを初期放出で放出する送達媒体内のBMPの一部とから生成されるrhBMP‐2など)の多相放出特性は急速放出(バーストリリース)または持続放出のみを実現する担体よりも効果的であるという、本発明者による新規発見を例示するものである。
本実施例では、rhBMP‐2を含み、持続放出特性で増殖因子を放出する担体を形成する方法を示す。
固有粘度が、1.33 dL/g のPLGA 75/25 (MW=205,000-210,000) をBirmingham Polymer Inc.(Birmingham、AL)から購入した。リン酸テトラカルシウム(TTCP)を太平化学産業株式会社(大阪、日本)から得て、第二リン酸カルシウム無水物(DCPA)およびジメチルスルホキシド(DMSO)をSigma Chemical Co. (MO、USA)から得た。糖粒子をTate & Lyle North America Inc. (Toronto、Canada)から購入した。
本実施例では、持続放出特性を有する、rhBMP‐2を含むリン酸カルシウムセメント(CPC)担体の形成方法を例示する。
リン酸テトラカルシウム(TTCP)を太平化学産業株式会社(大阪、日本)から得、第二リン酸カルシウム無水物(DCPA)をSigma Chemical Co. から得た。マクロ多孔性の二相性リン酸カルシウム顆粒(Eclipse)をCitagenix(Laval Qc、Canada)から購入した。組み換えヒトBMP‐2(rhBMP‐2、Induce biologics Inc.)を製剤バッファ(1.5 mg/ml、pH 4.5;5 ミリモルのグルタミン酸、2.5 %のグリシン、0.5 %のサッカロース、およびddH2Oを添加した0.01 %のTweenTM80)中で調製した。
[0093] マクロ多孔性の骨格としても機能するCPC系の持続放出性の担体を作製するために、CPC粒子(0.1 mm〜0.3 mm)をマクロ多孔性のリン酸カルシウム顆粒(1 mm〜2 mm)と1:1(w/w)の比で混合させた。
本実施例では、BMP結合コーティングを塗布して、持続放出特性を有する担体を形成する方法を示す。このような方法の1つでは、本発明者による同時係属中の出願である米国出願第13/002,444(前記米国出願を参照することにより、その開示内容全体を本明細書中に援用する)に記載の抗体またはBMP結合タンパク質で骨格をコーティングする。
精製した多クローン性ラビット抗ヒトBMP‐2抗体をCell Sciences(Canton MA.、カタログ#PA0025)から購入した。マクロ多孔性の二相性リン酸カルシウム(BCP)の顆粒(Eclipse)をCitagenix(Laval、Qc、Canada)から購入した。
[0098] 殺菌済のBCPの顆粒をバイオセーフティーキャビネット内で検量し、殺菌済のTPP管(Mandel Scientific、Guelph ON、Canada)内に投入した。抗体溶液をリン酸緩衝生理食塩水中で希釈して1 mlのPBS当たりの抗体の最終的な濃度を150 ng、300 ng、および600 ngとし、濾過滅菌し、上記骨格に1:1 (v/v)の比で塗布した。上記サンプルを凍結させて凍結乾燥させる前に、室温で少なくとも15分間撹拌させた。その後、上記顆粒にBMP溶液を塗布し、室温で少なくとも15分間吸収させ、その後に凍結させて再度凍結乾燥させた。
本実施形態は、F127を使用して、rhBMP‐2を含む送達媒体を調製する方法を示す。
以下の要領でポロキサマーを調製した。100 mlの蒸留水を4 ℃まで冷却させ、多様な量のポロキサマー407を撹拌させながら数時間掛けて徐々に添加させ、固体小球が全て溶解して最終濃度が12 %〜33 %の範囲となるようにした。その後、上記ポロキサマーの溶液を加圧滅菌器で殺菌(121 ℃、20分間、30 psi)した。殺菌後、上記ポロキサマーの溶液を使用時まで4 ℃で静置した。
本実施例では、rhBMP‐2を含み、これを多相放出特性で放出する3PS‐F127生体インプラントを形成する方法を示す。
担体5 mg当たりrhBMP‐2を0 μg、4.55 μg、または9.1 μg含む3PS担体(実施例1に記載)を調製してエッペンドルフチューブ内に貯蔵した。45.5 μlのF127(実施例4に記載の方法で調製)内に0 μg、4.55 μg、または9.1 μgのrhBMP‐2を含む送達媒体をエッペンドルフチューブ内に4℃で貯蔵した。使用直前に、上記F127を上記3PS担体上にピペットで移し、この担体を上記送達媒体内に混合させた。
本実施例では、実施例1〜5に記載の多様な生体インプラントからのrhBMP‐2の放出を測定する方法を記載する。
実施例1〜5の場合のように調製したrhBMP‐2を既知量含む生体インプラントをエッペンドルフチューブに移す。使用するrhBMP‐2の総量は、担体が5 mgおよびF127が45.5 μl当たり9.1 μgのrhBMP‐2、または100 μlのF127に対する10 mgの担体に対して20 μgのrhBMP‐2である。
BMPを含めなかった生体インプラントから回収した放出バッファでは何れも、検出可能なBMPが存在しなかった。rhBMP‐2を含めた上記担体のサンプルは、研究期間に渡ってrhBMP‐2の持続放出を示す一方で、上記送達媒体中のサンプルのみが「初期放出特性」で放出された。
本実施例は、生体インプラントから放出されたrhBMP‐2が活性を維持しているか否かについて決定する方法を記載する。放出されたrhBMPが生物学的に活性であることを実証するために、応答細胞を放出物中で培養し、増殖因子への応答を測定してもよい。このようなアッセイ法は当該技術分野において公知である(Peel et al., J. Craniofac. Surg. 2003, 14:284-291を参照)。
実施例1〜5に記載のようなrhBMP‐2を添加または無添加の材料を調製した。15 %のウシ胎仔血清および抗体を添加したアルファ最小必須培地(αMEM(alpha minimal essential medium)+15 %FBS(fetal bovine serum)+AB(antibiotics))をバッファに使用した以外は実施例3に記載の方法で放出物を調製した。
本実施例では、BMPを含む生体インプラントの骨誘導活性をインビボで決定する方法を記載する。生体インプラントの骨形成誘導能を評価するために、マウスマッスルパウチアッセイ法を使用した。このモデルでは、マウスの後肢に作製したマッスルパウチ内に生体インプラントを配置する。また、誘導された骨形成のサイズは試験対象のBMP量に比例するものである。このようなアッセイ法は当該技術分野において公知である(例えば、Barr et al., Oral Surg. Oral Med. Oral Pathol. Oral Radiol. Endod., 2010; 109:531-40を参照)。
実施例1〜5に記載のように生体インプラントを調製した。麻酔後、生後37日〜42日の雄のCD‐1マウスの後肢の大腿筋内に鈍的切開法により水平に並んだ2つの各パウチを作製した。その後、上記筋肉のパウチ内に配置されていた無菌状態のゼラチンカプセル内に上記生体インプラントが配置された。上記筋肉内の各パウチは互いに引き合わされ、ミッシェルクリップを用いて皮膚の縫合を行った。
実施例5に記載のように組み合わせられた担体および送達媒体
BMPの持続放出特性を有する3PS担体の内部にBMPが全て含まれる生体インプラントにより最も小さい骨の骨片が形成されること(図4;100‐0)、BMPの急速放出特性を有するF127送達媒体の内部にBMPが全て含まれる生体インプラントにより中間サイズの骨片が形成されること(図4;0‐100)、担体にBMPの50%が含まれ、送達媒体にBMPの50%が含まれ、BMPの多相放出特性を有する生体インプラントにより最も大きな骨の骨片が形成されること(図4;50‐50)が、マイクロCT分析によって示された。
本実施例では、実施例1、2、3、4、または5に記載の多様な生体インプラントを動物に移植後、この生体インプラントからのrhBMP‐2の放出を測定する方法を記載する。上記測定を行う方法は当該技術において周知である。例えば、Uludag et al. J Biomed Mater Res, 46, 193-202, 1999を参照。
組み換えhBMP‐2は、ヨウ素125(I-125)(Perkin Elmer)で放射標識されている。上記放射標識されたhBMP‐2(ホット)を非標識hBMP‐2(コールド)と混合して、1:100のホットおよびコールドの混合物を作製する。
本実施例では、持続性の短い放出特性で増殖因子を放出する担体を作製する手段を説明する。
マクロ多孔性の二相リン酸カルシウム(BCD)の顆粒(Eclipse)をCitagenix(Laval、Qc、Canada)から購入した。組み換えヒトBMP‐2(rhBMP‐2、Induce Biologics Inc.)を製剤バッファ(1.5 mg/ml、pH 4.5;5 ミリモルのグルタミン酸、2.5 %のグリシン、0.5 %のサッカロース、およびddH2Oを添加した0.01 %のTweenTM80)中で調整した。
F127にコーティングされていない担体(BCP)は、初日に放出されるBMPの量が最も大きく、その後の各時間点において放出されるBMPの量は減少するような急速放出特性を示した。F127中にBCPを混合(BCP−Pol)した結果、最初の4日目までの各日において回収したBMP量が類似する持続性の短い放出特性が得られた(図7)。
本実施例では、担体からの放出特性を変化させる手段を説明する。
固有粘度および分子量の異なるPLGAをBirmingham Polymers Inc.社(Birmingham、AL)から購入した。その後、実施例1のように、これらPLGAを使用して担体を作製した。上記担体からのBMPの放出特性を実施例6の方法で決定した。
全担体が持続放出特性を示した。しかし、放出されたBMPの量は、使用したPLGAの固有粘度/分子量に応じて異なるものであった。低粘度のPLGA(Pol‐1)で作製した担体は、高粘度のPLGA(Pol‐2)を用いて作製した担体と比較して、12週間の研究期間中により多くのrhBMP‐2を放出した(図8)。
Claims (20)
- 治療部位で増殖因子を多相放出するための組成物であって、
少なくとも1つの第1の増殖因子を含む送達媒体、および、
少なくとも1つの第2の増殖因子を含む担体を含み、
上記少なくとも1つの第1の増殖因子と上記少なくとも1つの第2の増殖因子とは骨形成タンパク質(BMP)であり、
上記送達媒体は、上記担体に塗布され、上記少なくとも1つの第1の増殖因子の少なくとも80%を72時間の第1の期間内に放出するように構成される、流動性を有する形態の重合体の液体またはゲルであり、
上記重合体はポロキサマー407であり、
上記担体は、上記少なくとも1つの第2の増殖因子を7日間以上の第2の期間に持続放出特性をもって放出するように構成された複数の粒子からなる、組成物。 - 上記担体は、複数のリン酸カルシウム粒子からなる請求項1に記載の組成物。
- 上記少なくとも1つの第1の増殖因子と上記少なくとも1つの第2の増殖因子は同一であるかまたは異なり、上記少なくとも1つの第1または第2の増殖因子は、形質転換増殖因子ベータのスーパーファミリー、インスリン様増殖因子(IGF)、繊維芽細胞増殖因子(FGF)、血小板由来増殖因子(PDGF)、および血管内皮増殖因子(VEGF)から選択される請求項1に記載の組成物。
- 上記少なくとも1つの第2の増殖因子はBMP−2またはBMP−7である請求項1に記載の組成物。
- 上記送達媒体は、上記少なくとも1つの第1の増殖因子の総量の少なくとも10%を送達し、上記担体は、上記少なくとも1つの第2の増殖因子の総量の少なくとも50%を送達する請求項1に記載の組成物。
- 増殖因子の多相送達用のキットであって、
送達媒体の要素、
上記送達媒体と関連付けられ、上記送達媒体によって送達されるように構成された少なくとも1つの第1の増殖因子、
担体の要素、および、
上記担体に関連付けられ、上記担体によって送達されるように構成された少なくとも1つの第2の増殖因子を含み、
上記少なくとも1つの第1の増殖因子と上記少なくとも1つの第2の増殖因子とは骨形成タンパク質(BMP)であり、
上記送達媒体は上記担体に塗布され、上記少なくとも1つの第1の増殖因子の少なくとも80%を72時間の第1の期間内に放出するように構成される、流動性を有する形態の重合体の液体またはゲルであり、
上記重合体はポロキサマー407であり、
上記担体は上記少なくとも1つの第2の増殖因子を7日間以上の第2の期間に持続放出特性をもって放出するように構成された複数の粒子からなる、キット。 - 上記少なくとも1つの第2の増殖因子はBMP−2またはBMP−7である請求項6に記載のキット。
- 上記少なくとも1つの第1の増殖因子は、キットに備えられる増殖因子の総量の少なくとも10%を含み、
上記少なくとも1つの第2の増殖因子は、キットに備えられる増殖因子の総量の少なくとも50%を含む請求項6に記載のキット。 - 上記複数の粒子は、高分子マトリクス内に分散されている請求項1に記載の組成物。
- 上記高分子マトリクスは、ポリ乳酸(PLA)または乳酸−グリコール酸の共重合体(PLGA)を含む請求項9に記載の組成物。
- 上記担体の上記少なくとも1つの第2の増殖因子は、上記粒子の表面に備えられる、または上記粒子内に混合される請求項1に記載の組成物。
- 上記担体は、複数のリン酸カルシウム粒子からなる請求項6に記載のキット。
- 上記複数の粒子は、高分子マトリクス内に分散されている請求項6に記載のキット。
- 上記担体の上記少なくとも1つの第2の増殖因子は、上記粒子の表面に備えられる、または上記粒子内に混合される請求項6に記載のキット。
- 上記少なくとも1つの第1の増殖因子と上記少なくとも1つの第2の増殖因子は同一であるかまたは異なり、上記少なくとも1つの第1または第2の増殖因子は、形質転換増殖因子ベータのスーパーファミリー、インスリン様増殖因子(IGF)、繊維芽細胞増殖因子(FGF)、血小板由来増殖因子(PDGF)、および血管内皮増殖因子(VEGF)から選択される請求項6に記載のキット。
- 上記少なくとも1つの第2の増殖因子はBMP−2またはBMP−7である請求項6に記載のキット。
- 上記送達媒体は、逆相重合体を含んでいる請求項1に記載の組成物。
- 上記送達媒体および上記担体を含んでいる上記組成物は、該担体が該送達媒体中に混合されるときにゲルまたはパテの形態である請求項1に記載の組成物。
- 上記送達媒体に対する上記担体の比は、1:1〜2:1(v:v)である請求項1に記載の組成物。
- 上記送達媒体は逆相重合体を含んでいる請求項6に記載のキット。
