本明細書で使用される用語「カドヘリン−11」、「Cad−11」および「OB−カドヘリン」は、天然または内因性カドヘリン−11(例えば哺乳類、例えばヒト)タンパク質、ならびに天然または内因性カドヘリン−11タンパク質の場合と同じアミノ酸配列を有するタンパク質(例えば、組換えタンパク質、合成タンパク質)を示す。したがって、用語「カドヘリン−11」、「Cad−11」および「OB−カドヘリン」は、本明細書中で同義的に使用され、例えば、哺乳動物(例えば、ヒト、非ヒト霊長類)において天然に生じる選択的スプライシングまたは他の細胞プロセスによって生成されるカドヘリン−11タンパク質(例えば、哺乳類、ヒト)の多形性または対立遺伝子変異体および他のアイソフォームを含む。好ましくは、カドヘリン−11タンパク質は、配列番号2のアミノ酸配列を有するヒトタンパク質である(Genbank登録番号NP001788および図15を参照)。
カドヘリンは、他のカドヘリンに同型的に結合することによって細胞接着を媒介するCa2+−依存性接着分子の大ファミリーに属する(M.J.WheelockおよびK.R.Johnson、Ann.Rev.Cell Dev.Biol.19:207−235頁(2003年))。古典的なカドヘリンは、5つの細胞外カドヘリン(EC)ドメイン(各々、約110アミノ酸長)、膜透過領域および保存された細胞質ドメインを有する1回膜貫通タンパク質である。カドヘリンは、ECドメイン間の相同度に基づき、I型またはII型カドヘリンのいずれかに分かれる。II型カドヘリンは、ヒトカドヘリン−5、−6、−8、−11、および−12、ならびにMN−カドヘリンを含む。細胞間結合の媒介における細胞外ドメインの各々の役割の相対的重要性は不明確である。
カドヘリン−11は、関節接合部(articulated joints)の滑膜表層内での滑膜細胞と滑膜細胞との結合を媒介する(Valenciaら、J.Exp.Med.200(12):1673−1679頁(2004年);KienerおよびBrenner、Arthritis Res Ther.7(2):49−54頁(2005年))。ヒトIgG2のヒンジ−CH2−CH3ドメインに融合された、ヒトカドヘリン−11の全部で5つの細胞外カドヘリンドメインからなる融合タンパク質は、インビトロで滑膜細胞表層の形成を阻害した(Kienerら、Am.J.Pathol.168頁(2006年))。さらに、マウスIgG2aのヒンジ−CH2−CH3ドメインに融合された、アンタゴニスト抗カドヘリン−11抗体およびマウスカドヘリン−11のEC1−5からなる融合タンパク質は、リウマチ様関節炎のマウスモデルにおける炎症および関節腫脹を阻害した(Leeら、Science 315:1006−1010頁(2007年))。
本発明のカドヘリン−11アンタゴニストは、カドヘリン−11タンパク質のEC1ドメインに特異的に結合する任意の作用物質でありえ、細胞内での1つ以上のカドヘリン−11媒介性活性を阻害する(例えば、低下させる、阻止する)。カドヘリン−11媒介性活性は、限定はされないが、細胞表面上でのカドヘリン−11を発現する細胞の凝集、および例えば、コラゲナーゼ、セリンプロテアーゼ、MMP1、MMP3、IL−6、IL−8またはRANKL/TRANCEなどの要素の発現または分泌を含む。作用物質は、特に、抗体、融合タンパク質、ペプチド、ペプチド模倣体、小分子、または核酸でありうる。
本明細書中に記載されるように、Cad−11(例えば配列番号3)のドナー配列およびカドヘリン結合ポケットを含む、ヒトカドヘリン−11のEC1ドメインのN末端部分内のエピトープに結合する抗体は、このタンパク質の他の領域内のエピトープに結合する抗体より有効にインビトロでのカドヘリン−11活性を遮断する(実施例1および2を参照)。
さらに、ヒトCad−11のEC1ドメイン(例えば、ヒトIgGの一部に融合されるヒトCad−11のEC1ドメイン)のみを有する免疫グロブリン融合タンパク質は、すべての5ECドメインを有する、Cad−11のEC領域のより大きい部分を含む融合タンパク質より有効にインビトロでのCad−11活性を阻害した。
本発明のカドヘリン−11アンタゴニストはまた、カドヘリン−11タンパク質のEC1ドメインに結合するペプチドでありうる。ペプチドは、任意の適切なL−および/またはD−アミノ酸、例えば一般的なα−アミノ酸(例えば、アラニン、グリシン、バリン)、非α−アミノ酸(例えば、β−アラニン、4−アミノ酪酸、6−アミノカプロン酸、サルコシン、スタチン))、および異常アミノ酸(例えば、シトルリン、ホモシトルリン、ホモセリン、ノルロイシン、ノルバリン、オルニチン)を含みうる。ペプチド上のアミノ、カルボキシルおよび/または他の官能基は、遊離(例えば未修飾)状態でありうるかまたは適切な保護基で保護されうる。アミノおよびカルボキシル基における適切な保護基、および保護基を付加または除去するための方法は、当該技術分野で公知であり、例えば、GreenおよびWuts、「Protecting Groups in Organic Synthesis」、John Wiley and Sons、1991年において開示されている。ペプチドの官能基はまた、当該技術分野で公知の方法を用いて誘導体化(例えばアルキル化)されうる。
カドヘリン−11アンタゴニストはまた、ペプチド模倣体でありうる。例えば、多糖類はペプチドと同じ官能基を有するように調製されうる。ペプチド模倣体は、例えば、ペプチド剤の三次元構造を、それが結合されるかまたは標的分子に結合することになる環境内で確立することによって設計されうる。ペプチド模倣体は、少なくとも2つの成分、1つ以上の結合部分および骨格または支持構造を含む。
カドヘリン−11アンタゴニストはまた、小分子でありうる。小分子の例として、有機化合物、有機金属化合物、無機化合物、および有機、有機金属または無機化合物の塩が挙げられる。小分子中の原子は、典型的には共有および/またはイオン結合を介して共に連結される。小有機分子中の原子の配列は、鎖(例えば、炭素−炭素鎖または炭素−ヘテロ原子鎖)を示しうるか、あるいは炭素原子を有する環、例えばベンゼンまたは多環系、または炭素およびヘテロ原子の組み合わせ、すなわちピリミジンまたはキナゾリンなどのヘテロ環を示しうる。小分子は任意の分子量を有しうるが、一般に約5,000ダルトン未満の分子を含む。例えば、かかる小分子は、約1000ダルトン未満でありえ、好ましくは約750ダルトン未満であるか、またはより好ましくは約500ダルトン未満である。小分子および他の非ペプチドカドヘリン−11アンタゴニストは、天然に見出され(例えば、同定され、単離され、精製され)、および/あるいは(例えば、従来の有機合成、生物媒介性(bio−mediated)合成、またはそれらの組み合わせによって)合成的に生成されうる。例えば、Ganesan、Drug Discov.Today、7(1):47−55頁(2002年1月);Lou、Drug Discov.Today、6(24):1288−1294頁(2001年12月)を参照のこと。天然小分子の例として、限定はされないが、ホルモン、神経伝達物質、ヌクレオチド、アミノ酸、糖類、脂質、およびそれらの誘導体が挙げられる。
本発明のCad−11アンタゴニストはまた、ヒトカドヘリン−11のEC1ドメインに結合する核酸分子(例えばオリゴヌクレオチド)でありうる。適切な核酸Cad−11アンタゴニストは、特定の目的の分子(例えばヒトカドヘリン−11のEC1ドメイン)に、古典的なワトソン−クリック塩基対合以外の相互作用を通じた高い親和性および特異性で結合可能なアプタマーを含む(TuerkおよびGold、Science 249:505頁(1990年);EllingtonおよびSzostak、Nature 346:818頁(1990年))。
小分子を含む、カドヘリン−11結合特異性を有する作用物質は、スクリーニング、例えば、化合物および/またはライブラリ(例えば、化学物質、ペプチド、核酸ライブラリ)のハイスループットスクリーニングにおいて同定されうる。
いかなる1つの理論に制約されるものではないが、Cad−11のEC1ドメインの最初の約35個のアミノ酸(例えば約33〜約37個のアミノ酸)が同型カドヘリン結合にとって必要とされ、Cad−11のこの領域に特異的に結合する作用物質がCad−11分子間の結合を有効に阻害しうると考えられる。したがって、かかる作用物質は、細胞(例えば滑膜細胞)におけるCad−11の発現または活性に関連した障害(例えば、炎症性障害、線維症、癌)の治療および予防において有用である。したがって、本発明の一態様は、哺乳動物被験体におけるカドヘリン−11媒介性障害を治療するための方法であって、ヒトカドヘリン−11のEC1ドメインペプチド(配列番号3)に結合する治療有効量のカドヘリン−11アンタゴニストを被験体に投与するステップを含む方法に関する。
材料および方法
ウエスタンブロッティング
タンパク質を、標準的方法を用い、SDS−PAGEによって分離し、ニトロセルロース(NC)膜に移した。つまり、NC膜をトリス緩衝生理食塩水−Tween(TBST)(8.8g/LのNaCl、0.2g/LのKCl、3g/Lのトリス塩基、500ul/LのTween−20、pHを7.4)ですすいだ。膜を、TBST中に溶解した4%BSAで、22℃で1時間ブロッキングした。NC膜をそれぞれTBSTで5分間にわたって3回すすいだ。マウス抗ヒトCad−11抗体をTBST中で0.5μg/mlに希釈し、NCを22℃で1時間インキュベートした。NC膜をそれぞれTBSTで5分間にわたって3回すすいだ。西洋わさびペルオキシダーゼ(HRP)と複合したヤギ抗マウスIg抗体をTBST中で1μg/mlに希釈し、NC膜を、二次溶液中、22℃の室温(RT)で最低時間の1時間インキュベートした。NC膜をそれぞれTBSTで5分間にわたって3回すすいだ。シグナルを、標準のHRP方法を用いて発生させた。
抗原(5μg/mlもしくは50μgのCad−11−EC−1−Fcまたは5μg/mlのカドヘリンペプチドのいずれか)を緩衝液中で希釈し、使用し、プレートを4℃で一晩コーティングした。プレートを洗浄し、次いで、PBS希釈緩衝液中、1.5%BSA、5%低脂肪乳粉末(PBS中、1.5%BSA、2.5%低脂肪乳粉末、0.1%Tween−20)でブロッキングした。次いで、プレートを、抗Cad−11ヒトfAbを有する細菌溶解物と共にインキュベートするか、または抗Cad−11ヒトfAbで1時間精製した。洗浄後、二次抗体(Cy5複合hu−Fab(1/100に希釈))を25分間適用した。次いで、プレートを洗浄し、得られた蛍光を読み取った。
先行的に報告されたカドヘリン−11に特異的な3つの抗体セットを、ヒトカドヘリン−11のEC1ドメインに対する結合能について試験した。これらの抗体は、カドヘリン−11ノックアウトまたは欠損マウスにおいてマウスカドヘリン−11−Fc融合タンパク質免疫原に対して産生された抗体(Leeら、Science 315:1006−1010頁(2007年))、CHO細胞内で既に生成されたヒトカドヘリン−11−Fc融合タンパク質免疫原に対する、カドヘリン−11野生型マウスにおいて産生された抗体(Valenciaら、J.Exp.Med.200(12):1673−1679頁(2004年))、および、ヒトカドヘリン−11のEC1−3ドメインを有する細菌によって生成されたタンパク質に対する、カドヘリン−11野生型マウスにおいて産生された抗体を含んだ。これらの抗体を、ウエスタン分析により、ヒトCad−11のEC1ドメインのみ(カドヘリン−11−EC1−Fc)、Cad−11のEC1およびEC2ドメイン(Cad11−EC1/2−Fc)またはCad−11の全部で5つのECドメイン(カドヘリン−11−EC1−5−Fc)を有する融合タンパク質に対する結合能について試験した。ウエスタンブロットに対し、試験した抗体の中で、EC1−FcまたはEC1−2−Fc融合タンパク質を認識するものは全くなかった(図1A)。しかし、3つの試験セットの各々に由来する抗体は、細胞外ドメイン1〜5を含むヒトCad−11−Fc融合タンパク質を認識した(図1B)。これらの結果は、試験抗体がヒトCad−11のEC1またはEC2ドメインに結合しなかったが、このタンパク質の細胞外領域内の他の部分のエピトープを認識したことを示す。

431−D細胞は、懸濁液中で成長し、通常はカドヘリンを全く発現せず、凝集しない。431−D−11細胞は、遺伝子組み換えにより、Cad−11が発現されている。431−D−11細胞を40分間にわたって培地中で単独でインキュベートし、それらが凝集し始める場合、ウェルの底に沈んだ細胞塊および懸濁液中の残りの非凝集細胞が測定され、凝集431−D−11の百分率が計算されうる。凝集アッセイにおいては、431−D−11細胞(D−11細胞)を、フラスコ内でサブコンフルエンスまで成長させ、次いで0.05%トリプシン+0.53mM EDTAを使用し、フラスコから除去した。約2×106個の431−D−11細胞を、2mlのSME培地(ダルベッコ改良イーグル培地(Dulbecco’s Modified Eagle’s Medium)−高グルコース、0.1Mヘペス、pH7.4および5U/mlのDNAse)に添加し、試験作用物質(例えば、抗体、Fab、融合タンパク質)の不在下または存在下のいずれかで、氷上で15分間プレインキュベートした。試験作用物質とのプレインキュベーション後、細胞を24ウェルプレート上の丸底ウェルに移し、ロータリーシェーカー上、30rpmで回転させながら、37℃でインキュベートした。細胞は、凝集するにつれて、ウェルの底に沈む。0分後および40分後、試料の中央から200μlをウェルから除去し、25μlの8%グルタルアルデヒドと混合し、細胞を固定した。200μlの固定した細胞の試料を9.8mlのCoulter Counter等張生理食塩溶液に添加し、Coulter Counterセットを使用し、8μm〜24μmの閾値でカウントした。3の細胞カウント/試料を記録した。40分後の細胞の減少または凝集した細胞の(0分時点での百分率に対する)百分率を計算した。
カドヘリン−11のEC1ドメインのN末端の35個のアミノ酸中のエピトープに対して結合特異性を有する候補Fab(クローンF9)は、MN−CadまたはCad−8のEC1ドメインに結合しないものであり、インビトロでのカドヘリン−11細胞凝集アッセイを使用し、Cad−11媒介性細胞凝集に対する阻害能について試験した。候補Fabは、細胞のカドヘリン−11媒介性凝集を1μg/ml以下の濃度で有意に阻害した(図4および5)。それに対し、EC1/2ドメイン外部のCad−11の細胞外領域内のエピトープに結合する13C2抗体から作製したFabは、10μg/mlの濃度のみでカドヘリン−11の凝集を阻害し、これは、F9 FabがCad−11の細胞外ドメインの他の部分に結合する抗体より低い濃度でCad−11活性をより効果的に阻害することを示唆する。
次のオリゴヌクレオチドプライマーを使用する、標準条件下で実施されるポリメラーゼ連鎖反応(PCR)を使用し、カドヘリン−11のEC1領域を、完全長ヒトカドヘリン−11 cDNAをコードするベクターから調製し(ヒトCad−11をInvitrogen pCEP4(登録商標)ベクターのNot1およびKpn−1部位にクローン化し)、EcoR1およびBglII部位(それぞれフォワードおよびリバースプライマー内の下線の配列を参照)を増幅産物に導入した。

マトリゲルプラグへの細胞浸潤/遊走
線維芽細胞様滑膜細胞(FLS)遊走活性を、マトリゲル ECMでコーティングされたトランスウェル内、FLS培地(ダルベッコ改良イーグル培地−高グルコース[Sigma#D7777]、10%ウシ胎仔血清[Benchmark#100−106]、1%ペニシリン−ストレプトマイシン[Gibco315140−122]、1% L−グルタミン[Gibco#25030]、0.5%ゲンタマイシン[Gibco#15710−064])中で評価した。1×104個の細胞を有するFLS培地中のヒトFLS細胞懸濁液を、0.750mLのFLS培地を有する24ウェルプレートのウェル内にセットした対照インサートまたはマトリゲルコーティングされたインサートに添加した。次いで、プレートを、37℃、5%の大気中CO2で加湿された組織培養インキュベーター内で22時間インキュベートした。
Cad−11−EC1−Fcは、3μg/mlの濃度で細胞凝集を有意に阻害した一方、ヒトCad−11の全部で5つのECドメインを有する完全長Cad−11−EC1−5−Fcタンパク質は、100μg/mlの濃度でCad−11媒介性凝集を阻害した(図13)。これらのデータは、Cad−11−EC1−Fc免疫グロブリン融合タンパク質がインビトロアッセイにおいてCad−11媒介性細胞凝集を有効に阻害することを示す。

Balb/cマウスを、BSAに共有結合した、ヒトCad−11 EC1ドメインの最初の33個のアミノ酸(GWVWN QFFVI EEYTG PDPVL VGRLH SDIDS GDG(配列番号10))に対応する0.01mgのペプチドで、1か月にわたり週2回、脚パッドに9回免疫した。このペプチドは本明細書中でペプチド4と称される。免疫マウス由来の脾臓を採取し、マウス融合パートナーP3X63−Ag8.653と融合し、抗体産生ハイブリドーマを作製した。ハイブリドーマを増殖させ、10、3または0.5細胞/ウェルのいずれかでサブクローン化し、ハイブリドーマ由来の抗Cad−11抗体含有培地を、細菌内で生成されるCad−11 EC1−2ドメインを含むタンパク質に対する特異的結合能について、ELISAでスクリーニングした。これらのペプチド4ハイブリドーマ由来の抗Cad−11抗体含有培地を、ヒトCad−8およびMN−カドヘリンのEC1−2ドメインを含むタンパク質への結合の不在について同時スクリーニングした。96ウェルEIAプレートを、各EC1−2 Cadタンパク質につき0.05mlの0.0〜0.3mg/mlで、4℃で一晩コーティングし、次いで生理食塩緩衝液で数回洗浄した。次いで、プレートを0.25mlのカゼイン−PBS緩衝液でブロッキングし、生理食塩緩衝液で数回洗浄した。抗Cad−11抗体を含有するハイブリドーマ培地を、各ウェル内で、22℃で1時間正確にインキュベートし、次いでPBS−Tween(0.05%)で2回洗浄した。ヤギ抗マウスIgG二次抗体の1/1000希釈物100μlを各ウェルに添加し、22℃で30分間インキュベートし、次いでPBS−Tween(0.05%)で2回洗浄した。100μl/ウェルの室温TMB(3,3’,5,5’−テトラメチルベンジジン)試薬を各ウェルに添加し、22℃で5分間着色させておいた。反応を100μlの室温2N硫酸で停止し、プレートを、Wallac 1420マイクロプレートリーダー上、450nmで読み取った。
ペプチド4ハイブリドーマ由来の抗Cad−11抗体含有培地は、Cad−11 EC1−2タンパク質(図16、HL対CAD−11)に結合したが、Cad−8およびMN−CadのEC1−2ドメインを含むタンパク質(図16、それぞれHL対CAD−8およびHL対MNCad)に結合しなかった。対照ハイブリドーマ培地は、試験したカドヘリンタンパク質のいずれにも結合しなかった(図16、Media対CAD−11、Media対Cad8、およびMedia対MNCad)。これらのデータは、ハイブリドーマにおいてペプチド4に対するCad−11特異抗体が存在することを示す。

ペプチド4 Cad−11 EC1抗体H1M1およびH14が結合するCad−11 EC1ドメイン内のエピトープを判定するため、EC1領域の最初の37個のアミノ酸を範囲とする4つの異なるペプチド(図22を参照)をELISAフォーマットで固定化し、4つのペプチドの各々に対するH1M1およびH14抗体の結合能を測定した。96ウェルReactibindプレートを、0.3ng/ウェルのペプチド1(Cad−11 EC1ドメインのアミノ酸G1〜P18)、0.3ng/ウェルのペプチド2(Cad−11 EC1ドメインのアミノ酸G15〜N34)、0.3ng/ウェルのペプチド3(Cad−11 EC1ドメインのアミノ酸V19〜Y37)、0.3ng/ウェルの免疫原ペプチド4(Cad−11 EC1ドメインのアミノ酸G1〜G33)、全EC1ドメイン(EFL)を含む20ngの融合タンパク質、または20ngの対照ヒトIg(Fc−ブロック)で、4℃で一晩コーティングした。ウェルを、PBS−Tween(0.05%)で2回洗浄し、dH2O中のカゼインで、22℃で3時間ブロッキングし、次いでPBS−Tween(0.05%)で2回、再洗浄した。異なるペプチド4 Cad−11 EC1ドメイン抗体の様々な希釈物を、ペプチド−またはタンパク質−コーティングウェルに移し、22℃で45分間インキュベートし、次いでPBS−Tween(0.05%)で2回洗浄した。ヤギ抗マウスIgG二次抗体(Jackson ImmunoResearch(West Grove,PA))の1/1000希釈物100μlを各ウェルに添加し、22℃で30分間インキュベートし、次いでPBS−Tween(0.05%)で2回洗浄した。100μl/ウェルの室温TMB試薬を各ウェルに添加し、22℃で5分間着色させておいた。反応を100μlの室温2N硫酸で停止し、プレートを、Wallac 1420マイクロプレートリーダー上、450nmの波長で読み取った。
1:11でのペプチド4抗Cad−11抗体H1M1(図21A)および1:23でのH14(図21B)の双方は、ELISAにおいてOD450プレート読み取り値の対照に対する増加によって示されるように、ペプチド4(PEP4)免疫原およびEC1ドメイン融合タンパク質(EFL)に結合した。これらの抗体のいずれも、ヒトIgG対照(Fcブロック)に結合しなかった。さらに、両抗体は、ELISAにおいて、ペプチド2(PEP2)に結合したが、ペプチド1(PEP1)またはペプチド3(PEP3)に結合しなかった(図21Aおよび21B)。

Cad−11抗体の、Cad−11媒介性細胞凝集に対する阻害能を評価するため、30μg/mlのH1M1ペプチド4抗体を、24ウェル丸底ポリプロピレンプレート内、0.5mlのDMEM−高グルコース、20mMヘペス(pH7.4)、10%FCSおよび10U/ml DNAse中で、75,000個のCad−11発現A−431−D類表皮癌細胞と共に培養した。24ウェルプレートを、約60rpmでの回転プラットフォーム上に配置し、5%CO2と共に37℃で一晩インキュベートした。翌日、細胞凝集を、(H1M1実験における)100倍または(H14実験における)40倍の倍率でプレートを撮影後、評価した。
対照アイソタイプ抗体(30μg/ml)の存在下で、Cad−11発現細胞は大きな塊を形成した一方(図23A)、親Cad−11陰性細胞は単一または2つの細胞集団として残存する(図23C)。H1M1で処置されたCad−11細胞は小さな細胞塊として残存し(図23B)、それは対照抗体を使用して得られる大きな塊を形成するまで至らなかった。

試験1−0日目および2日目、6週齢雄C57/Bl6マウスに150μlのKBN血清を注射した。KBN血清で処置したマウスは、生理食塩水の注射を受けたか(図25、白三角形)または異なる用量のH1M1抗Cad−11 EC1抗体で処置された。処置レジメンは、0日目での0.5mgの抗体/マウスと、その後、1日おき(q2d)での0.1mgの抗体/マウス(0.5mg+0.1mg)の投与(図25、黒三角形);0日目での0.5mgの抗体/マウス(0.5mg)の投与(図25、菱形);1日おき(q2d)での0.1mgの抗体/マウス(0.1mg+0.1mg)の投与(図25、四角形);または1日おき(q2d)での0.3mgの抗体/マウス(0.3mg+0.3mg)の投与(図25、円形)を含んだ。対照群はマウス5匹からなり、処置群はマウス7匹からなった。関節炎に随伴する関節腫脹を、キャリパー測定値を1日おきにとることによって測定した。
試験1−H1M1抗Cad−11抗体は、対照マウスに対して関節腫脹を阻害した。KBNで処置されたマウスに0.3mgのH1M1抗体を1日おきに投与することにより、関節炎に随伴する関節腫脹の最大の阻害を認めた(図25、円形)。

ペプチド3ハイブリドーマ由来の抗Cad−11抗体含有培地は、Cad−11 EC1−2タンパク質およびEC1−Fc融合タンパク質(図28、それぞれHL対Cad−11およびHL対Cad−11−EC1)に結合したが、Cad−8およびMN−CadのEC1−2ドメインを含むタンパク質(図28、それぞれHL対Cad8およびHL対MNCad)に結合しなかった。対照ハイブリドーマ培地は、カドヘリンタンパク質のいずれにも結合しなかった(図28、Media対Cad−11、Media対Cad8、およびMedia対MNCad)。
材料および方法
凝集アッセイ
このアッセイでは、ヒト線維芽細胞様滑膜細胞(FLS)系を、FLS培地(DMEM、10%FCS、1%ペニシリン−ストレプトマイシン、1%L−グルタミン、0.05%ゲンタマイシン、1%HEPES)中で、90〜100%コンフルエントになるまで培養した。アッセイの日に、FLSから培地を除去し、0.05%トリプシン−EDTAを使用して、フラスコから細胞を取り出し、MM培地(DMEM、10%FCS、1%ペニシリン−ストレプトマイシン、1%L−グルタミン、0.05%ゲンタマイシン、1%HEPES)で洗浄し、次いで、カルシウム含有HBS(0.137M NaCl、5.4mM、KCl 0.25mM Na2HPO4、0.44mM KH2PO4、1.3mM CaCl2、1.0mM MgSO4、および4.2mM NaHCO)で2回洗浄した。トリプシン処理されたFLSを、4×104細胞/mlでMM培地に再懸濁し、ウェル当たり2×104の細胞を、培地のみ、30μg/mlのアイソタイプ対照抗体、または種々の抗Cad−11抗体のいずれかを含有する24ウェルディッシュに入れた。プレートを、5%CO2インキュベーター中で24時間インキュベートし、翌日光学顕微鏡を用いてウェルを観察し撮影した。
種々の細胞外マトリックスタンパク質からなるマトリゲルへのFLS浸潤を阻害するH1M1抗体の能力を試験するために、マトリゲルスプリットウェルシステム(matrigel split well system)を用いる浸潤アッセイを使用した。FLS生物学のこのインビトロモデルは、関節接合部のヒト軟骨を分解し、その中に穴を開けるFLSの能力を模倣する。このアッセイでは、ヒトFLS系を、90〜100%コンフルエントになるまで、FLS MM培地中で培養する。アッセイ開始の前日に、無血清FLS培地中で24時間、FLSから血清を枯渇させた。アッセイの日に、血清枯渇FLSから培地を除去し、0.05%トリプシン−EDTAを使用して、フラスコから細胞を取り出し、MM培地で洗浄し、次いでカルシウム含有HBSで2回洗浄した。トリプシン処理されたFLSを、4×105細胞/mlでMM培地に再懸濁した。調製したマトリゲルコーティングインサートを、FLSに対して有系分裂促進物質として作用するFCSを含むMM培地を含有する24ウェルプレートに配置した。チャンバーの反対側では、抗体を含有しないかまたは作用濃度の2倍の処理抗体を含有するMM培地50μlを各ウェルの中に導入し、それに4×105細胞/mlの細胞懸濁液50μlを加えた。FLSおよび抗体を含有するチャンバーを、37℃インキュベーター中で18時間インキュベートした。
培地または対照抗体のいずれかと共にインキュベートされたFLSは、細胞凝集物、および細胞突起を介して結合する細胞の広範なネットワークを形成した(図30AおよびB)。これに対し、抗Cad−11抗体であるH1M1で処置された細胞は、細胞凝集物および結合をほとんど形成しなかった(図30C)。
材料および方法
抗Cad−11抗体であるH1M1を、関節炎のKBNモデルで試験した。この疾患は、0日目および2日目にKBN血清75μlを2回投与することにより誘導した。7匹の雄性C57BL/6マウス群に、0日目に開始して1日おきに10mg/kgのH1M1を腹腔内投与し、マウスの関節腫脹を毎日モニターした。関節腫脹すなわち足首の厚さを、ばね式ダイヤルキャリパー(Long Island Indicator Service,Hauppauge,NY)を使用して、足首を完全に屈曲した状態にして踵で測定した。
H1M1抗体は、対照群と比較して、関節腫脹を53%(p<0.001)抑制した(図32)。
材料および方法
3クローンのH1M1産生ハイブリドーマ細胞(H1、H17、およびH27)からRNAを抽出した。IgGVH、IgMVH、IgκVL、およびIGλVLそれぞれに対する定常領域プライマーに加え、マウスシグナル配列に対する縮重プライマープールを使用してRT−PCRを実施した。重鎖可変(V)領域RNAを、6つの縮重プライマープール(HA〜HF)1組を使用して増幅し、軽鎖V領域mRNAを、κクラスター(κA〜κG)に対する7つの縮重プライマープール1組およびλクラスターに対する1つの縮重プライマーを使用して増幅した(表1を参照のこと)。
ヒト化抗Cad−11抗体可変領域配列の設計
SYN0012抗体とも称される、本明細書に記載のマウスH1M1モノクローナル抗体の抗体可変(V)領域の構造モデルを、スイスタンパク質データベースを使用して作製し、SYN0012/H1M1抗体の結合特性にとって必須であると予測される、V領域の重要な「拘束」アミノ酸を同定するために分析した。CDR(KabatおよびChothiaの両定義を使用)内に含有される残基は、いくつかのフレームワーク残基と共に、重要であると考えられた。SYN0012/H1M1抗体のVHおよびVκ配列は両方とも、典型的なフレームワーク残基を含有し、そのCDR1、2、および3のモチーフは多くのマウス抗体と比較可能である。
上記の分析に基づいて、SYN0012ヒト化抗Cad−11抗体変異体の作製のために使用することができる配列セグメントの大きな予備セットを選択し、ヒトMHCクラスII対立遺伝子(Perryら 2009)に結合するペプチドのインシリコ分析に関し、公知の抗体配列に関連するT細胞エピトープ(Bryson C,Jones TD and Baker MP.Prediction of immunogenicity of therapeutic proteins:validity of computational tools.Biodrugs 2010,24(1):1−8)のTCEDTM(T Cell Epitope Database of Antitope LLC,Cambridge UK)を使用して分析した。ヒトMHCクラスIIに対する顕著な非ヒト生殖細胞系バインダーと同定されたか、またはTCEDTMに対して顕著なヒット得点を得た配列セグメントは廃棄された。これにより、縮小したセグメントセットが得られ、これらの組合せを、上記のように再度分析し、セグメント間の接合部に潜在的なT細胞エピトープが確実に含有されないようにした。次いで、選択されたセグメントを組み合わせて、合成のための重鎖および軽鎖V領域配列を作製した。SYN0012ヒト化抗Cad−11抗体変異体のために、図35A〜35Hに詳述されている配列を有する5つのVH鎖(VH1〜VH5;配列番号61、63、65、67、69)および3つのVκ鎖(Vκ1〜Vκ3;配列番号71、73、75)を設計した。
SYN0012/H1M1のための複合ヒト抗体VHおよびVκ領域の変異遺伝子をすべて、アニールされ、連結され、PCR増幅されて、完全長の合成V領域を与える一連の重複オリゴヌクレオチドを使用して合成した。IgG4(S241P)VH鎖およびVκ鎖のためのpANT発現ベクターシステム(Antitope,Cambridge UK)の中に直接、構築されたVH変異体をMlu IおよびHind III部位を使用してクローニングし、構築されたVκ変異体をBss HIIおよびBam H1制限部位を使用してクローニングした(図36)。構築物はすべて配列決定により確認された。
複合ヒト化IgG4(S241P)VHおよびVκ鎖の15の組合せをすべて、電気穿孔法によってNS0細胞の中に安定的に形質移入し、200nMメトトレキサート(Sigma Cat.No.M8407,Sigma Aldrich,UK)を使用して選択した。各構築物のメトトレキサート耐性コロニーを、IgG4のELISAを使用してIgG発現レベルについて試験し、最良の発現系を選択し、増殖させ、液体窒素中に凍結した。15のヒト化抗体変異体のそれぞれを発現する細胞を生成するのに成功した。

精製抗体の組換えヒトCad−11 EC1への結合
精製されたヒト化複合変異抗体(SDP011〜SDP151)の組換えCad−11 EC1ヒトIgG1融合タンパク質への結合を、競合結合ELISAで評価した。5μg/ml〜0.0022μg/mlの希釈系列のヒト化抗体または対照抗体(SYN0012/H1M1)を、一定濃度のビオチン化マウス抗Cad−11抗体のSYN0012/H1M1(0.1μg/ml、最終濃度)と前混合した後、PBSで希釈された0.25μg/mlの組換えヒトCad−11 EC1(R&D Systems Cat.No.1790−CA−050)で前コーティングされたNunc Immuno MaxiSorp 96ウェル平底マイクロタイタープレート(Fisher Cat.No.DIS−971−030J)上で室温にて1時間インキュベートした。ビオチン化mAbの結合は、ストレプトアビジン−HRP(Sigma Cat.No.S5512)およびテトラメチルベンジジン(TMB)基質(Invitrogen Cat.No.00−2023)を用いて検出した。
ヒト化抗Cad−11抗体のいずれがCad−11に対して最大の特異性を示すかを評価するために、Cad−11免疫原、またはカドヘリン−7、−8、−9、−18、−20、および−24から取り出した、Cad−11免疫原に最も関連した6つの配列に対応する配列を包含した、BSAに連結された37−merのアミノ酸長のペプチドに対する結合能について、ヒト化抗体をELISAフォーマットで試験した。
組換えhisタグ付きヒトCad−11に対するSDP051の親和性を決定するために、rhCad−11(R&D Systems)を単一濃度でチップに固定化し、SDP051について高解像度カイネティックスを測定し、2状態反応結合モデルをカイネティックデータに適合させるのに成功した。
凍結されたCad−11発現431D細胞(K.Johnson,U.Nebraska,Omaha)または非Cad−11発現親431D細胞(K.Johnson,U.Nebraska,Omaha)を解凍して洗浄し、2×106細胞/mlで再懸濁して、各ウェルに50μlずつ加えた。次いで、1ng/ml〜30μg/mlに濃度を増加させたヒト化SDP051またはSDP071抗体を用いて、氷上で45分間細胞を染色した。染色後、細胞を、1%ウシ胎児溶液(FBS)含有ハンクス緩衝生理食塩液(HBSS)で2回洗浄し、再懸濁し、マウス抗ヒトIgG4−PE(100倍希釈)で、光遮断下、氷上で45分間染色した。二次試薬で染色後、細胞を1%FBS含有HBSSで2回洗浄し、1%FBS含有HBSS中の2%ホルムアルデヒド200μlに再懸濁した後、BD FACSCaliburを用いて分析した。
凍結されたCad−11発現431D細胞または非Cad−11発現親431D細胞を解凍して洗浄し、2×106細胞/mlで再懸濁して、各ウェルに100,000細胞ずつ加えた。次いで、濃度を増加させた37−merのアミノ酸長のCad−11ペプチドまたはCad−7、Cad−8、Cad−9、Cad−18、Cad−20、もしくはCad−24ペプチドのいずれかと共に、氷上で45分間、0.5μg/ml SDP051を用いて細胞を染色した。染色後、細胞を、1%ウシ胎児溶液(FBS)含有ハンクス緩衝生理食塩液(HBSS)で2回洗浄し、再懸濁した後、100μlのヤギ抗マウスIgG−PE(100倍希釈)で、光遮断下、氷上で30分間染色した。二次試薬で染色後、細胞を1%FBS含有HBSSで2回洗浄し、再懸濁し、1%FBS含有HBSS中の2%ホルムアルデヒド200μl中で固定した後、BD FACSCaliburを用いて分析した。
ヒト化抗Cad−11抗体はすべて、Cad−11融合タンパク質への結合に対して、SYN0012と有効に競合した(図38A〜C)。これにより、それらの抗体がSYN0012と同じエピトープまたはCad−11上の重複エピトープに結合すること、およびそれらの抗体が、組換えヒトCad−11に対して、SYN012と少なくとも同様にまたはそれよりも良好に結合することが示された。曲線を使用して各抗体に対するIC50値を計算し、それらの値をSYN0012のIC50に対して規準化した。SYN0012は、プレート間の比較がなされうるように各ELISAプレートに含まれた(表3)。各細胞系の発現レベルおよび各抗体の計算等電点(pI)も示される(表3)。
材料および方法
抗体の精製
飽和するまで増殖した抗体発現細胞の5L培養物からSDP051を精製した。上清を細胞およびデブリから分離して、pH7.4に調整し、濾過滅菌し、0.5M NaOHであらかじめ衛生化した5mlのHi−Trap Mab Select SureプロテインAアフィニティカラム(GE Healthcare,Amersham,UK)に、5ml/分の流速で流した。カラムを100mlのPBS pH7.4で洗浄した。抗体を0.1Mクエン酸ナトリウムpH 3.0を用いて5ml分画で溶出し、各分画を直ちに0.25mlの1Mトリス−HClで中和した。各分画のタンパク質含量を280nmのUV吸収でモニターして、タンパク質含有分画をプールし、緩衝液を10mM酢酸ナトリウムpH5.5に交換して濃縮した。16/60Sephacryl S200カラム(GE Healthcare,Amersham,UK)を使用するサイズ排除クロマトグラフィーにより、抗体をさらに精製した。主要ピーク分画を集めてプールし、濾過滅菌して、エンドトキシンレベルについて、Endosafe(登録商標)−PTSTM(Charles River,Margate,UK)を使用して試験した。精製された抗体は、+4℃に保存した。最終濃度は、計算モル吸光係数、A280 1.0=1.51mg/mlを使用して、UV吸収により決定した。次いで、抗体をそれぞれ、AIMV培地(Invitrogen,Paisley,UK)中、100μg/mlに希釈した。
PBMCは、National Blood Transfusion Service(Addenbrooke’s Hospital,Cambridge,UK)から入手した健常ドナーバフィーコート(24時間以内に採血された血液に由来)から単離した。PBMCを、Lymphoprep(登録商標)(Axis−Shield,Dundee,UK)および密度遠心分離によりバフィーコートから単離し、CD8+RosetteSep(登録商標)(StemCell Technologies Inc.,London,UK)を使用して、CD8+T細胞を枯渇させた。Biotest SSP−PCRに基づいた組織適合試験キット(Biotest,Landsteinerstrasse,Denmark)を使用して、HLA−DRハプロタイプを同定すること、および「再現性」対照抗原(KLH:Pierce,Cramlington,UK)に対するT細胞応答を測定することにより、ドナーを特徴づけた。次いで、PBMCを凍結して、必要になるまで液体窒素に保存した。
増殖アッセイについては、SIが2以上(SI≧2.0)の経験的閾値が以前から確立されており、それによって、この閾値を上回る増殖応答を誘導する試料が陽性と見なされる(含まれる場合、境界線SI≧1.90が強調表示される)。広範なアッセイの展開および以前の研究により、これが、多数の偽陽性応答を検出することなく、最大の感度を可能にする最小の信号対雑音閾値であることが示されている。増殖データセット(n=3)については、陽性応答は統計的かつ経験的な閾値により以下のように定義された。
1.培地対照に対する試験ウェルのcpmを比較し、独立2試料スチューデントt検定を使用することによる応答の有意性(p<0.05)
2.2以上のSI(SI≧2.0)。
さらに潜在的な免疫原性を評価するために、ヒト化抗Cad−11 EC1抗体SDP051を、8日アッセイにおいて、25の異なるヒトT細胞ドナーからのCD8枯渇PBMC(CD4 T細胞)の増殖を誘導するその能力について試験した。マウスH1M1/SYN0012親抗体のマウス−ヒトIgG4 Fcキメラ型であるキメラSYN0014抗体は、ドナーの28%に増殖を誘導したが、SDP051抗体は、いずれのT細胞ドナーの増殖も誘導しなかった。これらの結果は、SDP051が低い免疫原性プロフィールを有するという以前のインシリコでの発見を支持する。
材料および方法
FLS浸潤アッセイ
初代ヒトFLS系を、FLS MM培地(DMEM、10%FCS、1%ペニシリン−ストレプトマイシン、1%L−グルタミン、0.05%ゲンタマイシン、1%HEPES)中で、90〜100%コンフルエントになるまで培養した。アッセイの開始前日に、無血清FLS培地中で24時間、FLSから血清を枯渇させた。アッセイの日に、血清枯渇FLSから培地を除去し、0.05%トリプシン−EDTAを使用してフラスコから細胞を取り出した後、カルシウム含有HBSで2回洗浄した。トリプシン処理されたFLSを、4×105細胞/mlでMM培地に再懸濁した。調製したマトリゲルコーティングインサートを、FLSに対して走化性因子として作用する5%胎児ウシ血清(FCS)を含むMM培地を含有する24ウェルプレートに配置した。チャンバーの反対側に、6μg/mlもしくは0.6μg/mlのSDP051(作用濃度の2倍)または6μg/mlの対照AC1−P抗体のいずれかを含有するMM培地50μlを入れ、次いで、4×105細胞/mlの細胞懸濁液50μlを各ウェルに加えた。FLSおよび抗体を含有するチャンバーを、37Cインキュベーター中で18時間インキュベートした。
Cad−11は、FLS生物学にとって必要とされる重要なカドヘリンである。その結果、Cad−11アンタゴニストは、2つのウェルを分離するマトリゲル層を分解し、その中に浸潤するヒト初代FLSの能力を阻害すると期待される。マトリゲルは種々の細胞外マトリックスタンパク質からなる。FLS生物学のこのインビトロモデルは、軟骨を分解し、その中に穴を空けるFLSの能力を含むFLS生物学の態様を再現する。
関節炎の動物モデルにおいてSDP051抗体を試験するために、D.MathisおよびC.Benoistにより開発された関節炎のK/BxN血清移入モデルが選ばれた(Korganow AS,Ji H,Mangialaio S,Duchatelle V,Pelanda R,Martin T,et al.From systemic T cell self−reactivity to organ−specific autoimmune disease via immunoglobulins.Immunity 1999;10:451−61)。このモデルは広範な関節滑膜炎を提供し、したがって、SDP051抗体の抗炎症および関節保護特性に対する良好な試験となった。
0および2日目に、KBN血清をマウスに投与した。0、2、4、および6日目に、10、3、もしくは1mg/kgのSDP051または対照AC3−1−PをマウスにIP投与した。足首の厚さの変化を図50に示す。足首の厚さの低下傾向が、3または10mg/kgのSDP051で処置されたマウスに観察された。10mg/kg群における足首の腫脹は、6、8、および10日目に有意に低下した。10日目では、この群の足首の腫脹は、対照群の腫脹よりも44%低下した。一連の試験を通して、10mg/kgのSDP051は、足首の腫脹を有意に低下させた。3mg/kgでは、足首の腫脹は低下傾向にあった。
1、2、4、および6日目に、10mg/kgのSDP051またはAC3−1−P対照抗体を尾静脈IVによりマウスに投与した。6日目において、SDP051群では、対照群よりも足関節腫脹が33%低下することが示された(表6)。これらのマウスから7日目に摘出した足首について、サイトカイン分析を以下のように実施した。足首関節を摘出して、皮膚および毛皮を除去し、関節を液体窒素中で即座に凍結した。次いで、ホモジナイゼーション緩衝液(2mM PMSF(Sigma part# P7626)および1Xプロテアーゼ阻害剤カクテル(Sigma part# P8340)、0.1Mインドメタシンを添加されたAffymetrix組織ホモジナイゼーション緩衝液(part# PC6002))中に関節を溶解して、遠心分離し、得られた上清を、複数アッセイ系(OceanRidge Biosciences)において、種々のサイトカインおよびケモカインのレベルについてアッセイした。
Claims (4)
- 癌の治療を必要とする哺乳動物被験体における癌の治療のための医薬の製造におけるヒト化抗体の使用であって、該抗体は、哺乳動物カドヘリン−11タンパク質のEC1ドメインに特異的に結合し、配列番号:69を含む抗体重鎖可変領域および配列番号:71を含む抗体軽鎖可変領域を含む、使用。
- 癌が、白血病、肺癌、食道癌、胃癌、結腸直腸癌、脳腫瘍、膀胱癌、乳癌、子宮頸癌、上皮癌、鼻咽腔癌、リンパ腫、子宮癌、肝癌、頭頸部癌、腎癌、雄性胚細胞腫瘍、悪性中皮腫、骨髄異形成症候群、卵巣癌、膵臓または胆汁癌、前立腺癌、甲状腺癌、および尿路上皮癌からなる群より選択される、請求項1記載の使用。
- 癌が乳癌である、請求項2記載の使用。
- 癌が前立腺癌である、請求項2記載の使用。
