Description
本発明は、有効成分としてTNP(N2−(m−トリフルオロベンジル),N6−(p−ニトロベンジル)プリン)又はその薬学的に許容される塩を含む、肝毒性の予防及び治療のための医薬組成物及びそれを含む健康機能食品に関する。
TNP(N2−(m−トリフルオロベンジル),N6−(p−ニトロベンジル)プリン)は、イノシトール五リン酸キナーゼ特異的阻害剤(非特許文献1)として知られる市販の化学物質である。TNPは5−イノシトールピロリン酸の生合成を阻害することで細胞内の5−イノシトールポリリン酸を下方制御し、それと同時に肝細胞のインスリンシグナル伝達を増大させる(非特許文献2)。骨髄由来の間葉系幹細胞をTNP処理すると、細胞の老化の進行を遅らせることができる(非特許文献3)。しかし、TNPが急性の肝毒性を予防又は治療する効果を有するかどうかについてはわかっていない。
ヒト胚性幹細胞を、内胚葉細胞を含む異なる株の細胞に分化、ヒト胚性幹細胞を、肝細胞に分化させることを調べるために、内胚葉細胞が誘導された(Cai, J.他、(2007) Hepatology 45(5):1229-1239)。
以下のようにコラゲナーゼかん流を介してマウス由来の肝細胞の培養を行った。
ヒト肝癌細胞株HepG2を、ゼラチンを予備被覆した6ウェルプレートに1×106cells/mlの密度にて分配した後、細胞接着のため24時間培養した。ここで培養培地は、10%FBS、10nMのデキサメタゾン、10nMのインスリン、及びペニシリン/ストレプトマイシン(100μg/ml)を補充したDMEM(Biowest)であった。細胞を、37℃のインキュベーターにおいて、温度及び湿度の制御下にてCO2を5%含有する混合ガスを供給しながら培養した。
TNP処理によるアセトアミノフェン起因性アポトーシスの変化を、実施例1に記載のものと同様の方法により培養したヒト胚性幹細胞由来の肝細胞において調べた。
TNP処理によるアセトアミノフェン起因性アポトーシスの変化を、実施例2に記載のものと同じ方法により培養したマウス由来の肝細胞において調べた。
TNP処理によるアセトアミノフェン起因性アポトーシスの変化を、実施例3に記載のものと同じ方法によって培養したヒト肝癌細胞株HepG2において調べた。
TNP処理によるアセトアミノフェンに対する肝細胞の代謝活性を調べた。
TNP処理による、肝細胞におけるアセトアミノフェンによって引き起こされたストレスに対する応答の制御を調べた。
TNP処理による、肝細胞内のアセトアミノフェンによって引き起こされたストレスに対する応答の制御を調べた。
本発明者らは、動物モデルにおいて、TNPがアセトアミノフェンによって引き起こされた毒性から肝細胞を保護する活性を有するかどうかを調べた。
実施例7の動物モデルにおけるアセトアミノフェン起因性肝毒性に対するTNPの予防効果を調べるため、組織、血液、及びJNKリン酸化を分析した。
Claims (6)
- 有効成分としてTNP(N2−(m−トリフルオロベンジル),N6−(p−ニトロベンジル)プリン)又はその薬学的に許容される塩を含む、肝毒性の予防及び治療用医薬組成物。
- 前記TNPが以下の式1:
- 前記TNPがアセトアミノフェン(AP)によって引き起こされるアポトーシスを特徴的に抑制する、請求項1に記載の肝毒性の予防及び治療用医薬組成物。
- 前記TNPが肝細胞内のグルタチオン(GSH)の濃度を特徴的に増加させる、請求項1に記載の肝毒性の予防及び治療用医薬組成物。
- 前記TNPがアセトアミノフェンにより増大したストレス応答を特徴的に抑制する、請求項1に記載の肝毒性の予防及び治療用医薬組成物。
- 有効成分としてTNP又はその薬学的に許容される塩を含む、肝毒性の予防及び改善用健康機能食品。
