滅菌される物品および上記滅菌インジケータを、滅菌媒体に曝露する工程と、
前記第1コンパートメントおよび前記第2コンパートメントの内容物を混合する工程と、
前記第1コンパートメントおよび前記第2コンパートメントの混合物を培養する工程と、
培養後に、検出可能な信号の有無を検出することにより前記滅菌プロセスの有効性を判定する工程とを含む、滅菌プロセスが提供される。
Claims (15)
- 遺伝子組換えされた生物学的インジケータを含有し、滅菌中に前記生物学的インジケータを滅菌媒体と接触させることができるように構成された第1のコンパートメント、および
少なくとも1つの酵素基質を含有し、滅菌中に前記酵素基質を前記生物学的インジケータから分離して維持し、前記生物学的インジケータが前記滅菌媒体に曝露された後に前記酵素基質が前記生物学的インジケータに接触することができるように構成された第2のコンパートメント、を備える滅菌インジケータであって、
前記遺伝子組換えされた生物学的インジケータは、少なくとも1つの供試生物およびインジケータ酵素を生成するのに適し、前記供試生物によって取り込まれた少なくとも1つのレポーター遺伝子を含み、
ただし、前記滅菌インジケータにおける供試生物は、前記供試生物中または滅菌インジケータに存在する場合に前記レポーター遺伝子の発現を抑制しうる活性リプレッサー遺伝子または活性化可能なリプレッサー遺伝子を含まず、かつ

滅菌される物品および請求項1〜10のいずれか1項に記載の滅菌インジケータを、滅菌媒体に曝露する工程と、
前記第1のコンパートメントおよび前記第2のコンパートメントの内容物を混合する工程と、
前記第1のコンパートメントおよび前記第2のコンパートメントの混合物を培養する工程と、
培養後に、前記検出可能な信号の有無を検出することにより前記滅菌プロセスの有効性を判定する工程とを含む、滅菌プロセス。 - 前記滅菌媒体は、蒸気、乾熱、放射線、プラズマ、1つ以上のガス状滅菌剤、および/または1つ以上の液状滅菌剤を含む、請求項11に記載の滅菌プロセス。
- 前記滅菌媒体は、電子ビーム放射線、電磁放射線、γ放射線、β放射線、エチレンオキシド、ガス状過酸化水素、液状過酸化水素、ホルマリン、グルタルアルデヒド、および/または過酢酸を含む、請求項11に記載の滅菌プロセス。
- 遺伝子組換えされた生物学的インジケータを含有し、滅菌中に前記生物学的インジケータを滅菌媒体と接触させることができるように構成された第1のコンパートメント、および
少なくとも1つの酵素基質を含有し、滅菌中に前記酵素基質を前記生物学的インジケータから分離して維持し、前記生物学的インジケータが前記滅菌媒体に曝露された後に前記酵素基質が前記生物学的インジケータと接触することができるように構成された第2のコンパートメント、を備える滅菌インジケータであって、
前記遺伝子組換えされた生物学的インジケータは、ゲオバチルス・ステアロサーモフィルスおよび/またはバチルス・アトロファエウスの芽胞、ならびにゲオバチルス・ステアロサーモフィルスおよび/またはバチルス・アトロファエウスの芽胞によって取り込まれた、インジケータ酵素を生成するためのlacZ、bgaB、xylE、およびcatから選択される少なくとも1つのレポーター遺伝子を含み、
ただし、前記滅菌インジケータにおける前記ゲオバチルス・ステアロサーモフィルスおよび/またはバチルス・アトロファエウスの芽胞は、前記供試生物中または滅菌インジケータに存在する場合に前記レポーター遺伝子の発現を抑制しうる活性リプレッサー遺伝子または活性化可能なリプレッサー遺伝子を含まず、かつ
前記インジケータ酵素および前記酵素基質は、前記酵素基質に対する前記インジケータ酵素の酵素作用により検出可能な信号が生じるように選択される、滅菌インジケータ。 - 前記インジケータ酵素は、β−D−ガラクトシダーゼ、β−D−グルコシダーゼ、α−D−グルコシダーゼ、アルカリホスファターゼ、酸性ホスファターゼ、ブチレートエステラーゼ、カプリレートエステラーゼリパーゼ、クロラムフェニコールアセチルトランスフェラーゼ、カテコール−2,3−ジオキシゲナーゼ、ミリステートリパーゼ、ロイシンアミノペプチダーゼ、バリンアミノペプチダーゼ、キモトリプシン、ホスホヒドロラーゼ、α−D−ガラクトシダーゼ、α−L−アラビノフラノシダーゼ、N−アセチル−β−グルコサミニダーゼ、β−D−セロビオシダーゼ、アラニンアミノペプチダーゼ、プロリンアミノペプチダーゼ、チロシンアミノペプチダーゼ、フェニルアラニンアミノペプチダーゼ、β−D−グルクロニダーゼ、脂肪酸エステラーゼ、またはそれらの2つ以上の混合物を含む、請求項14に記載の滅菌インジケータ。
