Description
a)放射性組成物のサンプルを高速液体クロマトグラフィー(HPLC)システムに与え、
b)分離された放射性組成物を得るため、段階(a)で与えられたサンプル中に存在する放射性化学種をHPLCによって分離し、
c)請求項1乃至請求項9のいずれか1項で定義されるような放射線検出器システムを使用して、放射線検出器システムのフローセルにおける所定数の複数の離散的に選択可能な流体導管を通過させられる、段階(b)で得られた分離された放射性組成物中に存在する、各放射性化学種が放出する放射線を検出することを含む。
放射線検出器と、
細長い流体通路をそれぞれ画成する複数の離散的に選択可能な流体導管を備え、流体導管がそれぞれフローセルを通って延在し、放射線検出器と放射線連通して位置付けられる、フローセルと、
フローセルの上流に位置し、入口ポート及び複数の出口ポートを含み、出口ポートがそれぞれ流体導管の一端と密閉された流体連通状態で位置し、入口ポートを流体導管の1つ以上の通路と流体連通させて配置することを可能にする、弁と
を備える。
a)放射性組成物のサンプルをHPLCシステムに適用し、
b)分離された放射性組成物を得るため、段階(a)で適用されたサンプル中に存在する放射性化学種をHPLCによって分離し、
c)本明細書で定義されるような本発明の放射線検出器システムを使用して、放射線検出器システムのフローセルにおける所定数の複数の離散的に選択可能な流体導管を通過させられる、段階(b)で得られた分離された放射性組成物中に存在する、各放射性化学種が放出する放射線を検出することを含む。
1.それらは、放射線検出器上の限定された線形範囲の問題に対する低コストの解決策を提供する。商用のPETセンターにおいて最も一般的に遭遇する放射線検出器は、各検出器ケーシング内でフローセルを設定することができるはずである。
2.フロースプリッタとは異なり、可変のフローセル容積の選択肢は、システムに対してさらなる背圧を追加しない。実際に、より大きい内径を有するより短い管材料によって、圧力はある程度低減される。さらに、フロースプリッタは、HPLCの勾配条件の変化(不均等な分割がもたらされる)による影響を受けることがあるが、可変のフローセル容量はクロマトグラフィー条件に依存しない。
3.フローセル容積の選択はRACに依存し、フローセル容積の選択肢の数は、切替え弁における位置の数及び検出器内の空間によってのみ限定される。本発明のフローセル内の3つの異なる流体導管は、求められる全ての柔軟性を提供するのに十分であろうことが考慮される。
4.ループ容積は、(放射性感知ユニット及び関連するエレクトロニクスの固有の範囲に加えて)約2桁分を超える容積に容易に及ぶことができる。このことは、広範囲のHPLCカラム及び流量を使用して、異なる化合物の範囲を生成する要件を満たすように、放射線検出器を設定できることを意味する。
5.本発明は、分析及び分取両方の放射線HPLCに適用することができ、全てのトレーサー及び放射性核種に対して適用可能である。
6.調節可能な容積を有する現在市販されている放射線検出器と比べて顕著な利点は、本発明によって、再適格化を要しない検出器の変更が可能になる点である。検出器内に複数の規定されたフローセル容積を含めることによって、異なるフローセル容積が選択されたとき、検出器の適格状態は不変のままである。放射線検出器内の流体導管は決して調節されないので、適格状態は不変であり、全ての選択は放射線検出器の外部で行われる。
7.検出器内のフローセルの容積は、クロマトグラフィーで分離されたピークの容積と比べて極めて小さいので、可変のフローセル容積を有することによって、最大ピーク高さにおいても、検出器内の活性レベルが可変になる。放射線検出器内の容積がHPLCカラムから出る「ピーク容積」よりも低いという事実は、比較的軽微な変更を行うことによって、検出器応答に対する変更が可能であることを意味する。
8.本発明のフローセルは、流量及びHPLCカラムの種類を変えること、並びに常に線形範囲内にあることを担保するように検出器容積を調整することを容易にする。
実施例1は、本発明に適したフローセルの製造について記載する。

実施例1:フローセルの製造
図5に示されるように、円形のプレキシガラスブロックの平坦面にチャネル3をエッチングした。3つの長さのPEEK管材料を、十分な管材料がUV検出器からの流出に付属し、十分な管材料が廃棄物コレクタに達するようにして、チャネル内に位置付けた。プレキシガラスの装着は、放射線検出器結晶の表面を、結晶を損傷する恐れがある管材料とのあらゆる接触から保護するように選択した。
[18F]フッ化物を注入することによって、検出器の線形性の調査を行った。最大検出器容量(緑PEEK管材料:9.2μL)及び最小検出器用量(赤PEEK管材料:0.25μL)について研究した。セル容積は、単に入口の管材料を切り替えることによって手動で切り替えられた。これは、切替え弁を使用することによって自動化する(かつ、クロメレオン(Chromeleon)などのクロマトグラフィーデータシステム内で動作させる)ことができる。プロット(図6)は、検出器の傾斜がフローセル容積にどのように正比例するかを示している。
検出器感度については、PETトレーサー18F−フルテメタモール(Nelissen et al 2009 J Nuc Med; 50(8): 1251-1259によって提示された方法に従って調製)の調製について分析した。オンカラム活性が研究した3つの検出器容積全てに対して同じであるように、注入量が変更された。注入容積は、赤(0.25μl PEEK管材料)、オレンジ(4.0μl PEEK管材料)、緑(9.2μl PEEK管材料)それぞれに対して(この順で)、20、25、及び35μLであった。容積は、サンプル崩壊に対して、ほぼ同じオンカラム活性が分析されることを担保するように調節された。合計の放射線検出器信号(総ピーク面積)は、フローセル容積に正比例することが見出された(図7a)。放射性不純物の検出に対する放射線検出器の感度は、放射線検出器容積を増加させるにつれて改善された(図7b)。
Claims (15)
- 放射線検出器システムであって、
放射線検出器と、
細長い流体通路をそれぞれ画成する複数の離散的に選択可能な流体導管を備えるフローセルであって、流体導管がそれぞれフローセルを通って延在し、放射線検出器と放射線連通して位置付けられる、フローセルと、
フローセルの上流に位置し、入口ポート及び複数の出口ポートを含む弁であって、出口ポートがそれぞれ流体導管の一端と密閉された流体連通状態で位置しており、入口ポートを流体導管の1つ以上の通路と流体連通させて配置することができる弁と
を備えており、
前記弁が、入口ポートと流体連通する複数の離散的に選択可能な流体導管の数を、所定数に基づいて調節するように制御され、離散的に選択可能な流体導管の所定数が、放射性組成物の放射線濃度(RAC)に基づいて決定される、放射線検出器システム。 - フローセルの離散的に選択可能な流体導管がそれぞれ、他の流体導管それぞれと同じ内径を有する、請求項1記載の放射線検出器システム。

- フローセルが、チャネルを横切って第1の主要面上に位置付けられる平坦な透明カバーをさらに備える、請求項4乃至請求項8のいずれか1項記載の放射線検出器システム。
- 放射性組成物の放射化学純度(RCP)を決定する方法であって、
a)放射性組成物のサンプルをHPLCシステムに与え、
b)分離された放射性組成物を得るため、段階(a)で与えられたサンプル中に存在する放射性化学種をHPLCによって分離し、
c)請求項1乃至請求項9のいずれか1項で定義されるような放射線検出器システムを使用して、放射線検出器システムのフローセルにおける所定数の複数の離散的に選択可能な流体導管を通過させられる、段階(b)で得られた分離された放射性組成物中に存在する、各放射性化学種が放出する放射線を検出することを含む、方法。 - 所定数の離散的に選択可能な流体導管が、放射性組成物の放射線濃度(RAC)に基づいて決定される、請求項10記載の方法。
