Description
また、テンニンカは、フトモモ科テンニンカ属(Rhodomyrtus)の常緑低木である。テンニンカ抽出物は、紫外線ダメージ回復効果があり、この回復効果の有効成分は、テンニンカ抽出物に含まれるピセアタンノールであることが報告されている(特許文献2を参照)。
ブラシノキはフトモモ科ブラシノキ属(Callistemon)の常緑の木本である。マキバブラシノキ(Callistemon rigidus)抽出物はMMP-2の阻害作用を有することが明らかとなっており、この阻害作用の有効成分の一つは、マキバブラシノキ抽出物に含まれるピセアタンノールであることが報告されている(非特許文献1を参照)。
本発明に係る、スチルベン単量体を含む溶液の製造方法、及びポリフェノール二量体に対するスチルベン単量体の濃縮方法は、スチルベン単量体及びポリフェノール二量体を含む、植物抽出物を含む溶液にゼラチンを添加する工程を含む。
パッションフルーツ(Passiflora edulis)の種子を破砕し、含水エタノール(水:エタノール=20:80(v/v))で抽出した。含水エタノール抽出液を、ロータリーエバポレーターにて溶媒を留去し、パッションフルーツ種子抽出物(固体)を得た。この抽出物を再度、水または20%エタノールに溶解させた。
ゼラチンとして、重量平均分子量が約3000のものを用い、表1及び表2に示した量(%(w/v))のゼラチンを添加した。
その後、3000rpm、10分間遠心分離を行った後に上清を回収することで澱を除去し、各上清について、ピセアタンノールの量及びスキルプシンBの量を、下記の条件のHPLCを用いて測定した。
・カラム:Mightysil RP-18 GP250-10 径10 mm、長さ250 mm(関東化学株式会社製)
・カラム温度:40℃
・溶出条件:流速3mL/min、0%メタノール−100%純水 → 30%メタノール−70%純水(グラジエント、10min)
・UV検出:280nm
本実施例では、実施例1の植物抽出液の代わりに、表3に示したスチルベン単量体及び二量体を含有する溶液を用いた。ゼラチンとして、重量平均分子量が約3000のものを用い、表4に示した量(%(w/v))のゼラチンを添加した。その後、15000rpm、5分間遠心分離を行った後に上清を回収することで澱を除去し、各上清について、表4に示した各スチルベン類化合物の量を、実施例1と同条件のHPLCを用いて測定した。そして、実施例1と同様に、各スチルベン類化合物の残存率を算出した。
Claims (5)
- レスベラトロール及び/又はピセアタンノールと、スキルプシンB及び/又はグネチンCとを含有する植物抽出物を含む溶液において、スキルプシンB及び/又はグネチンCに対して、ピセアタンノール及び/又はレスベラトロールを濃縮する方法であって、前記植物抽出物を含む溶液にゼラチンを添加する工程を含む、方法。
- 前記植物が、パッションフルーツ、テンニンカ、メリンジョ、リンゴンベリー、イタドリ、ブラシノキである、請求項1に記載の方法。
- 前記ゼラチンの濃度が、0.01%(w/v)以上である、請求項1または2に記載の方法。
- 前記ゼラチンの濃度が、0.05%(w/v)以上である、請求項1または2に記載の方法。
- 生成した澱を除去する工程をさらに含む、請求項1〜4のいずれか1項に記載の方法。
