Description
本発明に関する乳酸菌は、ラクトバチルス・ラムノーサス(Lactobacillus rhamnosus)に属する乳酸菌であり、今回新たに見出された2種の菌株である。これら菌株は、本発明者によって、独立行政法人製品評価技術基盤機構 特許微生物寄託センターに寄託されている。
う蝕菌や歯周病菌、カンジダ菌に対するOG4株及びOG5株の発育阻害効果を、特許文献1に開示された乳酸菌と比較しながら詳細に説明する。
MRS培地(Difco社)を121℃で20分間滅菌した後、OG4株及びOG5株のそれぞれを接種し、37℃で、48時間大気下で培養した後、蒸留水等で洗浄し、菌体を得た。なお、比較対象であるKO3株、YU3株及びYU4株についても、OG4株等と同じ条件で菌体を調製し、−80℃で保存した。
各菌株の菌体を、常温で解凍し、遠沈(3000rpm×5min)にて回収後、滅菌蒸留水で2回洗浄し、OD600で0.3とすることにより、所定の菌濃度(約1.0×109cfu/ml)に調整した。
被験菌株には、う蝕菌(mutans streptococci)として、ストレプトコッカス・ミュータンス(Streptococcus mutans Ingbritt)を、歯周病菌として、ポルフィロモナス・ジンジバリス(Porphyromonas gingivalis 381株)を、そして、カンジダ菌として、カンジダ・アルビカンス(Candida albicans MYA274)を用いた。
24穴プレートを用い、その各ウェルに、ストレプトコッカス・ミュータンスに対してはTSBY液体培地、ポルフィロモナス・ジンジバリスに対してはGAMまたはBHIの液体培地、カンジダに対してはサブロー液体培地のそれぞれを1ml添加した。
対応する各ウェルに、OG4株、OG5株、KO3株、YU3株及びYU4株の各培養上清1mlを個別に加え、対応する各被験菌株の菌懸濁液を個別に50μl接種し、テストサンプルとした。
OG4株及びOG5株の菌体、菌体培養物又はこれらの抽出物は、経口摂取する医薬品や食品などに利用できる。
OG4株、OG5株、及びKO3株が、それぞれ異なる菌株であること確認するために試験を行った。
Claims (4)
- ラクトバチルス・ラムノーサスOG4株(NITE P−1558)及びラクトバチルス・ラムノーサスOG5株(NITE P−1559)の乳酸菌のうち、少なくともいずれか1種の菌体、菌体培養物、又はこれらの抽出物を含有する、う蝕菌、歯周病菌、又はカンジダ菌に起因して口腔内で発症する口腔内疾患の予防、改善、又は治療用の材料。
- ラクトバチルス・ラムノーサスOG4株(NITE P−1558)及びラクトバチルス・ラムノーサスOG5株(NITE P−1559)の乳酸菌のうち、少なくともいずれか1種の菌体、菌体培養物、又はこれらの抽出物を含有する、う蝕菌、歯周病菌、又はカンジダ菌に起因して口腔内で発症する口腔内疾患の予防、改善、又は治療用の医薬品。
- ラクトバチルス・ラムノーサスOG4株(NITE P−1558)及びラクトバチルス・ラムノーサスOG5株(NITE P−1559)の乳酸菌のうち、少なくともいずれか1種の菌体、菌体培養物、又はこれらの抽出物を含有する、う蝕菌、歯周病菌、又はカンジダ菌に起因して口腔内で発症する口腔内疾患の予防、改善、又は治療用の食品。
- 請求項3に記載の食品において、
前記食品が、発酵乳又は発酵飲料である食品。
