Description
本発明は、幹細胞の生物学の分野に関する。
多能性幹細胞は培養中継続的に増殖する能力、および適当な増殖条件下で、内胚葉、中胚葉および外胚葉の3つの一次胚葉すべてを表す、系統を限定した細胞型に分化する能力の両方を有する(米国特許第5,843,780号;第6,200,806号;第7,029,913号;Shamblott et al., (1998) Proc. Natl. Acad. Sci. USA 95:13726;Takahashi et al., (2007) Cell 131(5):861;Yu et al., (2007) Science 318:5858)。特定の系統に限定した細胞型に適した増殖条件を規定することは、研究および治療的適用において用いるためにその細胞型を実質的に無制限に供給することになる。
特定の態様において、本発明は霊長類多能性幹細胞(pPS)の造血系統細胞へのインビトロ分化を提供する。pPS細胞は、ヒト造血系統細胞への分化に適したヒト多能性幹細胞であってもよい。造血系統細胞には、例えば、未熟樹状細胞(imDC)、成熟樹状細胞(mDC)、骨髄前駆細胞、単球が含まれうる。
以下に、本発明の基本的な諸特徴および種々の態様を列挙する。

特定の態様において、本発明は、pPS細胞の造血系統細胞へのインビトロ分化の改善された方法を提供する。したがって、本発明の特定の態様は、pPS細胞をDC(imDCおよびmDCを含む)などの造血系統細胞に分化させるのに適した最小数の外因性因子(サイトカインなど)を必要とする規定の条件を提供する。いくつかの態様において、本発明は、pPS細胞を最小数の外因性サイトカイン、例えば、7つ以下、6つ以下、5つ以下、4つ以下、3つ以下の外因性サイトカインを含む分化カクテルと接触させて、それにより造血系統細胞を生成することを提供する。一つの態様において、規定の条件は、4つ以下の添加外因性サイトカイン、例えば、BMP-4、GM-CSF、SCFおよびVEGFを含む分化カクテルを提供してもよい。他の態様において、既定の条件は3つ以下の添加外因性サイトカイン、例えば、a)BMP-4、GM-CSF、SCF;b)BMP-4、GM-CSF、VEGFを含む分化カクテルを提供してもよい。いくつかの態様において、それぞれのサイトカイン受容体に対するリガンドをそれぞれのサイトカインの代わりに用いてもよく、かつ/またはそれぞれのサイトカインに加えて提供してもよい。造血系統細胞がimDCである態様において、分化カクテルはIL-4をさらに含んでいてもよい。imDCはmDCを産生するために成熟カクテルとさらに接触させてもよい。
pPS細胞を造血系統細胞に分化させるための出発原料は、無血清、フィーダーフリーおよびストローマ細胞フリーで培養したpPS細胞を含む。pPS細胞をフィーダーフリーおよび無血清で培養するための条件が記載されており、例えば、Xu et al., (2001) Nat Biotechnol 19:971;Li et al., (2005) Biotechnol Bioeng 91:688を参照されたい。いくつかの態様において、pPS細胞を細胞凝集物、例えば、胚様体(EB)の形成に適した条件下で培養することが有利でありうる。EBの形成は以前に記載されており、例えば、米国特許広報第2006/0063255号およびPCT広報WO 01/51616を参照されたい。簡単に言うと、未分化pPS細胞をコラゲナーゼ処理によって回収し、細胞のクラスターまたは細片に分離し、非付着細胞培養プレートに凝集物として継代してもよい。回収したpPS細胞はいくつかの自然分化細胞を含んでいてもよい。自然分化細胞の数は、細胞がEBを形成し、次いで造血系統細胞に分化するにつれ、経時的に減少しうることが企図される。凝集物に適当な培地、例えば、X-VIVO 10;X-VIVO 15を供給してもよい。pPS細胞をフィーダーフリー、無血清およびストローマ細胞フリーで、EB形成の前および後の両方で増殖させてもよい。
特定の態様において、本発明は、pPS細胞を含む第一の細胞集団ならびに造血系統細胞および/または中胚葉細胞を含む第二の細胞集団を含む細胞培養物を提供する。造血系統細胞および/または中胚葉細胞は、pPS細胞の標的細胞型、例えば、中胚葉細胞、骨髄系前駆細胞、単球、樹状細胞などへの分化に有利な特異的増殖条件の結果として、培養物中に生じうる。増殖条件には、1つまたは複数の分化カクテル(以下に記載)およびいくつかの態様においては成熟カクテル(以下に記載)を提供することが含まれうる。細胞培養物は下記の1つまたは複数を含まなくてもよい:フィーダー細胞、ストローマ細胞、動物血清および/またはB27などの市販の血清代替品、ならびに外因性IL-3。
本発明は、インビトロでpPS細胞から分化した、すなわちpPS細胞のインビトロ子孫である、mDCまたはimDCを含む細胞集団を放射線照射する方法、ならびにインビトロでpPS細胞から分化したmDCまたはimDCを含む、放射線照射した細胞培養物を企図する。本発明の他の態様は、放射線照射した免疫調節製剤およびこれを作成する方法、ならびにmDCを含む放射線照射した細胞集団を用いて免疫応答を刺激する方法を企図する。本発明のさらに他の態様は、放射線照射したmDCを含むキットを企図する。
特定の態様において、本発明は、成熟樹状細胞のインビトロ産生のためのシステムであって、a)pPS細胞を含む第一の単離細胞集団、およびb)第一の細胞集団の一部のインビトロ子孫である、成熟樹状細胞を含む第二の単離細胞集団を含むシステムを企図する。
特定の態様において、本発明は、免疫応答を刺激するためのキットを提供する。一つの態様において、キットはpPS細胞およびDCを含む細胞培養物、ならびに1つまたは複数の容器を含む。任意に、キットは下記の1つまたは複数を含んでいてもよい:a)免疫応答を刺激するための説明書;b)DCを培養するための説明書;c)提供されるDCがimDCである場合、成熟カクテル;c)1つまたは複数の適当な培養容器;d)免疫応答を刺激するための1つまたは複数の抗原;e)1つまたは複数の免疫学的に適格な細胞;およびf)刺激された免疫応答を測定するのに適した試薬。キットは免疫応答をインビトロまたはインビボで刺激するために用いうる。DCはimDCまたはmDCでありうる。いくつかの態様において、DCを凍結して提供してもよい。細胞を液体窒素中で凍結し、約-140℃で保存してもよい。または、DCを包装し、冷蔵、例えば、約4℃で保存してもよい。成熟カクテルは、あらかじめ混合して、またはその各成分を別々に包装して供給してもよい。抗原はタンパク質もしくはペプチドとして、または抗原をコードする核酸、例えば、DNA、RNAとして提供してもよい。キットはDCに抗原を搭載するための説明書も提供しうる。搭載とは、DCを抗原と、抗原が免疫学的に適格な細胞に提示されるように接触させることを意味する。キットは、培養中でDCを増殖させるため、DCを抗原と接触させるため、DCを免疫学的に適格な細胞と接触させるための説明書をさらに含んでいてもよい。刺激された免疫応答を検出するのに適した試薬には、細胞増殖を測定するための3Hチミジン、抗原に対する免疫応答中に分泌される、IL-2、IFNなどのサイトカインに対する抗体が含まれうる。
本発明は、多数の造血および/または中胚葉系統の細胞を産生する方法を提供する。これらの細胞集団はいくつかの重要な研究、開発、および商業的目的のために用いることができる。
本発明の細胞を、そのような細胞およびそれらの様々な子孫の特徴に影響をおよぼす、因子(溶媒、低分子薬、ペプチド、オリゴヌクレオチドなど)または環境条件(培養条件または操作など)をスクリーニングするために商業的に用いることができる。特徴には、細胞の表現型または機能的形質が含まれうる。
特定の態様において、本発明は、抗原に対する免疫応答を刺激する方法であって、本発明の細胞、例えば、pPS細胞から分化したDCを抗原と接触させる段階を含む方法を提供する。抗原はタンパク質もしくはペプチドで構成されてもよく、または核酸、例えば、DNA、RNAで構成されてもよい。抗原がタンパク質またはペプチドである場合、樹状細胞はタンパク質またはペプチドを取り込み、それをMHCに関連して提示するために処理することになる。典型的に、処理には、抗原がMHCの溝に合うように、タンパク質分解が含まれる。抗原がタンパク質である場合、DC細胞はimDCであってもよい。抗原が全長タンパク質のペプチド断片である場合、DCはmDCであってもよい。抗原が核酸である場合、本発明は、核酸を細胞質中に送達するために、細胞膜を通過して核酸を輸送する当技術分野において公知の任意の手段を用いることを企図する。一つの態様において、細胞を電気穿孔して、核酸に細胞膜を通過させてもよい。細胞を抗原と接触させるために電気穿孔法を用いる態様において、適当な細胞はimDCかもしれない。また、細胞を抗原と接触させるために電気穿孔法を用いる他の態様において、適当な細胞はmDCかもしれない。細胞を、Gene Pulse Xcell(Bio-Rad Laboratories, Hercules, CA)を下記のパラメーターで用いて電気穿孔してもよい:300V、150uF、および100オーム。タンパク質発現レベルを、フローサイトメトリーまたはウェスタンブロット法によって判定してもよい。電気穿孔した細胞がimDCである場合、imDCがmDCに成熟するように、細胞を本明細書に記載の成熟カクテルと接触させてもよい。
本発明に従って作成した造血系統細胞を用いて、被験者において1つまたは複数の造血細胞集団を再構成してもよい。一例として、骨髄系前駆細胞を用いて、欠乏している細胞集団を再構成することにより、血球減少に関連する1つまたは複数の症状を改善してもよい。例えば、骨髄系前駆細胞を用いて、低血小板数、または低赤血球数の被験者の状態を改善してもよい。もう一つの例として、骨髄系前駆細胞などの造血系統細胞を用いて、凝固因子に関連する異常などの、遺伝的異常を有する被験者の1つまたは複数の症状を改善してもよい。したがって、本発明のいくつかの態様に従って作成した細胞を用いて、第VIII因子または第IX因子などの凝固因子のレベルを高めてもよい。他の態様において、造血系統細胞を用いて、HIV患者においてリンパ球集団、例えば、CD4リンパ球集団を再構成してもよい。
本発明に従って産生したmDCはPBMCから単離したmDCに機能的に匹敵する。例えば、本発明のmDCは抗原を取り込み、処理して提示し;特異抗原の提示に応答してT細胞増殖を刺激し、かつ抗原特異的T細胞が仲介する標的細胞の細胞溶解を誘導することができる。加えて、本発明のmDCの機能は放射線照射後でも維持される。したがって、本発明のmDCは特異抗原に対する免疫応答を刺激するためにヒト被験者に投与するためのDC源を提供する一方で、未分化細胞および病原性物質への曝露のリスクを最小限にとどめうる。
本発明の細胞を、他の系統からの細胞中で優先的に発現されるcDNAが比較的混入していないcDNAライブラリを調製するために用いてもよい。例えば、造血系統細胞を1000rpm、5分間の遠心により回収し、次いでmRNAを調製し、逆転写する。造血系統細胞の発現パターンを他の細胞型、例えば、pPS細胞と、Fritz et al., (2000)Science 288:316により総説が発表されているマイクロアレイ分析により比較してもよい。細胞は実質的に、それらが分化した元の親pPS細胞株と遺伝的に同一であるため、造血系統細胞の分化および成熟に関与する遺伝子を調べるために特に適した系を提供する。例えば、親細胞株および造血系子孫からの核酸ライブラリを調製し、差引きハイブリダイゼーションを用いて子孫細胞の分化および成熟において重要な遺伝子を単離してもよい。
本発明は、pPS細胞を造血系統細胞に分化させる方法を提供する。pPS細胞には任意の霊長類多能性細胞が含まれる。多能性細胞は、適当な増殖条件下で、中胚葉、内胚葉および外胚葉の3つの一次胚葉それぞれから少なくとも1つの細胞型を生成することができる。pPS細胞は前胚、胚もしくは胎児組織または成熟分化細胞から生じうる。または、樹立pPS細胞株は本発明を実施するのに適した細胞源でありうる。典型的には、pPS細胞は悪性の源由来ではない。pPS細胞は、免疫不全マウス、例えば、SCIDマウスに移植すると、奇形腫を形成することになる。
pPS細胞を、様々な基質、培地、および他の補充物質ならびに当技術分野において公知の因子を用いて培養してもよい。いくつかの態様において、適当な基質には、下記の1つまたは複数で構成されるマトリックスが含まれうる:ラミニン、コラーゲン、フィブロネクチン、ビトロネクチン、ヘパリン硫酸プロテオグリカン。いくつかの態様において、マトリックスはMatrigel(商標)(BD Biosciences, San Jose, CA)として市販されている、マウスEHS肉腫からの基底膜の可溶性抽出物を含みうる。他の態様において、マトリックスは、ヒト化された、またはマウス起源の、1つまたは複数のヒトの単離マトリックスタンパク質、例えば、CELLstart(商標)(Invitrogen, Carlsbad, CA)を含みうる。霊長類多能性幹細胞は、増殖を促進する一方で、分化を阻害する培養条件を用いて、培養中に継続的に増殖させることができる。例示的培地は、80%DMEM(Knock-Out DMEM, Gibcoなど)、20%の規定ウシ胎仔血清(FBS、Hyclone)または血清代替品(US 2002/0076747 A1, Life Technologies Inc.)のいずれか、1%非必須アミノ酸、1mM L-グルタミン、および0.1mMβ-メルカプトエタノールから作成してもよい。他の適当な培地には、X-VIVO(商標)10(Lonza, Walkersville, MD)などの無血清合成培地が含まれる。
本発明の実施において有用な一般的技術をさらに精巧にするために、当業者は標準の教科書および細胞生物学、組織培養、発生学、および免疫学の総説を参照することができる。
本発明の細胞を、分化の前または後のいずれかで遺伝子操作することにより、1つまたは複数の遺伝子改変を含むようにすることができる(US 2002/0168766 A1)。例えば、いくつかの態様において、細胞が発育が制限された系統の細胞または最終分化細胞に前進する前または後のいずれかで、テロメラーゼ逆転写酵素を発現するように細胞を遺伝子改変させることにより、それらの複製能力を高めるよう処理することができる(US 2003/0022367 A1)。
実施例1:分化カクテルを変化させることによるhES細胞のmDCへの分化
本実施例において、まずpPS細胞を分化カクテルと共に培養してimDCを得、次いでimDCを成熟カクテルと共にさらに培養することにより、pPS細胞をmDCに分化させた。分化カクテルは外因性サイトカインからなり、これらは細胞がimDCステージに分化するにつれ、実験の経過中に変化させた。(図1a)。ヒトES細胞株H1(Thomson et al., (1998)Science 282:1145)を、動物由来産物を含まない規定の無血清培地中、フィーダーフリーで培養した(Xu et al., (2001)Nat Biotechnol 19:971;Li et al., (2005)Biotechnol Bioeng 91:688)(図1b)。細胞を分化および成熟プロトコルの間、ストローマ細胞フリーでも培養した。
pPSをimDCに分化させるプロセスの間に生じる前駆細胞集団を特徴づけるために、実施例1に記載の細胞培養物を、RT PCRおよびリアルタイム定量的PCR(以下に記載のとおり)を用いて転写因子発現について経時的に評価し、フローサイトメトリー(前述のとおり)により細胞表面マーカーの発現について経時的に評価した。
hES由来imDCの抗原を処理する能力を試験するために、蛍光色素DQ-OVA(Invitrogen, Carlsbad, CA)をPBSに1mg/mlで溶解し、実施例1に記載のhES由来imDCに100μg/mlで加えた。タンパク質をpH非感受性BODIPY-Fl色素で標識した。色素は、タンパク質が完全な状態であれば自己消光するが、タンパク質が変性されているか、またはタンパク質分解を受けていれば、明るい緑色蛍光を発する。細胞を37℃または4℃(背景蛍光のための対照として)のいずれかでインキュベートし、フロー緩衝液で2×洗浄した。データをFL1においてFACSCalibur(商標)(Becton Dickinson, San Jose, CA)で収集した。処理した細胞は蛍光を発することが判明し、タンパク質がimDCによってタンパク質分解されたことを示していたが、対照細胞は蛍光を発しなかった(図5a)。
定性的サイトカインアレイ分析を、Human Cytokine Array IIIおよびVキット(Raybiotech, Norcross, GA)を用いて、実施例1記載の方法に従って得たimDCおよびmDC両方で実施した。検定を製造者の説明書に従って実施した。mDCは以下の炎症誘発性サイトカインを産生することが判明した:IL-6、IL-7、IL-8、およびIL-10。IL-7はT細胞生存にとって重要であると考えられる。IL-8は走化性刺激であると考えられる。サイトカインIL-6、IL-10、およびIL-12を、BD Cytometric Bead Array(BD Biosciences, San Jose, CA)を用い、製造者の説明書に従って定量した。hES由来の未熟および成熟DCからの上清を回収し、Amicon Ultra-15 10,000 NMWL(Millipore, Bedford, MA)遠心チューブを用いて濃縮した。上清をヒトIL-6、IL-10、およびIL-12ビーズフレックスセット(BD Biosciences, San Jose, CA)に加え、25℃で1時間インキュベートした。PE(BD Biosciences, San Jose, CA)に結合した抗体検出試薬を加え、室温でさらに2時間インキュベートした。試料を1×洗浄し、洗浄緩衝液(BD Biosciences, San Jose, CA)に再懸濁し、FACSCaliber(商標)(Becton Dickinson, San Jose, CA)でのフローサイトメトリーにより回収した。サイトカイン濃度をFCAP Array Software(BD Biosciences, San Jose CA)を用いて定量した。結果を図6Aに示し、これは3つのサイトカインすべての顕著なレベルがmDCによって産生されることを示していた。
AIM-V培地(Invitrogen, Carlsbad, CA)を、孔径8.0uMのインサートを含むTranswell 24穴プレート(Corning, Corning, NY)の上部および下部チャンバーに加え、37℃、5%CO2で終夜インキュベートした。各ウェルから培地を除去した後、ケモカインMIP3β(100ng/ml)を含む、または含まないAIM-V培地0.6mlを下部チャンバーに加えた。hES由来成熟DC(実施例1に記載のとおり)を回収し、AIM-V培地で2×洗浄した。細胞をAIM-V培地に2.0×106細胞/mlで再懸濁し、0.1mlを上部チャンバーに加えた。トランスウェルプレートを37℃、5%CO2で2時間インキュベートした。下部チャンバーに遊走した細胞の数を、血球計数器を用いて求めた。結果を図6Dに示し、これらは本発明のmDCがMIP3βに応答して遊走することを示していた。
実施例1の方法に従って産生したmDCの、免疫応答を刺激する能力を特徴づけるために、いくつかの検定を実施した。第一に、mDCが強い未処置アロ応答を刺激する能力を有することを示すために、混合リンパ球反応(MLR)検定を行った。
hES細胞をimDCおよびmDCに増殖および分化させるための培養条件を、様々な外因性サイトカインを含む分化カクテルを比較するために、用いる分化カクテルを変える以外は、実施例1に記載のとおりに実施した。hES細胞をimDCに分化させる能力について、7、5、4および3つのサイトカイン(増殖因子)の様々な組み合わせを試験した。表IIは、分化カクテルが7、5、および4つの外因性サイトカインを含む実験の詳細を示す(プラス記号は因子の存在を示し、マイナス記号はその因子を使用しなかったことを示す)。表の下半分の数値は、パーセンテージの真上に示す対応するカクテルで得られた、各細胞マーカーのパーセンテージを示す。表IIIは、分化カクテルが4および3つの外因性サイトカインを含む実験の詳細を示す(プラス記号は因子の存在を示し、マイナス記号はその因子を使用しなかったことを示す)。対応するカクテルについて得られたマーカーのパーセンテージに関する一式を表の下半分に示し、パーセンテージは数値データの上に示すカクテルに対応している。表IV〜VIは、実験の時間経過中の分化カクテルの組成(表IIおよび表IIIに記載のとおり)に関する詳細を示す(「X」は因子が明記された期間存在したことを示す)(「d」はこれらの表では「日」の略語として用いる)。
本発明の様々な態様に従ってimDCに分化させたpPS細胞を、サイトカイン/因子の異なる組み合わせを用いてmDCに成熟させた。細胞濃度0.05×106細胞/ウェルを96穴プレートに播種し、表VIIIに示すサイトカイン/因子の様々な組み合わせを補充したX VIVO-15培地中で24時間培養した。用いた因子の濃度は上の表VIIに示したとおりであった。Poly I:Cには10ug/mlを用い;PGE2には1ug/mlを用い;iL-6には10ng/mlを用いた。試験したすべての成熟カクテルは50ng/mlのGM-CSFを含んでいた。imDCのmDCへの成熟の指標として、24時間の時点の上清からのIL-12およびIL-10のレベルを、BD(商標) Cytometric Bead Array(BD Biosciences, Franklin Lakes, NJ)を用いて測定した。結果は、4つという少ないサイトカインでimDCのmDCへの成熟を刺激しうることを示唆するものであった。
PBMCを健常ヒト供与者のHLA-A2+軟膜から、Ficoll Plaque-Plus(GE Healthcare Bioscience AB, Piscataway, NJ)分離法を用いて単離した。imDCを生成するために、HLA-A2+PBMCからの単球をCD14+マイクロビーズ(Miltenyi, Aurburn, CA)を用いて単離し、rhGM-CSF(1000U/ml)(Berlex, Richmond, CA)およびrhIL-4(1000U/ml)(R&D systems, Minneapolis, MN)を含む無血清AIM-V培地(Invitrogen, Carlsbad, CA)に移し、37℃、5%CO2で5日間インキュベートした。DCをTNFα(10ng/ml)(R&D systems)、IL1β(10ng/ml)(R&D systems)、IL-6(10ng/ml)(R&D systems)、およびPGE2(1ug/ml)(R&D systems)を含むサイトカインカクテルを加えることにより24時間成熟させた。mDCを回収し、AIM-V培地で2×洗浄し、200ulのAIM-V培地に再懸濁し、hTERTタンパク質のアミノ酸540で始まる九量体である540 hTERTペプチド(100ug/ml)(AnaSpec Inc, San Jose, CA)を37℃、5%CO2で2時間適用した。
hES由来mDC(実施例1)を200ulの無血清AIM-V培地(Invitrogen, Carlsbad, CA)に再懸濁し、540 hTERTペプチド(100ug/ml)を37℃、5%CO2で2時間適用した。非適用mDCを対照として用いた。非適用hES由来mDCを対照として用いた。mDCをAIM-V培地で2×洗浄し、AIM-V培地に再懸濁し、抗IFN-γAb(10ug/ml)(Mabtech, Mariemont, OH)でコーティングしたELISpotプレートに540 hTERT T細胞株と共に刺激細胞と応答細胞の比1:10で播種した。検定プレートを37℃、5%CO2で16〜24時間放置し、製造者が提供する説明書に従って展開した。スポットをCTL Analyzer(Cellular Technology Limited, Decatur, IL)を用いてカウントした。結果を図8に示し、これらはhESから分化したmDCがhTERT抗原に対する特異的T細胞応答を刺激することを示している。
540 hTERT T細胞株をあらかじめ温めたPBS/0.1%BSAに1.0×106/mlで再懸濁した。CFSE(Invitrogen, Carlsbad, CA)を細胞に、最終濃度2uMで加え、37℃で10分間インキュベートした。染色を、10%FBS(Clonetech, Mountain View, CA)を含む、あらかじめ冷却したAIM-V培地を加えることにより停止した。細胞を2×洗浄した後、検定の準備をした。成熟hES由来DC(実施例1)に10μg/mlの540 hTERTペプチド(Anaspec, San Jose, CA)を37℃、5%CO2で2時間適用し、AIM-V培地で2×洗浄した後、96穴U底Falcon(商標)プレート(BD, San Jose, CA)に2×104/ウェルで播種した。CFSE標識540 hTERT T細胞株を2×105細胞/ウェルで播種した。非適用hES由来mDCを対照として用いた。第4日に、細胞を回収し、APC(ProImmune, Bradenton, FL)に結合した540五量体試薬で染色した。細胞をFACS緩衝液で2×洗浄し、7AADで染色した後、FACSCaliber(商標)(Becton Dickinson, San Jose, CA)を用いて回収した。分析をFlowJoソフトウェア(Tree Star, Ashland, OR)を用いて行った。結果を図9に示し、これらはhES細胞から分化したmDCがHLA-A2に関連してhTERT抗原を提示し、抗原特異的T細胞増殖を刺激しうることを示している。
成熟樹状細胞を、実施例1に記載の方法に従い、インビトロでpPS細胞から分化させた。インビトロでhES細胞から分化させた樹状細胞(hESC-DC)における放射線照射の効果について取り組むために、放射線照射および非放射線照射hESC-DCのT細胞の抗原特異的エフェクター応答を刺激する能力を比較した。CMVペプチドpp65(アミノ酸配列495〜503)を用いて、CD8+リンパ球へのhESC-DC抗原提示を示した。HLA-A2に複合したpp65を認識するCD8+T細胞を含む、特徴づけられたPBMC応答細胞(Cellular Technology Limited, Decatur, IL)を応答細胞として用いた。pp65特異的抗原提示のために、成熟hESC-DCを10μg/ml pp65ペプチドを補充した、または補充なしの無血清AIM-V培地(Invitrogen, Carlsbad, CA)150ulに再懸濁した。細胞を37℃、5%CO2で2時間インキュベートし、次いでAIM-V培地(Invitrogen, Carlsbad, CA)で2回洗浄した。pp65適用および非適用hESC-DCの一部に、Torrex 150D X線検査システム(EG&G Astrophysics Research Corporation, Long Beach, CA)を用いて、2,000radで4分14秒間X線照射した。X線照射および非照射hESC-DCをELISPOTプレートに1×104細胞/100ul/ウェル、応答細胞と刺激細胞の比10:1で播種した。特徴づけられたPBMC応答細胞を37℃の水浴中で解凍し、2回洗浄し、AIM-V培地(Invitrogen, Carlsbad, CA)中に再懸濁し、ELISPOTプレート(Millipore, Bedford MA)に1×105細胞/100ul/ウェルで播種した。ELISPOTプレートを10ug/mlの抗IFNγAb(Mabtech, Mariemont, OH)で終夜(16〜24時間)コーティングした。検定プレートを37℃、5%CO2で16〜24時間放置し、Mabtechが提供する説明書に従って展開した。スポットをCTL Analyzer(Cellular Technology Limited, Decatur, IL)を用いてカウントした。図10に示す結果は、放射線照射hESC-DCが抗原特異的様式でIFNγ産生を刺激する能力を維持していたことを示している。
成熟樹状細胞を実施例12で用いたものと同じプロトコルに従って分化させた。放射線照射および非放射線照射mDC(hESC-DC)の走化性リガンドMIP3β(図11のMIP3b)存在下で遊走する能力を、インビトロトランスウェル検定を用いて調査した。AIM-V培地(Invitrogen, Carlsbad, CA)を、孔径8.0uMのインサートを含むTranswell 24穴プレート(Corning, Corning, NY)の上部および下部チャンバーに加え、37℃、5%CO2で終夜インキュベートして膜を平衡化した。mDCを回収し、AIM-V培地で2回洗浄した。細胞をAIM-V培地に1.5×106細胞/mlで再懸濁し、これらの細胞の一部に、Torrex 150D X線検査システム(EG&G Astrophysics Research Corporation, Long Beach, CA)を用いて、2,000radでX線照射した。トランスウェルから培地を除去した後、ケモカインMIP3β(100ng/ml)を含む、または含まないAIM-V 0.6mlを下部チャンバーに加えた。X線照射および非照射mDC(0.15×106細胞)の0.1ml量を上部チャンバーに加えた。トランスウェルプレートを37℃、5%CO2で2時間インキュベートした。下部チャンバーに遊走した細胞の数を、血球計数器を用いて求めた。結果を図11に示し、これらは放射線照射がmDCのMIP3βに応答して遊走する能力に影響をおよぼさないことを示していた。
2つの市販の培地、mTeSR(商標)無血清培地(Stem Cell Technologies, Vancouver, BC, Canada)およびXVIVO-10(商標)(Lonza, Walkersville, MD)のmDC細胞収量に対する影響を調べた。H1 hESC培養方法及び分化方法を実施例1に記載のとおりに実施した。各回収時の成熟DCの数を、XVIVO-10またはmTeSR培養物から生成したhESCの間で比較した、図12。最初の分化から合計3回の回収を実施した。培地はいずれもインビトロでhESから分化したmDCをうまく産生したが、結果はmTeSR中で培養したhESCはX VIVO-10よりも良好な細胞収量を提供することを示唆している。
成熟DCはT細胞応答を刺激する能力を有し;したがって、hESC由来DCの成熟プロセスを最適化することが望ましい。(例えば、Chiara et al, 2007参照)。本発明者らは実施例1に記載のとおりにH1 hESCを分化させたが、未熟および成熟hESC由来DCを生成する段階の間、Cellgro DC培地を用いた。24時間の成熟後、細胞を回収し、1)細胞表面マーカー発現、2)遊走、および3)IL-12発現に基づき、成熟DC表現型の存在について評価した。
H1 hESCを、hESCを培養するためにmTeSR(商標)を用い、hESC由来未熟および成熟DCを生成するためにCellgro(商標) DCを用いる以外は、実施例1に記載の方法に従って分化させた。2.0〜4.0e6の間のhESC由来成熟DCに、0.4cmキュベット(Biorad, Hercules, CA)中、Biorad Gene Pulser Xcell(Biorad, Hercules, CA)を用い、以下のパラメーターを用いて、hTERT-LAMPまたはGFPをコードするRNAを電気穿孔した:300V、150uF、および100Ω。電気穿孔した細胞をCellgro(商標) DC培地で1×洗浄し、細胞を成熟培地にさらに6時間移した。実施例9および10に記載のとおり、GFP-およびhTERT-LAMP RNAを電気穿孔したhESC由来成熟DCを回収し、540 hTERT T細胞株と同時インキュベートして、IFNγの発現を検出した。結果は、hTERT-LAMP RNAを電気穿孔したhESC由来成熟DCは、GFP-RNAをトランスフェクトしたhESC由来DCと比べて、540 hTERT特異的T細胞株からのIFNγの高いレベルを刺激することを示した(図16)。このデータは、hESC由来DCはhTERT抗原を処理および提示する能力を有することを示唆している。
Claims (7)
- インビトロの細胞培養物であって、
(a)霊長類多能性幹細胞を含む第一の細胞集団と、
(b)該第一の細胞集団の一部のインビトロで分化した子孫である未熟樹状細胞を含む第二の細胞集団と、
(c)顆粒球-マクロファージコロニー刺激因子(GM-CSF)および骨形成タンパク質4(BMP-4)を含む培養培地と、
を含む細胞培養物。 - 血管内皮増殖因子(VEGF)、幹細胞因子(SCF)、胎児肝キナーゼリガンド(FLT3L)、トロンボポエチン(TPO)、インターロイキン4(IL-4)、およびインターロイキン3(IL-3)からなる群から選択される1または複数の因子をさらに含む、請求項1記載の細胞培養物。
- CD40リガンド(CD40L)をさらに含む、請求項2記載の細胞培養物。
- 成熟樹状細胞をインビトロで生産するためのシステムであって、
(a)霊長類多能性幹細胞を含む第一の単離細胞集団と、
(b)該第一の単離細胞集団の一部のインビトロで分化した子孫である成熟樹状細胞を含む第二の単離細胞集団と、
(c)顆粒球-マクロファージコロニー刺激因子(GM-CSF)および骨形成タンパク質4(BMP-4)を含む培養培地と、
を含むシステム。 - 前記成熟樹状細胞の少なくとも5%が、CD86およびCD83から選択される1または複数のマーカーを発現する、請求項4記載のシステム。
- 前記CD86およびCD83から選択される1または複数のマーカーを発現する成熟樹状細胞が、MHC IIおよびCCR7から選択される1または複数のマーカーをさらに発現する、請求項5記載のシステム。
- 前記第二の単離細胞集団の成熟樹状細胞が、有糸分裂不活性である、請求項4記載のシステム。
