(a)電位依存性T型カルシウムチャネルの発現量が異なる2種類の神経細胞を調製する工程;
(b)電位依存性T型カルシウムチャネルの発現量が低い細胞(細胞A)を用いたアッセイ系(アッセイ系A)に被検化合物を添加し、添加による細胞内のカルシウムイオンレベルの変化を測定する工程;
(c)電位依存性T型カルシウムチャネルの発現量が高い細胞(細胞B)を用いたアッセイ系(アッセイ系B)に被検化合物を添加し、添加による細胞内のカルシウムイオンレベルの変化を測定する工程;
(d)アッセイ系Bにおけるカルシウムレベルの上昇がアッセイ系Aにおける上昇よりも大きい化合物を選抜する工程
を含む、前記スクリーニング方法。
式(II)の化合物は、塩基存在下、例えば室温下、クロロホルムなどの適当な溶媒中で、クロロアセト酢酸エステル(例えば、クロロアセト酢酸エチルエステルなど)と反応させることにより、式(III)の化合物に変換することができる。式(III)の化合物は、例えば過熱下(例えば溶媒還流下)、適当な溶媒(例えばクロロホルムなど)の溶媒中で、アミンと反応させることにより、式(IV)の化合物に変換することができる。所望の化合物に対応する式(II)の化合物を用いることにより、または式(III)もしくは式(IV)の化合物の置換基を変換することのより、所望の式(I)の化合物を合成することができる。また必要に応じて保護基の導入および脱離を行ってもよい。
試験において以下の試薬を使用した:
雄性マウスC57BL/6(10-12週齢)の脳を矢状断で切断し、海馬 CA1 領域を含む冠状断脳スライス (400 μm) を作製した。海馬スライスを95% O2/5% CO2 ガスで飽和させた人工脳脊髄液 (126 mM NaCl, 5 mM KCl, 26 mM NaHCO3, 1.3 mM MgSO4-7H2O, 1.26 mM KH2PO4, 2.4 mM CaCl2-2H2O, 10 mM グルコース) 中において、34 ℃で2時間回復させた。海馬スライスを測定チャンバーに移し、ST101 (100 pM)、SAK1 (100 pM)、SAK3 (100 pM) を人工脳脊髄液に添加して、灌流適用した。その後、海馬CA1領域に入力するSchaffer 側枝と交連線維を刺激電極で刺激し、それにより惹起される CA1 領域における シナプス後集合電位(fEPSP) の測定を行った。測定後、スライス切片から海馬 CA1 領域のみを切り出し、免疫ブロットによる測定用に、-80 ℃にて凍結保存した。
上記の凍結マウス脳スライス切片 (海馬 CA1 領域) を用いて、ホモジナイジングバッファー (50 mM Tris-HCl pH 7.5、0.5 % Triton X-100、4 mM EGTA、0.5 M NaCl、10 mM EDTA、1 mM Na3VO4、30 mM ピロリン酸ナトリウム、50 mM NaF、1 mM DTT、100 nM カリクリンA、50 μg/mL ロイペプチン、25 μg/mL ペプスタチンA、50 μg/mL トリプシン阻害剤) を1サンプルあたり150 μL 添加し、氷上でホモジナイズした。その後、冷却遠心分離機 (14,000×g、10 分、4 ℃) で遠心分離後に、上清100 μL を採取し、6×Laemmli buffer を20 μL 加え100 ℃ で3分間煮沸し、免疫ブロット用のサンプルとした。なお、上清の一部を用いてBradford法により蛋白質濃度を測定した。同量の蛋白質を含む試料を、10% SDS- ポリアクリルアミド電気泳動ゲル (SDS-PAGE gel) へ添加し、ゲル1枚当たり80 mA で180分間、SDS-PAGE を行った。泳動分離後、PVDF メンブレンに70 V で120分間転写を行った。抗体の非特異的結合を防ぐために、スキムミルクを5 % の濃度で溶解した Tween、Tris-bufferd saline (TTBS:50 mM Tris-HCl, pH 7.5, 150 mM NaCl および 0.1 % Tween 20) 中にメンブレンを浸し、常温でブロッキングを 1 時間行った後、次に示す一次抗体を4 ℃で一晩反応させた。一次抗体には、抗リン酸化CaMKII抗体 (1:5000, Fukunaga et al., J. Biol. Chem. 267, 22527-22533, 1992) 及び 抗CaMKII抗体 (1:5000, Fukunaga et al., J. Neurochem. 51, 1070-1078, 1988) を用いた。TTBS 溶液で洗浄後、メンブレンを 西洋ワサビペルオキシダーゼ (HRP)-標識化抗ウサギ IgG 抗体 (1:5000, GE healthcare, Buckinghamshire, UK) の二次抗体と常温で 60分間反応させた。TTBS 溶液による洗浄後、二次抗体に標識された HRP を ECL detection system (GE healthcare) により発光させ、X 線フィルム (Fuji Medical X-ray Film, Fuji Film) に感光させることによりバンドの検出を行った。免疫反応物の定量解析には NIH image 解析ソフトを用いた。
マウス神経芽腫細胞株 Neuro2A 細胞は、Human Science Research Resources Bank (#IFO50081, Osaka, Japan) から購入した。Neuro2A 細胞は、10% 牛胎児血清を含むダルベッコ変法イーグル培地(DMEM) に、37 ℃/5% CO2の条件下で培養した。その後 Neuro2A 細胞を、ガラスボトムディッシュに1〜2 x 106 cell/φ35-mm ディッシュの濃度で播種し標準培地に24時間培養した後、Lipofectamine 2000 (Invitrogen, Carsbad, CA, USA) を用いてT型カルシウムチャネル(Cav3.1)遺伝子を含んだプラスミドベクター (Lipofectamine 2000 : DNA=1μl : 1μg、1.5 ml の無血清培地) を用いてトランスフェクト(遺伝子導入)した。T型カルシウムチャネル(Cav3.1)遺伝子はOriGene Technologies, Inc.から、プラスミドベクターpCMV-SPORT6 は、Thermo Fisher Scientific Ltd.から、Lipofectamine 2000はInvitrogenから購入し、前述のLipofectamine 2000とプラスミドベクターとの比率調整以外は、Lipofectamine 2000に添付のプロトコルに従いトランスフェクトした。トランスフェクトから6時間後に無血清培地を標準培地に置換し、さらに48時間培養した後、Cav3.1発現を確認後に、カルシウムイメージング(カルシウム濃度測定)を行った。
実験には、10週齢のDDY 雄性マウス (Nippon SLC, Hamamatsu, Japan) を使用した。動物は東北大学大学院薬学研究科の動物実験施設において一定条件下 (温度22±2 ℃、12h:12h明暗サイクル) で、水・飼料を自由に与えて飼育を行った。尚、本論文中に記載した動物の取り扱い及び動物実験は、東北大学実験動物委員会の承認を得て、東北大学動物実験指針に基づいて行った。
海馬内で遊離するアセチルコリンの量をマイクロダイアリス法により測定した。ガイドカニューレ及び透析プローブ (AG-4; Eicom, Kyoto, Japan) の設置は、1.5% ハロタン (Takeda Chemical Industries Ltd., Osaka, Japan) で麻酔を維持した条件でマウスを脳定位固定装置に固定し、左側海馬 (AP=+3, L=+3, V=-1.8) にガイドカニューレを設置し、ガイドカニューレに透析プローブを挿入した。マウス海馬からの脳内 ACh 試料採取は、プローブ設置手術の翌日に行った。手術からの回復を確認したマウスを、無麻酔、自由行動下でプローブをマイクロポンプ (ESP-64; Eicom) に接続して、リンゲル液 (1.3 mM CaCl2, 3mM KCl, 146 mM NaCl および 1mM MgSO4) を毎分2μL の流速で流した。ダイアリシス試料は19.5分毎に採取し、オートインジェクター (EAS-20; Eicom) を用いてアセチルコリン分離用カラムに注入し、分析には微量生体資料分析システム (HTEC-500; Eicom) を用いた。SAK3は、試験開始40分前(図8のグラフで−40分の時点)に腹腔内投与した。ニコチン誘発性 ACh 遊離は、3mM ニコチンを含むリンゲル液をプローブから試験開始から20分間(図20のグラフの0〜20分の間)灌流投与し、その後は通常のリンゲル液に置換え測定した。
マウス大脳皮質の分散培養は既報の方法(Fukunaga et al., J. Biol. Chem. 267, 22527-22533, 1992)に従って行った。3週間培養後にインキュベーションメディウム(KRH)に 100pMのSAK3, SAK8, SAK9を添加して、30分間インキュベーションした。刺激後メディウムを除去、細胞は凍結保存した。凍結した大脳皮質細胞は試験例2における方法と同様の方法により、ホモジナイズ後に免疫ブロット法にて、CaMKIIの活性化状態(自己リン酸化反応)を定量分析した。

- 基−NR15R16が、C1−6アルキルアミノ、ジ(C1−6アルキル)アミノ、[(C1−6アルコキシカルボニル)C1−6アルキル]アミノ、もしくは[(C6−10アリール)C1−6アルキル]アミノを表し、または基−NR15R16が、1−ピロリジニル、1−ピペリジニル、1−ピペラジニル、4−モルホリニル、もしくは1−ホモピペリジニルから選択される含窒素ヘテロ環基を表し、ここで該含窒素へテロ環基は請求項1に定義された1以上の置換基により置換されていてもよい、請求項11に記載の電位依存性T型カルシウムチャネル活性化剤。
- R3、R4、R5、およびR6が、それぞれ独立に、水素原子、およびC1−6アルキルから選択される、請求項9〜12のいずれか1項に記載の電位依存性T型カルシウムチャネル活性化剤。
- 式(I)で表される化合物または医薬として許容なその塩が、請求項6に記載の化合物および医薬として許容なそれらの塩から選択される、請求項9〜13のいずれか1項に記載の電位依存性T型カルシウムチャネル活性化剤。
