Abstract
Description
上記のbAVMおよび脳動脈瘤に対し、カテーテルを用いる塞栓術式は、次のような問題を抱えている。bAVMに対するOnyx塞栓術には、Onyxを構成する成分に起因する複数の理由により、患者への使用上、著しく危険な特性がある。最大の理由はOnyxの有効成分が、合成有機高分子であるエチレンビニルアルコール(EVOH)コポリマー(EVALともいう)であり、かつ、生体内にこれを注入するための溶媒としてジメチルスルホキシド(DMSO)を採用しているという点である。Onyx塞栓術においては、多くの場合、大腿部血管から脳動脈内患部付近に挿入されたマイクロカテーテル先端から、Onyxの有効成分であるEVOHのDMSO溶液が患部付近に注入される。このとき、DMSOは血中水分により徐々に希釈され、水不溶性のEVOHコポリマー小塊が血管内に沈殿する結果として、標的血管塞栓が起こる。
発明者らの基礎研究結果によると、本種のシルクタンパク質を構成する主要なアミノ酸は、グリシン、アラニン、セリン等の疎水性アミノ酸である。この特徴は、カイコのシルクフィブロインタンパク質と同様のアミノ酸組成であることが注目される。本種のシルクタンパク質が持つ特徴は、上記アミノ酸の他にも、チロシンおよびプロリンの含量の高さであり、カイコシルクでは組成比5%に満たないアミノ酸を、その倍の10%程度含む。さらに、上記のセリンの側鎖水酸基の多くがホスホリル化を受けているという点が最大の特徴である。ホスホリル化を受けたセリン残基をホスホセリンという(以降、「pSer」と記す)。セリン側鎖上ホスホリル基は、水中で1価または2価のマイナスイオンとなり、2価のプラスイオンであるカルシウムイオン等との静電結合を経て、シルクタンパク質分子は互いに凝集をして、水不溶性のシルク繊維に変化する。これは、カイコのシルク繊維形成機構とは全く異なり、本種のシルクタンパク質固有のタンパク質化学的性質なのである。本種のシルクタンパク質のアミノ酸組成については後述する(実施例6)。
本発明の血液凝固剤溶液の製造方法はまた、ヒゲナガカワトビケラの5齢幼虫のシルク繊維ネットを糸状粉末とし、第1段階として、この粉末をpH 6を超えpH 9未満の抽出用緩衝液であって、尿素または臭化リチウムを含む抽出用緩衝液に浸してインキュベートし、遠心上清を採取し、抽出残渣を得、次いで、第2段階で、第1段階で得られた抽出残渣を、カルシウムイオンと結合するキレート剤を添加した第1段階抽出用緩衝液に浸してインキュベートし、得られた抽出液からキレート剤と結合したカルシウムイオンを除いた後、36 ± 3 % のlesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)の規格化リン酸化率を有するSmsp-1およびSmsp-4であって、lesp-400kDa(Smsp-1):lesp-14kDa(Smsp-4)の重量比が9:1で存在するSmsp-1およびSmsp-4を有する液体の血液塞栓性タンパク質固有組成物E2Pを得、血液凝固を促進加速させる前記液体の血液塞栓性タンパク質固有組成物E2Pを有効成分とし、この液体の血液塞栓性タンパク質と、リン酸イオンを含まない、体液(恒温動物の血液)と等張な緩衝液とからなる血液凝固剤溶液を製造することを特徴とする。
本発明で得られる液体の血液塞栓性タンパク質固有組成物E2Pに係わる実施の形態によれば、液体の血液塞栓性シルクタンパク質固有組成物E2Pは、水生昆虫のシルク繊維ネット、好ましくは水生昆虫における酸化酵素活性が上昇する前の水生昆虫5週齢幼虫のシルク繊維ネットから抽出される、カルシウムイオンを含まないものであり、カルシウムイオンが除去されたpSerを含むlesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)からなるタンパク質であり、カルシウムと結合することで血管を塞栓する機能を有し、前記水生昆虫が、例えば、ヒゲナガカワトビケラ幼虫等からなる毛翅類、および近縁種である双翅目から選ばれる。そして、この液体塞栓性タンパク質E2Pは、標的血管部位の血液凝固を促進加速させる。
S. marmorataの5齢幼虫をガラス製の筒状飼育容器(直径13mm、長さ95mm)に入れて3-5日間流水飼育し、ガラス飼育容器内に吐糸されたシルク繊維ネットを回収し、蒸留水を用いてきれいに洗浄し、メタノールに浸漬した。シルク繊維ネットをメタノールでも洗浄し、その後、減圧乾燥した。試料のメタノール洗浄により、シルク繊維ネットに付着する微生物等を除去できる。なお、回収するシルク繊維ネットに夾雑物が混入しないように、虫飼育中のS. marmorata 5齢幼虫には、一切の給餌をせず、摂食させなかった。
lespic E2Pの収率を向上させるための次のような技術的な工夫を凝らした。収率を上げるための必須成分は、5-9Mの尿素または7-10Mの臭化リチウムの添加が効果的であった。しかし、こうした条件下での処理であっても、収率は最大50%程度までしか上昇しなかった。これは、S. marmorata 5周齢幼虫の生理状態が、蛹への変態準備に入る時に、lespic E2Pとして可溶化される有効成分であるlesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)の性質を変化させ、第2段階の抽出に用いる液体へと溶けにくくなるためである。本発明者らが実施した基礎的な研究結果によると、この現象には、ある酸化酵素活性上昇が関与することが分かっている。従って、lespic E2P収率向上のための好適手段は、この酵素活性が上昇するに先だって、S. marmorata 5周齢幼虫を捕集後、実験室飼育によりシルク繊維ネットを回収することである。このシルク繊維ネットを処理する際に、本発明の製造方法に従って5-9 M 尿素や 7-10 M 臭化リチウムを用いた場合には、収率が50%以上に向上し、製造効率はより良好、かつ、より安定となった。S. marmorata 5周齢幼虫シルク繊維ネットを含め、天然由来の原料はえてしてそのロットにより製造効率は不安定であるのが通常であるが、上記の成分をこのような範囲で添加し、本発明記載の製造法を用いることで、血液塞栓性タンパク質固有組成物E2Pの収率は50%以上に向上するだけでなく、繰り返し操作することによっても結果が再現され、40%以下に低下することはなかった。これにより、lespic E2Pの収率を、50%から、さらに増加させることが可能である。酸化酵素活性の上昇に関しては、S. marmorata 以外の水生昆虫も同様であり、その測定法は従来公知の方法であればよい。本実施例のような濃度の尿素や臭化リチウムを添加した場合に得られたlespic E2Pは規格化リン酸化率36 ± 3 %を有していた。なお、上記したように尿素や臭化リチウムを添加しなければ、キレート剤が効果的な量共存していても、E2P収率は、10%以下まで低下し、酷いときには、実験室飼育で得られたシルク繊維ネットから、目的とするE2Pの収率は全くと言っていいほど回収できないこともある。
SDS-ポリアクリルアミド電気泳動(SDS-PAGE)法によりlespic E2Pのタンパク質成分を分析した。SDS-PAGEは公知の手法で分析できるが、アクリルアミドの最適濃度は14-16%であり、特にSmsp-4のバンドが明瞭に観察できる最適条件を試行錯誤で探索したところ解明されたものである。電気泳動後のゲルを公知の手法たるクマシーブリリアントブルーを用いて染色を行った結果、lesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)の既知分子量に相当するタンパク質のバンド2本のみが観察された(図1)。これらのバンドのそれぞれに対応するタンパク質種、すなわち、本発明に関わる有効成分であるlesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)の更なる同定のため、質量分析を行い、その結果を実施例6に示す。
実施例5記載の2本のバンドを従来公知の方法による質量分析を行った。すなわち、ゲル内で酵素トリプシンを用いて消化する方法を採用した。lesp-400kDa(Smsp-1)バンド、およびlesp-14kDa(Smsp-4)バンドのゲル小片から、それぞれ、lesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)のトリプシン消化断片を回収した。それぞれのタンパク質断片を試料とし、既知の手法たるMatrix Assisted Laser Desorption Ionization/Time of Flight(MALDI-TOF)型の質量スペクトルを得た。イオン化のためのマトリックスとして公知のα−シアノ桂皮酸を用いた。この分析結果と、本発明者らが次世代シーケンシング手法を用いた幼虫絹糸腺トランスクリプトーム網羅的解析結果により構築した再構成済コンティグデータベースに収録されているタンパク質断片アミノ酸配列データを互いに照合した結果、実施例3における高分子量側のバンドがlesp-400kDa(Smsp-1)であり、低分子量側のバンドがlesp-14kDa(Smsp-4)であると同定した。lesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)のアミノ酸組成は以下の表2(朝倉哲朗編, Biotechnology of Silk;p. 115, Table 6.1, 2013参照)記載の組成と一致した。かくして、本発明で得られたlesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)のアミノ酸組成は、表2の通りであった。しかし、この表2記載の抽出方法と本発明の製造方法とは異なるが、得られるタンパク質は同じである。ただし、この文献記載の抽出方法で得られる抽出物(=タンパク質組成物)を溶液にしようとすると、組織毒性のある変性剤を使用しなければならず、結果として「液体塞栓材料」として利用することはできない。
lespic E2P(規格化リン酸化率36 ± 3 %)15mgを以下の緩衝液に溶解し、これを試料とするゲル浸透クロマトグラフィー(GPC)法によりlesp-400kDa(Smsp-1)とlesp-14kDa(Smsp-4)とをそれぞれ単離・回収した。ここでいう「緩衝液」の組成とは、50mM リン酸カリウム(pH 8.0)、0.15M 塩化カリウム、9.0M 尿素、5mM EDTA、0.5mM DTTであり、リン酸イオンとEDTAがカルシウムイオンとpSerとの相互作用を、塩化カリウムが静電相互作用を、また、尿素が水素結合相互作用を抑制するという、タンパク質分子間に働く相互作用全てを消去できる諸成分を含むため、かくして、lesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)の分離が可能となる。GPC法は既知の手法であるが、lesp-400kDa(Smsp-1)とlesp-14kDa(Smsp-4)との良好な分離のため、本発明者らは、カラムとしてShodex KW803(内径8mm、カラム長30cm)、操作流速0.5mL/min、さらに、カラムオーブン温度40℃を使用した。この条件が最適条件であった。上記では、lesp-400kDa(Smsp-1)とlesp-14kDa(Smsp-4)とをそれぞれ単離するために、あえてリン酸緩衝液を移動層として用いたが、lesp-400kDa(Smsp-1)、lesp-14kDa(Smsp-4)を血液凝固剤として用いるときにはリン酸緩衝液は使用できない。
実施例7に記載したGPC法における操作条件を用いてlesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)を分離し、それぞれを固体として回収した。その後、lesp-14kDa(Smsp-4)固体を、溶離用緩衝液1(溶離用緩衝液1の組成:20mM Tris HCl(pH 8.0)、9.0M 尿素、2mM 塩化カルシウム二水和物、および、0.5mM DTTを含む。)に溶解し、溶離用緩衝液1を用いて平衡化したカラムに注入した。この時の操作流速およびカラムオーブン温度は実施例7と同様である。この溶離用緩衝液2mMは塩化カルシウム二水和物を含むため、凝集したlesp-14kDa(Smsp-4)の保持時間15-16 minとなった。SDS-PAGEによる分析の結果、保持時15-16 minの画分には、14 kDaのポリペプチドのみが存在することが判明した。かくして、カルシウムイオンに応答して凝集するタンパク質として Smsp-4が精製された。この結果は、Smsp-4がpSerを含むことを強く示唆するものであるが、pSerを対象とした分析ではなく、従って直接的な証拠にならない。このことに加え、精製されたlesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)に含まれるセリン(Ser)残基のうち、どれくらいの割合がpSerに変化すると「血液塞栓」効果が発揮されるのかという検証が必要である。この目的のため、ホスホアミノ酸分析により、lesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)におけるpSer 含量を実証した。
実施例8において回収したlesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)を試料とし、文献(Long-range periodic sequence of the cement/silk protein of Stenopsyche marmorata: purification and biochemical characterization, Kousaku Ohkawa, Yumi Miura, Takaomi Nomura, Ryoichi Arai, Koji Abe, Masuhiro Tsukada, Kimio Hirabayashi, Volume 29, Issue 4, 2013, pages 357-367)に記載の手法に従い、減圧溶封したガラスアンプル内で5.7 M HCl中、105℃、2時間加水分解処理を行い、その後、ガラスアンプルを開管し減圧乾固した。上記文献記載の分析装置・分析操作を行い、lesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)の試料重量あたりに含まれるpSerのモル百分率を求めた。なお、上記文献には、Stenopsyche marmorata幼虫絹糸腺から精製されたlesp-400kDa(Smsp-1)がpSerを含むことを示すためのホスホアミノ酸分析手法が記載されているが、定性分析結果のみが示されているのみであり、本発明における「血液塞栓」効果を立証するための、pSer定量結果は一切記載されていない。
実施例8において回収したlesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)を試料とし、上記文献記載の手法に従い、減圧溶封したガラスアンプル内で5.7 M HCl中、105℃、24時間加水分解処理を行い、その後、ガラスアンプルを開管し減圧乾固した。ホスホアミノ酸分析では加水分解処理時間を2時間とした。通常アミノ酸分析では公知常用されるのは24時間であるが、上記処理によりpSer全量がSer残基へと変化した。なお、通常アミノ酸分析自体は公知の手法であり、タンパク質試料のアミノ酸組成 (アミノ酸配列ではないことに留意) を求める実験に常用される。lesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)加水分解物を試料とし、各4回以上通常アミノ酸分析操作を行った。その結果、Smsp-1のSer残基含量は205 ± 15 nano mol/mgとなった。この値は、1 mgのlesp-400kDa(Smsp-1)が平均205 ± 15 nano molのSer残基を含むということを意味する。lesp-14kDa(Smsp-4)ではSer残基含量は22 ± 0.1 nano mol/mgであった。
通常アミノ酸分析から求まるSer残基含量を分母、かつ、ホスホアミノ酸分析から求まるpSer含量を分子とする百分率が規格化リン酸化率となる。lesp-400kDa(Smsp-1)の場合は、100 x ((75 ± 1 nano mol/mg)/(205 ± 15 nano mol/mg)=37 ± 3 %であり、同様な計算をlesp-14kDa(Smsp-4)について行うと、100 x (6.1 ± 0.4 nano mol/m)/22 ± 0.1 nano mol/mg)=28 ± 2となる。これらの値は、本発明において製造したlespic E2Pが「血液塞栓性」を発揮するのに必要な特徴を示すものである。本発明において製造したlespic E2Pは、後述の実施例に示すように、動物実験において血液塞栓性をもつことが実証されているため、これ以降、血液塞栓性E2Pと表記する。
本発明記載の製造方法を経て得られる血液塞栓性E2P(規格化リン酸化率36 ± 3 %) を公知の緩衝液系に濃度5 mg/mLで溶解した。ここに、市販の脱リン酸化酵素(フォスファターゼなど)を数マイクログラム添加した。なお、上記の「公知の緩衝液系」とは、無機リン酸塩成分を含まず、かつ、脱リン酸化酵素を失活する成分を含まないことが条件である。脱リン酸化酵素は、血液塞栓性E2Pが含むpSerを脱リン酸し、Ser残基に変える働きをする。かような処理により、血液塞栓性E2Pの規格化リン酸化率は低下する。脱リン酸化反応条件を例示すると、血液塞栓性E2P濃度5 mg/mL、公知の緩衝液系として10 mMトリス緩衝液(pH 8.0)、脱リン酸化酵素として市販プロテインフォスファターゼ濃度10 マイクログラム/mL として、37℃で8時間インキュベートする。その後、上記の酵素反応混合物に塩化カルシウム二水和物水溶液を添加し[Ca2+] = 2.0 mM としても、E2Pの凝集・沈殿形成は直ちには起こらなかった。これは、脱リン酸化処理により、血液塞栓性E2Pの効能が低下し、マイクロカテーテル術式のための血液塞栓材料たる条件のひとつ「血流への注入時に瞬時に凝集・沈殿形成する」を満たさなくなったことを意味している。
本実証実験、すなわち、GPC法による実証においては、公知の緩衝液を使用する代わりに、最適な溶離用緩衝液として、本発明に関わる新規組成を持つ溶離用緩衝液1を見出した。溶離用緩衝液1の組成は、20mM Tris HCl(pH 8.0)、9.0M 尿素、2mM 塩化カルシウム二水和物、および、0.5mM DTTを含む。この溶離用緩衝液1を用いて検証することにした。溶離用緩衝液1に含まれる2mM 塩化カルシウム二水和物はpSerを所定mol%含むlesp-400kDa(Smsp-1)とlesp-14kDa(Smsp-4)とを凝集させる。ただし、凝集沈殿をすると、溶液状態が必須条件であるGPCでの分析が実施不能となるため、lesp-400kDa(Smsp-1)、lesp-14kDa(Smsp-4)凝集体の沈殿を抑制するため、緩衝液系に9.0M 尿素を添加した。以上が、当該緩衝液成分の効用である。この溶離用緩衝液1にlespic E2P 15mgを溶解し、[0066]記載の好適条件においてGPC操作を実施した。SDS-PAGE法により保持時間毎のタンパク質組成を調べた結果、lesp-14kDa(Smsp-4)の保持時間は15-16minとなった。2mM 塩化カルシウム二水和物非存在下での実施例7では、lesp-14kDa(Smsp-4)の保持時間は18-20minであったので、以上の結果は、lesp-14kDa(Smsp-4)の分子サイズが2mM 塩化カルシウム二水和物存在下で上昇したことを意味する。すなわち、カルシウムイオンを介したlesp-400kDa(Smsp-1)とlesp-14kDa(Smsp-4)との凝集が誘起されたことが本実施例13において実証された。本実施例で使用したlespic E2Pは、上記したようにして製造した規格化リン酸化率36 ± 3%を有するものであった。なおpSer を規格化リン酸化率で 8.1 ± 0.7 mol% 含む実施例12の「脱リン酸化処理済みE2P」について、実施例13に記載の操作を実施し、カルシウムイオンを介したlesp-400kDa(Smsp-1)とlesp-14kDa(Smsp-4)との凝集が誘起されたかを検討した。その結果、脱リン酸化処理済みE2Pでは、凝集が誘起されておらず、Smsp-4の保持時間は18-20minであった。この結果は、規格化リン酸化率を低下させる処理を施した「脱リン酸化処理済みE2P」では、pSer含量の絶対値が足らないために、カルシウムイオン存在化でも凝集することができないことを示している。
lespic E2P(規格化リン酸化率 36 ± 3%)2mgを、例えば、150mM NaClおよび20mM Tris HCl(pH 8.0)を含む緩衝液0.5mLに溶解した。この例の緩衝液に含まれる、いわゆる「緩衝能」を提供する成分、および中性無機塩は、カルシウムイオンとpSerとの間の相互作用を阻害し、故に、カルシウムイオン添加時のlesp-400kDa(Smsp-1)とlesp-14kDa(Smsp-4)との迅速な凝集・沈殿、すなわち、本発明により提供される機能を妨げる物質およびその濃度を除き、上記の例に限定されるものではない。上記例のような緩衝液は、一般に、タンパク質の溶解にしばしば用いられ、新規性は無いが、このようなごく普通の緩衝液系に可溶なlespic E2Pの性質は、[0043]記載の要求事項たる(2)溶媒が体液と等張な緩衝液であること、を満しており、lespic E2Pの物質新規性を示す証拠となる。なぜなら、S. marmorata 5齢幼虫の体内絹糸腺を原料とし、既に特許査定された当該発明者らの特許文献1記載の手法を経て製造されるタンパク質画分は、9M程度の尿素が共存する緩衝液系にしか溶解しないからである。かような高濃度尿素溶液に、仮に、脳内血管塞栓の有効成分を含んでいたとしても、カテーテル先端からの血管内注入後、有効成分の凝集固化を尿素が阻害するだけでなく、尿素の有する極めて強力なタンパク質変性作用により、注入部位周囲の血管内壁細胞の機能不全、あるいは、著しい程度の血球破壊を誘発し、結果として、患者を重篤な状態にする危険性があるからである。従って、特許文献1により製造されるタンパク質画分は、本発明において主張する血液凝固機能を提供しない。
本発明に関わるlespic E2P製造方法において、[0065]に記載したシルク繊維ネットの粉末、[0066]に記載した第1段階抽出後に溶け残った残渣(E1P残渣という)、および[0067]に記載した第2段階抽出後に溶け残った残渣(E2P残渣という)を試料として用いた。これらを公知の走査型電子顕微鏡(Scanning Electron Microscope, SEM)用試料台にカーボンテープを用いて貼付け、装置に装填し、次に、公知の手法によりエネルギー分散X線元素分析(Energy Dispersion X-ray spectral elemental analysis, EDX)を実施した。リン元素(P、実施例17において後述)およびカルシウム元素(Ca)について上記3種の試料の元素マッピングを行った結果、シルク繊維ネット粉末およびE1P残渣におけるCaシグナルのEDX画像は、対応するSEMの画像と一致した。しかし、E2P残渣では、Caシグナルが検出されず、EDX画像が得られなかった。以上の結果は、EDTAを用いる第2段階の抽出により、E1P残渣に含まれるカルシウムイオンが除去されることで、lesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)が可溶化されてlespic E2Pが製造されることの証拠である。E2P残渣については、酸化酵素によりタンパク質分子間に化学結合が形成されており、緩衝液に溶解しないため、血液凝固剤としては使用できない。
[0058]記載の方法により、lespic E2PのSDS-PAGEを行った後、クマシーブリリアントブルーによるタンパク質バンドの染色を行わずに、ゲル内に展開されたタンパク質、すなわち、lesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)を、一般にエレクトロブロットと呼ばれる公知の手法により、市販のポリフッ化ビニリデン膜に転写した。その後、市販の抗pSer抗体を用いて、一般に免疫染色(またはウェスタンブロットという)と呼ばれる公知の手法により、抗pSer抗体と結合するタンパク質を調べた。免疫染色の操作は市販の染色キットを用いて行った。その結果、lesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)はともに、抗pSer抗体と結合することが確かめられた。かくして、lesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)がともに、抗pSer抗体のエピトープ(一般に、抗原認識部位ともいう)たるpSerを含むことが実証された。かくして製造されたlespic E2P(lesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)は、規格化リン酸化率36 ± 3%を有するものであった。
lespic E2P(規格化リン酸化率 36 ± 3%)試料3mgを公知の手法、すなわち、真空溶封したガラス容器内、5.7M HCl水溶液を用いて、105℃において2時間加水分解した。試料を減圧乾固してHClを除き、その後、公知の手法、すなわち、市販のイオン交換カラムを用いるホスホアミノ酸分析を行った(例えば、Kousaku Ohkawa, Yumi Miur, Takaomi Nomura, Ryoichi Arai, Koji Abe, Masuhiro Tsukada & Kimio Hirabayashi, Long-range periodic sequence of the cement/silk protein of Stenopsyche marmorata: purification and biochemical characterization, Biofouling 29(4), 357-367 (2013)参照)。標準試料として、pSer、ホスホスレオニン(pThr)、およびホスホチロシン(pTyr)の当量混合物を用いて分析を行い、pSer、pThr、およびpTyrのクロマトグラム上の保持時間と、lespic E2Pの加水分解産物に含まれるホスホアミノ酸の保持時間を比較した。その結果、lespic E2Pの加水分解産物には、pSerのみが含まれていることが確かめられた。さらに、lespic E2P試料重量あたりのリン元素重量の割合を、ホスホアミノ酸分析において検出されたpSerのモル数から計算し、実施例16、すなわち、[0109]記載のEDXによるリン元素(P)の重量比と比較した結果、±0.05wt%の精度において一致した。かくして、lesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)がともに、pSerのみを含み、その他のホスホアミノ酸、すなわち、pThrおよびpTyrは含まないことが実証された。
市販の病理検査用ウマ脱フィブリン血液を供試し、血液凝固機能を検討した。ウマ脱フィブリン血液とは、採血したウマ血液をガラス棒等で穏やかに撹拌し、血液の異物反応たるフィブリンタンパク質の重合を刺激することで、これらを取り除いて凝固できなくする処理を加えた血液標品のことを指す。一般に、ヒト、ウマ、および、ブタ等の恒温動物の血中カルシウムイオン濃度は2.5-4.0mMの間で変化する。ウマ脱フィブリン血液の場合、これを製造する過程で起こる血液の異物反応に伴い、血中の血小板由来のカルシウムイオンは、トロンビン等のカルシウム結合タンパク質の作用により、脱フィブリン処理を施さない血液よりも若干低濃度となっている。
[0104]記載のlespic E2P(規格化リン酸化率 36 ± 3%) 3mgを含む緩衝溶液0.5mLを用いた。従来技術により全身麻酔した動物実験用ブタ(体重30kg;三元ブタ)大腿部から腰骨付近の腹部皮下2.0-2.5cmに、間隔をあけて計3箇所にlespic E2P 3mgを含む緩衝溶液0.5mLを注入した(これら3箇所を実験区という)。この動物実験用ブタを2週間飼育した。その後、実験区の皮下組織摘出前に、表皮注入部外観を目視で観察した結果、実験区と周辺組織の間には有意な差は見られず、治癒過程における異常は観察されなかった。この結果は、lespic E2Pが生体に対する組織反応性が極めて低いことを意味している。
血管撮影装置(シーメンス製Artis Zee Floor型)を用い、人間に施行する方法と同様の脳血管撮影法をブタ(上記と同様の三元ブタ)に採用した。なお、この装置は、2014年現在、脳血管撮影装置としては、ほぼ最新型、かつ、最も優れた解像度を有している。上記の血管撮影装置を装備した手術台に、[0114]記載と同一の全身麻酔済動物実験用ブタを固定した後、右大腿動脈よりカテーテル(Boston Scientific社製マイクロカテーテルExcelsiorTM 1018TM、150cm×6cm)を進め、左の浅頚動脈(血管直径約2-3mm)内にマイクロカテーテルを留置した。本発明による新規物質のlespic E2P 3mgを含む[0104]記載の緩衝溶液0.5mLをマイクロカテーテル先端から注入し、直後にヨード系造影剤を用いて再度血管撮影し、lespic E2P緩衝溶液の血管閉塞性能を調べた。
(1)ヒゲナガカワトビケラの5齢幼虫のシルク繊維ネットからのタンパク質抽出液からSmsp-2とSmsp-3とが除去され、さらにカルシウムイオンが除去されてなる、規格化リン酸化率が、36 ± 3 %のlesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)からなる組成物であり、lesp-400kDa(Smsp-1):lesp-14kDa(Smsp-4)の重量比が9:1で存在する組成物からなる液体の血液塞栓性タンパク質固有組成物E2Pであって、血液凝固を促進加速させる液体の血液塞栓性タンパク質固有組成物E2Pが有効成分として含まれ、この液体の血液塞栓性タンパク質固有組成物E2Pと、リン酸イオンを含まない、体液と等張な緩衝液との溶液からなる血液凝固剤溶液を、カテーテルを通して標的血管内に注入し、標的部位の血流(患部血流)を塞栓することを特徴とする標的部位の血流の塞栓方法。
(2)標的血管内の標的部位の血流(患部血流)の塞栓に用いるための物質であって、その物質が、ヒゲナガカワトビケラの5齢幼虫のシルク繊維ネットからのタンパク質抽出液からSmsp-2とSmsp-3とが除去され、さらにカルシウムイオンが除去されてなる、規格化リン酸化率が、36 ± 3 %のlesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)からなる組成物であり、lesp-400kDa(Smsp-1):lesp-14kDa(Smsp-4)の重量比が9:1で存在する組成物からなる液体の血液塞栓性タンパク質固有組成物E2Pであって、血液凝固を促進加速させる液体の血液塞栓性タンパク質固有組成物E2Pが有効成分として含まれ、この液体の血液塞栓性タンパク質固有組成物E2Pを、リン酸イオンを含まない、体液と等張な緩衝液に溶解してなる溶液からなる血液凝固剤溶液であることを特徴とする物質。
(3)標的血管内の標的部位の血流(患部血流)の塞栓に使用するための薬剤の製造のための物質であって、その物質が、ヒゲナガカワトビケラの5齢幼虫のシルク繊維ネットからのタンパク質抽出液からSmsp-2とSmsp-3とが除去され、さらにカルシウムイオンが除去されてなる、規格化リン酸化率が、36 ± 3 %のlesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)からなる組成物であり、lesp-400kDa(Smsp-1):lesp-14kDa(Smsp-4)の重量比が9:1で存在する組成物からなる液体の血液塞栓性タンパク質固有組成物E2Pであって、血液凝固を促進加速させる液体の血液塞栓性タンパク質固有組成物E2Pが有効成分として含まれ、この液体の血液塞栓性タンパク質固有組成物E2Pと、リン酸イオンを含まない、体液と等張な緩衝液との溶液からなる血液凝固剤溶液である薬剤の製造のため物質の使用。
Claims (7)
- ヒゲナガカワトビケラの5齢幼虫のシルク繊維ネットのタンパク質由来のSmsp-1およびSmsp-4を有し、36 ± 3 % のlesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)の規格化リン酸化率を有する組成物であって、lesp-400kDa(Smsp-1):lesp-14kDa(Smsp-4)の重量比が9:1で存在する組成物である液体の血液塞栓性タンパク質固有組成物E2Pであり、血液凝固を促進加速させる液体の血液塞栓性タンパク質固有組成物E2Pを有効成分として含んでなり、この液体の血液塞栓性タンパク質固有組成物E2Pと、リン酸イオンを含まない、体液と等張な緩衝液との溶液からなることを特徴とする血液凝固剤溶液。
- ヒゲナガカワトビケラの5齢幼虫のシルク繊維ネットのタンパク質由来のSmsp-1およびSmsp-4を有し、36 ± 3 % のlesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)の規格化リン酸化率を有する組成物であり、lesp-400kDa(Smsp-1):lesp-14kDa(Smsp-4)の重量比が9:1で存在する組成物である液体の血液塞栓性タンパク質固有組成物E2Pであって、血液凝固を促進加速させる液体の血液塞栓性タンパク質固有組成物E2Pを得、この有効成分としての前記液体の血液塞栓性タンパク質固有組成物E2Pと、リン酸イオンを含まない、体液と等張な緩衝液との溶液からなる血液凝固剤溶液を得ることを特徴とする血液凝固剤溶液の製造方法。
- ヒゲナガカワトビケラの5齢幼虫のシルク繊維ネットを糸状粉末とし、第1段階として、この粉末をpH 6を超えpH 9未満の抽出用緩衝液であって、尿素または臭化リチウムを含む抽出用緩衝液に浸してインキュベートし、遠心上清を採取し、抽出残渣を得、次いで、第2段階で、第1段階で得られた抽出残渣を、カルシウムイオンと結合するキレート剤を添加した第1段階抽出用緩衝液に浸してインキュベートし、得られた抽出液からキレート剤と結合したカルシウムイオンを除いた後、36 ± 3 % のlesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)の規格化リン酸化率を有するSmsp-1およびSmsp-4であって、lesp-400kDa(Smsp-1):lesp-14kDa(Smsp-4)の重量比が9:1で存在するSmsp-1およびSmsp-4を有する液体の血液塞栓性タンパク質固有組成物E2Pを得、血液凝固を促進加速させる前記液体の血液塞栓性タンパク質固有組成物E2Pを有効成分とし、この液体の血液塞栓性タンパク質と、リン酸イオンを含まない、体液と等張な緩衝液とからなる血液凝固剤溶液を製造することを特徴とする血液凝固剤溶液の製造方法。
- 前記第1段階および第2段階におけるインキュベート温度が40-60℃、時間が30-60分であることを特徴とする請求項3に記載の血液凝固剤溶液の製造方法。
- ヒゲナガカワトビケラの5齢幼虫のシルク繊維ネットを糸状粉末とし、第1段階として、この粉末をpH 6を超えpH 9未満の抽出用緩衝液であって、尿素または臭化リチウムを含む抽出用緩衝液に浸してインキュベートし、遠心上清を採取し、抽出残渣を得、次いで、第2段階で、第1段階で得られた抽出残渣を、カルシウムイオンと結合するキレート剤を添加した第1段階抽出用緩衝液に浸してインキュベートし、得られた抽出液からキレート剤と結合したカルシウムイオンを除いた後、規格化リン酸化率が36 ± 3 % のlesp-400kDa(Smsp-1)およびlesp-14kDa(Smsp-4)であるSmsp-1およびSmsp-4であって、lesp-400kDa(Smsp-1):lesp-14kDa(Smsp-4)の重量比が9:1で存在するSmsp-1およびSmsp-4を有する液体の血液塞栓性タンパク質固有組成物E2Pを製造することを特徴とする液体の血液塞栓性タンパク質固有組成物E2Pの製造方法。
- 前記第1段階および第2段階におけるインキュベート温度が40-60℃、時間が30-60分であることを特徴とする請求項5に記載の液体の血液塞栓性タンパク質固有組成物E2Pの製造方法。
- 前記第1段階における抽出用緩衝液が、尿素または臭化リチウムを含むTris HCl、SDS、およびDTTからなる緩衝液であり、前記第2段階における抽出用緩衝液が、この第1段階抽出用緩衝液にカルシウムイオンと結合するキレート剤を添加した緩衝液であることを特徴とする請求項5または6記載の液体の血液塞栓性タンパク質固有組成物E2Pの製造方法。
