本明細書において使用する場合、用語「微小区画」とは、脂質層、および、その内部の水層から構成される閉じられた微小な空間をいう。「微小区画」としては、例えば、リポソームが挙げられるがこれに限定されない。
本発明は、あらゆる膜タンパク質に適用可能である。代表的な膜タンパク質としては、例えば、トランスポーター、レセプターなどが挙げられるがこれらに限定されない。必要に応じて、本発明の膜タンパク質をコードする配列は、タンパク質を膜に挿入するためのリーダー配列を含んでもよい。
本発明の膜タンパク質は、トランスポーターであっても、そうでなくてもよい。本発明のトランスポーターとしては、細胞において物質輸送に関るタンパク質(例えば、EmrEタンパク質)、脂質二重膜を透過しない物質を透過させるタンパク質(例えば、ヘモリシン)が挙げられるがこれらに限定されない。
本発明において使用する一枚膜リポソームは、実施例に記載する遠心沈降法を用いて調製することができるが、調製法は、これに限定されない。例えば、遠心沈降法の他にも、静置水和法(P.Mueller and T.F.Chien,Biophys.J.,1983,44,375−381)、および、エレクトロフォーメーション法(Miglena I.Angelove and Dimiter S.Dimitrov,Faraday Discuss.Chem.Soc.,1986,81,303−311)を利用することが可能である。
一枚膜リポソームの製造において使用される脂質の成分/組成は、特に限定されることはないが、リン脂質およびコレステロールを含むことが好ましい。脂質としては、例えば、L−アルファ−ホスファチジルコリン、コレステロール、L−アルファ−ジラウロイルホスファチジルコリン、L−アルファ−ジラウロイルホスファチジルエタノールアミン、L−アルファ−ジラウロイルホスファチジルグリセロールナトリウム、L−アルファ−モノミリストイルホスファチジコリン、L−アルファ−ジミリストイルホスファチジルコリン、L−アルファ−ジミリストイルホスファチジルエタノールアミン、L−アルファ−ジミリストイルホスファチジルグリセロールアンモニウム、L−アルファ−ジミリストイルホスファチジルグリセロールナトリウム、L−アルファ−ジミリストイルホスファチジン酸ナトリウム、L−アルファ−ジオレイルホスファチジルコリン、L−アルファ−ジオレオイルホスファチジルエタノールアミン、L−アルファ−ジオレオイルホスファチジルセリンナトリウム、L−アルファ−モノパルミトイルホスファチジルコリン、L−アルファ−ジパルミトイルホスファチジルコリン、L−アルファ−ジパルミトイルホスファチジルエタノールアミン、L−アルファ−ジパルミトイルホスファチジルグリセロールアンモニウム、L−アルファ−ジパルミトイルホスファチジルグリセロールナトリウム、L−アルファ−ジパルミトイルホスファチジン酸ナトリウム、L−アルファ−ステアロイルホスファチジルコリン、L−アルファ−ジステアロイルホスファチジルコリン、L−アルファ−ジステアロイルホスファチジルエタノールアミン、L−アルファ−ジステアロイルホスファチジルグリセロールナトリウム、L−アルファ−ジステアロイルホスファチジルグリセロールアンモニウム、L−アルファ−ジステアロイルホスファチジン酸ナトリウム、L−アルファ−ジエルコイルホスファチジルコリン、1−パルミトイル−2−オレオイルホスファチジルコリン、ベータ−オレイル−ガンマ−パルミトイル−L−アルファ−ホスファチジルエタノールアミン、ベータ−オレイル−ガンマ−パルミトイル−L−アルファ−ホスファチジルグリセロールナトリウム、スフィンゴミエリン、ステアリルアミンが挙げられるがこれらに限定されない。
マグネシウムの濃度は、好ましくは15mM〜50mM、より好ましくは18.88mM〜42.48mM、さらにより好ましくは28.32mM〜37.76mM、最も好ましくは33.04mMである。
本発明において使用する核酸分解酵素としては、例えば、RNA分解酵素、および、DNA分解酵素が挙げられるがこれらに限定されない。使用する核酸分解酵素の供給源は、特に限定されることはない。核酸分解酵素としてDNaseを使用する場合、使用する酵素活性は、100μLのリポソーム溶液あたり、1U〜20U、より好ましくは、5U〜15U、もっとも好ましくは、約12.5Uである。核酸分解酵素としてRNaseを使用する場合、使用する酵素活性は、100μLのリポソーム溶液あたり、1μg〜20μg、より好ましくは、5μg〜15μg、もっとも好ましくは、約10μgである。当業者は、使用する酵素量を容易に決定することができる。
例えば、リポソームに含ませる遺伝情報がDNAである場合、そのDNAに、タンパク質のコード配列、ならびに、このコード配列に作動可能に連結された翻訳調節配列、および、このコード配列に作動可能に連結された転写翻訳調節配列を含ませる。
本発明のリポソームを分子進化工学に利用することができる。
以下に記載の遠心沈降法により、一枚膜リポソームを調製した。
・10mgの脂質(ホスファチジルコリン:コレステロール=9:1)を100μlクロロホルムに溶解し、2mlの流動パラフィンと混合した。
・80℃で30分インキュベーションした。
・リポソーム外液(333mM グルコース、無細胞蛋白質合成系から翻訳タンパク質群とtRNAを除いたもの)、内液(330mM スクロース、1μM Transferrin Alexa 647、無細胞蛋白質合成系、40U/μl RNaseインヒビター(Promega)、0.4μM リボソームS1サブユニット、50pM DNAを調製した。DNAとして、EmrE−myc−his配列(配列番号1:EmrE遺伝子のC末端にmycタグおよびhisタグを含む配列)を含むDNA、または、GUS配列(配列番号3:GUS配列およびmyc配列を含むネガティブコントロール)を含むDNAを用いた。この条件は、各リポソームに1分子のDNAが封入される条件である。使用した無細胞蛋白質合成系の組成は、以下のとおりである:
各0.3mM アミノ酸(アラニン、グリシン、ロイシン、イソロイシン、バリン、セリン、スレオニン、プロリン、トリプトファン、フェニルアラニン、グルタミン、グルタミン酸、アスパラギン、アスパラギン酸、リシン、アルギニン、ヒスチジン、メチオニン、システイン、チロシン)、3.6μg/μl tRNA、2mM ATP、2mM GTP、1mM CTP、1mM UTP、14mM 酢酸マグネシウム、50mM Hepes−KOH (pH7.8)、100mM グルタミン酸カリウム、2mM スペルミジン、20mM クレアチンリン酸、2mM ジチオスレイトール、10ng/μl 10−ホルミル−5.6.7.8.−テトラヒドロ葉酸、翻訳タンパク質群(2500nM IF1、411nM IF2、728nM IF3、247nM RF1、484nM RF2、168nM RF3、485nM RRF、727nM AlaRS、99nM ArgRS、420nM AsnRS、121nM AspRS、100nM CysRS、101nM GlnRS、232nM GluRS、86nM GlyRS、85nM HisRS、365nM IleRS、99nM LeuRS、115nM LysRS、109nM MetRS、134nM PheRS、166nM ProRS、99nM SerRS、84nM ThrRS、102nM TrpRS、101nM TyrRS、100nM ValRS、588nM MTF、926nM MK、465nM CK、1307nM NDK、621nM Ppiase2、1290nM EF−G、2315nM EF−Tu、3300nM EF−Ts、529nM Tig、22nM HrpA、1440nM TrxC)。
・脂質を溶解した流動パラフィン400μlにリポソーム内液20μlを入れ、氷上に1分置いた。
・ボルテックスミキサーの最大強度で40秒攪拌しエマルションを調整後、氷上で10分置いた。
・新しいチューブにリポソーム外液150μlをいれ、その上に調製したエマルションを積層し、氷上で10分置いた。
・14k×g、4℃で30分遠心した。
・チューブの底に穴を開け、底に溜まったリポソーム懸濁液80μlを回収した。
・リポソーム懸濁液に2μlの5U/μl DNase、または、4mg/ml RNaseを添加した。
・リポソーム懸濁液を37℃で3時間インキュベーションし、タンパク質合成を行った。
・PBS+1% BSA溶液で抗体(Alexa Fluor 488で標識した、抗Mycタグ抗体(マウスIgG1))を希釈し終濃度5μg/mlになるようにリポソーム懸濁液に加えた。(リポソーム溶液9μlに50g/mlの抗体を1μl加えた。)
・室温30分放置後、抗体を顕微鏡観察で観察した(Ex:470〜490 Em:510〜550)。
リポソームに、EmrE−myc−his配列(配列番号1:EmrE遺伝子のC末端にmycタグおよびhisタグを含む配列)を含むDNA、または、GUS配列(配列番号3:GUS配列およびmyc配列を含むネガティブコントロール)を含むDNAそれぞれ5nM、および、PURE systemを内封し、37℃2時間インキュベートしEmrE−myc−hisとGUS−mycを発現させた。リポソーム調製後、外液1から、EtBr 5μg/mlを含む外液2に交換した。一分間ごとに蛍光を測定し、EtBrの取り込みを観測した。その後、同じサンプルを蛍光顕微鏡で観察した(Ex:520〜550 Em:580〜)。
リポソームに、ヘモリシン配列を含むDNA5nM、ハロタグタンパク質および、PURE systemを内封した。この際、リポソーム内液及び外液のMg濃度を18.88、23.6、28.32、33.04、37.76、42.28、47.2、51.92、56.64mMの9条件でリポソーム調整を行った。リポソーム調製後、37℃、16時間インキュベートし、ヘモリシンを発現させた。発現したアルファヘモリシンの機能を測定するためにリポソーム外液にハロタグAlexa Fluor 488リガンド1μMを加え、3時間後にリポソーム内部に蓄積されたハロタグAlexa Fluor 488リガンドの蛍光量を測定した。その結果、33.04mMのMg濃度値で調製したリポソームで最も多くのハロタグAlexa Fluor 488リガンドの蓄積がみられた。したがって、ヘモリシンの活性検出には、好ましくは18.88mM〜23.6mM、より好ましくは23.6mM〜28.32mM、最も好ましくは28.32〜42.48mMの条件を用いる事が良い、と判明した。
実施例1で用いたEmrE−myc−his配列のかわりにヘモリシンをコードする配列(配列番号5)をトランスポーターを発現するために用いた。さらに、ヘモリシンが輸送するリガンドであるハロタグAlexa Fluor 488リガンドに結合する因子として、ハロタグタンパク質(配列番号7)を用いた。ヘモリシンは、膜にポアを作る膜タンパク質であり、3kDaよりも小さい物質を透過させる。そのため、ヘモリシンが発現すると、リポソームにポアが生じ、その結果、脂質膜を透過できないハロタグAlexa Fluor 488リガンドを透過させる。ポアを介して透過したハロタグAlexa Fluor 488リガンドは、ハロタグタンパク質と結合し、その結果、リポソーム内に移動したハロタグAlexa Fluor 488リガンドはリポソーム内に蓄積する。
(実施例5:脂質成分/組成の検討−2)
次に、実施例4と同様の手法で、種々の脂質を用いてリポソームを合成し、発現した膜タンパク質の活性を比較した。結果を図4に示す。
野生型ヘモリシン(配列番号5)と致死変異型ヘモリシン(配列番号8)を用い、実施例4と同様の手法を用いて、実験を行った。野生型と致死変異型との比率は、1:12として、10倍以上多くの致死変異型を用いた。37℃で160分間培養し、膜タンパク質を発現させ、その後、セルソーターで輸送活性を示したリポソームを選択し、リポソームに含まれる野生型遺伝子と変異型遺伝子との割合を決定したところ、野生型:変異型=8:1であった。この結果は、本発明のスクリーニング/選択によって100倍の濃縮がされたことを実証するものである。
以下の手順を用いて、進化実験を行った。
脂質組成はPOPC:Chol=1:1 (wt/wt)を用いた。内液組成には、実施例1に記載の無細胞蛋白質合成系と同様の組成(ただし、酢酸マグネシウム濃度を33.04mMに変更)を用いた。さらに、100nM T7 RNAポリメラーゼ、200mM スクロース、5mM β−グルクロニダーゼ結合体化ハロペプチド、1mM トランスフェリン結合体化alexa fluor 647、5pM DNA(T7プロモーター制御下にヘモリシンのORFを配置)を用いた。外液組成には、実施例1に記載の無細胞蛋白質合成系と同様の組成(ただし、酢酸マグネシウム濃度を33.04mMに変更)の小分子のみに加え、200mM グルコースを含む溶液を用いた。

Claims (24)
- 一枚膜リポソームであって、以下:
(a)プロモーター配列、翻訳開始配列、および、トランスポーターである膜タンパク質をコードする配列を含むDNA;
(b)RNAポリメラーゼ;
(c)リボヌクレオチド;、
(d)無細胞蛋白質合成系;ならびに、
(e)該膜タンパク質が輸送するリガンドに結合する因子、
を含む、一枚膜リポソーム。 - 請求項1に記載の一枚膜リポソームであって、核酸分解酵素によって処理された一枚膜リポソーム。
- 前記核酸分解酵素が、RNA分解酵素、および、DNA分解酵素からなる群から選択される、請求項2に記載の一枚膜リポソーム。
- 前記核酸分解酵素が、RNA分解酵素である、請求項3に記載の一枚膜リポソーム。
- 複数の一枚膜リポソームを含むライブラリーであって、該一枚膜リポソームは、以下:
(a)プロモーター配列、翻訳開始配列、および、トランスポーターである膜タンパク質をコードする配列を含むDNA;
(b)RNAポリメラーゼ;
(c)リボヌクレオチド;
(d)無細胞蛋白質合成系;ならびに、
(e)該膜タンパク質が輸送するリガンドに結合する因子、
を含む、ライブラリー。 - 請求項5に記載のライブラリーであって、前記一枚膜リポソームが核酸分解酵素によって処理された一枚膜リポソームである、ライブラリー。
- 前記核酸分解酵素が、RNA分解酵素、および、DNA分解酵素からなる群から選択される、請求項6に記載のライブラリー。
- 前記核酸分解酵素が、RNA分解酵素である、請求項7に記載のライブラリー。
- 一枚膜リポソームであって、以下:
(a)翻訳開始配列、および、トランスポーターである膜タンパク質をコードする配列を含むRNA;(d)無細胞蛋白質合成系;ならびに、(e)該膜タンパク質が輸送するリガンドに結合する因子、、
を含む、一枚膜リポソーム。 - 請求項9に記載の一枚膜リポソームであって、核酸分解酵素によって処理された一枚膜リポソーム。
- 前記核酸分解酵素が、RNA分解酵素、および、DNA分解酵素からなる群から選択される、請求項10に記載の一枚膜リポソーム。
- 前記核酸分解酵素が、RNA分解酵素である、請求項11に記載の一枚膜リポソーム。
- 複数の一枚膜リポソームを含むライブラリーであって、該一枚膜リポソームは、以下:
(a)翻訳開始配列、および、トランスポーターである膜タンパク質をコードする配列を含むRNA;(d)無細胞蛋白質合成系;ならびに、(e)該膜タンパク質が輸送するリガンドに結合する因子、
を含む、ライブラリー。 - 請求項13に記載のライブラリーであって、前記一枚膜リポソームが核酸分解酵素によって処理された一枚膜リポソームである、ライブラリー。
- 前記核酸分解酵素が、RNA分解酵素、および、DNA分解酵素からなる群から選択される、請求項14に記載のライブラリー。
- 前記核酸分解酵素が、RNA分解酵素である、請求項15に記載のライブラリー。
- 核酸分解酵素によって処理された一枚膜リポソームの製造方法であって、以下:
(1)以下:
(a)プロモーター配列、翻訳開始配列、および、トランスポーターである膜タンパク質をコードする配列を含むDNA;
(b)RNAポリメラーゼ;
(c)リボヌクレオチド;
(d)無細胞蛋白質合成系;ならびに、
(e)該膜タンパク質が輸送するリガンドに結合する因子、
を封入した一枚膜リポソームを調製する工程;ならびに、
(2)工程(1)で調製された一枚膜リポソームを、核酸分解酵素によって処理する工程、
を包含する、方法。 - 前記核酸分解酵素が、RNA分解酵素、および、DNA分解酵素からなる群から選択される、請求項17に記載の方法。
- 前記核酸分解酵素が、RNA分解酵素である、請求項18に記載の方法。
- 核酸分解酵素によって処理された一枚膜リポソームの製造方法であって、以下:
(1)以下:
(a)翻訳開始配列、および、トランスポーターである膜タンパク質をコードする配列を含むRNA;
(d)無細胞蛋白質合成系;ならびに、
(e)該膜タンパク質が輸送するリガンドに結合する因子、
を封入した一枚膜リポソームを調製する工程;ならびに、
(2)工程(1)で調製された一枚膜リポソームを、核酸分解酵素によって処理する工程、
を包含する、方法。 - 前記核酸分解酵素が、RNA分解酵素、および、DNA分解酵素からなる群から選択される、請求項20に記載の方法。
- 前記核酸分解酵素が、RNA分解酵素である、請求項21に記載の方法。
- 一枚膜リポソームのライブラリーを用いたスクリーニング方法であって、以下:
(i)請求項5〜8のいずれか一項に記載のライブラリーを提供する工程;
(ii)該ライブラリーから、所望の特徴を有する一枚膜リポソームを選択する工程;
(iii)該一枚膜リポソームに含まれるDNAを増幅する工程;および、
(iv)該増幅したDNAを単離する工程、
を包含する方法。 - 一枚膜リポソームのライブラリーを用いたスクリーニング方法であって、以下:
(i)請求項13〜16のいずれか一項に記載のライブラリーを提供する工程;
(ii)該ライブラリーから、所望の特徴を有する一枚膜リポソームを選択する工程;
(iii)該一枚膜リポソームに含まれるRNAに逆転写酵素を作用させて、DNAを生成する工程;
(iv)該生成したDNAを増幅する工程;および、
(v)該増幅したDNAを単離する工程、
を包含する方法。
