Description
本発明の更なる目的は、細胞移植及びin vitro薬物試験に好適である、ヒト肝細胞の少なくとも1つの特徴を有する細胞を提供することである。
本発明の好ましい実施形態の幾つかは、従属クレームの主題を形成する。
(a)培養容器中の細胞培養培地にヒト単球を播種するステップ;
(b)インターロイキン3(IL−3)及びアデノシンを含む細胞培養培地中でその単球を培養するステップ;及び
(c)その細胞を、ステップ(b)と同時に又はステップ(b)の後で、線維芽細胞増殖因子4(FGF−4)を含む細胞培養培地中で培養するステップ。
本発明の更なる好ましい実施形態では、アデノシンは、10〜300nMの濃度でステップ(b)の細胞培養培地に存在する。
本発明の更に別の実施形態では、ステップ(b)及び/又は(c)の細胞培養培地容積は、培養容器の40〜90μl/cm2である。
本発明の更に別の実施形態では、細胞は、本発明の方法のステップ(c)において、少なくとも7日間培養される。
好ましくは、ヒト肝臓から単離された肝細胞は、除外される。
更に、本発明は、in vitro毒性試験又は細胞療法のための、本発明の細胞の使用に関する。
下記で例示的に説明されている本発明は、任意の要素(複数可)の非存在下で、本明細書で具体的に開示されていない制限(複数可)下で、好適に実施することができる。
用語「含む」は、本明細書及び請求項で使用される場合、他の要素を除外しない。本発明の目的では、用語「からなる」は、用語「含む」の好ましい実施形態であるとみなされる。これ以降、ある群が、少なくともある数の実施形態を含むと定義される場合は、これも、好ましくはこれら実施形態のみからなる群を開示すると理解されるべきである。

− ヘキソキナーゼ/グルコース−6−リン酸デヒドロゲナーゼ法(Bondarら(1974年)Clin Chem 20巻/5号:586〜590頁)を使用してNADPHの光度測定法により測定される、グルカゴン刺激後のグルコース分泌

− 例えば、蛍光染料ローダミン123又はHOECHST33342の輸出により測定される(Brown CD(2008年)、Toxicol.Appl.Pharmacol.233巻(3号):428〜38頁)、ABC輸送体ファミリーのタンパク質等の生体異物輸送体の発現;及び
− 例えば、様々なCYP P450遺伝子及びE2F等の転写因子の遺伝子発現パターン、並びにRAR、RXR及びPPARファミリーの遺伝子発現パターン。
本発明の方法により産生される細胞は、上記に列挙した特徴の少なくとも1つ、好ましくは2つ又は3つ、より好ましくは4つ又は5つ、更により好ましくは6つ、7つ、又は8つ、最も好ましくは全てを示す。
これら単球は、通常、ヘパリン、EDTA、又はクエン酸塩等の抗血液凝固剤で処理された全血から得られる。全血は、単球を単離するまで、4℃で最長24時間保管することができる。
しかしながら、本発明のプロセスで使用される培地は、L−グルタミン、ペニシリン、及びストレプトマイシン等の他の従来の細胞培養培地補完物を含有していてもよい。
IL−3は、多能性造血幹細胞の骨髄前駆細胞への分化を刺激し、単球を含む骨髄系統の全ての細胞の増殖を刺激するサイトカインである。好ましくは、ヒトIL−3が使用され、最も好ましくは、組換えヒトIL−3が使用される。野生型ヒトIL−3は、例えば、R&D Systems社(ミネアポリス、USA)、GenWay社(サンディエゴ、USA)、又はMiltenyi Biotech社(ベルギッシュ グラートバッハ、ドイツ)から得ることができる。
M−CSF(CSF−1とも呼ばれる)は、単球、繊維芽細胞、及び内皮細胞により産生される、その特異的c−Fms受容体(CSF−1Rとも呼ばれる)に結合すると単球の分裂を誘導するサイトカインである。好ましくは、ヒトM−CSFが使用され、最も好ましくは、組換えヒトM−CSFが使用される。細胞培養培地に使用されるM−CSFは、R&D Systems社(ミネアポリス、USA)又はSigma Aldrich社(ダイセンホーヘン、ドイツ)等の好適な供給業者から得ることができる。
第1及び第2の培養ステップで使用される細胞培養培地は、胆汁酸又は胆汁酸塩、例えばタウロコール酸、グリココール酸、コール酸、ケノデオキシコール酸、デオキシコラート、及びリトコール酸;カフェイン;及び/又はヒドロコルチゾン及びデキサメタゾン等のグルココルチコイド等の更なる化合物を含んでいてもよい。デオキシコラートが培地中に存在する場合、デオキシコラートは、1〜30μM、好ましくは2〜25μM又は3〜20μM、より好ましくは4〜18μM又は5〜15μM、最も好ましくは10μMの濃度で存在する。カフェインが培地中に存在する場合、カフェインは、5〜100μM、好ましくは10〜80μM又は20〜70μM、より好ましくは30〜65μM又は40〜60μM、最も好ましくは50μMの濃度で存在する。
本発明の細胞は、例えば薬物スクリーニングで物質の毒性を研究するためにin vitroで使用することができる。例えば、細胞は、基本培地中で8〜24時間饑餓状態にし、その後様々な量の試験物質又は対照用の対応する媒体を含有する基本培地でインキュベートすることができる。最長72時間のインキュベーションの場合、培地は変更されず、より長いインキュベーションの場合は、培地試料を取得し、培地を週3回変更する。インキュベーション期間の終わりに、培地試料を取り出し、対応する溶解緩衝液(例えば、Triton X100/MOPS)で細胞を溶解する。溶解した後、溶解物中のタンパク質含有量及び酵素活性(DPP−IV、GGT、LDH、ALT)を決定することができる。
細胞療法の場合、細胞は、脾臓内、門脈内、肝臓内、又は末梢静脈内注射、又はそれらの組み合わせのいずれかにより、無菌PBS、0.9%NaCl、又はリンゲル液中の懸濁物として投与される。
健常ドナーに由来する全血をヘパリンで処理し、血液対フィコールの比率が2:1のフィコールを使用して勾配遠心分離(400g;室温;30分間)で分離した。中間層を、1:3〜1:4の細胞懸濁物対PBSの比率で氷冷無菌PBSに再懸濁し、その後400gで5分間4℃にて遠心分離した。最初の洗浄後、上清を廃棄し、細胞ペレットを、氷冷無菌PBSに再懸濁した(50ml全血からのPBMCの場合、20〜30mlの氷冷無菌PBSを使用する)。可視的な赤血球の混入が存在した場合、この洗浄ステップは、無菌溶血緩衝液(8.29g/lのNH4Cl、1g/lのKHCO3、0.037g/lのEDTA)で10分間、暗所で室温にてインキュベーションした後で行う。再びPBSに再懸濁した後、細胞を300gで5分間遠心分離した(4℃)。もう一度上清を廃棄し、細胞をPBSに再懸濁し、その後250gで5分間4℃にて最後の遠心分離を行った。

100nMアデノシン(Sigma社、ダイセンホーヘン、ドイツ)
0.5ng/ml組換えヒトM−CSF(R&D Systems社)
細胞を、250,000細胞/cm2の密度で、細胞播種の30〜120分前に培地と共に予めインキュベートしたペトリ皿に播種した。細胞を96ウエルプレートに播種した場合、外側ウエルは、空のままにしておき、細胞培養培地のみで満たし、温度並びにO2及びCO2分圧の違いをなくした。
細胞を、IL−3、M−CSF、及びアデノシンを含む培地でインキュベートした後、培地を、以下の組成を有する同じ容積の培地と取り換えた:
DMEM/Ham’s F12(1:1)
10%FCS(標準条件)又は2%ヒトアルブミン(無血清条件)
2mMグルタミン
100μg/mlストレプトマイシン
1000IEペニシリン
3ng/ml組換えヒトFGF−4
10ng/ml組換えヒトEGF
5nM組換えヒトグルカゴン
0.28IE組換えヒトインスリン
5IE/mlヘパリン
細胞を、少なくとも7日間、又は図及び図の凡例に示されている期間、この培地でインキュベートした。第2の培養ステップ中に、形態が再び変化し、細胞器官が豊富な細胞質を有する、大型で単核又は多核の多角形細胞、metahepsのコンフルエント又はサブコンフルエント層がもたらされる。この形態は、培養培地中の血清の存在又は非存在とは無関係に観察される。
タンパク質含有量は、ビシンコニン酸(BCA)アッセイ(Sigma社、ダイセンホーヘン、ドイツ)で決定した。細胞を、培養容器で増殖させ、基本培地で24時間饑餓状態にし、表示されている実験用化学薬品と共にインキュベートした。培養培地を除去した後、細胞を、Triton X−100(登録商標)/MOPSで溶解し、10μl等量の溶解物を、200μlのBCA試薬と共に37℃にて30分間インキュベートした(50部の試薬+1部の4%CuSO4)。560nmの吸光度をマルチラベルカウンター(Dynatech社製MR7000)で測定した。タンパク質含有量は、ウシ血清アルブミンを基準として使用して計算した。
a)LDH活性
LDH活性は、ピルビン酸塩及びβ−NADHを乳酸塩及びβ−NDA+にするLDH触媒反応から生じるβ−NAD+を、標準的プロトコール(Bergmeyer H.U.(Hrsg.)「Methoden der enzymatischen Analyse」3.Aufl.1974、Band 1、Verlag Chemie Weinheim/Bergstr)を使用して、又は製造業者の説明書に従いCyto Tox96(登録商標)キット(Promega社)を使用して測定することにより分光光度的に決定した。
ALTの活性は、ALTによるL−アラニン及びα−ケトグルタル酸塩からピルビン酸塩及びL−グルタミン酸塩の生成、及びLDH依存性のβ−NAD+形成によるピルビン酸塩生成のその後の定量化からなる2段階反応により測定した(Schumann Gら(2002年)Clin Chem Lab Med 40巻(7号):718〜724頁)。試料(13,000U/分で4℃にて遠心分離した後の、Triton X−100(登録商標)/MOPS中の細胞溶解物)を、α−ケトグルタル酸塩の非存在下で、反応混合物(100mmol TRIS、500mmol L−アラニン、0.18mmol β−NADH、0.1mmolピリトキサル−5’−ホスフェート、1700U/l L−乳酸デヒドロゲナーゼ、pH7.15)と共に37℃にて300秒間インキュベートした。15mmol α−ケトグルタル酸塩を添加することにより反応を開始し、90秒間インキュベーションした後、β−NADH吸光度の減少を180秒間モニターした。或いは、Randox Chemicals社製のALTキットを製造業者の説明書に従って使用した。
ガンマ−グルタミル−トランスペプチターゼ活性は、L−γ−グルタミル−p−ニトロアニリン及びグリシルグリシンからγ−グルタミル−グリシルグリシド及びp−ニトロアニリンへのγ−GT触媒反応から生じるp−ニトロアニリンの光度決定を使用して測定した(Szasz G:Gamma−glutamyl−transpeptidase、in Methoden der Enzymatischen Analyse、第3版(1巻)Bergmeyer HU編、Weinheim/Bergstr.、Verlag Chemie、1974年、757〜762頁)。
DPP−IV活性は、発色基質Gly−L−Pro−p−ニトロアニリドのDPP−IV触媒切断から生じるp−ニトロアニリンの光度決定を使用して測定した。
CYP P450活性を測定するために、P450Glo(商標)アッセイ(Promega社)を製造業者の説明書に従って使用した。手短に言えば、細胞を基本培地中で饑餓状態にし、様々な誘導因子と共にインキュベートした(CYP1A2の場合は、0.1〜10μM、典型的には1μMの濃度の3−メチルコラントレン;10μMのデキサメタゾン;10〜100μM、典型的には50μMのリファンピシン;CYP3A4の場合は、5〜10μM、典型的には10μMのフェニトイン;及び2C9の場合は、50μMリファンピシン及び10μMフェニトインで24時間〜72時間)。典型的には、誘導は、48時間のインキュベーション時間を使用して実施した。誘導した後、細胞を氷冷PBSで3回すすぎ、製造業者の説明書に従って、対応するルシフェリン基質を含有する基本培地でインキュベートし、30分間インキュベーションした後、製造業者の説明書に従いルシフェリン検出試薬を用いて上清の発光を測定した。発光測定値は、BCA法により決定された細胞溶解物中のタンパク質含有量に正規化した。
毒性アッセイは、細胞を基本培地で24時間飢餓状態にした後、様々な濃度のAPAPを基本培地に溶解させて24時間インキュベーションすることにより実施した。培地及び溶解物中のLDH活性を、Promega社製のCyto Tox96(登録商標)キットを使用して上述のように決定し、LDH放出(%)を、100×(培地中のLDH活性/(培地中のLDH活性+溶解物中のLDH活性)で計算した。
a)HOECHST33342染色によるアポトーシス
細胞を、様々な濃度の抗APO(Alexis AXXORA Biochemicals社、レラッハ、ドイツ)/プロテインA(BioVision Research Products社、マウンテンビュー、USA)と共に24〜48時間インキュベートした。インキュベーション期間後、細胞をHOECHST33342(10μM)で15分間染色し、PBSで3回洗浄した後、蛍光顕微鏡を使用して分析した。
細胞溶解物中のカスパーゼ−3活性は、Benesicら(2006年)Kidney Blood Press Res.29巻(5号):280〜293頁に記載のように発色カスパーゼ−3基質Ac−DEVD−pNA(終濃度40μM)を使用して決定した。p−ニトロアニリンを用いた標準曲線を使用して活性を評価し、BCA法により測定されたタンパク質含有量に対して正規化した。
DNA断片化によりアポトーシスを評価するために、ヨウ化プロピジウムで染色した細胞のFACS分析を実施した。細胞を基本培地で24時間饑餓状態にし、基本培地又は表示濃度の抗APO(Alexis AXXORA Biochemicals社、レラッハ、ドイツ)を含有する基本培地でインキュベートした。24時間インキュベーションした後、Accutase(登録商標)を使用して細胞を剥離し、250gで5分間(4℃)遠心分離し、0.2%ウシ血清アルブミンを含有する氷冷PBSに再懸濁した。連続撹拌下で純粋エタノールをPBS/BSAにゆっくりと添加して、エタノールの終濃度を75%にした。細胞を−20℃で凍結して、一晩固定させた。その後、細胞を400gで5分間(4℃)遠心分離した。上清を廃棄し、PBS中の50μlヨウ化プロピジウムで少なくとも30分間暗所にて細胞を染色し、その後FACS分析した。アポトーシスは、サブG1事象を評価することにより評価した。
尿素合成は、細胞を基本培地で24時間飢餓状態にし、その後、様々な濃度の塩化アンモニウム(0〜100mM)で24時間インキュベーションした後で測定した。上清を収集し、製造業者の説明書に従いQuantiChrome(商標)尿素アッセイキット(Clonagen社)を使用して尿素含有量を決定した。培地の尿素含有量は、BCA法により測定された細胞溶解物中のタンパク質含有量に対して正規化した。
第VII因子分泌は、Espositoら(2000年)Kidney Int.58巻:123〜130頁;Ziyadehら(1990年)Am.J.Physiol.259巻:F704〜F714頁)と同様に、直接ELISAにより測定した。飢餓状態にした後に、細胞を、表示されている様々なメナジオン濃度で48時間インキュベートした。培地を収集し、100μ等量を、免疫吸着プレート(Nunc社)にて200μl Voller緩衝液(1.59g Na2CO3+2.93g NaHCO3を100mlの再蒸留水(aqua bidest)に添加)で一晩4℃にてインキュベートした。プレートを、200μlのPBS/0.05%Tween(登録商標)−20で3回すすぎ、PBS/0.05%Tween(登録商標)−20中200μlの2%BSAで2時間ブロッキングした。その後、プレートを空にし、2%−BSA−PBS/0.05%Tween(登録商標)−20中で50μlのウサギ抗第VII因子抗体(Abeam社、ケンブリッジ、英国)1:1000と共に1〜2時間4℃にてインキュベートした。200μlのPBS/0.05%Tween(登録商標)−20で3回洗浄した後、プレートを、2%−BSA−PBS/0.05%Tween(登録商標)−20中で50μlの抗ウサギペルオキシダーゼ−Ak1:5000(Santacruz社、ハイデルベルグ、ドイツ)と共に暗所で室温にて1時間インキュベートした。200μlのPBS/0.05%Tween(登録商標)−20で3回すすいだ後、その後、100μlのHRP基質(再蒸留水中の0.5mg/mlのo−フェニレンジアミン、11.8mg/mlのNa2HPO4・2H2O、7.3mg/mlのクエン酸、0.015%のH2O2)と共に、暗所にて15及び45分間インキュベーションを行った。マルチウエルリーダで490nmの吸光度を測定し、その後波長補正した(630nm)。吸光度を、BCA法により決定された細胞溶解物タンパク質含有量で正規化した。
第VIII因子は、Espositoら(2000年)Kidney Int.58巻:123〜130頁;Ziyadehら(1990年)Am.J.Physiol.259巻:F704〜F714頁と同様に、サンドイッチELISAで測定した。飢餓状態にした後、細胞を24時間基本培地でインキュベートした。培地を収集し、100μl等量を、1μg/mlヤギ抗ヒト第VIII因子(Santacruz社、ハイデルベルグ、ドイツ)が事前にコーティンされている免疫吸着プレート(Nunc社)中で2時間室温にてインキュベートした。プレートを、200μlのPBS/0.05%Tween(登録商標)−20で3回すすいだ。その後、プレートを空にして、1%−BSA−PBS/0.05%中で50μlウサギ抗第VIII因子(Santacruz社、ハイデルベルグ、ドイツ)1:1000と共に室温にて1時間インキュベートした。200μlのPBS/0.05%Tween(登録商標)−20で3回洗浄した後、プレートを、1%−BSA−PBS/0.05%Tween(登録商標)−20中で50μl抗ウサギペルオキシダーゼ−Ak1:5000(Santacruz社)と共に暗所で室温にて1時間インキュベートした。200μlのPBS/0.05%−Tween(登録商標)−20で3回すすいだ後、その後、100μlのHRP基質(再蒸留水中の0.5mg/mlのo−フェニレンジアミン、11.8mg/mlのNa2HPO4・2H2O、7.3mg/mlのクエン酸、0.015%のH2O2)で、暗所にて15及び45分間インキュベーションを行った。マルチウエルリーダで490nmの吸光度を測定し、その後波長補正した(630nm)。吸光度を、BCA法により決定された細胞溶解物タンパク質含有量で正規化した。
同じドナーのMetaheps及び初代ヒト肝細胞(pHH)の遺伝子発現を、cRNA−マイクロアレイ(Kat.Nr.OHS 401、Human Toxicology and Drug Resistance SA Biosciences社)で解析した。このアレイで解析される遺伝子は、下記の表1に示されている。
製造業者の説明書に従って、mRNA試料を、ビオチン化cRNAに転写し、アレイ膜上でハイブリダイズした。その後、SA Biosciences社製の化学発光検出キットを使用して化学発光検出を実施した。
11)本発明の細胞の長期毒性
本発明の細胞が長期毒性研究に適合することを示すために、細胞を上述のように産生し、低濃度のアセトアミノフェン(APAP)及びジクロフェナク(diclo)と共に15日間インキュベートした。インキュベーションの1、3、6、8、10、13、及び15日目に、培地試料を取り出し、培地中のLDH活性を、Promega社製の細胞毒性キットを使用して決定した。LDH放出を、100×(培地中のLDH/LDH培地+LDH溶解物)として%で計算した。
12)結果の考察
提示されたデータは、本発明の細胞の肝細胞様特徴を示す。重要な知見は、1A2、2C9、及び3A4等のCYP P450酵素の発現及び活性により要約され、また、これらは、典型的物質(例えば、アリール炭化水素受容体(AHR)、プレグナン−X−受容体(PXR)、構成的アンドロスタン受容体(CAR)、及びグルココルチコイド受容体(GR)のリガンド)を使用して誘導可能である。肝臓で最も豊富なCYP P450酵素であるCYP 2E1は、APAP毒性により、及びタンパク質レベルで間接的に示すことができた。更に、アンモニア依存性尿素合成並びに凝固因子VII及びVIIIの合成は、ELISAにより示すことができた。DPP−IV、γ−GT、及びLDHの特異的酵素活性が存在し、初代肝細胞又は肝細胞由来細胞(Huh−7又はHepG2等)と類似している。したがって、本発明の細胞は、肝細胞様の代謝、遺伝子発現、合成機能、同様に酵素活性を示す。更に、本発明の細胞は、カスパーゼ−3活性、HOECHST染色、及びFACS分析により示されるように、CD95結合により誘導されるアポトーシスを起こすことが可能である。これは、治療介入に潜在的に使用することができる細胞の重要な特徴である。
Claims (12)
- ヒト肝細胞の少なくとも1つの特徴を有する細胞を産生するための方法であって、前記方法は、
(a)培養容器中の細胞培養培地にヒト単球を播種するステップと;
(b)0.6〜1.6ng/mlのインターロイキン3(IL−3)、0.3〜2.5ng/mlのマクロファージコロニー刺激因子(M−CSF)及び50〜200nMのアデノシンを含む細胞培養培地中で前記単球を培養するステップと;
(c)ステップ(b)と同時に又はステップ(b)の後で、1〜5ng/mlの線維芽細胞増殖因子4(FGF−4)を含む細胞培養培地中で前記細胞を培養するステップと、
を含む方法。 - ステップ(c)の前記細胞培養培地が、上皮増殖因子(EGF)、グルカゴン、インスリン、及びヘパリンからなる群から選択される少なくとも1つの化合物を更に含む請求項1に記載の方法。
- ステップ(c)の前記細胞培養培地が、EGF、グルカゴン、インスリン、及びヘパリンを含む、請求項2に記載の方法。
- ステップ(a)、(b)及び(c)のうちの少なくとも一つの前記細胞培養培地が、無血清である、請求項1乃至3のいずれか一項に記載の方法。
- ステップ(a)、(b)及び(c)のうちの少なくとも一つの前記細胞培養培地が、アルブミンを含む、請求項4に記載の方法。
- 前記ヒト単球を播種した後1時間未満の時点で、前記細胞培養培地を変更するステップを更に含む、請求項1乃至5のいずれか一項に記載の方法。
- ステップ(b)及び(c)のうちの少なくとも一方において、前記細胞培養培地の容積が、前記培養容器の40〜90μl/cm2である、請求項1乃至6のいずれか一項に記載の方法。
- ステップ(b)において、前記細胞が、少なくとも4日間培養される、請求項1乃至7のいずれか一項に記載の方法。
- ステップ(c)において、前記細胞が、少なくとも7日間培養される、請求項1乃至8のいずれか一項に記載の方法。
- ヒト単球に由来する細胞であって、請求項1乃至9のいずれか一項に記載の方法により取得可能なヒト肝細胞の少なくとも1つの特徴を有する細胞において、前記細胞は、その代謝特性及び生存能を失わずに少なくとも15日間培養中で維持することができ、前記細胞は、CD95結合抗体と共に24時間インキュベートされるとアポトーシスを起こすことが可能であり、かつ前記細胞は、500μMのカフェインで細胞を処理することによって阻害することができないシトクローム活性を示す細胞。
- in vitro毒性試験における、請求項10に記載の細胞の使用。
- 細胞療法において使用するための請求項10に記載の細胞。
