Description
・CAPD(連続携行式腹膜透析)、すなわち、処方箋に従って1日あたり4袋の透析液を通過させることに基づく治療、
・APD(自動腹膜透析)、すなわち、処方箋に従って8時間あたり約15リットルの透析液に対応する連続夜間治療。
・水混和性溶媒を用いてマルトデキストリンの分別沈殿を行うこと、
・または適切なカットオフもしくは排除閾値を有する種々の膜を介して上述のマルトデキストリンの分子濾過を行うこと、のいずれか
で構成される。
(i)生物学的溶液中に微量で存在する細菌汚染物質を検出するのは困難であり、
(ii)相乗効果があるのでPGNおよびLPSの検出に限定しないことが重要であり、
(iii)感度の高いかつ再現性のある新しい検出方法を開発することが必要であり、しかも
(iv)汚染物質の性質を特徴付けうる感度の高いかつ再現性のある検出方法を用いることが有利である。
・細胞株は、0.5〜1×106細胞/ml、好ましくは0.7〜0.8×106細胞/ml、さらにより好ましくは約0.75×106細胞/mlの密度で使用され、かつ/または
・グルコースポリマー濃度は、50mg/ml未満、好ましくは約25mg/mlであり、かつ/または
・サイトカイン産生は、RANTESでは20時間の刺激後および/もしくはTNF−αでは8時間の刺激後に測定され、かつ/または
・MDPは、1〜100μg/ml、好ましくは約10μg/mLの濃度でグルコースポリマー調製物に添加される。
・MDP(好ましくは1〜100μg/mlの濃度)の存在下で、約20時間にわたり、炎症誘発性汚染物質を含有する可能性のあるグルコースポリマーの調製物の存在下にマクロファージを配置することと、ここで、グルコースポリマー濃度は、50mg/ml未満、好ましくは約25mg/mlであり、かつマクロファージは、0.5〜1×106細胞/ml、好ましくは0.7〜0.8×106細胞/ml、さらにより好ましくは約0.75×106細胞/mlの密度を有する、
・CCL5/RANTESの産生を測定することと、ここで、CCL5/RANTESの産生は、炎症反応を引き起こしうる汚染物質が調製物に含有されることの指標となる、
を含む。
・HEK−Blue(商標)hTLR2株(TLR2アゴニストに特異的に反応する系)、
・HEK−Blue(商標)hNOD2株(PGN解重合生成物および関連分子(MDP、L18−MDPなど)に効果的に反応する系)、ならびに
・Raw−Blue(商標)株(アルカリホスファターゼを発現するようにトランスフェクトされたマウスマクロファージ系)
からなる群から選択可能である。Raw−Blue(商標)株は、先天性免疫レセプター、特定的にはTLR2、TLR4、およびNOD2レセプターを発現する。
・細胞株は、96ウェルプレートにHEK−Blue(商標)hTLR2またはRaw−Blue(商標)では約50000細胞/ウェル、および96ウェルプレートにHEK−Blue(商標)hNOD2では10000細胞/ウェルの密度で使用され、かつ/または
・試験されるグルコースポリマー調製物は、HEK−Blue(商標)hTLR2および/またはHEK−Blue(商標)hNOD2細胞を使用する場合、5〜50mg/mlのグルコースポリマー濃度、好ましくは約37.5mg/mlのグルコースポリマー濃度、ならびにRaw−Blue(商標)細胞を使用する場合、約50mg/mlのグルコースポリマー濃度を有し、かつ/または
・グルコースポリマー調製物を細胞に接触させる処理は、約16〜24時間継続され、かつ/または
・SEAPレポーター遺伝子のシグナルは、好ましくは少なくとも60分間、理想的には60分間のインキュベーションの後、20:180、好ましくは50:150の培養上清:SEAP基質比で検出される。
(a)以上の第1の実施形態に記載されるように、炎症誘発性汚染物質を含有する可能性のあるグルコースポリマーの調製物の存在下に細胞株の細胞、好ましくはマクロファージを配置することと、炎症の急性期のサイトカイン、特定的にはTNF−α、IL−1β、および/またはケモカインたとえばCCL5/RANTESの産生を測定することと、に依拠する少なくとも1つのin vitro炎症反応試験を行うことと、(ここで、これらのサイトカインの産生は、炎症反応を引き起こしうる汚染物質が調製物に含有されることの指標となる、)および/または
(b)以上の第2の実施形態に記載されるように、1種の先天性免疫レセプターまたは複数種の先天性免疫レセプターの活性の検出を可能とする細胞株を調製物の存在下に配置して、レセプターの活性またはこのレセプターに関連するレポーター遺伝子のシグナルを検出することと、(ここで、この活性またはこのシグナルの検出は、レセプターのアゴニストである汚染物質が調製物中に存在することの指標となる、)
に依拠する工程を含む。
(a)炎症誘発性汚染物質を含有する可能性のあるグルコースポリマーの調製物の存在下に細胞株の細胞、好ましくはマクロファージを配置することと、炎症の急性期のサイトカイン、特定的にはTNF−α、IL−1β、および/またはケモカインたとえばCCL5/RANTESの産生を測定することと、に依拠する少なくとも1つのin vitro炎症反応試験を行うことと、(ここで、これらのサイトカインの産生は、炎症反応を引き起こしうる汚染物質が調製物に含有されることの指標となる、)
(b)調製物が汚染物質を含有する場合、以上に記載したように、1種の先天性免疫レセプターまたは複数種の先天性免疫レセプターの活性の検出を可能とする細胞株を調製物の存在下に配置して、レセプターの活性またはこのレセプターに関連するレポーター遺伝子のシグナルを検出することと、(ここで、この活性またはこのシグナルの検出は、レセプターのアゴニストである汚染物質が調製物中に存在することの指標となる、)
に依拠する工程を含む。
・HEK−Blue(商標)hTLR4系:この株は、TLR4アゴニストに特異的に反応する。それは、特定的には、LPSのアッセイを可能にする。
・HEK−Blue(商標)hTLR2株:この株は、TLR2アゴニストに特異的に反応する。それは、特定的には、PGNのアッセイを可能にする。
・HEK−Blue(商標)hNOD2株:この株は、NODSアゴニストに特異的に反応する。それは、特定的には、MDPのアッセイを可能にする。
・HEK−Blue(商標)Null2株:これは対照株であり、その使用は、グルコースポリマー溶液が内因性機序を介して酵素の産生を誘発しないことを検証するために必要である。

・糖脂質およびリポペプチドに対しては、それらを回収して濃縮するために、それらの両親媒性に基づく分別手順が行われる。
本発明に係るグルコースポリマーを得るための原材料は、以下の方法でモチトウモロコシデンプンから生成される。
・トウモロコシ全粒を排他的に保持するためにトウモロコシを清浄化し、
・粒を軟化させるために、こうして清浄化されたトウモロコシを乳酸の存在下で浸漬し、
・湿式ミル処理してから、種々の成分、すなわち、胚芽、セルロース外皮、タンパク質、およびデンプンを分離し、
・物理化学的にも細菌学的にもデンプンを精製するために、精製水を用いて向流モードでデンプンを清浄化し、
・デンプンの遠心分離および乾燥を行い、
・40%の最終乾物含有率および45℃〜50℃の温度で精製水中にデンプンを懸濁させ、
・pH<2のHClの添加によりデンプン懸濁液を酸性化して、6〜8分間で温度を115〜120℃に上昇させる、
・このpHでタンパク質および脂肪を凝集させ、
・懸濁液をpH5に中和し、
・懸濁液を珪藻土に通して濾過し(残留するタンパク質、脂肪、およびセルロースを保持するために)、
・強カチオン性樹脂および弱アニオン性樹脂を用いて脱塩し、
・70〜80℃の温度および4〜4.5のpHで活性炭により処理して、着色不純物の除去および微生物学的不純物のレベルの低減を行う。
乾量基準で0.2%〜0.5%の濃度で添加された活性炭粉末は、あらかじめフィルター剤が充填された10μmセラミックフィルター上に保持される。
・流下膜式蒸発器に通して濃縮し、
・Niro社により販売されているMSDスプレー乾燥機で濃縮溶液をスプレー乾燥させる。
pH:10%の溶液で4.0〜7.0、

1)水調製/水質
・3μmの濾過による水の精製、活性炭による処理、陽イオンおよび陰イオン交換樹脂による脱塩、ならびに再濾過(UA)、
・使用した2つのタンク:
デンプン加水分解物の溶解ならびにスプレー乾燥式の濯ぎおよび清浄化のために10m3
主要プロセス(タンクの清浄化、それへの活性炭の懸濁、およびクロマトグラフィー)のために60m3。
3)クロマトグラフィー
・60〜85℃の温度で35〜45%の乾物含有率を得るための、精製水を用いたデンプン加水分解物の可溶化、
・ΔP<3barで0.45μm次いで0.22μmに通して行われるデンプン加水分解物の滅菌濾過、
・6連の二重プレートで構成されるそれぞれ1m3の樹脂の連続システムを用いて行われるサイズ排除クロマトグラフィー(SEC)分離。使用される樹脂は、Purolite社により販売されているPCR145Kである。
・酵母およびカビ:0/g
・好気性微生物:0/g
・エンドトキシン(終点ゲルクロットLAL試験):≦0.3EU/g
・ペプチドグリカン:<3ng/g
・B.アシドカルダリウス(B.acidocaldarius):1/g
材料および方法
THP−1細胞(88081201、ECACC)を実験室で慣例に従って培養する。
・LAL試験:大腸菌(E.coli)O55B5 LPS
・標準SLP試験:M.ルテウス(M.luteus)PGN−Wako
・SLP−HS試験:S.アウレウス(S.aureus)PGN−Wako。
第1の試験は、分化THP−1細胞による炎症誘発性サイトカインの産生に対するMDPとf−MLPさらにはPGN/LPS組合せとの相乗能の試験に依拠するものであった。
・分化THP−1細胞の最終濃度:0.75×106細胞/ml、
・感作剤:10μg/mLのMDP(S.アウレウス(S.aureus))、
・最終グルコースポリマー濃度:25mg/ml、
・反応:20時間の刺激後のRANTES産生のELISAアッセイ。
材料および方法
HEK−Blue(商標)細胞株(InvivoGen)は、先天性免疫レセプターをコードするベクターを用いて安定なトランスフェクションにより改変された株である。これらの細胞はまた、同一細胞株内で発現されるレセプターに関連するシグナリング経路の直接制御下で合成が行われる分泌型アルカリホスファターゼ(SEAP:分泌型胎盤アルカリホスファターゼ)を産生するレポーター遺伝子と共トランスフェクトされる。
・HEK−Blue(商標)hTLR2株(HEK−TLR2):この株は、TLR2アゴニスト(特定的にはPGNならびに大多数の糖脂質およびリポペプチド)に特異的に反応する、
・HEK−Blue(商標)hNOD2株(HEK−NOD2):この株は、MDPおよび関連分子単量体たとえばPGNに特異的に反応する、
・HEK−Blue(商標)Null2株(HEK−Null):これは対照株であり、その使用は、グルコースポリマー溶液が内因性機序を介して酵素の産生を誘発しないことを検証するために必要である、
・Raw−Blue(商標)株:これは、SEAPでトランスフェクトされたマウスマクロファージ株である。実質上すべての先天性免疫レセプターを天然で発現するこの株は、試験で陽性対照として使用される。
HEK−Blue細胞は、凍結バイアルの形態で入手される。炎症反応試験を開始する前、細胞の増殖の再開および炎症性分子に反応するそれらの能力を確認することが必要である。さらに、これらのトランスフェクト細胞は、複数回の継代後、急速に変性し、その結果、先天性免疫レセプター用の発現ベクターさらにはSEAPをコードするベクターが損失する可能性がある。
・最終グルコースポリマー濃度:HEK−Blue細胞に対して37.5mg/mlおよびRaw−Blue細胞に対して50mg/ml、
・酵素反応:50μlの調整培地+150μlのSEAP試薬、
・最小反応時間:60分間。
・THP−1:高いLPS反応性を有する任意の炎症性汚染物質。
・Raw−Blue:高いPGN反応性を有する任意の炎症性汚染物質。
・HEK−TLR2:高いPGN反応性を有するTLR2アゴニストに特異的。
・HEK−NOD2:MDPつまりPGN分解産物に特異的。THP−1細胞に関しては、PGNおよび/またはLPSのレベルと細胞反応の大きさとの間に比例関連が存在しないことにも、注目すべきである。
実施例2および3は、感作THP−1株、HEK−TLR2株、HEK−NOD2株、およびRaw−Blue株が、グルコースポリマーサンプル中の微量の炎症性汚染物質に効果的であることを示している。TLR4およびNOD2レセプターを介して存在が検出されるLPSおよびMDPに加えて、サンプル中に存在しうる他の汚染物質は、主に、TLR2リガンド(PGN、LTA、およびリポペプチド)である。したがって、これらの株により、TLR2特異的反応におけるPGNの寄与を確定することはできない。しかしながら、PGNは、約100kDa〜数百万Daの重量範囲の高分子である。これとは対照的に、LTAおよびリポペプチドは、15kDa未満の低重量を有する。したがって、限外濾過工程(30kDa)の導入により、PGNを保持して小サイズのTLR2リガンドに対する反応のみを測定することが可能である。
この実施例に関する実験のために、実施例2および3に示された以下の5つの細胞株が使用される。
・THP−1単球株:高いLPS反応性を有する任意の炎症性汚染物質に対する反応、
・Raw−Blue(商標)株:高いPGN反応性を有する任意の炎症性汚染物質に対する反応、
・HEK−Blue(商標)hTLR2株:高いPGN反応性を有するTLR2アゴニストに特異的、
・HEK−Blue(商標)hNOD2株:PGN全解重合生成物(MDP)に特異的、
・HEK−Blue(商標)Null株:陰性反応対照。
図20に示された結果は、限外濾過の前および後の種々のサンプルの存在下で得られたHEK−TLR2細胞およびRaw−Blue細胞の反応を示している。
I−10.01:汚染されていないサンプル。
I−10.02:PGNで強く汚染されたサンプル。THP−1細胞およびHEK−NOD2細胞ならびに濾液の反応が不在であることから、他の汚染物質の寄与は無視しうることが示唆される。
I−10.03:おそらく小サイズの分解産物に由来する残分で弱く汚染されたサンプル。実際に、全反応および濾液反応は、すべての試験でバックグラウンドノイズ限界にある。
I−11.12:PGNで強く汚染されたサンプル。また、THP−1細胞の弱い反応は、微量のLPSを示唆する。
MM−10.04:LPSおよび小サイズのTLR2活性化分子(LTA、リポペプチド)で弱く汚染されたサンプル。また、MDPの存在は、PGN分解産物を示唆する。
MM−10.05:PGNおよび小サイズの他の分子で汚染されたサンプル。他の試験の弱い反応は、微量のLPSおよび他のTLR2リガンドの存在を示唆する。
MP−10.06:多数の小分子で汚染されたサンプル。一方、弱いTHP−1およびHEK−TLR2反応から、PGNおよびLPSの不在が示唆される。
MP−10.07:LPSおよびMDPの割合が高い多数の小分子で汚染されたサンプル。
このアッセイは、TLR2レセプターを発現する株によるPGNの特異的認識と、TLR2に関連するシグナリング経路の活性化を介して測定可能な酵素活性の生成と、に基づく。
このアッセイに関する実験のために、以下の2つの株が使用される。
・HEK−Blue(商標)hTLR2株、
・HEK−Blue(商標)Null2株。
標準S.アウレウス(S.aureus)PGNを用いて、用量−反応曲線を作成した(図21)。
・(A)部は、TLR2の効果的活性化を与える濃度未満の低濃度のPGNを用いて得られる反応に対応する。したがって、この非線形ゾーンは、この方法の閾値検出限界に対応する。この方法の変動性を含めると、この検出閾値は、バックグラウンドノイズの値(刺激の不在下で得られる反応)の3倍であると推定される。
・(B)部は、線形反応が観測されるので、最も興味深い。この効果的反応ゾーンにより、細胞反応とPGNレベルとの直接的関係を決定することが可能である。したがって、それはアッセイゾーンである。
・(C)部は、高すぎるPGN濃度の存在下での細胞反応の飽和に対応する。実際に、TLR2レセプターの飽和が存在する。
PGNアッセイは、グルコースポリマー溶液を用いて行われる。PGN定量と必要とするサンプルをHEK−Blue(商標)hTLR2細胞と共にインキュベートし、生成された酵素活性を定量することにより細胞反応を測定する。サンプル中に含有されるPGNの量は、用量−反応曲線を参照することにより決定可能である。
未処理ポリマーの存在下でのHEK−TLR2細胞の反応は弱く、このことは、実施例3と比較してポリマー濃度が低いことを考えれば、予想されたことである。
Claims (17)
- グルコースポリマーに含まれる炎症誘発性汚染物質を検出する方法であって、前記汚染物質が、単独または組合せで、炎症反応を引き起こすことが可能であり、少なくとも1種の炎症反応因子の検出を可能とする細胞株を用いた少なくとも1つのin vitro炎症反応試験を含み、前記細胞株が、マクロファージ細胞株もしくはマクロファージ分化細胞株のいずれかであり、前記炎症反応試験が、炎症誘発性汚染物質を含有する可能性のあるグルコースポリマーの調製物の存在下に前記細胞株の細胞を配置することと、炎症の急性期のRANTESの産生を測定することと、に依拠し、前記RANTESの産生が、炎症反応を引き起こしうる汚染物質が前記調製物に含有されることの指標となることを特徴とする、方法。
- 前記グルコースポリマーが、腹膜透析、経腸栄養および非経口栄養、ならびに新生児の栄養のためのグルコースポリマーの群から選択されることを特徴とする、請求項1に記載の方法。
- 前記細胞株が、マクロファージ分化THP−1株であることを特徴とする、請求項1または2に記載の方法。
- 前記細胞株が、0.5〜1×106細胞/mlの密度で使用されることを特徴とする、請求項1に記載の方法。
- 前記RANTES産生が、20時間の刺激後に測定されることを特徴とする、請求項1に記載の方法。
- ムラミルジペプチド(MDP)および関連分子(L18−MDP、グリコリル−MDP)、f−MLP(ホルミル−Met−Leu−Pheトリペプチド)タイプのホルミル化微生物ペプチド、ならびにリポポリサッカリド(LPS)から選択される分子が、前記グルコースポリマー調製物に添加されることを特徴とする、請求項1〜5のいずれか一項に記載の方法。
- MDPおよびLPSから選択される分子が前記グルコースポリマー調製物に添加されることを特徴とする、請求項1〜6のいずれか一項に記載の方法。
- MDPが、1〜100μg/mlの濃度で前記グルコースポリマー調製物に添加されることを特徴とする、請求項7に記載の方法。
- 前記方法が、以下の特徴、すなわち、
・前記細胞株は、0.5〜1×106細胞/mlの密度で使用され、かつ/または
・前記グルコースポリマー濃度は、50mg/ml未満であり、かつ/または
・前記RANTES産生は、20時間の刺激後に測定され、かつ/または
・MDPは、1〜100μg/mlの濃度で前記グルコースポリマー調製物に添加される、
の1つ以上の特徴を有することを特徴とする、請求項1〜8のいずれか一項に記載の方法。 - ペプチドグリカン(PGN)および/またはLPSによる前記グルコースポリマーの汚染を検出可能にすることを特徴とする、請求項1〜9のいずれか一項に記載の方法。
- 試験される前記グルコースポリマー調製物が、5〜50mg/mlのポリマー濃度を有することを特徴とする、請求項1〜10のいずれか一項に記載の方法。
- 前記調製物を前記細胞と共にインキュベートする前にムタノリシンで前記グルコースポリマー調製物を処理する工程を含むことを特徴とする、請求項1〜11のいずれか一項に記載の方法。
- 30kD〜150kDのカットオフ閾値で前記グルコースポリマー調製物を濾過する工程をさらに含み、その濾液を前記細胞と共にインキュベートし、前記濾液に関する結果を前記グルコースポリマー調製物に関する結果と比較することを特徴とする、請求項1〜12のいずれか一項に記載の方法。
- 本発明に係る方法が、
(a)炎症誘発性汚染物質を含有する可能性のあるグルコースポリマーの調製物の存在下にマクロファージを配置することと、RANTESの産生を測定することと、に依拠する少なくとも1つのin vitro炎症反応試験を行うこと(ここで、これらのRANTESの産生は、炎症反応を引き起こしうる汚染物質が調製物に含有されることの指標となる)、及び
(b)1種の先天性免疫レセプターまたは複数種の先天性免疫レセプターの活性の検出を可能とする細胞株を前記調製物の存在下に配置して、前記レセプターの活性またはこのレセプターに関連するレポーター遺伝子のシグナルを検出すること(ここで、この活性またはこのシグナルの検出は、前記レセプターのアゴニストである汚染物質が調製物中に存在することの指標となる)、
に依拠する工程を含むことを特徴とする、請求項1〜13のいずれか1項に記載の方法。 - TLR2レセプターの活性の検出を可能とする細胞を用いた試験(b)を行うことを含むことを特徴とする、請求項14に記載の方法。
- さらに、NOD2レセプターの活性の検出を可能とする細胞を用いた試験(b)および/またはTLR2、TLR4、およびNOD2レセプターの活性の検出を可能とする細胞を用いた試験(b)を行うことを含むことを特徴とする、請求項15に記載の方法。
- 炎症反応を引き起こしうる汚染物質を同定または定量することに依拠する工程をさらに含むことを特徴とする、請求項1〜16のいずれか一項に記載の方法。
