Description
本発明は、特に乳児、子供、又は高齢者向けの食品、並びに医療及びダイエット用食品及びその他の食品用途のための乳(milk)由来のシアリルラクトース濃縮物に関する。本発明はまた、シアリルラクトース濃縮物の調製方法を含む。
人体において細胞間コミュニケーションは中心的かつ普遍的な重要性を有する。このプロセスのキープレーヤーは、特定の物質、例えば、いろいろな細胞膜分子、サイトカイン、又は病原体と結合する細胞外炭水化物部分である。このような炭水化物部分のひとつの重要な成分はシアル酸であり、これはほとんどのヒト及び動物組織に存在する9炭素原子の単糖類である。シアル酸の濃度は、処理や相互作用が高い速度で行われる脳などの組織において高くなる。シアル酸はすべての人体体液に存在し、その含有量は乳で特に高い。シアル酸は人体が合成できる。
本発明は、乳製品中の天然由来(naturally occurring)のシアリルラクトースに富んだ乳製品由来の濃縮物であって、シアリルラクトース含有量が乾物量(dry matter)に基づいて0.32〜25重量%である前記濃縮物に関する。この濃縮物は乾燥させることができる。天然の反芻動物に由来する乳源から得られるこのようなシアリルラクトース濃縮物粉末は、例えば乳児用調製乳、その他の乳児用栄養食品、子供用栄養食品、機能性食品、及び医療用及びダイエット用食品などを非制限的に含む様々な種類の食品に、組み込むことを意図している。
本発明の濃縮物は、乾物量に基づいて0.32〜25重量%、好ましくは0.4〜25重量%、1〜25重量%、5〜25重量%、10〜15重量%のシアリルラクトース含有量を有する。
本発明では、シアリルラクトース濃度は、UV検出システムとShodexカラムを備えた高性能液体クロマトグラフィー(HPLC)を用いて測定されたが、許容される精度を有する任意の技術を用いることができる。

この生成物を以下の実施例でシアリルラクトース濃縮物という。
1600kgの分画されたホエー透過物を、18 GH3840-30D限外濾過膜(GE Osmonics)を用いて、送給温度10℃、圧力5〜7barで限外濾過した。
3000kgの分画されたホエー保持物を、18 GH3840-30D限外濾過膜(GE Osmonics)を用いて、送給温度10℃、圧力5〜7barで限外濾過した。
実施例1で調製されたシアリルラクトース濃縮物を、混合バット中で、80wt/wt%の蛋白質を含むホエー蛋白質濃縮物(Lacprodan 80,Arla Foods,Denmark)と、完全に溶解するまで混合する。ホエー蛋白質濃縮物を、シアリルラクトース濃縮物の再循環の流れと結合された粉末添加漏斗を通して加える。シアリルラクトース濃縮物とホエー蛋白質濃縮物は33%のシアリルラクトース濃縮物と66%のホエー蛋白質濃縮物という組み合わせで混合される。

本発明の実施例において、ミルクをベースとする開始乳児用調製乳がシアリルラクトース濃縮物で強化されるが、他の調製乳、例えば、フォローオン調製乳、グローイングアップ調製乳、早期調製乳、又はダイズ・ベース調製乳も、同様の仕方で強化できると思われ、それ故、本実施例に含まれる。本実施例は、脂肪、蛋白質、炭水化物、及び灰分の濃度に関するEU規制(乳児用調製乳及びフォローオン調製乳に関する欧州委員会指令91/321/EEC)を満たすために作成される。
Claims (15)
- 天然由来のシアリルラクトースを含む乳製品からシアリルラクトース濃縮物を調製する方法であって、以下のステップ:
i)限外濾過膜を使用して、天然由来のシアリルラクトースを含む乳製品を限外濾過し、限外濾過保持物を得;
ii)限外濾過膜を使用して、前記限外濾過保持物をダイアフィルトレーションし;
iii)逆浸透を行い、場合により乾燥を行い、シアリルラクトースの含有量が、乾物量に基づいて1〜40重量%である、シアリルラクトース濃縮物を得ること

- 前記限外濾過、及びダイアフィルトレーションが、4℃〜15℃の温度、及び5〜20barの送給圧力で行われる、請求項6に記載の方法。
- 前記限外濾過、及びダイアフィルトレーションが、約10℃の温度、及び5〜7barの送給圧力で行われる、請求項6に記載の方法。
- シアリルラクトースが、前記限外濾過保持物中で得られる、請求項1に記載の方法。
- 前記組成物が、逆浸透、結晶化、クロマトグラフィー、乾燥又はそれらの組み合わせ、あるいは1又は2以上の担体と共に乾燥することによってさらに処理される、請求項1〜9のいずれか1項に記載の方法。
- 前記乳製品が、ホエーからラクトースが調製されるときに得られる母液である、請求項10に記載の方法。
- 前記シアリルラクトース濃縮物が、単独で、あるいは油脂、ホエー、脱ミネラル・ホエー、ホエー蛋白質濃縮物、ホエー蛋白質単離物、その他のホエー分画、ホエー又はミルク透過物又は濃縮物、スキムミルク、全乳、半スキムミルク、ミルク分画、マルトデキストリン、ショ糖、ラクトース、天然デンプン及びα化デンプン、グルコース・シロップ、カゼイン、及びカゼイン分画から成る群から選択される1又は2以上の担体と共に乾燥される、請求項1〜11のいずれか1項に記載の方法。
- 前記限外濾過膜が、クロスフロー渦巻き形膜、クロスフロー濾過、デッドエンド濾過、プレート・アンド・フレームシステム、カートリッジシステム、振動システム、フラットシート膜、渦巻き形膜、繊維膜、及びチューブ状膜からなる群から選択される構造を有する、請求項1〜12のいずれか1項に記載の方法。
- 使用される前記膜がクロスフロー渦巻き形膜である、請求項13に記載の方法。
- 使用される前記膜が、GE Osmonics GHシリーズの膜である、請求項14に記載の方法。
