アニオン脂質の塩で構成された非磁性および磁性の安定化ナノ粒子の調製
非磁性のオレイン酸カルシウム系ナノ粒子を調製するため、オレイン酸(100mg)を14.8mgの水酸化ナトリウムを含有する5mlの水溶液に、40℃まで穏やかに加熱しながら溶解させることによりオレイン酸ナトリウムを形成した。この水性オレイン酸ナトリウムにアルブミン溶液(10%、0.4ml)を添加した。塩化カルシウム水溶液(5ml中111mg)を滴下すると、チンダル効果による特徴的な青みがかったオパール色を示すナノ粒子形成がもたらされた。ナノ粒子径は、光子相関分光法により測定すると230±20nmであった。
磁性ナノ粒子への親油性の中性薬剤の組込み
この実施例は、タキソールなどの親油性の中性薬剤を、小容量の生体適合性の水混和性溶媒中のその濃縮溶液を、アニオン脂質と安定剤を含有する磁性流体に添加することによる、該薬剤の磁性ナノ粒子に組込むことを示す。
全トランス型レチノイン酸負荷磁性ナノ粒子の調製
上記の処方方法は、イオン性化合物をその水不溶性複合体として含むことによるナノ粒子の調製に適している。抗癌剤である全トランス型レチノイン酸(atRA)ナトリウム塩は、カルシウムと容易に複合体を形成するアニオン脂質であり、ナノ粒子に組み込むことができるイオン性物質の一例を示す。
アデノウイルス含有磁性ナノ粒子の調製
アデノウイルスが封入された磁性ナノ粒子は、ナノ粒子の形成工程前に、オレイン酸ナトリウム溶液含有磁性流体、またはCa2+もしくはZn2+溶液のいずれかにアデノウイルスを添加することにより作製され得る。以下の実施例において、緑色蛍光タンパク質(GFP)をコードする50μlのアデノウイルス(5×1012粒子/ml)を磁性流体に添加し、該粒子内へのアデノウイルスの取込みを行なった。
カチオン性ペプチドの存在下での磁性ナノ粒子の形成
形質導入効率のさらなる改善は、1種類以上のカチオン性ペプチドまたはタンパク質、特に、アルギニン高含有ポリペプチドと、ポリ−L−アルギニンまたはプロタミンなどのタンパク質を、塩化カルシウムまたは塩化亜鉛の溶液に共添加することによって達成され得る。このようなペプチドとタンパク質を用いたナノ粒子の修飾により、細胞内取込みが助長され、したがって、封入されたアデノウイルスのより効率的な内在化がもたらされ、導入遺伝子発現の増大がもたらされる。ポリ−L−アルギニン塩酸塩(分子量は約70,000Da)をナノ粒子に組み込むと、培養ラット大動脈平滑筋細胞およびウシ大動脈内皮細胞のどちらにおいても、アデノウイルス遺伝子導入の用量依存的増大がもたらされる。
ナノ粒子の磁気勾配標的化
赤色蛍光標識を有するアルブミン修飾磁性ナノ粒子を、6mm長のグレード304ステンレス鋼ステントを既に留置しておいたラットの尾静脈に注射した(図6A)。Grade 304 Stainless Steel(「304スチール」)は、移植可能なデバイスにおける使用の歴史を有する。304スチールで作製された市販のステントはないが、ステント設計を行ない、医療用デバイスの企業と契約して実験における使用のための一連のこのようなステントが製作された。したがって、ここの報告する試験のすべてで、現在商業的に使用されているステントを用いたわけではなかった。
により、初期投与だけでなく、反復投与でもステント留置動脈壁へのベクターの磁場媒介性局在が利用できる可能性のため、大きな有望性がもたらされる。この結果は、電磁場とともに全身性の静脈内送達を行なった群では、「磁場なし」対照と比べてステント上および隣接動脈組織上へのナノ粒子の堆積が有意に高いことを明白に示す。非ステント留置動脈では、磁場ありまたはなしでナノ粒子の局在は示されなかった。
残留ナノ粒子および細胞の磁性トラップおよび除去
この実施例は、外部磁気応答性スチールフィルターでの残留ナノ粒子および細胞の除去を示す(「磁性トラップ」)。図7は、循環系からの磁性ナノ粒子または細胞の回収をモデル設計するために使用される回収システム108(図4)を模式的に要約したフローシステム400を示す。図7に示すように、フローシステム400は、磁性トラップ402(残留ナノ粒子を捕捉するための430ステンレス鋼メッシュを有するエッペンドルフ型)、磁場を発生させるための電磁石404、蠕動ポンプ406、撹拌器408、および流れをサイクルAまたはサイクルBに指向するための蛇口410を含む。適当な蠕動ポンプ406、撹拌器408および蛇口410(一般的には、アフェレーシス装置用のもの)は、本明細書の記載から当業者に理解されるであろう。
アルブミン安定化磁性ナノ粒子のTEMおよび磁化曲線
次に図11Aおよび11Bを参照すると、それぞれ、実施例6に関して上記のアルブミン安定化磁性ナノ粒子(MNP)の透過電子顕微鏡検査の結果および磁化曲線(磁場に対する磁気モーメント)が示されている。小径で多数の個々のオレイン酸コートマグネタイト粒がMNPポリマーマトリックス中に分布されていることに注目のこと(図11A)。MNPは、有意なヒステリシスは示されず、それぞれの示される残留磁気が飽和磁化値0.5%程度である超常磁気的挙動を示す(図11B)。
MNP細胞負荷
次に図12A〜12Bを参照すると、インビトロMNP細胞負荷試験が示されている。特に、図12Aは、MNP用量およびインキュベーション時間の関数としてのウシ大動脈内皮細胞(BAEC)によるMNP取込みの動態を示し;図12Bは、MNP用量およびインキュベーション時間の関数としての細胞バイアビリティを示し;図12Cは、MNP自体で観察されたような超常磁気的挙動を示すMNPが負荷された細胞の磁化曲線を示す。ナノ粒子取込みは、内在化されたMNPの蛍光によって測定した。細胞の生存率はAlamar Blueアッセイによって測定した。
遺伝子導入効率および細胞毒性
該粒子を、培養状態の細胞のトランスフェクションについて試験した。3種類の磁気応答性粒子を調製し、DNAと異なるPEI:DNA比で複合体形成させた。実験ではすべて、ナノ粒子を0.25μgのGFPコードDNAプラスミド/ウェルと5%グルコース中で30分間、複合体形成させ、次いで、10%ウシ胎仔血清(FBS)を補充した細胞培養培地と1:4で混合し、磁場を伴って細胞に10分間適用した。そのトランスフェクション効率、ならびにナノ粒子の取込みおよび毒性を、培養ラット大動脈平滑筋およびウシ大動脈内皮細胞(それぞれ、A10およびBAEC)において、非磁性粒子を対照として用いて試験した。遺伝子発現、NP取込みおよび細胞の生存率を、2日目の時点で、それぞれ、蛍光を485/535nm、620/670nmで測定することにより、およびAlamar Blueアッセイ(540/575nm)により測定した。結果を図13A〜Fに示す。磁気応答性処方物では、非磁性ナノ粒子とは対照的に(図13A〜F)、その細胞内取込みと相関して(図13CおよびD)高レベルの遺伝子産物がもたらされた。すべての処方物で、調べた量範囲の細胞培養物において示された毒性は低かった(図13およびF)。
siRNA含有ナノ粒子の磁性標的化
磁性ナノ粒子を、以下プロトコルを用いて処方した:300mgのFeCl3六水和物と150mgのFeCl2四水和物を含む5.5mlの水溶液を、NaOHの4.75mlの水溶液(1.0M)とすばやく混合した。得られた析出物を磁石で分離した。オレイン酸(150mg)を滴下し、析出物を2mlのエタノールに懸濁させ、混合物をアルゴン中で脱気した。内容物を90℃まで水浴中で5分間、数回の撹拌を伴って加熱した。4mlの水を滴下して穏やかに撹拌すると、オレイン酸コート酸化鉄が磁石上に析出され、液相を注意深く吸引した。析出物を4mlのエタノールで洗浄して過剰のオレイン酸を除去し;磁石上への沈降後、エタノールを吸引した。この析出物を5mlのクロロホルムに再懸濁させた。
スチールステント表面への磁性ナノ粒子負荷内皮細胞の高い磁場勾配での標的化
ナノ粒子の処方および特性評価。塩化第二鉄と塩化第一鉄(それぞれ、300mgおよび150mg)から水性水酸化ナトリウムでのアルカリ析出によって調製したマグネタイトを磁気により分離し、2mlのエタノールに再懸濁させ、アルゴン下で90℃まで水浴中にて5分間加熱することによりオレイン酸(200mg)でコーティングした。4mlの水の滴下によって過剰のオレイン酸を相分離し、脂質コートマグネタイトをエタノールで2回洗浄した。親油性マグネタイトを6mlのクロロホルム中に分散させると、安定な磁性流体が形成された。得られた酸化鉄の有機分散液を用いてPLAを溶解させ、かくして有機相を形成した。この有機相を、氷浴内での超音波処理によって水性アルブミン溶液(1%)中で乳化させた後、有機溶媒をエバポレーションした。該粒子を未結合アルブミンから反復磁性の沈降/再懸濁サイクルによって分離し、10%(w/v)グルコースを抗凍結剤として用いて凍結乾燥させた。凍結乾燥MNPを−20℃で維持し、使用前に脱イオン水に再懸濁させた。
磁性ナノ粒子負荷細胞を316Lステントに標的化させるための均一なMRI磁場の使用
磁性ナノ粒子(MNP)を負荷した細胞のスチールステントへの標的化が、磁気共鳴イメージング(MRI)システム内に存在する均一な磁場を用いて実現可能であることを実証するためのモデル実験を行なった。該イメージング装置内に存在する磁場は1.5テスラ(T)であり、これは、他の試験で使用される最も高い磁場(1000ガウス、すなわち0.1テスラ)よりも15倍大きい。さらに、先の試験では、304スチールステントを使用し、これは0.1テスラの磁場で磁化され、一方、316Lスチールステントはこのレベルの磁場強度では磁化されなかった。さらに、現在臨床使用されているスチールステントのほとんどは316Lスチールであることに注意されたく、したがって、このモデル実験は臨床的に特に重要である。

治療目的のための核酸の非ウイルス送達は、主に、比較的低い有効性のために依然として難題である。本発明者らは、磁気による標的化特性を有する非ウイルス遺伝子担体を使用することにより、この課題に取り組んだ。核酸の磁力ターゲテッド送達が達成可能となることにより、有効で無毒性の遺伝子導入のための臨床的に実行可能な解決策がもたらされ得る。本発明者らの先の研究において、本発明者らは、分枝ポリエチレンイミン(PEI 25K)で表面修飾した、ポリラクチド(PLA)系の生分解性ナノ粒子(NP)の処方物を開発した。最近の科学文献により、線状PEIの市販の調製物を脱アシル化すると、プロトン化可能な窒素の数が増加し、これにより、おそらく、より密な核酸の凝縮およびPEI/核酸複合体のより良好なエンドソーム逸出がもたらされることにより遺伝子送達効率が劇的に刺激されることが示された。本発明の試験では、脱アシル化された線状PEIを用いて処方した超常磁性NP磁気駆動性送達によって、非ウイルス遺伝子導入が向上され得るという仮説を調べた。200kDaのポリ(2−エチル−2−オキサゾリン)の酸触媒型加水分解によって合成してpH7に調整した線状PEIを使用し、改良した乳化−溶媒エバポレーション方法論によって酸化鉄負荷NPを処方した。重量基準で35%の酸化鉄を含有するNPは、平均粒径360±25nm、ゼータ電位43±3mVを有し、超常磁性(磁気飽和値の0.5%未満の残留磁気)を示すものであった。線状PEI−NP処方物が核酸を送達する能力を、インビトロで、培養A10ラット大動脈平滑筋細胞(SMC)およびウシ大動脈内皮細胞(BAEC)において、緑色蛍光タンパク質(GFP)をレポーター遺伝子として用いて調べた。分枝PEIを用いて処方したNPを比較のために使用した。核酸と複合体形成させたNPを細胞に15分間、磁場(500G)下で血清含有細胞培養培地中にて適用した。ある組の実験では、GFPコードプラスミドDNAを細胞に送達し、トランスフェクション効率を、処理の2、4および8日後に蛍光定量的に測定した。細胞内NPレベルは、どちらのPEI処方物でも、調べたNP濃度範囲において直接的に用量依存性であった。GFP発現は、どちらのPEI処方物でも4日目に最大レベルに達し、線状PEI−NP処方物でトランスフェクトした両方の細胞型で2.5〜3倍高いGFPレベルがもたらされた。別の組の実験では、強化型GFP(eGFP)低分子干渉RNA(siRNA)を細胞に送達し、レンチウイルス形質導入平滑筋細胞および内皮細胞におけるeGFP発現の抑制ならびに細胞バイアビリティ(AlamarBlueによる)を、処理の5日後に蛍光定量的に測定した。GFPサイレンシング実験では、分枝または線状いずれかのPEIを用いて処方した磁性NPを使用すると、効率的なeGFP抑制が達成された。A10細胞におけるeGFP抑制は、どちらの処方物型でも、NP用量に直接的に依存した。eGFPの抑制は、線状PEI−NPの場合は直接siRNA用量依存性であり、分枝PEI−NPでは逆依存性であり、どちらのNP型でも40%の最大抑制がもたらされた。BAECでは、eGFP抑制は、分枝PEI−NPの場合のみNP用量に直接的に依存した。どちらのNP型でもeGFP抑制はsiRNA用量に依存性でなく、どちらのNP処方物でも50%の最大抑制がもたらされた。試験したNP/siRNA複合体により細胞の生存率は有意に障害されず、NPおよび細胞型のどちらの場合も処理の5日後、最大NP用量で90%より多くの生存細胞が示された。磁気応答性線状PEI−NPにより、分枝PEI−NPと比べてプラスミドDNAの送達効率の増大が示され、一方、遺伝子サイレンシング実験では、どちらのNP型でも遺伝子抑制能力に差は観察されなかったと結論付けられる。
磁気により指向させたカタラーゼ含有オレイン酸カルシウムナノ粒子は、インビトロで酸化的ストレスから内皮細胞を効率的に保護する
大型の酵素ナノ担体の設計の考慮事項は数多くある。粒子の合成およびその後の精製の条件は、酵素活性を維持したままのタンパク質質量を効率的に負荷することが可能なものであるのがよい。カタラーゼなどの大型酵素のナノ担体が有効なターゲテッド抗酸化性治療薬であるためには、担体は、活性酵素を負荷する能力を有するだけでなく、タンパク質分解からの保護ももたらすものであるのがよい。有機溶媒または高剪断(sheer)乳化法を伴わずにタンパク質が効率的に負荷され得る安定な生体適合性のナノ担体を考案することが望ましい。タンパク質の活性を保持させるため、水性媒体中でオレイン酸カルシウム系MNPが形成され得、剪断、溶媒曝露および油と水の界面による損傷へのタンパク質の曝露が排除され得る水溶性構成要素を使用した。
MNPの物理化学的特性(粒子径)。多数のTEM画像(図18Aに例)のデジタル解析によって求めたカタラーゼ負荷MNPの粒子径では、303+/−38nmの平均粒径が示された。同等の処方物の動的光散乱(DLS)では、340+/−29nmの平均粒子径が示された(図18Cおよび18D)。動的光散乱の測定自体では、SOD負荷粒子は350+/−10nmの流体力学的直径を有していた。
インビトロおよびインビボでのカタラーゼ含有磁性ナノ粒子の抗体標的化
この試験は、このような磁性のナノ担体(MNP)が、インビトロおよびインビボで、活性な抗酸化酵素を内皮標的に負荷および送達する能力を試験するために行なった。
Claims (26)
- 磁気材料または磁化可能な材料、コロイド安定剤、治療用タンパク質、ならびに、1種以上のアニオン性脂質と1種以上の生体適合性多価カチオンとの間で形成される析出した複合体を含む治療用粒子であって、該治療用タンパク質が該析出した複合体の中に封入される、治療用粒子。
- 前記磁気材料または前記磁化可能な材料が超常磁性材料である、請求項1に記載の治療用粒子。
- 前記1種以上のアニオン性脂質がオレイン酸またはその塩を含む、請求項1に記載の治療用粒子。
- 前記タンパク質が抗酸化酵素である、請求項1に記載の治療用粒子。
- 前記抗酸化酵素がカタラーゼ、スーパーオキシドジスムターゼ、またはグルタチオンペルオキシダーゼである、請求項4に記載の治療用粒子。
- 前記安定剤がビオチン化されている、請求項1に記載の治療用粒子。
- さらに、抗体を含む、請求項1に記載の治療用粒子。
- 前記抗体が前記安定剤に連結されている、請求項7に記載の治療用粒子。
- 前記抗体が、内皮細胞の表面上の抗原に特異的に結合し得るものである、請求項7に記載の治療用粒子。
- 細胞を酸化的損傷から保護するためのシステムであって、該システムは:
請求項1に記載の治療用粒子であって、前記治療用タンパク質が少なくとも1種類の抗酸化酵素を含む、治療用粒子;ならびに
該細胞に近位の該磁気材料または該磁化可能な材料を、該細胞が該粒子を内在化させることを可能にするのに充分な期間、磁化させ得る均一磁場を発生させるための磁場源
を含むシステム。 - 前記細胞が内皮細胞である、請求項10に記載のシステム。
- 前記内皮細胞が血管内皮細胞である、請求項11に記載のシステム。
- 前記少なくとも1種類の抗酸化酵素が、カタラーゼ、スーパーオキシドジスムターゼ、またはグルタチオンペルオキシダーゼである、請求項10に記載のシステム。
- 前記粒子が、さらに、前記細胞表面上の抗原に特異的に結合し得る抗体を含む、請求項10に記載のシステム。
- 前記細胞が哺乳動物の体内に存在している、請求項10に記載のシステム。
- 治療用タンパク質を移植可能なデバイスへ磁気的に標的化させるためのシステムであって、該システムは:
(a)被験体内への移植のためのデバイス;
(b)複数の請求項1に記載の治療用粒子;
(c)磁化可能な材料を磁化させ得る均一磁場を発生させるための磁場源であって、該均一磁場は、該デバイスの近位に磁場勾配を発生させる、磁場源;ならびに
(d)該デバイスに送達されなかった該複数の粒子のうちの1つ以上を該被験体から除去するための手段
を含み、ここで、
該デバイスが第2の磁気材料または磁化可能な材料を含み、
該勾配により、該複数の粒子の1つ以上が該デバイスに標的化される、
システム。 - 前記タンパク質が抗酸化酵素である、請求項16に記載のシステム。
- 前記抗酸化酵素が、カタラーゼ、スーパーオキシドジスムターゼ、またはグルタチオンペルオキシダーゼである、請求項17に記載のシステム。
- 前記複数の粒子の各々が請求項1に記載の粒子である、請求項16に記載のシステム。
- 前記デバイスがステントである、請求項16に記載のシステム。
- 前記ステントが304ステンレス鋼で形成されている、請求項20に記載のシステム。

- 前記複数の粒子が前記第1の磁気材料または磁化可能な材料およびパクリタキセルを含むものである、請求項16に記載のシステム。
- さらに、パクリタキセルを含む、請求項1に記載の治療用粒子。
