Description
分離腫瘍細胞による周囲脳組織の侵襲は、多型性神経膠芽腫(GBM)の有効治療に対する主要な障害の1つを示す。神経膠腫は、中枢神経系(CNS)において生じる多数の腫瘍を含む。成人において、最も一般の腫瘍は、星状膠細胞又は乏突起膠細胞に由来する高グレードの新生物である。世界保健機関は、これらの悪性腫瘍をそれらの退形成の程度によって、グレードII(び漫性星状細胞腫)、グレードIII(退形成性星状細胞腫)、及びグレードIV(GBM)に分類している1。
本発明は、CD95活性を高グレードの神経膠腫を有する個体に対して中和する薬剤を投与することを含む、高グレードの神経膠腫を有する個体を治療する方法に関する。
(a)(i)CD95又はそれらの機能的断片、
(ii)候補薬剤
を含有する混合物を、CD95又はそれらの機能的断片が対照活性を有する条件下でインキュベートする工程、
(b)CD95又はそれらの機能的断片の活性を検出して、前記薬剤の存在下で活性を決定する工程、

を含む、CD95の活性を調整する/CD95の活性に作用する、有利には中和する薬剤のためのスクリーニング法に関する。

原発星状細胞腫、乏突起細胞腫及び神経膠芽腫の細胞系におけるCD95表面発現のFACS分析は、低い継代(≦4)を有する。DNAの断片化のために、細胞を、LZ−CD95L(CD95L)の指示投与量で、24時間及び48時間処理し、そしてFACSによって分析した。

原発神経膠芽腫の細胞系(NCH342、354、357、378、417、419及び2421)におけるCD95表面発現のFACSを分析した。CD95Lに対する中和抗体(Nok1、10μg)によって阻害されうる、γ−放射線照射に対する、それら原発NCH細胞において、遊走を誘発した。結果は、平均±標準誤差、*P<0.05、**P<0.01、***P<0.005として示され、かつ2つの独立した実験を示す。

CD95Lは、SrcファミリーメンバーのYes、及びPI3Kのp85サブユニット(その2つのサブユニットp85及びp110によって本明細書に示される)の漸増を、CD95に誘発し、それによってAKTを活性化する。活性化AKTは、GSK3βをリン酸化及び不活性化し、核中へβ−カテニンの転位を可能にし、その際、それはMMPの転写を誘発する。このシグナル経路を、Yesに対してsiRNA、もしくはMMP−2及び−9、PI3K特異性阻害剤(Ly290059)によって、又はGSK3βの優性活性変異体(S9A)でのレンチウィルス感染によって阻害することができた。

詳細な説明
CD95/CD95L複合体が、アポトーシスを誘発することが、一般的に評価されている18。しかしながら、CD95が、アポトーシス非依存法、例えば増殖、血管形成、線維化、及び炎症を媒介することができることが、成長の証拠である19,20。ルイス肺癌腫細胞におけるCD95の過剰発現は、インビボでの腫瘍細胞の生存の利点をもたらす21。同様の点で、CD95のトリガーは、神経膠腫細胞における細胞循環のプログレッションを動因することが報告されている22。悪性星状細胞腫において、CD95結紮は、炎症促進性ケモカインの発現及び血管形成を促進した23-25。本明細書で、神経膠芽腫におけるCD95のトリガーが、最終的に侵襲性を増加することを導くシグナル伝達現象のカスケードを誘導することを報告している。CD95で誘発される遊走を、培養された尿細管細胞中で最初に観察し26、そして卵巣、乳房、肺及び腎臓の腫瘍細胞に関して最近報告している13。後者の研究において、血清勾配を、細胞遊走を起こしかつ方向付けるために、CD95刺激に加えた13。一方、神経膠芽腫細胞は、それらの侵襲する傾向のある表現型によって特徴付けられ、かつ付加刺激の不在下で遊走する。
実施例1−試薬及び一般的な方法
次の抗体、抗ヒトCD95L G247−4(1:200)、CD95Lに対する中和抗体(Nok1)、抗ネズミCD95(Jo2)、抗ネズミCD95L(MFL3)を使用し、そして適切なアイソタイプコントロールのハムスターIgG1、λ1を、Becton Dickinsonから得た。CD95(α−Apo−1)に対する抗体を、以前に記載されているように生成し46、リン酸化GSK3β(P−Ser9−GSK3β、1:1000)、リン酸化AKT(P−Ser473−KT、1:1000)、合計AKT(T−AKT、1:1000)、Srcファミリーキナーゼ(Src、1:1000)、リン酸化Src(P−Tyr416、1:50)及び合計β−カテニン(1:1000)を、New England Biolabs社から得た。CD95(CD95、1:1000)、合計GSK3β(T−GSK3β、1:1000)、リン酸化ERK1/2(P−ERK、1:1000)、合計Yes(Yes、1:1000又は1:200)、合計Fyn(1:200)、及び合計ERK(T−ERK、1:1000)に対する抗体を、Santa Cruz社から得た。抗GFAP(1:200)を、Chemicon社から得て、抗PI3K(p85 N−SH2ドメイン、1:1000)、抗FADD(FADD、1:1000)及び活性−β−カテニン(P−Ser37又はP−Thr41、1:800)を、Upstate社から得て、抗MMP9 GE−213(1:100)をOncogene社から得て、抗GFP(1:400)を、Molecular Probes社から得て、抗GADPHを、Abeam社から得て、かつ抗−カスパーゼ−8抗体(1:10)を、ハイブリドーマの上清から研究所で製造した47。抗PED抗体(1:2000)は、Dr.Herve Chneiweissによって、寄贈された。組織学的染色に対する特異的抗体の視覚化のために、Dako社から得られたストレプトアビジン−アルカリホスファターゼ/FastRedの双方を、使用した。免疫蛍光法の研究のために、モノクローナル抗体Alexa Fluor 488(1:500、Molecular Probes社)及び抗ローダミン(1:200、Dianova社)を、使用した。
NCH腫瘍の組織検体を、患者のインフォームドコンセント及び地域の倫理委員会の承認後に手術中に得た。新鮮な組織を、原発腫瘍培養を確立するための1部分と、RNA抽出のための他部分の2つの部分に分けた。腫瘍分類、手術時の年齢、及び性別についてのそれぞれの患者の臨床データを、表1(図22)で要約する。
動物実験は、German Cancer Research Centerの動物管理使用委員会(institutional animal care and use committee)及びRegierungspraesidium Karlsruheによって承認された。頭蓋内投与のために、8〜12週齢の近交系Vm/Dkマウスを使用した。5000個のSMA−560細胞を、トリプシン処理によって回収し、1μlダルベッコ変法イーグル培地(10%胎児ウシ血清(FCS)、1%ペニシリン/ストレプトマイシン(PS)、及び1%L−グルタミン200mMで補足したDMEM)中で再懸濁し、そして10μlをFlexilfilシリンジ(WPI社、ドイツベルリン)中に置いた。穿頭を、ブレグマに対する外側2.75mmで孔あけし、そして針を、3mmの深さまで挿入した。マウスを、投与の7日、14日又は18日後に屠殺した。
腫瘍接種の14日後に、マウスを屠殺し、そして腫瘍を抽出した。そして腫瘍外植を、1時間、培地、培地とアイソタイプコントロール抗体(10μg/ml)、又はMFL3(10μg/ml)を有する培地中でインキュベートした。コラーゲン中へ外植の包埋後に、細胞侵襲を、72時間にわたって、微速度顕微鏡(オリンパス、ドイツ)で記録した。
確立された神経膠芽腫細胞系A172、T98G及びLN18、並びに原発神経膠芽腫細胞(NCHs)を、DMEM(10%FCS及び1%PSで補充)中で、CO2インキュベーターで、36.5℃及び湿度90%で培養した。NCH細胞系は、記載されているように、C.Herold−Mendeの研究所で確立されている48。生化学及び分子分析のために、1×106個の細胞を、培地中で10cmの培養皿上で培養し、そして前記に記載されているようにインキュベートした。スフェロイドを、以前に記載されているように製造した49。簡単に言えば、T98G及びLN18細胞(2−3×104)を、DMEM10mlを含有する10cmの培養皿の蓋の上に懸滴(20μl)で培養した。48時間後に、細胞凝集物を回収し、そして培地で満たした基剤で被覆された2%寒天皿上に転位した。さらに48時間後に、スフェロイドを、3次元コラーゲンゲル中で侵襲分析のために包埋した。
調査侵襲のための生理学的モデルは、3次元コラーゲンゲルアッセイである。スフェロイドを、コラーゲンゲル溶液(Vitrogen3mg/ml(原液溶液)、最終濃度2.4mg/ml)中へ包埋する1時間前に、α−Apo−1(2μg/ml)、LZ−CD95L(5ng/ml)又はNok1(50ng)/LZ−CD95L(5ng/ml)で処理した。コラーゲンゲルの重合(37℃で3060分)後に、DMEMを、ゲルの上層においた。コラーゲンマトリックス中への細胞の侵襲を、微速度顕微鏡(オリンパス、ドイツ)で記録した。スフェロイド境界までのシグナル侵襲細胞の距離(スフェロイド毎にn=10、処理毎に3スフェロイド)を、Image J1.34ソフトウェア(NIH Imageに基づく)で決定した。
FACS分析のために、腫瘍を、指示時点で取り除き、37℃で20分間トリプシン処理し、PBS/10%FCS中で3回洗浄し、ガラスパスツールピペットで粉砕し、100μmナイロンメッシュ(BD Falcon社)を通して濾過し、そして蛍光標示式細胞分取器(Fluorescence Activated Cell Sorter(FACS、Becton Dickinson社))分析のためにPBS/10% FCSで再懸濁した。
インビボで神経膠腫細胞の遊走を、コラーゲンIで被覆された(Chemicon社)transwell inserts(Falcon社)による遊走によって測定した。5×104個の細胞を、孔サイズ8μmを有するコラーゲンで被覆された(50μg/ml)transwell inserts上で培地200μl中で培養した。細胞を、培養前にγ−放射線照射し、又は培養後に、α−Apo−1(2μg/ml又は0.1μg/ml+架橋のためのタンパク質A)、LZ−CD95L(5ng/ml)、Nok1(50ng)/LZ−CD95L(5ng/ml)、Nok1(50ng)LZ−上清(SN、5ng/ml)、CD95L(10ng/ml)及びCD95L−T34で処理した。有利には、培養24時間後に、細胞を、基礎的なDMEMで、それらが処理されるさらに24時間前に、殺す。完全に郵送された細胞の数を、処理後12時間、24時間、及び36時間で計測した。全ての実験において、三回、それぞれの処理に関して測定した。
本来の及び再発性神経膠腫におけるCD95Lのスコアリングを、それぞれのCD95Lで染色された腫瘍から3つの領域の分析(250倍の倍率)によって実施した。ポジティブ細胞を、二重盲検法で、及びポジティブ細胞の数によって指定されたスコアで計測した。
2.5×105個の細胞を、放射線照射の12時間前に、6cmの培養皿上で培養した。細胞を、1、3、10及び50Gyで、室温で、137Cs源(ガンマ細胞1000、Atomic Energy of Canada,Ltd.、ON)を使用して、10.23Gy/分で放射線照射した。細胞を、放射線照射後すぐに、Nok1(10μg/ml)で処理し、又は未処理のままにした。その後、細胞を、遊走アッセイ、RNA抽出、又はNicolettiアッセイのために使用した。
遊走及びmRNA発現データの統計的分析を、ノンパラメトリックStudent t検定を使用して実施し、処理群とコントロールとの差異を比較した。信頼区間を、95%で決定し、*P値<0.05、**P値<0.01、***P値<0.005を、統計上有意であるとみなした。
2×107個の細胞を、10、500及び5μg/mlのCD95L−T4で、1及び5分間(特に指示されない限り)37℃でそれぞれ処理し、又は処理せず、PBS+リン酸塩阻害剤(NaF、NaN3、pNPP、NaPPi、β−グリセリンリン酸のそれぞれ10mM及びオルトバナジン酸塩2mM)中で2回洗浄し、続いて緩衝液A(20mM Tris/HCl、pH7.5、150mM NaCl、2mM EDTA、1mMフッ化フェニルメチルスルホニル、プロテアーゼ阻害剤カクテル[Roche社]、1% Triton X−100[Serva、Heidelberg、ドイツ]、10%グリセロール、並びにリン酸阻害剤[NaF、NaN3、pNPP、NaPPi、β−グリセリンリン酸のそれぞれ10mM、及びオルトバナジン酸塩2mM])中で溶解し(刺激条件)、又は処理せずに溶解した(未刺激条件)。タンパク質濃度を、BCAキット(Pierce社)を使用して決定した。タンパク質1mgを、前記に記載されたような抗カスパーゼ−858、5μgのα−Apo−1、又は2.5μgの抗p58、及び30μlのタンパク質Aセファロースで、一昼夜免疫沈降した。ビーズを、5回、溶解緩衝液の20容量で洗浄した。その免疫沈降物を、それぞれ15%又は7.5%のSDS−PAGE上で分析した。続いて、ゲルを、Hybondニトロセルロース膜(Amersham Pharmacia Biotech社、Freiburg、ドイツ)に転写し、PBS/Tween(PBS+0.05%Tween 20)中で2%BSAで1時間阻害し、そして一次抗体で、2%BSA PBS/Tween中で、4℃で、一昼夜インキュベートした。ブロットを、製造者のプロトコル(PerkinElmer Life Sciences社、Rodgan、ドイツ)に従って、化学発光法で顕色した。
細胞外染色:
シグナル細胞の表面上でCD95の発現を、FACSによって分析した。10%胎児ウシ血清を含有するリン酸緩衝食塩水(PBS 10%FCS)中で1×106細胞/mlを、α−Apo−1(0.01μg/μl)で、20分間氷上でインキュベートし、続いて二次抗体(1:30ヤギ抗マウスフィコエリトリン共役;Dianova社)で、30分間インキュベートした。フローサイトメトリー分析を、Cell Quest Softwareを使用してFACSCalibur(Becton Dickinson社)で実施した。試料毎に最低10000個の細胞を分析した。
PTENの過剰発現効率、Yesのノックダウン効率、又はFADD、Fyn及びYesの基礎的なレベルをそれぞれ測定するために、細胞内FACS染色を実施した。細胞をトリプシン処理し、上清を捨て、そしてペレットをPB緩衝液中で4%パラホルムアルデヒド中で、15分間氷上で再懸濁した。インキュベーション後に、固定された細胞を遠心分離し(3000rpm、4℃、5分)、上清を捨て、そしてペレットを、2回PBS/0.1%サポニン及び10%FCSで洗浄した。試料を、一次抗体(α−HA1:1000 Roche社、α−Yes1:200又はα−Fyn1:200 Santa Cruz社)で30分間氷上でインキュベートし、2回のPBS/0.1%サポニン及び10%FCSでの洗浄工程に続いて、二次抗体(α−マウス−PE1:33 Pharmingen社、又はα−ウサギ−Alexa488(登録商標)1:250 Molecular Probes社)の添加前に、さらに20分間インキュベートした。染色された細胞を、2回、PBS/0.1%サポニン及び10%FCSで洗浄し、そしてそのペレットを、同様の緩衝液で再懸濁した。そして細胞を、FACSによって分析した。値を、特異性抗原に関して正規化平均蛍光強度(MFI)として示した。
DNA断片化を量化するために、トリプシン/EDTA(Gibco社)で分離された細胞を、200×gで遠心分離し、そして70%エタノールで、−20℃で、1時間固定した。固定した細胞を、ヨウ化プロピジウム溶液(50μg/ml、0.0025%クエン酸ナトリウム及び0.0025%Triton−X−100)で1時間又は一昼夜4℃で染色し、そしてFACSによって分析した。
未処理又は処理(10ng/ml及び20ng/mlのCD95L−T4)したT98G細胞の調整培地を、非還元条件下で、ゼラチン1mg/mLを含有する10%SDS−ポリアクリルアミドゲル上に置いた。電気泳動及びTritonX−100(2.5%(v/v)、30分2回)でのゲル洗浄後に、そのゲルを、MMP反応緩衝液[Tris−HCl(pH7.8)50mmol/L、NaCl200mmol/L、CaCl25mmol/L]中で、37℃で16時間インキュベートした。ゼラチン分解活性を、クーマシーブリリアントブルーG−250溶液での染色、及び脱色溶液(10%酢酸、20%エタノール)中でのインキュベーションをして、透明なバンドとして検出した。
T98G細胞を、4%PFAで、37℃で15分間固定し、50mM塩化アンモニウムでインキュベートし、そしてPBS中で0.1% TritonX−100で5分間浸透処理した。遮断後に、細胞を、それぞれの一次抗体でインキュベートし、そして免疫反応を、ローダミン又はフルオレセインイソシアネート(FITC)に結合させたモノクローナル又はポリクローナル抗体で視覚化した。
T98G及びLN18細胞を、レンチウィルスベクターpEIGW及びpEIGW−GSK3βS9Aで、5の感染多重度(MOI)で感染させた。そのプラスミドを、EF1aプロモーターと、pIRES2−eGFPからのIRES−eGFPカセット(Clontech社、ドイツ)を有するpWPTSeGFP(ジュネーブのD.Tronoによって寄贈された)におけるWPRE要素との間のeGFP配列をスプライシングすることによって構築した。組換えレンチウィルスベクターpEIGW−GSK3βS9Aを、pcDNA3 HA−GSK3βS9A(Trevor C.Daleによって寄贈された)を使用して構築した。このベクターは、残基9で、セリンからアラニンの置換を含む構成的な活性GSK3β突然変異体(GSK3βS9A)をコード化する。全てのレンチウィルスを、前記に記載されている方法41を使用して増殖した。全ての導入遺伝子の発現を、感染させた細胞中で、GFP発現のFACS分析によって検証した。感染細胞のパーセンテージは、80〜90%であった。

PTEN(pBP−PTEN−HA)に関する過剰発現プラスミド及び空のベクター(pBP)は、Frank Furnari(San Diego、USA)59によって寄贈された。T98G細胞を、JetPeiを使用して、pBP−PTEN−HA及びpBP(6μg)で形質移入した。形質移入した細胞を、処理後さらに48〜72時間、培養した。
ノックダウン実験を、取扱説明書に従ってリポフェクタミン2000(登録商標)(Invitrogen Life Technologies社)で一時的に形質移入することによって実施した。遊走実験を、Yes、MMP−2及びMMP−9のための有効shRNAmir−pGIPZベクターのネガティブコントロール又はプール、並びにネガティブコントロールとしての非標的shRNAmir−pGIZベクター(それぞれ、RHS 4430−98843955、−98820654、−99161516、−98514235、−98709361 、−99137419、−99291751 、−99298712、−99138418及びRHS4346−OB、Open Biosysterns社、USA)を使用して、Yesに対する有効siRNA(Qiagen社 SI00302218)、及びSrcファミリーキナーゼFynの種々のメンバーを標的とする第二のsiRNA(Qiagen社 SI00605451)を使用して実施した。種々のsiRNAs細胞での一時的な形質移入を、CD95L−T4(10ng/ml及び20ng/ml)での処理の72時間前に培養した後に、遊走を、2次元遊走アッセイでの処理の24時間後に分析した。そのノックダウンを、定量リアルタイムPCR及びFACSによって制御した。Yes−siRNAの適切でない効果を除くために、細胞を、Yes、Yes過剰発現プラスミド(p−CMV−Yes)、又は双方に対するsiRNAで形質移入し、そして遊走プレートに形質移入する48時間前に培養した。さらに48時間後に、細胞を、CD95L−T4(10ng/ml及び20ng/ml)で処理した。遊走を、2次元遊走アッセイで、処理の24時間後に測定した。
ルシフェラーゼレセプターベクターは、次の源から寄贈された:pTOPFLASH及びpFOPFLASH(Randall T.Moon、Howard Hughes Medical Institute of Washington;USA)並びに6個のNFκB−結合部位を有するNFκBプラスミド(Min Li−Weber、German Cancer Research Center Heidelberg、ドイツ)。形質移入実験を、製造業者の指示書によって、リポフェクタミン2000(登録商標)試薬(Invitrogen Life Technologies社)を使用して実施した。細胞を、形質移入する24時間前に、ウェル毎に5×104個の細胞の密度で24−ウェルプレートに播いた。ホタルルシフェラーゼ構成体を、CMVウミシイタケルシフェラーゼプラスミド(10ng)で同時形質移入して、ルシフェラーゼ値を標準化した。ルシフェラーゼ活性を、構造に依存する形質移入の24時間後に、Promega (Madison社、WI、USA)から市販されているキットを使用することによって、測定した。発光を、Ascient96ウェルマイクロプレート照度計を使用して定量化した。全ての形質移入を、少なくとも2つの独立した場合に4回実施し、かつ誤差バーは、標準誤差を示した。
ヒトCD95−リガンド−4(CD95L−T4)の遺伝子操作
TRAIL/DR5複合体及びTNF−α構造を、ヒトCD95L−レセプター結合ドメイン(CD95L−RBD)のための発現戦略の開発のためのモデルとして使用した。三量体ヒトCD95L−RBDの構造が、原則としてTNF−α−又はTRAIL−RBD−構造(PDBエントリー:それぞれ1TNF及び1D0G/1DU351,55,62)に類似していることを前提として、次の観測を、考慮に入れた:
1.TRAIL及びTNF−αからのRBDのN−及びC−末端のアミノ酸が、逆平行β−ストランドを形成する。
2.このβ−ストランドの末端アミノ酸が、TRAIL−RBD三量体の重心軸に近い分子の同一部位で隣接して配置される(図19を参照)。
これは、位置的理由のために、タンパク質ドメインの融合のため(例えば、安定化モチーフ又はタグの付加のため)の同一分子におけるN−及びC−末端使用が、互いに排反であることを意味する。理想的な安定化モチーフは、リガンド/レセプター相互作用部位との干渉のその危険度を最小化するために安定化モチーフの反対の部位でCD95L三量体のN−及びC−末端を有するCD95L三量体の重心軸の近くに配置された、小さく、十分に定義された三量体であるべきである。これらの基準を満たす適切な三量体タンパク質ドメインは、バクテリオファージT4のフィブリチンからのT4フォルドンモチーフである61,65。前記の考慮によって、T4フォルドンを、ヒトCD95L−RBD(CD95LのGlu142−Leu281)に対してC末端に融合した。CD95L−RBDとT4フォルドンとの間で、可撓性リンカー要素(GSSGSSGSSGS)を配置し、そしてヘキサヒスチジンタグ及びstrepタグ−II(HHHHH−HSAWSHPQFEK)を、C末端に付加した。このアフィニティタグを、可撓性リンカー要素(SGPSSSSS)によってT4フォルドンに結合した。分泌を基礎とする発現を可能にするために、ヒトIgκからのシグナルペプチドを、N末端(GIu142)に融合した。IgκリーダーとCD95LRBDとの融合によって形成された提案されたシグナルペプチド開裂部位は、ヒトCD95LのGlu142に対応するN末端に位置するグルタミンを有する最終生成物を放出することを予想された。図19Cで示されるCD95L−T4構成体のアミノ酸配列を、逆翻訳し、そしてそのコドン使用法を、哺乳動物細胞を基礎とする発現のために最適化した。遺伝子合成を、ENTELECHON GmbH (Regensburg、ドイツ)によって行った。最終発現カセットを、プラスミドの特有のHind−III−及びNot−I−部位を使用して、pCDNA4−HisMax主鎖中へサブクローンした。全ての前記に記載された特徴を含む図の要約を、TRAIL−T4−DR5−複合体(図19D)のために例示的に示す。
10%FBS、ペニシリン100unit/ml及びストレプトマイシン100μg/mlで補足したDMEM+GlutaMAX(GibCo社)中で成長させたHek 293T細胞を、CD95L−T4をコード化するプラスミドで一時的に形質移入した。組換えCD95L−T4を含む細胞培養物の上清を、形質移入後3日間培養し、そして300gで遠心分離し、続いて0.22μm滅菌フィルターを介して濾過することによって透明にした。アフィニティ精製のために、ストレプタクチンセファロース1ml(IBA GmbH、Goettingen、ドイツ)をカラムに詰め、そして緩衝液W(100mM Tris−HCl、150mM NaCl、pH8.0)15mlで平衡化した。その細胞培養物の上清を、流量4ml/分でカラムに適用した。続いて、そのカラムを、緩衝液Wで洗浄し、そして結合CD95L−T4を、7×1ml緩衝液E(100mM Tris−HCl、150mM NaCl、2.5mMデスチオビオチン、pH8.0)の添加によって段階的に溶出した。溶出画分のタンパク質含有量を、SDS−PAGE及び銀染色によって分析した(図19E)。続いて、画分E2〜E5を、限界濾過によって濃縮し、そしてさらにサイズ排除クロマトグラフィー(SEC)によって分析した。SECを、Aektaクロマトグラフィーシステム(GEHealthcare社)を使用してSuperdex 200カラム上で実施した。そのカラムを、リン酸緩衝食塩水で平衡し、そして濃縮させた、ストレプタクチンで精製したCD95L−T4(E2〜E5)を、SECカラム上に、流量0.5ml/分で置いた。CD95L−T4の溶出を、280nmでの吸光度で監視した。精製されたCD95L−T4の見かけの分子量を、ゲル濾過標準タンパク質(図19F及びG)(Bio−Rad GmbH、Muenchen、ドイツ)で、Superdex200カラムの検量線を基に決定した。
Jurkat A3永久ヒトT−細胞系(cat.番号CRL2570、ATCC)での細胞アッセイを使用して、CD95L−T4の活性を誘発するアポトーシスを定量した。Jurkat細胞を、10%FBS(Biochrom社)、ペニシリン100unit/ml及びストレプトマイシン100μg/ml(GibCo社)で補足したRPMI 1640−培地+GlutaMAX(GibCo社)を有するフラスコ中で成長した。アッセイ前に、100000個の細胞を、96ウェルマイクロタイタープレート中へウェル毎に播いた。CD95L−T4の種々の濃度のウェルへの添加(最終容量200μl)を、37℃で3時間のインキュベーションによって実施した。細胞を、溶解緩衝液(250mM HEPES、50mM MgCl2、10mM EGTA、5%Triton−X−100、100mM DTT、10mM AEBSF、pH7.5)20μlを加えることによって溶解し、そしてプレートを、30分間氷上に置いた。アポトーシスを、カスパーゼ−3及びカスパーゼ−7の増強させた活性によって平行する。従って、特異的カスパーゼ−3/−7基質Ac−DEVD−AFC(Biomol社)の開裂を使用して、アポトーシスの限界を決定した。実際に、カスパーゼ活性は、ヨウ化プロピジウム及びHoechst−33342で細胞を染色した後に、形態学的に決定されたアポトーシス細胞のパーセンテージと関係がある(データは示されていない)。カスパーゼ活性アッセイのために、細胞溶解物20μlを、黒い96ウェルマイクロタイタープレートに移した。50mM HEPES、1%スクロース、0.1%CHAPS、50μM Ac−DEVD−AFC及び25mM DTT、pH7.5を含有する緩衝液80μlの添加後に、そのプレートを、Tecan Infinite F500マイクロタイタープレートリーダーに移し、そして蛍光強度における増加を、監視した(励起波長400nm、放出波長505nm)(図19H)。
長期のヒト悪性神経膠腫細胞系において、最初に、CD95のトリガーに対するアポトーシスの誘発を検査した。ロイシンジッパー(LZ)−CD95Lでの処理は、種々の神経膠腫細胞系における変化しやすい効果を誘発した。LZ−CD95Lは、A172細胞中でアポトーシスを誘発せず、T98G細胞における高い投与量でのみアポトーシスを生じ、又はLN18細胞において低い投与量ですぐにアポトーシスを媒介した(図1a)。LZ−CD95Lで誘発される死の特異性を、CD95Lに対する抗体(Nok1:データは示されていない)によるアポトーシスの中和によって証明した。CD95で誘発されるアポトーシスに対するA172の耐性を、CD95表面発現の低いレベルに帰することができた(図1b)。しかしながら、LN18及びT98G細胞系は、同等に高レベルのCD95表面発現を示す間に、アポトーシスに対する種々の感受性を呈した(図1b)。
神経膠腫細胞の侵襲は、既に概説されているように、マトリックス金属タンパク質分解酵素(MMP)によって細胞外マトリックス構成成分の開裂を要求する。従って、MMP9及びMMP−2のmRNAレベルは、遊走の傾向があるT98GにおけるCD95トリガーに対して大いに増加したが、アポトーシス耐性LN18細胞においては増加しなかった(図2)。インテグリン結合キナーゼ(ILK)は、グリコーゲン合成キナーゼ−3β(GSK3β)の阻害によるMMP9発現を媒介することが最近報告されている9,10。GSK3βの、そのセリン−9(ホスホ−ser9)残基でのリン酸化による阻害は、LZ−CD95L又はα−Apo−1抗体での処理に対するT98G細胞において観察された(図3a及び図8)。GSK3βのリン酸化は、LN18細胞においても見出されるが、異なる反応速度を有する(図3a及び図8)。遊走の傾向があるT98G細胞は、CD95のトリガーに対して、より高い基礎的なホスホ−ser9−GSK3βレベル、及び徐々に増加する長期にわたるGSK3βのリン酸化を呈した(図3a及び図8)。アポトーシスの傾向があるLN18細胞は、CD95のトリガーに対して一時的な(5〜10分)GSK3βのリン酸化を示した(図3a)。構造的に活性のあるGSK3β突然変異体(GSK S9A)の過剰発現は、T98G細胞のCD95で誘発された遊走を阻害した(図3b)。GSK S9A発現T98G細胞及びそれらの野生の相当物は、CD95で誘発されたアポトーシスに対する感受性の及び成長速度の比較できるレベルを呈した(図3c〜d)。従って、T98G細胞における構造的に活性のあるGSK3βによる遊走の阻害は、種々の増殖速度に寄与されなかった。結果として、ILK阻害剤KP−SO−1での前処理は、T98G細胞のCD95で媒介された遊走を、基礎的な遊走を影響することなく阻害した(図3e)。ILKは、タンパク質キナーゼB(PKB/AKT)を活性化し、かつホスファチジルイノシトール−3−キナーゼ(PI3K)依存法におけるGSK3β活性を阻害する11。従って、細胞外レセプターキナーゼ(ERK)のリン酸化状態を変化することなく、LY294002によるPI3Kの阻害は、CD95で誘発されたAKT活性及びT98G細胞におけるGSK3βのser9リン酸化を阻害した(図3f)。
他の系列の実験において、患者の腫瘍由来の短時間の神経膠腫培養物を使用した。び漫性星状細胞腫(WHO III)からの細胞は、高いCD95表面発現を呈し、そして相対的にCD95で媒介されるアポトーシスに対して感受性があった(図4)。対照的に、乏突起細胞腫(WHO III)又は神経膠芽腫(WHO IV)から生じる細胞は、高いCD95表面発現にもかかわらず、CD95で媒介されるアポトーシスに対してより高い耐性がある(図4及び図9)。本明細書で試験されたどの原発GBM由来の培養物も、侵襲の傾向があるT98G細胞系における観察と比較して、高いCD95表面発現(n=18)、及び同様の又はより高いレベルのCD95で誘発されたアポトーシスに対する耐性(n=8)を呈した(図9)。CD95表面発現のレベルとCD95で媒介されたアポトーシスに対する耐性との双方は、培養物における継代数によって影響されなかった(データは示されていない)。さらに、CD95で誘発されるアポトーシスに対して相対的に(NCH125)又はより高い(NCH89及びNCH270)耐性がある、3つのGBM由来の培養物を試験した。NCH125及びNCH270におけるCD95のトリガーは、MMP9及びMMP−2の発現、及び続く遊走を増加した(図5a〜c)。NCH89細胞におけるCD95の刺激は、MMP9及びMMP−2の遊走又は発現を増加しなかった(図5b〜c)。従って、CD95投与に対する遊走反応は、厳密に言えば、アポトーシスに対する耐性の程度と関連しない。同様の点で、CD95及びCD95LのmRNAの発現は、より高い侵襲性の試験された原発GBM腫瘍の間で非常に異なった(図9)。
臨床の場において、手術を回避する侵襲細胞は、放射線療法及びアジュバント化学療法の標的である。γ−放射線照射は、CD95及びCD95Lの発現を増加し、そしてそれによってアポトーシスを誘発することが報告されている3。現在のデータを考慮すると、放射線照射で誘発されたCD95及びCD95Lが、神経膠腫細胞の侵襲性も増加することができるかどうかを対処することを必要とする。最初に、T98G細胞の放射線照射が、CD95及びCD95LのmRNAの発現を増加することを示した(図6a)。最も高いCD95及びCD95LのmRNAの発現は、3グレイ(Gy)の投与量で見出された。同様の投与量で、MMP−2のmRNAは、著しく誘発された(図6b)。MMP9のmRNAは、3及び10Gyで、しかし低い程度まで、著しく上方制御された(図6b)。最も重要なことに、MMP発現を、CD95Lの中和によって回復することができる、放射線照射させた細胞のより高い遊走率によって再現した(図6c)。原発GBM培養物は、放射線照射後に、より侵襲性の表現型も呈した(図6d及び図10)。放射線照射で誘発される遊走は、完全にCD95Lに依存した(図6d)。興味深いことに、CD95の直接的なトリガー後に、侵襲性の表現型を呈さないNCH89培養物においてさえ、10Gyの放射線照射は、CD95Lによる細胞の遊走数を増加した(図6d)。放射線照射は、本明細書で試験されたGBM由来の原発培養物を十中八九、CD95依存法において遊走を著しく誘発した(図6d及び図10)。CD95刺激に対して著しく遊走する傾向を呈することに失敗した培養物のみが、より低いCD95表面発現レベルを有した(NCH417、図10)。さらに、原腫瘍の手術及び放射線照射後に生じる、再発性腫瘍におけるそれらの分子の発現を試験した。腫瘍内でのCD95Lの発現レベルは、0〜4と評価された(図6e)。最初に検出された神経膠腫におけるレベルが決して0ではない(領域毎に1〜24個のCD95Lポジティブ細胞)場合に、放射線治療後のCD95L発現の劇的な増加を、研究された9つの再発性腫瘍のうち8つにおいて検出した(図6e)。CD95Lを、GFAPポジティブ腫瘍細胞において検出した(図6f)。CD95及びMMP9の付加発現を、連続した切片における同様の領域で検出した(図6f)。重要なことに、アポトーシス細胞は、CD95Lを発現する細胞の近くで観察されなかった(データは示されていない)。
Srcは、共免疫沈降実験によって示されたように、CD95をPI3K活性に関連させる(図14A〜C)。むしろ、T98G及びLN18細胞の、CD95L−T4での処理は、Src、及びCD95に対するPI3Kのp85サブユニットの漸増を誘発した。p85とCD95との会合は、CD95又はp85を免疫沈降することによって試験した。p85とCD95との会合の程度は、T98G細胞におけるCD95−T4の濃度と、反比例して関連した(図14B)。しかしながら、LN18細胞において、CD95に対するp85の漸増を、高濃度のCD95L−T4でのみ検出した(図14A)。CD95の免疫沈降は、低濃度のCD95L−T4での処理後5分でSrcファミリーメンバーの検出を可能にした(図14A及びB)。Srcの会合は、より高い濃度で減少した(図14A及びB)。従って、低濃度のCD95L−T4で、Srcとp85との双方は、T98G細胞においてCD95で検出できるレベルで会合するが、しかしLN18細胞においては、Srcのみが検出された。さらに、いくつかのSFK、例えばFyn、Lyn、pp60及びYesに対する抗体でのスクリーニング後に、CD95をPI3Kに連結するSrcファミリーメンバーとしてYesを識別した(図14C)。神経膠腫細胞の遊走におけるYesの役割を検証するために、ノックダウン実験を実施した。Yes siRNAで形質移入された細胞において、FACS及びqRT−PCRによって評価されたようなYesの発現を、Fyn発現、他のSrcファミリーメンバーが、影響されないままである場合に低減した(図14E)。FynではなくYesに対するsiRNAは、T98Gの、及びNCH125細胞のCD95L−T4で誘発された遊走を著しく破壊した(図14D)。この遊走の阻害を、T98G及びLN18細胞におけるYesの過剰発現によって低減した(図14F)。PI3K阻害剤LY290059のように、Yesに対するsiRNAは、AKTのCD95で誘発されたリン酸化も阻害した(図14G)。
PI3K経路リプレッサーPTEN(MMAC1、TEP1)の役割を試験した。アポトーシスの傾向があるLN18細胞が、完全なPTENを有する場合に、T98G細胞は、1つの対立遺伝子、及びPTENの第二の対立遺伝子の欠如における点突然変異(コドン42CTT〜CGT;グリシン〜グルタミン)、並びにある染色体10の全損失を媒介する57,59。しかしながら、PTEN過剰発現は、CD95で媒介されたアポトーシスに対してT98G又はNCH125細胞を感作しなかった(図15A)。
多形性神経膠芽腫に苦しむ患者におけるCD95Lの発現は、試験された全ての腫瘍においてCD95Lの三角様分布を示した(図7a、17A)。腫瘍の内部で、少量のCD95Lのみが発現した(図7a.1、17A.a)。腫瘍柔組織境界面で増加された発現(図7a.2)は、腫瘍に隣接する脳柔組織においてピークに達し(図7a.3、17A.b)、かつ神経膠腫との距離が増加するにつれて再び減少した(図7a.4、17A.c)。CD95Lを、神経膠腫細胞、ニューロン及びマクロファージにおいて検出した(データは示されていない)。腫瘍内でのCD95Lの付加発現を、腫瘍血管を囲む神経膠腫細胞において観察した。同様に、Srcファミリーキナーゼのリン酸化(pScr)及びYesの発現を、腫瘍の侵襲における役割を示唆する、全ての試験された試料における腫瘍宿主の感染で一貫して見出した(図17B)。充実性腫瘍領域内で、Yesの発現は、腫瘍試料間で、散乱腫瘍細胞において非常に高い発現からわずかな発現まで、大いに様々であった。このより高いYes発現領域において、Srcのリン酸化は、検出されず、むしろ制限された(図17B)。
Claims (2)
- CD95レセプター活性を中和する薬剤を含み、WHOグレードIIIの神経膠腫を有する個体を、CD95レセプターをCD95Lへの結合から妨げ、又はCD95レセプター/CD95L複合体を破壊することによって治療するための医薬であって、
該薬剤が
(a)阻害性抗−CD95L−抗体及び
(b)細胞外CD95ドメイン及びヒトIgG1−Fcドメインを含む阻害性融合タンパク質
からなる群から選択されるCD95Lに結合する化合物である、前記医薬。 - 前記の化合物が、CD95Lに結合する抗体である、請求項1に記載の医薬。
