Description
本発明はインビトロ診断の分野に属する。この分野において、本発明は、特に、定性および/または定量の目的のための内部対照核酸ならびに水性バッファ中の少なくとも1つの外部対照核酸を用いた、少なくとも1つの液体試料中に存在し得る少なくとも第1の標的核酸の単離、増幅および検出に関する。
分子診断の分野において、数多くの供給源からの核酸の増幅は、かなり重要である。核酸の増幅および検出の診断適用の例は、ヒト乳頭腫ウイルス(HPV)、西ナイルウイルス(WNV)などのウイルスの検出、あるいはヒト免疫不全ウイルス(HIV)、B型肝炎ウイルス(HBV)および/またはC型肝炎ウイルス(HCV)の存在に関する献血のルーチンのスクリーニングである。さらに、前記増幅技術は、ミコバクテリアなどの細菌標的または腫瘍学マーカーの分析に適している。

第1の局面において、本発明は、少なくとも1つの液体試料中に存在し得る標的核酸を単離し、増幅するための方法であって、該方法は、
a. 内部対照核酸を前記液体試料に添加すること
b. 固相支持体材料と前記液体試料を、標的核酸および内部対照核酸を含む核酸が固相支持体材料上に固定化されることを可能にするのに充分な期間かつ充分な条件下で、槽内で一緒に合わせること
c. 分離ステーションにおいて、該液体試料中に存在するその他の物質から該固相支持体材料を単離すること
d. 前記分離ステーション内の核酸を精製し、固相支持体材料を洗浄バッファで1回以上、洗浄すること
e. 精製された標的核酸および精製された内部対照核酸を少なくとも第1の槽内で、ならびに水性バッファ中の少なくとも1つの外部対照核酸を少なくとも第2の槽内で、1種類以上の増幅試薬と接触させること
f. 前記反応槽内で、前記精製された標的核酸、前記精製された内部対照核酸および前記水性バッファ中の少なくとも1つの外部対照核酸を、前記1種類以上の増幅試薬とともに、前記標的核酸および前記対照核酸の有無を示す増幅反応が起こるのに充分な期間かつ充分な条件下でインキュベートすること

工程e.〜f.での増幅のための条件は、前記精製された標的核酸、前記内部対照核酸および前記水性バッファ中の少なくとも1つの外部対照核酸について同一である、方法に関する。

・ 全プロセス対照(すなわち、核酸精製、増幅、および好ましくは検出)として作用する
・ 特定の試料の組成に関連する標的反応に対して障害を生じさせる効果のモニターとして作用する:
・ 水性バッファ中の外部対照核酸
・ アッセイ特異的試薬の完全性をモニタリングする
・ 標的特異的シグナルを検出するシステムの能力をモニタリングする
・ 全プロセス対照として作用する必要はなく、したがって、試料マトリックス内で製造される必要はない。
・ Tris: 好ましくは、1、2、3、4、5、6、7、8、または9mMの値から、12.5、15、17.5、20、30、40、50、60、70、80、90、または100mMの値までの濃度で。最も好ましくは、濃度は約10mMである。
・ EDTA: 好ましくは、0.01、0.02、0.03、0.04、0.05、0.06、0.07、0.08、または0.09mMの値から、0.125、0.15、0.175、0.2、0.3、0.4、0.5、0.6、0.7、0.8、0.9、または1mMの値までの濃度で。最も好ましくは、濃度は約0.1mMである。
・ アジ化ナトリウム: 好ましくは、0.005、0.01、0.02、0.03、または0.04%(w/v)の値から、0.06、0.07、0.08、0.09、0.1、または0.5%(w/v)の値までの濃度で。最も好ましくは、濃度は約0.05%(w/v)である。
・ ポリ(rA)RNA: 好ましくは、1、2.5、5、7.5、10、12.5、15、または17.5mg/lの値から、22.5、25、27.5、30、40、50、75、100、150、175、または200mg/lの値までの濃度で。最も好ましくは、濃度は約20mg/lである。
g. 前記標的核酸の量を測定する工程
を含む、上記の方法である。
i)反応の有効性をモニタリングする。
ii)力価計算において参照として機能し、そのために阻害の影響を補償し、調製および増幅手順を調節して、より正確な定量を可能にする。
そのため、標的-陰性試験においてのみ陽性でなければならない定性的試験における定性的内部対照核酸とは対照的に、定量的試験における定量対照核酸は、2つの機能:反応対照および反応較正を有する。そのため、該対照核酸は、標的-陰性反応および標的-陽性反応の両方において陽性かつ有効でなければならない。該対照核酸はさらに、高い核酸濃度の計算について信頼性の高い参照値を提供することに適している必要がある。したがって、内部定量対照核酸の濃度は比較的高くある必要がある。
・ 実験が失敗した場合のトラブル処理が容易なこと:例えば、内部対照核酸だけ、または外部対照核酸だけしか使用されない場合、増幅の失敗は、実施が有効でないという結論に至るにすぎず、なぜこうなったかの結論には至らない。対照的に、両方の型の対照が使用され、例えば、外部対照核酸のみが増幅される場合、これは、増幅試薬が外部対照核酸に対して作用するため、試料中にインヒビターが存在している可能性がある。この相乗効果は、陰性対照を含めた好ましい態様では、さらに増強され、それ以降、存在し得る所望でない、したがって夾雑性の核酸が前記陰性対照の陽性結果によって示され得る。
・ 外部対照核酸に対して水性バッファの使用が可能なこと:上記でさらに記載のように、分析する液体試料に内部対照核酸を含めることにより、外部対照核酸に対する液体試料のマトリックスと類似または同一の流体マトリックスを使用する必要が排除される。液体試料マトリックスは、内部対照核酸が全プロセス対照として機能を果たし、液体試料中に存在し得るインヒビターをモニタリングすることができるため、模倣される必要はない。
・ 上記のような複雑な流体マトリックス中に外部対照核酸を提供することを求める臨床的分子診断における規制要件が、本発明の方法のように外部対照核酸を水性バッファ中で使用する場合であっても満たされ得る(Guidance for Industry and FDA Staff - Assayed and Unassayed Quality Control Material、2007; Section IV、B、1. Matrix Effects)。
工程aにおいて、前記内部対照核酸を水性バッファ中の少なくとも1つの外部対照核酸に添加する工程

- 列をなして配列された頂部に開口部を有する多数の槽を含む頂部表面。槽は上部分、中央部分および底部分を含む。上部分はマルチウェルプレートの頂部表面につながっており、2つの長辺(longer side)と2つの短辺(shorter side)を含む。中央部分は、2つの長辺と2つの短辺を有する実質的に長方形の断面を有する;

オリゴマー化合物の別の特定の亜群に属し、モノマー単位として1つ以上のヌクレオチドと1つ以上の改変ヌクレオチドを有する。したがって、用語「改変オリゴヌクレオチド」(または「オリゴヌクレオチドアナログ」)は、オリゴヌクレオチドと実質的に類似した様式で機能を果たす構造物をいい、本発明との関連において互換的に使用され得る。合成の観点から、改変オリゴヌクレオチド(またはオリゴヌクレオチドアナログ)は、例えば、リン酸主鎖、リボース単位またはヌクレオチド塩基の適切な改変によってオリゴヌクレオチドの化学的改変によって作製され得る(Uhlmann and Peyman、Chemical Reviews 90(1990)543; Verma S.,and Eckstein F.,Annu. 定量的なHIVアッセイの感度を示すLOD値を、実施例2に記載の核酸パネルと類似の様式で測定した。結果は、HIVアッセイの感度は、両方の試料マトリックスについての95%信頼区間の重複で明らかなように、両方の試料マトリックスで同等であることを示す(表19および表20参照)。
NHPに基づくHCV試料およびICバッファに基づくHCV試料についての長期安定性試験により、RMCの安定性は、6ヶ月貯蔵後の安定なヒット率、CT値およびRFI(相対蛍光強度)値で明らかなように両方の対照マトリックスで同等であることが示された(表22参照)。
Claims (15)
- 少なくとも1つの液体試料中に存在し得る少なくとも1つの標的核酸を単離、増幅および検出するための方法であって、該方法は、
a.液体試料に内部対照核酸を添加する工程
b. 固相支持体材料と前記液体試料を、槽中で、標的核酸および内部対照核酸を含む核酸が固相支持体材料に固定化されるのに充分な時間および条件下で一緒に合わせる工程
c. 分離ステーション中で、液体試料中に存在する他の物質から固相支持体材料を分離する工程
d. 前記分離ステーション中で核酸を精製し、固相支持体材料を洗浄バッファで1回以上洗浄する工程
e. 精製された標的核酸および精製された内部対照核酸を少なくとも第一の槽内で、ならびに水性バッファ中の少なくとも1つの外部対照核酸を少なくとも第二の槽内で、1つ以上の増幅試薬と接触させる工程、ならびに
f. 前記反応槽内で、前記精製された標的核酸、前記精製された内部対照核酸および前記水性バッファ中の少なくとも1つの外部対照核酸を、前記1つ以上の増幅試薬と、前記標的核酸および前記対照核酸の存在または非存在を示す増幅反応が生じるのに充分な時間および条件下でインキュベートする工程
の自動化された工程を含み、工程e.〜f.における増幅の条件が、前記精製された標的核酸、前記内部対照核酸および前記水性バッファ中の少なくとも1つの外部対照核酸について同じであり、工程a.の後の工程に、前記水性バッファ中の少なくとも1つの外部対照核酸が供され、前記少なくとも1つの外部対照核酸が処理の前には前記水性バッファ中に含まれ、前記水性バッファはヒト血漿を含まない、方法。 - 該水性バッファが、6〜12のpHを有し、
・Tris: 1〜100mM
・EDTA: 0.01〜1mM
・アジ化ナトリウム: 0.005〜0.5%(w/v)、および
・ポリ(rA)RNA: 1〜200mg/l
を含む、請求項1記載の方法。 - 該水性バッファが8のpHを有し、
・Tris: 10mM
・EDTA: 0.1mM
・アジ化ナトリウム: 0.05%(w/v)、および
・ポリ(rA)RNA: 20mg/l
を含む、請求項2記載の方法。 - 工程a.の後の全ての工程に、陰性対照が供される、請求項1記載の方法。
- 少なくとも1つの液体試料が臨床試料である、請求項1〜4いずれか記載の方法。
- 以下:
g. 前記標的核酸の量を測定する工程
をさらに含む、請求項1〜5いずれか記載の方法。 - 少なくとも1つの液体試料中に存在し得る標的核酸を増幅するための分析システム(440)であって、該システムは、
・反応槽を含む増幅ステーション、ここで、前記反応槽は、増幅試薬、精製された標的核酸および精製された内部対照核酸、ならびに水性バッファ中の少なくとも1つの外部対照核酸を含む、
を含み、前記水性バッファ中の少なくとも1つの外部対照核酸が、前記精製された標的核酸とは異なる槽に含まれ、前記少なくとも1つの外部対照核酸が処理の前には前記水性バッファ中に含まれ、前記水性バッファはヒト血漿を含まない、分析システム。 - ・固相支持体材料を含む分離ステーション、ここで、該分離ステーションは、前記少なくとも1つの液体試料中に含まれる核酸を分離および精製するように構築および配列されている、
をさらに含む、請求項7記載の分析システム。 - 前記増幅ステーションに含まれる前記反応槽が、一体型配列で配列される、請求項7または8記載の分析システム。
- 前記一体型配列がマルチウェルプレートであり、前記水性バッファ中の少なくとも1つの外部対照核酸が前記精製された標的核酸とは異なるウェルに含まれる、請求項9記載の分析システム。
- 前記増幅ステーションが、前記精製された標的核酸および前記水性バッファ中の外部対照核酸とは異なる槽中に、陰性対照をさらに含む、請求項7〜10いずれか記載の分析システム。
- 前記陰性対照が、前記外部対照核酸の液体マトリックスとして機能するものと同じバッファである、請求項11記載の分析システム。
- ・分析物により引き起こされるシグナルを検出するための検出モジュール
・シーラー
・試薬および/または使い捨て品のための貯蔵モジュール、ならびに
・システム構成部分を制御するための制御ユニット
からなる群より選択される1つ以上の要素をさらに含む、請求項7〜12いずれか記載の分析システム。 - 1つより多くの外部対照核酸を使用する、請求項1〜6いずれか記載の方法。
