Description
1.細胞周期依存性のRhoGTP分解活性タンパク質(RhoGAP)を阻害しうる物質を有効成分として含有する新規抗腫瘍剤。
2.細胞周期依存性のRhoGAPを阻害しうる物質が、ARHGAP11Aを阻害しうる物質である、前項1に記載の新規抗腫瘍剤。
3.ARHGAP11Aを阻害しうる物質が、ARHGAP11Aをコードする遺伝子に対するアンチセンスオリゴヌクレオチド又はRNA干渉を誘導するオリゴヌクレオチドであり、或いはARHGAP11Aをコードする遺伝子の転写産物若しくは翻訳産物に結合し得る低分子化合物、天然高分子から選択されるいずれかである前項2に記載の新規抗腫瘍剤。
4.ARHGAP11Aを阻害しうる物質が、当該ARHGAP11Aをコードする遺伝子に対するアンチセンスオリゴヌクレオチドであって、オリゴヌクレオチドを構成する塩基が14塩基〜200塩基であり、当該塩基配列中に人工核酸を少なくとも1以上含むことを特徴とする前項3に記載の新規抗腫瘍剤。

21.前項1〜7のいずれかに記載の新規抗腫瘍剤を用いる悪性腫瘍の治療及び/又は予防方法。
さらに、生体検体中のARHGAP11Aの発現を例えばmRNAにより検査した結果、がんの進行度(病期ステージ分類)や、無再発生存期間との関係で有意な違いが認められた。このことから、生体検体中の細胞周期依存性のRhoGAPを定量することで、悪性腫瘍を検査することができる。
B)対照の系で同手法により測定された特定のRhoGAPをコードする遺伝子の発現量を測定する工程;
C)前記A)及びB)の工程により測定された特定のRhoGAPをコードする遺伝子の発現量と、比較する工程;
D)A)の工程により測定された遺伝子の発現量が、B)の工程により測定された遺伝子の発現量に比べて低い場合の候補物質を選別する工程。
まずはじめに、外科的に切除された大腸がん組織Geminin(細胞周期:S/G2/Mステージ)に分類される細胞を抗Geminin(S/G2/M)ポリクローナル抗体を用いて組織免疫染色法により染色したところ、非がん部位(A)、がん細胞浸潤先端部位(B)及びがん中心部位(C)について確認した。その結果、(B)の部位でGemininが最も強く染色され、S/G2/M期の細胞がん細胞浸潤先端部位に多く存在することが確認された(図1)。
がん組織の生体イメージング実験系を用いて、HCT116(ヒト大腸がん細胞株)が正常組織に浸潤していく様子をリアルタイムで解析した。S/G2/MとG1の細胞を分取してマイクロアレイ解析を行うことにより、細胞周期に依存する遺伝子を抽出し、その中で可動性に関連するARHGAP11AがS/G2/M期の細胞に高発現していることを初めて見出した。
E2Fファミリーは、細胞周期依存性の転写調節因子であることが知られている。ARHGAP11A(chr15: 32907691)の転写開始部位から-20-28b(chr15: 32907663-32907671)の部位にE2Fsが結合する配列があることに気づき、ARHGAP11Aの発現は、細胞周期依存性の転写因子であるE2Fによって制御されていることを、ルシフェラーゼレポーターアッセイ(図5A)及びクロマチン免疫沈降法(ChIP)により確認した。ARHGAP11Aの発現を阻害すると、がん細胞の遊走・浸潤能が低下することが確認され、治療標的として有望であることが示された(図5B)。
本参考例では、本発明の新規抗腫瘍剤が標的とするがん細胞を確認した。即ちARHGAP11Aが発現することで、転移や浸潤が誘導される可能性のあるがん細胞が、本発明のARHGAP11Aを阻害しうる物質を有効成分とする新規抗腫瘍剤が標的とするがん細胞であると考えられた。ヒト大腸がん細胞(HCT116)、ヒト肝臓がん細胞(HepG2)、リンパ管線維芽細胞(Human Lymphatic Fibroblasts :HLF)、ヒト膵臓腺がん細胞(PSN1、MiaPaca2、Panc1)について、ARHGAP11Aの発現を定量的リアルタイムPCRの方法で確認した。βアクチンを内部コントロールとし、βアクチンの発現量を1としたときの相対値を示した。その結果、ARHGAP11Aの発現を認めたヒト大腸がん細胞やヒト膵臓腺がん細胞に由来するがん細胞(HCT116、PSN1)においてARHGAP11Aの発現が認められた(図6)。よって、これらのがん細胞に対して、本発明の新規抗腫瘍剤が有効に作用しうると考えられる。
また、GeneNoteのGeneCardsID:GC15P032907を参照したところ、マイクロアレイによるARHGAP11A発現が、大腸がん、すい臓がん、前立腺がん、乳がん、頭頸部がん、黒色腫(メラノーマ)、卵巣がん、肺がん、脳がん、膵臓がん、肝細胞がん、腎細胞がん、皮膚がん等に由来する各がん細胞において認められており、これらのがん細胞に対して、本発明の新規抗腫瘍剤が有効に作用しうると考えられる。なお、GeneNoteのGeneCardsID:GC15P032907では、各がん細胞において発現が認められたことが掲載されているのみであり、ARHGAP11Aの発現によるがん細胞に及ぼす影響や、ARHGAP11Aを阻害する場合の効果については一切言及されていない。
本実施例では、細胞周期依存性RhoGAPのうちARHGAP11Aの機能を確認し、本発明の新規抗腫瘍剤の効果を確認した。以下の配列番号1又は2に示す塩基配列からなる2種類のshRNA(SH)を含むレンチウイルスベクターを作製し、HCT116(ヒト大腸がん細胞株)に各々を導入し、ARHGAP11A発現阻害株(SH#1、SH#2)を構築した。コントロールとして、以下の配列番号3に示す塩基配列からなるオリゴヌクレオチドを含むレンチウイルスベクターを作製し、同手法によりHCT116に導入した。それぞれのARHGAP11A発現阻害株(SH#1、SH#2)について、細胞増殖能及び細胞浸潤を確認した。

本実施例では、in vivoでのARHGAP11A阻害作用による抗腫瘍効果を確認した。
免疫不全マウス(NOD/SCID)に、レンチウイルスベクターを導入していない野生株のHCT116を1×106cell局注し、移植した。
その後、実施例1で作製した配列番号1又は2に示す塩基配列からなるshRNA(SH)を含むレンチウイルスベクター(SH#1、又はSH#2)をin vivo siRNA導入キットであるAteloGene(R)(Koken) を用いて局注した(siRNA投与群)。対照群として、(a)実施例1で作製した配列番号3からなる塩基配列からなるshRNAを含むレンチウイスルベクター(コントロールshRNA群)又は(b)遺伝子組換えしていないレンチウイルスベクター1をAteloGene(R)(Koken) を用いて上記の如く導入した(遺伝子導入試薬コントロール群)さらに、(c)レンチウイルスを局注していない系(コントロール群)についても対照とした。移植4週間経過後の腫瘍細胞の拡大を、腫瘍サイズの大きさで確認した。ARHGAP11Aに対するshRNAを腫瘍に投与すると、腫瘍の拡大を有意に抑制することができた(図8)。
大腸がん摘出手術を施行した患者について、手術標本よりがん組織部位をレーザーマイクロダイセクション法(laser microdissection)にて分取し、これよりmRNAを分離してARHGAP11Aの発現をマイクロアレイ法にて確認した。大腸がん及び正常大腸粘膜の手術切除標本について比較すると、がん部位でARHGAP11Aの発現が上昇しており、さらに大腸がんの病期分類によりステージが進行するにつれて、ARHGAP11Aの発現が上昇していることが確認された。例えば、がんが組織に浸潤し固有筋層に達するT1(UICCのTNM分類による)と比較してT2,T3,T4期で発現が上昇することが明らかとなった(図9)。
大腸がん摘出手術を施行した患者について、がん摘出手術(根治的手術)が可能であった症例に対して、高発現群と低発現群に分類し、無再発生存期間を解析した。手術標本よりがん組織部位をレーザーマイクロダイセクション法(laser microdissection)にて分取し、これよりmRNAを分離してARHGAP11Aの発現をマイクロアレイ法にて確認した。その結果、ARHGAP11Aの発現が平均より低い患者群(n=38)と高い患者群(n=26)での、無再発生存期間を確認したところ、ARHGAP11Aが低い患者群のほうが有意に生存率が高く、高発現群で予後が悪い事が確認された(図10)。これにより、ARHGAP11Aの発現量を確認することで、がんの予後予測が可能と考えられる。なお実施例3及び4の内容は、国立大学法人九州大学の倫理委員会の了承を得、同大学の生体防御研究所でなされた試験結果に基づくものである。
本実施例では、人工核酸であるBNAを含む35種類のアンチセンスオリゴヌクレオチド((以下、「アンチセンスBNA」という。)について、HCT116(ヒト大腸がん細胞株)でのARHGAP11Aに対する発現抑制効果を確認した。
以下に示す各配列において、N(L)は人工核酸BNA、5は5-mC(メチルシトシン)、5(L)はL-mC(メチル化人工核酸BNA)、T(L)は人工核酸チミジン、T(A)は人工核酸アデニンを示す。ここで使用するBNAは、株式会社ジーンデザインにより作製されたものである。

本実施例では、実施例5でARHGAP11Aの発現抑制効果を認めたアンチセンスBNAについて各種がん細胞でのARHGAP11A発現抑制効果を確認した。
アンチセンスBNAは、ARHGAP-625-BNA-16(#625:配列番号10)、ARHGAP-969-BNA-16(#969:配列番号13)、ARHGAP-1344-BNA-16(#1344:配列番号16)、ARHGAP-1447-BNA-16(#1447:配列番号17)、ARHGAP-1748-BNA-16(#1748:配列番号19)、ARHGAP-1931-BNA-16(#1931:配列番号21)又はARHGAP-2032-BNA-16(#2032:配列番号22)を用いた。コントロールは配列番号40に示すARHGAP-1931-BNA-16-cont2を用いた。また、アンチセンスBNAやコントロールとしてのオリゴヌクレオチドを加えないものを野生型(wt)とした。がん細胞は、DLD1(ヒト結腸腺がん細胞株)、HT29(ヒト結腸腺がん細胞株)、Panc1(ヒト膵臓腺がん細胞株)、PSN1(ヒト膵臓腺がん)の4種類の各種がん細胞について確認した。
各々のアンチセンスBNAを実施例5に記載の方法と同手法により、リポフェクタミン試薬を用いて各がん細胞に導入し、ARHGAP11Aの発現抑制効果をウエスタンブロッティングにより確認した。
その結果、各細胞においてアンチセンスBNAによるARHGAP11Aの発現抑制が認められ、特にARHGAP-1748-BNA-16(#1748:配列番号19)により強い発現抑制が認められた(図12)。
本実施例では、FITC標識アンチセンスBNA(ARHGAP-1748-BNA-16:#1748)について、リポフェクタミン試薬を用いてHCT116内への取り込み量を確認した。コントロールとしては、実施例1で作製した配列番号40からなる塩基配列からなるオリゴヌクレオチドを使用した。
その結果、ARHGAP-1748-BNA-16を導入した細胞では、ARHGAP11Aの発現が抑制されることが確認された(図13(A))。また、ARHGAP-1748-BNA-16の細胞内取込量はコントロールと同等であった(図13(B))。
本実施例では、アンチセンスBNA(ARHGAP-1748-BNA-16:#1748)によるin vivoでの抗腫瘍効果を確認した。免疫不全マウス(NOD/SCID)に実施例2と同手法により、野生株のHCT116を移植したのちに、FITC標識ARHGAP-1748-BNA-16を実施例2と同手法によりAteloGene(R)(Koken) を用いて、腫瘍周囲に局注した。ARHGAP-1748-BNA-16の投与により、腫瘍の拡大を有意に抑制することが確認された(図14)。さらに、ARHGAP-1748-BNA-16の投与後14日目に、マウスの各臓器(脳:brain、肝臓:liver、脾臓:spleen、肺:lung、腎臓:kidney、胃:intestine)及び腫瘍部位へのFITC標識ARHGAP-1748-BNA-16の取り込みを確認した。その結果、ARHGAP-1748-BNA-16は腫瘍部のみに到達し、その他の臓器には到達していないことが確認された(図15)。
本実施例では、アンチセンスBNA(ARHGAP-1748-BNA-16:#1748)によるin vivoでの悪性腫瘍細胞の転移抑制効果を確認した。ルシフェラーゼをコードする遺伝子を含むレンチウイルスを作製し、HT1080(ヒト線維肉腫細胞)に感染させルシフェラーゼ遺伝子を導入し、ルシフェラーゼ導入細胞(HT1080/Luc)を作製した。ヌードマウスにHT1080/Lucを5×106cell静脈注射した。HT1080/Luc注射後14日間経過後に、アドリアマイシン8mg/kg又はアンチセンスBNA10mg/kgを静脈注射した。さらに7日経過後、ルシフェリンを150mg/kg投与し、HT1080/Lucの肺転移を発光イメージングで確認した(図16)。その結果、アドリアマイシン投与群(n=3)よりアンチセンスBNA投与群(n=3)の方が腫瘍は小さかった(図17)。また、肺の重量を測定した結果、各マウスについて、肺の重量はアンチセンスBNA投与群のほうがアドリアマイシン投与群に比べて軽量であった。一方、体重については両群に差を認めなかった(図18)。このことから、アンチセンスBNA投与群のほうがアドリアマイシン投与群に比べて腫瘍細胞の転移が抑制されていることが示唆された。
さらに、生体検体中の細胞周期依存性のRhoGAP、例えばARHGAP11Aの発現を、例えばmRNAにより検査した結果、がんの進行度(病期ステージ分類)や、無再発生存期間との関係で有意な違いが認められた。このことから、生体検体中の細胞周期依存性のRhoGAPを定量することで、悪性腫瘍を検査することができる。さらに、生体検体中の細胞周期依存性のRhoGAPの発現、例えばARHGAP11AのmRNA量によれば、悪性腫瘍の進行度(病期ステージ分類)や、無再発生存期間との関係で有意な違いが認められる。これにより、生体検体中の細胞周期依存性のRhoGAP、例えばARHGAP11Aの発現を測定することで、がんの進行度及び/又は予後の予測が可能と考えられ、有用である。
Claims (7)
- ARHGAP11Aを阻害しうる物質を有効成分として含有する新規抗腫瘍剤であって、
当該ARHGAP11Aを阻害しうる物質が、当該ARHGAP11Aをコードする遺伝子に対するアンチセンスオリゴヌクレオチドであり、当該アンチセンスオリゴヌクレオチドを構成する塩基が14塩基〜200塩基であり、当該塩基配列中に人工核酸を少なくとも1以上含むことを特徴とする、新規抗腫瘍剤。 - アンチセンスオリゴヌクレオチドが、以下の1)〜13)のいずれかに示す塩基配列を含むオリゴヌクレオチドである、請求項1に記載の新規抗腫瘍剤:

15)ARHGAP11A #2 (TRCN0000047282):CCGGCCTTCTATTACACCTCAAGAACTCGAGTTCTTGAGGTGTAATAGAAGGTTTTTG(配列番号2)。 - 抗腫瘍剤が、悪性腫瘍の転移阻害作用及び/又は浸潤阻害作用を有することを特徴とする、請求項1〜3のいずれか1項に記載の新規抗腫瘍剤。
- 生体検体中のARHGAP11Aを定量し、ARHGAP11Aの発現が平均より高い場合に悪性腫瘍細胞を検出することを特徴とする、悪性腫瘍の検査方法。
- 悪性腫瘍が、大腸がんやすい臓がん、前立腺がん細胞、乳がん、頭頸部がん、黒色腫(メラノーマ)、卵巣がん、肺がん、脳がん、膵臓がん、肝細胞がん、腎細胞がん、皮膚がんから選択される一種又は複数種である、請求項5に記載の悪性腫瘍の検査方法。
- 悪性腫瘍の検査が、がんの進行度及び/又は予後を予測するための検査である、請求項5又は6に記載の悪性腫瘍の検査方法。
