Description
−リグノセルロース系材料または他のポリマー炭水化物バイオマスを含む材料の処理に関するエネルギーコストが、市販の酵素ならびに熱機械的および化学的処理の代わりに発酵油脂工程それ自身からの酵素を使用することによって低減される。
−発酵油脂産生発酵からの液体の残余物からの酵素タンパク質の除去は、発酵油脂産生から遊離される発酵液の生物学的酸素消費量を減少させる。
−細胞物質の除去の後、実質的に細胞を含まない発酵廃棄溶液は、再使用により適切である。
−発酵油脂工程の炭素バランスは、工程流において生物学的負荷を引き起こす代わりに、酵素タンパク質が減少し、または、発酵廃液から除去され、触媒的目的のために再使用され、または発酵油脂産生工程もしくは別の工業的もしくはとりわけバイオ技術的工程において栄養素として使用される場合に改善される。
−発酵溶液から回収される酵素は、微生物のポリマー化合物または微生物の構成成分の加水分解のために使用され得る。
−本発明の方法は、公知のおよび試験されたユニット操作ならびに工業的に応用可能な微生物種によって実行され得る。
−本発明は発酵油脂産生のためのリグノセルロース系材料の経済的および技術的有用性に寄与する。
「バイオ燃料(biofuel)」は、主にバイオマスまたはバイオ廃棄物由来の固体、液体、ガス状の燃料を意味し、有史以前の植物および動物の有機遺物由来である化石燃料とは異なる。
脂質を含む微生物は、例えばろ過または傾斜法などを用いることによってなど、任意の公知の方法によって、培養培地から分離され得る。代わりに、大容積容量の産業規模の工業用遠心分離器を用いた遠心分離が、所望の製品を分離するために使用されてもよい。
1.酵素を産生する方法であって、前記方法が、
−発酵油脂産生工程において、発酵油脂産生および酵素産生に適切な条件下で、発酵油脂および酵素の両方を産生することのできる微生物を培養し、そして前記微生物によって発酵油脂および酵素を産生する工程、
−酵素調製物としての、または酵素源としての使用のために、発酵油脂および酵素を含む微生物培養物を得、そして、少なくとも一部の(i)微生物培養物、(ii)微生物培養物から分離された上澄みおよび/または微生物細胞、(iii)上澄みからの富化されたタンパク質画分、および/または(iv)細胞からのタンパク質画分を回収する工程、および
−微生物細胞から発酵油脂を回収する工程
を含む方法。
糖の規定:
溶液の糖濃度を規定するために、溶液は、適切な希釈液とされ、HPLC分析に先立ち、0.2μmを通してろ過された。
サンプルの脂肪酸成分がSuutariら(1990)によって記載される方法で測定された。サンプル中の脂質は、初めに遊離脂肪酸へと加水分解され、そのナトリウム塩へとケン化され、そしてその後、メチルエステルへとメチル化された。脂肪酸メチルエステルはガスクロマトグラフィーを用いて分析された。
培養物ブロースのタンパク質濃度が、Whatman3ろ紙を通してブロースをろ過した後に分析された。タンパク質濃度は、Bio−Rad Protein Assay(ブラッドフォード法に基づく)によって分析された。
使用済み培養物ブロースは、加水分解試験前にWhatman3ろ紙を通してろ過された。
脂質産生微生物は、通常、例えばATCC、DSMなどの複数の公認の微生物培養物(株)保存機関によって一般に公開されている。本発明の様々な実施態様が、以下の実施例において、以下の微生物株を使用することにより議論される。アスペルギルス オリザエ(Aspergillus oryzae) DSM 1861、アスペルギルス オリザエ(Aspergillus oryzae) DSM 1864、アスペルギルス テレウス(Aspergillus terreus) DSM 1958。

Claims (15)
- 酵素を産生する方法であって、前記方法が、
発酵油脂産生工程において、発酵油脂産生および細胞外酵素産生に適切な条件下で、発酵油脂および細胞外酵素の両方を産生することのできる微生物を培養し、そして前記微生物によって発酵油脂および酵素を産生する工程、
酵素調製物としての、または酵素源としての使用のために、発酵油脂および細胞外酵素を含む微生物培養物を得、そして、上澄みからの富化されたタンパク質画分の少なくとも一部を回収する工程、および
微生物細胞から発酵油脂を回収する工程
を含む方法。 - 前記微生物培養物もしくはその一部または前記上澄みもしくはその一部が、工程に再循環される請求項1記載の方法。
- 前記工程から遊離されるプロセス水が、酵素回収なしの発酵油脂工程のプロセス水と比較して、少なくとも5%少ないタンパク質を含む請求項1または2記載の方法。
- 前記工程から遊離されるプロセス水が、酵素回収なしの発酵油脂工程のプロセス水と比較して、少なくとも10%少ないタンパク質を含む請求項1または2記載の方法。
- 前記微生物培養物の水相または上澄みが濃縮される請求項1〜4のいずれか1項に記載の方法。
- 酵素産生が、微生物培養物中に酵素誘導物質を添加することによって開始および/または維持されるか、または、酵素産生が構成的に起こる請求項1〜5のいずれか1項に記載の方法。
- 前記微生物が、炭素源としてポリマーバイオマスを含む培地上で培養される請求項1〜6のいずれか1項に記載の方法。
- 前記ポリマーバイオマスが、リグノセルロースまたはその画分である請求項7記載の方法。
- 前記酵素が、ヒドロラーゼ、酸化還元酵素、リアーゼ、イソメラーゼ、トランスフェラーゼもしくはリガーゼまたはそれらの任意の混合物を含む請求項1〜8のいずれか1項に記載の方法。
- 前記酵素がグリコシド結合を加水分解する請求項1〜9のいずれか1項に記載の方法。
- 前記微生物が、真菌または酵母である請求項1〜10のいずれか1項に記載の方法。
- 前記真菌が、アスペルギルス、トリコデルマ、リゾプスおよびフミコーラの群より選択される属に属している真菌である請求項11記載の方法。
- 前記真菌が、アスペルギルス属に属する真菌である請求項12記載の方法。
- 前記酵母が、クリプトコッカス属に属する酵母である請求項11記載の方法。
- 産生された前記酵素が同じまたは別の工業的工程において使用される請求項1〜14のいずれか1項に記載の方法。
