Description
本発明の第一の局面は、a)動的核分極(DNP)法により過分極活性基質の過分極前駆体を製造し、
b)該過分極前駆体を適切な担体と接触させてそれを過分極活性基質に変換する
工程を含む磁気共鳴研究法に用いるための過分極活性基質の製造方法に関する。
a)MRIシステム中に乗せ、上記方法に従って前駆体から得た過分極活性基質で処置した対象を、該活性基質の非ゼロ核スピン核中の核スピン遷移を励起するために選択した電磁波の周波数にかけて、
b)該励起した核からのMR信号を記録する
工程を含むMRIシステムの操作方法に関する。
a)MRIシステム中に乗せ、上記方法に従って前駆体から得た過分極活性基質で前処置した対象を、該活性基質の非ゼロ核スピン核中の核スピン遷移を励起するために選択した電磁波の周波数にかけて、
b)該励起した核からのMR信号を記録する
工程を含むMRIシステムの操作方法に関する。
本発明は、生物学的興味がある分子、特に、代謝経路、例えばトリカルボン酸(TCA)回路(クエン酸回路としても知られる)、解糖、β酸化、尿素サイクル、およびケト体代謝経路の部分である分子(代謝物)のex vivo過分極の代替法を提供する。
a)動的核分極(DNP)法により過分極活性基質の過分極前駆体を製造し、
b)該過分極前駆体を適切な担体と接触させて活性過分極基質に変換する
工程を含む、過分極活性基質の製造方法に関する。
1a)
[式中、Halはハロゲン原子(F、Cl、Br、I)である;Rは、C1-C6アルキルもしくはアルケン、シクロアルキルもしくはシクロアルケン、アリールアルキルもしくはヘテロアリールアルキルラジカル(これらは所望により分枝状、および/または1またはそれ以上のさらなる官能基、例えば、-OH、COOH、-NH2、-NHCH3、-N(CH3)2、SH、SCH3で置換されている)であるか、またはRは、式-CO-Zのカルボニル基である(ここで、Zは、水素、-OH、所望により置換されたC1-C6アルキル、C1-C6アルコキシ、アリール、またはベンジル基である)]。
1b)
[式中、Rはポイント1a)と同意義である;R’は、所望により置換されたアルキル、アリール、シクロアルキル、アリールラジカル(またはそのあらゆる組み合わせ)である。
これは、加水分解によってRCOOHおよびR’COOHの放出をもたらし、R’COOHはRCOOH(対称無水物)と同じであるか、または異なる生理学的に許容されるカルボン酸(混合無水物)である。この後者の場合、それは種々の活性基質、医薬活性化合物、または単に無毒性基質(ここで、本明細書に記載の「無毒性基質」は、LD50>0.1mmol/Kgの化合物を表す。)でありうる。

7)下記加水分解反応式に従ったケト酸特異的前駆体:
7a)下記一般式のケト酸のエノール形:
[式中、Rはポイント1a)と同意義である。n=0〜2である。]、および1またはそれ以上の生物適合性金属イオンを有するか、または好ましくはポリヒドロキシアミンまたはポリヒドロキシ酸で塩化されたその対応する塩(「エノレート」)。
7b)下記一般式のケト酸の金属キレート(後者はその脱プロトン化ケト酸または脱プロトン化エノール):

酪酸無水物:
R-COOH(ここで、Rは、C1-C10アルキル、C1-C10オキソ-アルキル、C1-C10ヒドロキシ-アルキル、C1-C10アミノ-アルキル、C1-C10アルケンを表す)、
R-(COOH)2(ここで、Rは、C1-C10アルキル、C1-C10オキソ-アルキル、C1-C10ヒドロキシ-アルキル、C1-C10アミノ-アルキル、C1-C10アルケンを表し、カルボキシル基はRの2つの異なる炭素原子と結合している)、
R-(COOH)3(ここで、Rは、C1-C10アルキル、C1-C10オキソ-アルキル、C1-C10ヒドロキシ-アルキル、C1-C10アミノ-アルキル、C1-C10アルケンを表し、カルボキシル基はRの3つの異なる炭素原子と結合している)。
過分極前駆体を、従来技術の方法に従って過分極させ、適切な媒質に溶解させる。そうして得られた過分極前駆体含有溶液を、先に記載のごとく酵素的加水分解のために最適化させた(緩衝液のタイプ、pH、および温度)。次に、該過分極前駆体を含む溶液を、所望の酵素活性を有する酵素溶液または固定化酵素と混合し、加水分解が完結した後、所望により該酵素を反応媒質から除去した。
(a)均一な磁場の存在下、低温で高T1前駆体と分極剤(DNP調製物)を接触させ、
(b)該分極剤に電子スピン遷移を引き起こすために適切に選択された周波数の超音波照射に該分極剤を暴露し、
(c)該DNP調製物を適切な担体(過分極溶液)に溶解し、所望により
(d)該過分極溶液から分極剤を除去する。

所望により置換された直鎖または分岐鎖C1-C6アルキル基(所望により硫黄または酸素原子が割り込んでいる)、所望により置換された脂肪族または芳香族C3-C8環状基またはヘテロ基から選ばれる。]
a)i)該基質の過分極前駆体を調製し、
ii)該過分極前駆体を適切な担体と接触させて該前駆体を過分極活性基質に変換することを含む方法により過分極活性基質を調製し、
b)該過分極活性基質を該試料に投与し、
c)該試料を該過分極活性基質中で核スピン遷移をもたらすように選択した周波数の放射線に暴露し、
d)該活性物質からMR信号を検出し、
e)所望により、該検出した信号から画像、生理学的データ、または代謝データを得る。

- 該加水分解が該前駆体の少なくとも75%の変換をもたらす請求項1記載の方法。
