Description
a) 機能的精細胞を一つ有する花粉を、中央細胞ではない胚嚢細胞と受精させる工程;
b) 中央細胞を増殖させて胚乳とする工程; および
c) 該胚乳から倍加半数体植物を再生する工程。
突然変異体花粉による受粉および胚乳培養
CDC2A 遺伝子は、植物の有糸分裂細胞周期において中心的な役割を果たす。CDC2A 領域における負の(negative)突然変異は、雄原細胞の有糸分裂がうまく起こらない花粉をもたらし、その結果、精細胞を一つだけ有する花粉がもたらされる (Nowack et al, Nature genetics 38: 63 (2006))。
Claims (13)
- 以下の工程を含む、倍加半数体植物を作出する方法:
a) 機能的精細胞を一つ有する花粉を、中央細胞ではない胚嚢細胞と受精させる工程;
b) 中央細胞を増殖させて胚乳とする工程; および
c) 該胚乳から倍加半数体植物を再生する工程。 - 機能的精細胞を一つ有する花粉が突然変異体花粉である、請求項1に記載の方法。
- 突然変異体花粉が、化学的突然変異、核酸を用いる形質転換、または照射によって得ることができるものである、請求項2に記載の方法。
- 化学的突然変異が、EMS、EES、BMS、PMS、MES または MMS からなる群より選択される化学物質による種子の処理によってもたらされるものである、請求項3に記載の方法。
- 照射が、UV照射、X線、ガンマ線または電離放射線である、請求項3に記載の方法。
- 核酸が、一過性に発現するものであるか又は安定に組み込まれるものである、請求項3に記載の方法。
- 形質転換が、アグロバクテリウム・ツメファシエンスまたは微粒子銃を用いて行われる、請求項3に記載の方法。
- 核酸が、第2の精細胞の形成を調節する遺伝子の発現を阻止する RNAi であるか又は該 RNAi をコードするものである、請求項3に記載の方法。
- 花粉が、第2の精細胞の形成の阻害または停止に関与する遺伝子が突然変異した花粉である、請求項3に記載の方法。
- 突然変異した遺伝子が、CDC2A もしくはサイクリン依存性キナーゼタンパク質(CDK)ファミリーの別のメンバーまたは KRP タンパク質ファミリーの遺伝子の負の突然変異体である、請求項9に記載の方法。
- 花粉粒の一つの精細胞が破壊されている、請求項1に記載の方法。
- 機能的精細胞を一つ有する花粉を生産する植物が eco-tilling によって得ることができるものである、請求項1に記載の方法。
- 機能的精細胞を一つ有する花粉が、胚嚢細胞または卵細胞を提供するアクセプター植物とは別の種に属するドナー植物のものである、請求項1〜12のいずれかに記載の方法。
