Abstract

Description
本发明涉及产生双单倍体植物的新方法。本发明进一步涉及由此获得的植物以及这些植物的后代、细胞、组织和种子。
自从Guha和Maheshwari在1964年(Nature 204:497)发现植物可再生自单倍体孢子,已完成了许多研究来获得关于其它物种的相似认识(参阅例如″In vitro Haploid production in Higher plants″Vol.1,2,3,4,5,Eds:S.Jain,S.Sopory and R.Veilleux(1996)KluwerAcademic Publishers)。
现在,为了加速产生基因纯系并且也为了评估和监测难于了解的特性诸如那些由多基因/等位基因编码的特性,使用双单倍体(doublehaploid,DH)的现代植物育种已成为非常有用的工具。
在繁殖农作物中产生和使用DHs对许多物种而言是众所周知的(参阅例如Thomas W.et al.(2003),In:Doubled Haploid productionin crop plants.A Manual.Eds.M.Maluszynski,K.Kasha,B.Forsterand I.Szarejko.Kluwer Academic Publishers,pp 337-349)。现在为止,DHs可获自雄性或雌性器官的孢子。来自雄性器官的孢子被称为小孢子,而体外培养物被称为小孢子培养物。长期以来在芸苔中已很好地建立了典型小孢子培养物(参阅例如Keller et al.(1984)In:K.Giles,S.Sen(eds.),Plant Cell Culture in Crop Improvement pp169-183.Plenum Pub.Corp.,New York)。来自雌性器官的孢子被称为大孢子,并且这些孢子的体外培养通常被称为雌核生殖(gynogenesis)。
对于例如糖甜菜以及黄瓜而言雌核生殖是一项成熟的技术(参阅例如Hosemans D.and Bossoutrot,Z.Pflanzenzuchtg.91:74-77(1983);EP 0 374 755)。
尽管有许多技术进步,雌核生殖和小孢子培养二者的成功仅限于一些顺应性较高的基因型。不仅存在对于产生DHs而言成功率低的植物种类诸如西瓜(Sari N.,Hort.Science 1994,vol.29(10),1189-1190)和南瓜(Kurtar E.S.et al.,Euphytica,Volume 127(3),2002,335-344(10),一些物种对DHs的诱导完全抵制。
这意味着DHs的巨大利益不能被用于每一种期望的植物种类。
因此本发明的目标是提供适于产生DHs的新方法。此目标通过包含以下步骤的用于产生双单倍体植物的方法得以实现:
a)用有一个功能性精细胞的花粉对非中央细胞的胚囊细胞授精;
b)使中央细胞增殖成胚乳;
c)从胚乳再生双单倍体植物。
本发明没有通过直接使用小孢子或大孢子获得DH植物。相反,所述DH植物再生自雌性配子体的中央细胞。
被子植物中有性生殖的特点是被称为双重受精的独特过程。这意味着来自花粉粒的两个精细胞进入雌配子体。然后第一个精细胞将使单倍体卵细胞受精,而第二个精细胞将使包含两个核的中央细胞受精。从受精卵细胞将发育出二倍体胚芽,而从中央细胞将增殖出三倍体胚乳。不使中央细胞受精以及/或者没有来自受精卵细胞的引发,中央细胞通常不会增殖成胚乳。唯一的例外是可产生自主胚乳发育的fis和fie突变体。
不过,在本发明中,其中精细胞之一缺失或失活的突变体花粉将只使卵细胞受精。在缺失第二个精细胞时中央细胞将保持未受精,并因此保持在二倍体阶段,它在本质上是双单倍体。卵细胞受精将引发未受精的中央细胞增殖成胚乳。由此可使用对于许多植物种类可广泛获取的适于从胚乳再生三倍体植物的技术(参阅T.D.Thomas&R.Chaturvedi,Plant Cell Tissue and Organ Culture 93:1)来从未受精中央细胞产生双单倍体植物。
因此本发明涉及利用突变体花粉使得只有卵细胞受精,这将引发未受精双单倍体中央细胞的发育。
在某一实施方案中,用EMS或类似EMS的化学药品诸如EES,BMS,PMS,MES,或MMS通过化学诱变产生具有唯一一个功能性精细胞的花粉。
在某一实施方案中,通过用UV光、X-射线、γ射线辐照或致电离辐射诱变产生只带一个功能性精细胞的花粉。
在某一实施方案中,可用eco-tilling针对在生殖细胞中细胞分裂被抑制的适当突变筛选诱变植物。
在某一实施方案中,可用eco-tilling筛选具有只含一个功能性精细胞之花粉的天然群体。
在某一实施方案中,抑制生殖细胞分裂的分子在花粉发育期间瞬时表达,例如通过存在于质粒上的核酸实现。抑制分子可以是核酸或蛋白质,其通过来自质粒的组成型表达而在花粉或小孢子中产生。
在某一实施方案中,所述抑制生殖细胞分裂的分子表达自稳定掺入花粉基因组的核酸。该细胞分裂抑制分子(可以是核酸或蛋白质)通过组成型表达在花粉或小孢子中产生。
依照本发明的某一实施方案,通过用核酸转化可获得只包含一个功能性精细胞的花粉。转化可以任何适当的方式进行,诸如通过根癌土壤杆菌的方法或通过粒子轰击(基因枪)的方法。
这些转化技术是众所周知的。用根癌土壤杆菌方法转化植物细胞是一项成熟技术,并且例如综述于文献De Ia Riva et al.,EJB Vol.1(3)(1998),and Bent,Plant Physiol.124:1540-1547(2000)中。
最近,人们发现植物的遗传转化不仅仅局限于土壤杆菌,其它的细菌也具有转化植物的能力(Broothaerts et al.,Nature 433,629-633(2005),收编于此作为参考)。通过获取非致瘤Ti质粒(disarmed Tiplasmid)和合适的二元载体使这些植物相关的共生细菌适合于进行基因转移。这样的转化体系也适用于本发明。
基因枪转化也是本领域技术人员众所周知的,并且用于此应用的工具多年来已是商品化了的(Ralph Bock,In:QiagenNews,Issue No.5,1997)。用于本发明的合适的技术也可参阅Barinova等的文章(J Exp Bot.53(371):1119-29(2002)),其中显示了在小孢子水平的DNA递送及其在金鱼草(Antirrhinum ma jus)中的瞬时表达,或者参阅Ramaiah等人关于紫花苜蓿(Medicago sativa.L.)的文章(Current Science73:674-682(1997))。在烟草中用基因枪轰击转化小孢子或花粉的方法可参阅Baubak Bajoghli,(Matrikel number:9802743,University ofVienna,Experimentelle Genetic III.Plant Biotechnology byAlisher Touraev,July 2001)。文献Van der Leede-Plegt,et al.,Transgenic Research 4(2):77-86(1995)描述了通过微粒轰击方法将DNA直接递送入烟草(粘毛烟草(Nicotiana glutinosa))的花粉内。这些和其它技术可用于本发明所用花粉或小孢子的转化。
在某一实施方案中,花粉和小孢子由于核酸存在而因此包含细胞分裂抑制分子。被引入的核酸可以是细胞分裂抑制分子本身,或者可以编码细胞分裂抑制分子。在后一种情况下,抑制分子是蛋白质或肽。在第一种情况中,抑制分子是核酸。所述核酸可抑制其自身或者它可阻断其它核酸被表达。
例如,核酸可以是或编码抗CDK蛋白家族或KRP家族的成员的RNAi。
本发明基于只有一个精细胞通过转化或天然突变体花粉被递送入胚囊或卵细胞。能抑制生殖细胞中的细胞分裂的基因构建体或分子是本身已知的,并且可用于本发明的新方法中。
在某一实施方案中,随后花粉粒被转移至来自同一物种或其中可发生所说花粉/小孢子细胞的花粉卸载的物种的植物雌蕊上。后者被称为异源授粉。异源授粉的一个例子是使用属于茄科的物种作为花粉供体而以西红柿作为受体。其它例子可参阅de Martinis,D et al.Planta 214(5):806-812(2002)和Dore C et al.,Plant Cell Reports 15:758-761(1996)。通常,适于异源授粉的物种属于同一植物科内。
本发明进一步涉及产生只含一个功能性精细胞的花粉的植物和来自所述植物或其后代的小孢子、卵细胞、种子、细胞或组织。
最后本发明涉及可通过本发明方法获得的双单倍体胚乳以及再生自所述双单倍体胚乳的植物、所述植物的后代、来自所述植物或其后代的种子、细胞、组织、小孢子和卵细胞。
在所有的实施方案中,所述花粉包含能成功使卵细胞受精的一个功能性精细胞或生殖细胞。
附图描述了本发明的方法。胚囊细胞1包含三个反足细胞2、一个双核中央细胞3和两侧是两个助细胞5和6的单倍体卵细胞4。当用具有两个功能性精细胞9和10的野生型花粉8进行受精7时,在胚囊细胞1中形成了受精的三倍体中央卵细胞11和受精的二倍体卵细胞12。通过发芽,由胚芽形成了二倍体植物13。用只包含一个功能性精细胞15的突变体花粉14进行受精16后,中央细胞17未发生受精。未受精的中央细胞17是双单倍体。受精后卵细胞18是二倍体。随后可从中央细胞17再生出双单倍体植物19。
本发明将在以下实施例中进行进一步说明。这些实施例仅供例证说明的目的,而不应解释为以任何方式限制了本发明。
实施例
用突变体花粉授粉和胚乳培养
CDC2A基因在植物的有丝分裂细胞周期中起核心作用。CDC2A区域中的阴性突变产生了其中生殖细胞的有丝分裂失败的花粉,这导致了花粉只含有一个精细胞(Nowack et al,Nature genetics 38:63(2006))。
用实施例1中获自西红柿植物的已转化突变体花粉对西红柿花朵进行去势和授粉。在授粉后,子房扩张形成类果实的形体。这种嫩的果实样结构在植物上保持2-4周。植物在受控气候条件下生长(22℃白天,18℃晚上)。
收获果实并将胚乳与其余的胚芽细胞分开。然后将胚乳细胞在通常用于胚乳再生的培养基中孵育(参阅T.D.Thomas&R.Chaturvedi,Plant Cell Tissue and Organ Culture 93:1(2008)及其中的参考文献)。用成功生成的植物的叶子通过流式细胞术方法确定植物的倍性(K.E.Arumuganathan&E.D.Earle Plant Molecular Biology Reporter9:229)。再生自胚乳的大部分小植株具有与二倍体西红柿植株相似的核DNA含量,意味着这些植物事实上是双单倍体并且没有发生中央细胞的受精。
Claims (16)
1.用于产生双单倍体植物的方法,包括以下步骤:
a)用含一个功能性精细胞的花粉使非中央细胞的胚囊细胞受精;
b)使中央细胞增殖成胚乳;和
c)从胚乳再生出双单倍体植物。
2.权利要求1的方法,其中含一个功能性精细胞的花粉是突变体花粉。
3.权利要求2的方法,其中所述突变体花粉可通过化学突变、用核酸转化或辐照获得。
4.权利要求3的方法,其中所述化学突变是通过用选自EMS,EES,BMS,PMS,MES,或MMS的化学试剂处理种子来实现的。
5.权利要求3的方法,其中所述辐照是UV辐照、X-射线、γ-射线或致电离辐射。
6.权利要求3的方法,其中所述核酸是瞬时表达的或稳定掺入的。
7.权利要求3的方法,其中所述转化通过根癌土壤杆菌(Agrobacterium tumefaciens)或粒子轰击方法进行。
8.权利要求3的方法,其中所述核酸是或者编码阻断调控第二个精细胞形成的基因表达的RNAi。
9.权利要求3的方法,其中所述花粉在涉及抑制或阻止第二个精细胞形成的基因中被突变。
10.权利要求9的方法,其中所述突变基因是CDC2A或周期蛋白依赖性激酶蛋白(CDK)家族的另一成员或KRP蛋白家族基因的阴性突变体。
11.权利要求1的方法,其中所述花粉粒的一个精细胞被破坏。
12.权利要求1的方法,其中产生含一个功能性精细胞的花粉的植物可通过eco-tilling获得。
13.权利要求1-12中任一项的方法,其中含一个功能性精细胞的花粉来自就提供胚囊细胞或卵细胞的受体植物而言属于另一物种的供体植物。
14.产生依照权利要求1至13的只含一个功能性精细胞的花粉的植物或来自所述植物的后代、种子、细胞或组织。
15.通过权利要求1至13中任一项的方法可获得的双单倍体植物或胚乳。
16.来自权利要求15的胚乳或植物的后代、种子、细胞或组织。
