- 請求項3に記載の宿主細胞を、変異DNAポリメラーゼをコードする核酸の発現のために適切な条件下で培養することを含んで成る、DNAポリメラーゼの生成方法。
- 請求項1に記載のDNAポリメラーゼと、プライマー、ポリヌクレオチド鋳型及び遊離ヌクレオチドとを、前記プライマーの拡張のために適切な条件下で接触し、それにより、拡張されたプライマーを生成することを含んで成る、プライマー拡張を行うための方法。
- 前記ポリヌクレオチド鋳型がRNA又はDNAである、請求項5に記載の方法。
- 前記遊離ヌクレオチドが、生来的でないヌクレオチドを含んで成り、当該生来的でないヌクレオチドがリボヌクレオチド又はラベルされたヌクレオチドを含んで成り、前記生来的でないヌクレオチドが、生来のdNTP、rNTPもしくはddNTPに比べてリボース糖の2'位置で修飾されているヌクレオチド、リボヌクレオチドの2'-PO4類似体、ホスホロチオエートdNTP([[α]-S]dNTP)、5'-[α]-ボラノ-dNTP、[α]- メチル-ホスホネートdNTP、又はリボヌクレオシド三リン酸(rNTP)である、請求項5又は6に記載の方法。
- 前記DNAポリメラーゼと、プライマー対、前記ポリヌクレオチド鋳型及び前記遊離ヌクレオチドとを、前記ポリヌクレオチドの増幅のために適切な条件下で接触することを含んで成る、請求項5〜7のいずれか1項に記載の方法。
- 請求項1に記載のDNAポリメラーゼを供給する少なくとも1つの容器、並びに
(a)プライマー拡張条件下で、予定されるポリヌクレオチド鋳型にハイブリダイズできるプライマーを供給する容器;
(b)遊離ヌクレオチドを供給する容器;及び
(c)プライマー拡張のために適切な緩衝液を供給する容器、
から成る群から選択された1又は複数の追加の容器、
を含んで成る、拡張されたプライマーを生成するためのキット。
