Description
The present invention relates to the field of ophthalmic agents. More particularly, the present invention relates to a technique that makes it possible to improve a decrease in pH over time caused by chlorobutanol in ophthalmic agents.
Ketotifen fumarate is one of the drugs known as an antihistamine and has histamine release inhibitory action and histamine receptor antagonistic action. It is also known to have leukotriene production / release inhibition and leukotriene antagonism. And since ketotifen fumarate improves allergic symptoms based on the above-mentioned mechanism of action, it is widely used clinically as eye drops and nasal drops for the purpose of prevention and improvement of allergic symptoms.
However, eye drops containing ketotifen fumarate are known to cause eye irritation when instilled. Accordingly, several methods for improving the occurrence of eye irritation upon instillation have been disclosed. For example, there are a method of blending a vasoconstrictor and chlorobutanol with ketotifen fumarate (see Patent Document 1), and a method of blending menthol and chlorobutanol with ketotifen fumarate (see Patent Document 2). However, eye drops containing chlorobutanol are known to have a pH that decreases with time due to decomposition of chlorobutanol, which causes problems such as instability and precipitation of the ingredients.
Conventionally, as a method for suppressing a decrease in pH of a preparation, a method of adding a phosphate buffer (sodium dihydrogen phosphate-disodium hydrogen phosphate), a borate buffer (boric acid-borax), or the like is known. However, the suppression effect is not sufficient (see Non-Patent Document 1).
This invention makes it a subject to suppress the pH fall by having mix | blended chlorobutanol in the eye drop which mix | blended ketotifen fumarate for the eye irritation | stimulation at the time of eye drop.
As a result of various studies to solve such problems, the present inventors have found that by adding boric acid and a sugar alcohol to an eye drop containing ketotifen fumarate and chlorobutanol, a decrease in pH of the preparation can be suppressed. The headline and the present invention were completed.

(Test example)
While stirring 5 mL of the eye drops obtained in Examples and Comparative Examples, 500 μL of 0.05N HCl was dropped, and stirring was stopped to check the pH value. Moreover, the pH of the preparation when the same preparation was stored at 65 ° C. for 7 days was measured.
The results are shown in Tables 1-1 to 1-5. The numerical value of the prescription was shown as “mg / 100 mL”.
In the eye drops of Examples 1 to 11 according to the present invention, the decrease in pH was suppressed as compared with the eye drops of the comparative example. The sugar alcohol used in the present invention is a component used as a tonicity agent, but the effect of stabilizing the pH was not obtained with other isotonic components, and the effect was specific to the sugar alcohol. In addition, the eye drop of the present invention had a higher effect of stabilizing the pH than a boric acid buffer (boric acid-borax) which is a general buffer.

(Reference example)
Four healthy subjects instilled the eye drops of the examples, and the irritation feeling was evaluated according to the following evaluation criteria. The eye irritation score was expressed as an average value of 4 people.
Evaluation criteria:
0: No stimulation
1: I feel some stimulation
2: Feeling the stimulus
3: Strong stimulation
4: Table 2 shows the results of feeling the stimulation quite strongly. The numerical value of the prescription was shown as “mg / 100 mL”.
Eye drops containing boric acid and sugar alcohol for the purpose of suppressing pH decrease did not affect the eye irritation suppression effect of chlorobutanol.
From the above, it was found that the eye drop of the present invention is useful as an eye drop because it has a stable pH and suppresses eye irritation as compared with the eye drop of the comparative example.
According to the present invention, it has become possible to provide an extremely useful eye drop that can suppress a decrease in pH caused by blending ketotifen fumarate and chlorobutanol.
