Description
この目的は、本発明による、請求項1に記載の多糖類マトリックス、請求項13および14に記載のその調製のためのプロセス、ならびに本発明による物質分離のための請求項18に記載の膜の使用によって達成される。
これらの2つの態様において、多糖類は、セルロースエステル、セルロースエーテル、セルロース水和物、アガロース、キチン、キトサン、デキストランまたはその組み合わせからなる群から選択される。任意に、前述の多糖類は、架橋していてもよい。
R1R2N−B−NR3R4 (I)
で表される対称の、または非対称のポリアミンであり、ここで、基R1R2Nのみ、または他に基NR3R4が、グラフトオンポリマーのエポキシ基との開環反応に進入するかを、架橋Bの長さおよび構造によって制御することが可能である。
最も好ましくは、第1の配位子はスルホン酸基であり、第2の配位子はスルファニル酸である。
A)少なくとも1つのカルボン酸基および/または、式中X=−O、−Sまたは−Nである少なくとも1つの酸性XH基を有する有機酸で処理した、多孔質の多糖類出発マトリックスを提供すること、ならびに
B)ステップA)において提供したマトリックスに、遷移金属またはランタニド化合物および官能基を有する少なくとも1種のエチレン系モノマー化合物を含む混合物をグラフトして、官能基を有するポリマーを生成し、それを、多糖類出発マトリックス上にグラフトすること、
を含む。
A)遷移金属またはランタニド化合物で処理した多孔質の多糖類出発マトリックスを提供すること、ならびに
B)ステップA)において提供したマトリックスに、少なくとも1つのカルボン酸基および/または、式中X=−O、−Sまたは−Nである少なくとも1つの酸性XH基を有する有機酸、ならびに官能基を有する少なくとも1種のエチレン系モノマー化合物を含む混合物をグラフトして、官能基を有するポリマーを生成し、それを多糖類出発マトリックス上にグラフトすること、
を含む。
本発明を、例によってより詳細に以下に説明するが、例は、いかなる方法によっても本発明を限定しない。
例におけるCA膜のすべての言及は、ポリエステル不織布で補強され、約3μmの孔径(”Capillary Flow Porometer 6.0” Coulter Porometer, CAPWIN Software System, Porous Materials Inc.を用いて測定したもの)および600〜700ml/(分*bar*cm2)の水流量を有する、1種の酢酸セルロース膜を指す。本発明の膜および比較膜についてのすべての流量値を、pH=7.3における20mMトリス(ヒドロキシメチル)アミノメタン/HCl(TRIS/HCl)緩衝液に対して、ml/(分*bar*cm2)において報告する。すべての静的結合能値を、mg/cm2において報告する。
他に明記しない限り、百分率は重量による。
240cm2の合計面積を有する8枚の円形のCA膜シートを、逆浸透(RO)水で湿潤させ、RO水で10分間洗浄し、次に、1000gの、水中、表1に列挙した有機酸0.1mmolの溶液に10分間浸漬した。その後、膜を、循環空気乾燥キャビネット中で、80℃にて15分間乾燥した。有機酸で処理した240cm2のCA膜を、22℃の温度に調整した反応容器のガス空間中に固定した。14.6gのグリシジルメタクリレートおよび0.45gのArlatone(登録商標)G(Atlas)を、最初に反応容器に投入し、30分間膜と共に窒素をガス供給した。
表1の比較膜を、例1に従って調製したが、グラフトを行う前に、表1からの有機酸の1種でのCA膜の処理を、行わなかった。
240cm2の合計面積を有する8枚の円形のCA膜シートを、RO水で湿潤させ、RO水で10分間洗浄し、次に、1000gの、水中のクエン酸0.1mM溶液に10分間浸漬した。その後、膜を、循環空気乾燥キャビネット中で、80℃にて15分間乾燥した。240cm2のCA膜を、22℃の温度に調整した反応容器のガス空間中に固定した。14.5gのグリシジルメタクリレート、0.07gのエチレングリコールジメタクリレートおよび0.45gのArlatone(登録商標)G(Atlas)を、最初に反応容器に投入し、30分間膜と共に窒素をガス供給した。
240cm2の合計面積を有する8枚の円形のCA膜シートを、RO水で湿潤させ、RO水で10分間洗浄し、1.5gの硫酸セリウム(IV)、5.0gの20%強度の硫酸および478gのRO水を含む500gの溶液に5分間浸漬した。その後、膜を、循環空気乾燥キャビネット中で、60℃にて15分間乾燥した。240cm2のCA膜を、22℃の温度に調整した反応容器のガス空間中に固定した。10.0gのグリシジルメタクリレート、0.05gのエチレングリコールジメタクリレート、0.05gのクエン酸および490gのRO水を含む溶液に、反応容器のガス空間中の膜と共に、30分間窒素をガス供給した。
240cm2の合計面積を有する8枚の円形のCA膜シートを、RO水で湿潤させ、RO水で10分間洗浄し、次に、1000gの、水中のクエン酸0.1mM溶液に10分間浸漬した。その後、膜を、循環空気乾燥キャビネット中で、80℃にて15分間乾燥した。240cm2のCA膜を、22℃の温度に調整した反応容器のガス空間中に固定した。13.5gのグリシジルメタクリレート、0.07gのエチレングリコールジメタクリレート、1.0gのベンジルメタクリレートおよび0.45gのArlatone(登録商標)G(Atlas)を、最初に反応容器に投入し、30分間膜と共に窒素をガス供給した。
例1、2および3からのグラフト膜を、トリメチルアミンの10%強度の水溶液で30℃にて35分間、および5%強度の硫酸水溶液で室温にて5分間連続的に処理し、次に流水で10分間洗浄した。
例1からの膜を、380gのNa2SO3、40.0gのNa2HPO4・H2O、100gのテトラブチルアンモニウム二硫酸塩および1200gの水を含む溶液中で、32%強度の水酸化ナトリウム水溶液でpH 8.0に調整し、80℃にて45分間処理し、次に流水で10分間、35gの1%強度の塩酸水溶液で5分間、30gの1M NaCl水溶液で各々の回において2×5分間、500gの5%強度の硫酸水溶液で5分間、および流水で10分間洗浄した。
18重量%のNa2SO3、1重量%のアニリン、5重量%のテトラブチルアンモニウム二硫酸塩、2重量%のNa2HPO3・2H2O(32%強度のNaOHでpH8に調整したもの)を含む312gの水溶液を、35℃に加熱した。例1からの膜を加え、反応溶液を、35℃にて16時間撹拌した。膜を、次に、流れるRO水で10分間、200gの5%強度の硫酸水溶液で15分間、200gの50mMのNa2SO4水溶液で2×10分間、流れるRO水で5分間、300gのエタノールで2×10分間および最後に流れるRO水で5分間洗浄した。
例2からの膜を、例6に記載したように反応させた。
配位子(スルファニル酸基)を導入するポリマー類似反応
RO水中のスルファニル酸(4−アミノベンゼンスルホン酸)の2%強度の溶液を、32%強度のNaOHでpH 11.3に調整した。例2からの膜を加え、反応溶液を65℃にて3時間撹拌した。膜を、流れるRO水で10分間、200gの5%強度の硫酸水溶液で15分間、200gの50mMのNa2SO4水溶液で2×10分間、流れるRO水で5分間、300gのエタノールで2×10分間および最後に流れるRO水で25分間洗浄した。
例6に記載したスルホン化溶液を、溶解した構成成分の出発濃度の2/3までRO水で希釈し、次に、希釈したスルホン化溶液中のスルファニル酸含量が2重量%または4重量%となるまで、スルファニル酸(Aldrich S5263)と混合した。スルファニル酸が溶解した後、pHを、32%強度のNaOHで11.3に調整した。例2からの膜を加え、反応溶液を65℃にて3時間撹拌した。膜を、流れるRO水で10分間、200gの5%強度の硫酸水溶液で15分間、200gの50mMのNa2SO4水溶液で2×10分間、流れるRO水で5分間、300gのエタノールで2×10分間および最後に流れるRO水で25分間洗浄した。
例1の膜を、10重量%のトリメチルアミンおよび2.5重量%のジアザビシクロオクタンを含む水溶液中で30℃にて35分間、および5%強度の硫酸中で室温にて5分間処理し、次に流水で10分間洗浄した。
例1からの膜を、N−ジエチル−N’−ジメチル−1,3−プロパンジアミンの20%強度の水溶液中で60℃にて1時間、および5%強度の硫酸中で室温にて5分間処理し、次に流水で10分間洗浄した。
例2からの膜を、N,N−ジメチルベンジルアミンの1%強度の水溶液中で60℃にて1時間、および5%強度の硫酸中で室温にて5分間連続的に処理し、次に流水で10分間洗浄した。
得られた膜を、以下に記載するように評価した。
1)流量決定
12.5cm2の活性膜面積を有する膜を、各々ケーシング中に取り付け、100mlの緩衝液(10mMのKPi緩衝液(KH2PO4溶液をK2HPO4溶液と混合することにより調製した、pH=7.0を有する0.01mol/リットルのリン酸カリウム緩衝液)および1MのNaCl水溶液を含む混合物であり、7.0のpHに調整したもの)を濾過する時間を測定した。配位子と反応した膜についての表2に示した流量値は、pH=7.3における対応するTRIS/HCl緩衝液に関連する。同一の緩衝液を、以下に記載する結合能決定に関して用いた。
各々の場合において17.6cm2の活性膜面積を有する膜を、35mlの20mMのTRIS/HCl(pH7.3)中で3×5分間、毎分約80回転(rpm)にて振盪した。その後、膜を、20mMのTRIS/HCl(pH=7.3)中の2mg/mlのウシ血清アルブミン(BSA)溶液(Kraeber, Ellerbeck, 注文番号041 80 10 900、ロット番号49 09 20 52)の溶液35ml中で、12〜18時間、20〜25℃にて約80rpmにて振盪した。その後、膜を、各々の場合において35mlの20mMのTRIS/HCl(pH=7.3)で2×15分間洗浄した。その後、膜試料を、20mMのTRIS/HClの溶液および1Mの塩化ナトリウム水溶液を含む20mlの混合物中で、pH=7.3にて振盪した。溶出したウシ血清アルブミンの量を、280nmにて光学濃度(OD)を測定することにより決定した。
各々の場合において17.6cm2の活性膜面積を有する膜を、35mlの10mMのKPi緩衝液(pH7.0)中で、3×5分間約80rpmにて振盪した。その後、膜を、10mMのKPi緩衝液(pH7.6)中の2mg/mlのリゾチーム(Sigma Aldrich Chemie, Taufkirchen, 注文番号L6876、ロット番号114K0626)の溶液35ml中で、12〜18時間、20〜25℃にて約80rpmにて振盪した。その後、膜を、2×15分間、各々の場合において35mlの10mMのKPi緩衝液(pH7.0)で洗浄した。その後、膜を、10mMのKPi緩衝液および1M塩化ナトリウム水溶液を含む20mlの混合物(pH=7.3)中で振盪した。溶出したリゾチームの量を、280nmにて光学濃度(OD)を測定することにより決定した。
比較例2による、ならびに例8および9による膜の静的結合能を、以下の方法によって決定した:各々の場合において17.6cm2の活性膜面積を有する膜を、35mlの20mMの緩衝酢酸溶液(pH=5.0、150mMまたは300mMのNaCl濃度に選択的に調整したもの)中で、3×5分間約80回転/分にて振盪した。その後、膜を、20mMの緩衝酢酸溶液(pH=5.0、150mMまたは300mMのNaCl濃度に選択的に調整したもの)中の1mg/mlのグロブリンG-5009 Sigma(Sigma Aldrich Chemie、由来:ウシ血液)の溶液35ml中で、12〜18時間、20〜25℃にて約80回転/分にて振盪した。
膜の3つの層を、膜ホルダー中に固定した。積層膜(membrane stack)は、15cm2の膜面積、5cm2の流量面積および750μmの層高(bed height)(積層膜の厚さ)を、膜ホルダーに有していた。膜ホルダー中の膜を、20mMのTRIS/HCl緩衝液(pH=7.3)で浸し、空気を置換し、次にGeneral Electric Health Careからの「Aekta(登録商標) Explorer 100」FPLCシステムに接続した。
1.pH=7.3における10mlの50mMの20mMのTRIS/HCl緩衝液との膜の平衡
2.UV検出器中の濃度が出発濃度の10%となるまで、pH=7.3における20mMのTRIS/HCl緩衝液中の1mg/mlのBSAで膜に負荷をかける。
** 10%DBC=10%破過における動的結合能
+ 膜の厚さを考慮した、cm2での膜面積を基準とした容量の、mlでの膜容積を基準とした容量への換算
Claims (17)
- 多孔質であり、非粒子状であり、対流的に透過性の多糖類マトリックスであって、表面上に、官能基を有する少なくとも1種のエチレン系モノマー化合物から由来するグラフトオンポリマーが固定され、
・多孔質であり、非粒子状であり、対流的に透過性の多糖類出発マトリックスに、
・少なくとも1つのカルボン酸基および/または式中X=−O、−Sもしくは−Nである少なくとも1つの酸性XH基を有する有機酸、ならびに
・遷移金属またはランタニド化合物
の存在下で、
・官能基を有する少なくとも1種のエチレン系モノマー化合物を
グラフトして、
官能基を有するポリマーを生成し、それを、多糖類出発マトリックス上にグラフトし、
グラフトの後に、多糖類出発マトリックス上にグラフトしたポリマーの官能基を、流体中に存在する吸着質と相互作用することが可能である少なくとも1種の配位子と反応させる
ことによって調製される、前記多孔質であり、非粒子状であり、対流的に透過性の多糖類マトリックス。 - 多糖類出発マトリックスが多糖類からなる、請求項1に記載の多糖類マトリックス。
- 多糖類出発マトリックスが多糖類コーティングを有する多孔質担体である、請求項1に記載の多糖類マトリックス。
- 多孔質担体が、ポリアミド、ポリ(エーテル)スルホン、ポリビニリデンジフルオリド、ポリアクリロニトリル、ポリ塩化ビニル、ポリプロピレン、ポリエチレン、ポリテトラフルオロエチレン、それらのコポリマーまたはその混合物からなる群から選択されたポリマーからなる、請求項3に記載の多糖類マトリックス。
- 多糖類が、セルロースエステル、セルロースエーテル、水和セルロース、アガロース、キチン、キトサン、デキストランまたはその組み合わせからなる群から選択される、請求項1〜4のいずれか一項に記載の多糖類マトリックス。
- 少なくとも1種のエチレン系モノマー化合物が、(メタ)アクリル酸誘導体であり、エチレン系モノマー化合物の官能基が、イオノゲン性基、疎水性基および/またはエポキシ基である、請求項1〜5のいずれか一項に記載の多糖類マトリックス。
- 遷移金属またはランタニド化合物が、セリウム、マンガン、鉄、ニッケル、コバルト、バナジウム化合物またはその組み合わせからなる群から選択される、請求項1〜6のいずれか一項に記載の多糖類マトリックス。
- 少なくとも1つのカルボン酸基および/または少なくとも1つの酸性XH基を有する有機酸が、クエン酸、リンゴ酸、酒石酸、フマル酸、マレイン酸、コハク酸、シュウ酸、マロン酸、アスコルビン酸、グルクロン酸、乳酸またはその組み合わせからなる群から選択され、ここでX=−Oである、請求項1〜7のいずれか一項に記載の多糖類マトリックス。
- 配位子が、カチオン性、アニオン性および/または疎水性基を含む、請求項8に記載の多糖類マトリックス。
- グラフトの後に、多糖類出発マトリックス上にグラフトしたポリマーの官能基を、2種の異なる配位子と反応させ、そのうち、第1の配位子がアニオン性基を含み、第2の配位子が疎水性基を含む、請求項9に記載の多糖類マトリックス。
- 第1の配位子が、スルホン酸基またはスルホン酸基に変換可能な基を含み、第2の配位子が、アニリンまたは4−アミノベンゼンスルホン酸である、請求項10に記載の多糖類マトリックス。
- 請求項1〜11のいずれか一項に記載の多糖類マトリックスを調製するプロセスであって、以下のステップ:
A)少なくとも1つのカルボン酸基および/または、式中X=−O、−Sまたは−Nである少なくとも1つの酸性XH基を有する有機酸で処理した、多孔質の多糖類出発マトリックスを提供すること、ならびに
B)ステップA)において提供したマトリックスに、遷移金属またはランタニド化合物および官能基を有する少なくとも1種のエチレン系モノマー化合物を含む混合物をグラフトして、官能基を有するポリマーを生成し、それを、多糖類出発マトリックス上にグラフトすること
C)ステップB)におけるグラフトの後に、多糖類出発マトリックス上にグラフトしたポリマーの官能基を、流体中に存在する吸着質と相互作用することが可能である少なくとも1種の配位子と反応させること
を含む、前記プロセス。 - 請求項1〜11のいずれか一項に記載の多糖類マトリックスを調製するプロセスであって、以下のステップ:
A)遷移金属またはランタニド化合物で処理した多孔質の多糖類出発マトリックスを提供すること、ならびに
B)ステップA)において提供したマトリックスに、少なくとも1つのカルボン酸基および/または、式中X=−O、−Sまたは−Nである少なくとも1つの酸性XH基を有する有機酸、ならびに官能基を有する少なくとも1種のエチレン系モノマー化合物を含む混合物をグラフトして、官能基を有するポリマーを生成し、それを多糖類出発マトリックス上にグラフトすること
C)ステップB)におけるグラフトの後に、多糖類出発マトリックス上にグラフトしたポリマーの官能基を、流体中に存在する吸着質と相互作用することが可能である少なくとも1種の配位子と反応させること
を含む、前記プロセス。 - 多糖類が、セルロースエステル、水和セルロース、セルロースエーテル、アガロース、キチン、キトサン、デキストランおよび/またはその組み合わせからなる群から選択される、請求項12または13に記載のプロセス。
- 多糖類がセルロースエステルであり、マトリックスをさらに、ステップB)の後、ステップC)の間またはステップC)の後に塩基性媒体で処理する、請求項14に記載のプロセス。
- 請求項1〜11のいずれか一項に記載の多糖類マトリックスの、物質分離のための使用。
- 請求項11に記載の多糖類マトリックスを、カチオン交換体として生体分子を精製するために用いる、請求項16に記載の多糖類マトリックスの使用。
