Description
本発明は、分子生物学的方法、および該方法を実施するための産物に関する。より具体的には、本発明は、モジュラータンパク質をコードする核酸をクローニングする方法であって、モジュールが所定の位置で連続的に付加されうる方法に関する。
組換えDNA技術および核酸をクローニングする分子生物学的方法は、当技術分野において公知である。そのような方法は、制限エンドヌクレアーゼ、リガーゼ、ヌクレオチド/ヌクレオシドキナーゼ、およびホスファターゼ、ポリメラーゼ、ならびに、所望の組換え核酸を生成するための他の分子生物学用ツールを用いて核酸を操作する方法を含む。いくつかの実験マニュアルが、分子生物学研究者のための参考書として書かれている。これらの参考マニュアルの例は、Sambrook, J., E. SEQ ID NO:1は、核酸モジュールの例である。本モジュールは、以下の5'‐3'構造を有する:5'‐所定の制限酵素部位‐関心対象のポリペプチドのオープンリーディングフレーム(ORF)‐所定の制限酵素部位‐スタッファー(stuffer)‐所定の制限酵素部位‐3'。所定の制限酵素部位およびスタッファーはまた、哺乳動物においてアミノ酸をコードし、かつ関心対象のポリペプチドとインフレームであり、そのために、モジュール全体が複合的なオープンリーディングフレームである。SEQ ID NO:1は、図11に示されている。
本発明は、モジュラーキメラタンパク質発現産物を作製する方法、および該方法において利用される組成物に関する。特に、本発明は、ポリペプチドモジュールをコードするポリヌクレオチドの連続的な方向性のクローニングに関する。各々のクローン化可能な要素は、所定の制限酵素部位が隣接するオープンリーディングフレームを含む。本方法は、組換えDNA技術のための開始材料として、クローン化可能な要素およびこれらの要素を含むベクターを用いる段階を含む。本発明の一つの利点は、各々の次のクローニング段階を設計および評価する必要なく、融合タンパク質の多くの変形物を、迅速かつ容易に作製することを可能にすることである。
多くのタンパク質が、天然においてモジュール式になっている。例えば、核受容体は、DNA結合ドメイン、リガンド結合ドメイン、二量体化ドメイン、および活性化ドメインを含むドメインを有する。ドメインの機能性が研究され得るため、ならびに/または、新たな研究および治療ツールが作製され得るために、受容体の間で機能ドメインを交換することによってキメラ受容体を作製することがしばしば望ましい。新規の細胞活性または治療活性を有する融合タンパク質を合成することもまた、望ましい。例えば、プロテインキナーゼD活性を調節するキメラポリリガンドが、様々な異種のタンパク質源から設計され、合成された(60/728,259参照)。
モジュラー融合タンパク質は、以下の方法を用いて、所定の制限酵素部位が隣接する関心対象のオープンリーディングフレーム(ORF)を各々含む二つの試薬から開始して作製される。関心対象のORFに隣接する制限酵素部位の例は、制限酵素NgoM IVおよびCla I;または、Xma IおよびCla Iによって認識される配列である(図1参照)。図1を参照すると、本方法の一つの態様は、二つの核酸開始試薬を利用する。一つの試薬は、5'末端にNgoM IV部位が隣接し、かつ、3'末端にXma I部位およびCla I部位が順に隣接する、関心対象のオープンリーディングフレーム(ORF 1)を含む環状DNAである。換言すれば、第一の試薬は、5'から3'へ以下の特性を有するDNAを含む:-- NgoM IV‐ORF 1‐Xma I‐スタッファー‐Cla I --。図1をさらに参照すると、第二の試薬は、5'末端にNgoM IV部位が隣接し、かつ、3'末端にXma I部位およびCla I部位が順に隣接する、関心対象のオープンリーディングフレーム(ORF 2)を同様に含む環状DNAである。スタッファーは、2種類の異なる制限酵素がDNAに結合して切断するのに十分な空間を可能にするヌクレオチドに相当する。「スタッファー」という用語は、「スペーサー」と同義であり、この二つの用語は本明細書において互換的に使用される。一般に、スタッファーまたはスペーサーは、隣接するオープンリーディングフレームとインフレームであるべきであり、そのため、コドンが3塩基の長さであるために3の倍数の長さを有するべきである。例示的なスタッファーは、GlyGlyGlyをコードするGGAGGCGGAであると考えられる。スタッファー/スペーサーは、他のアミノ酸組成を有してもよい。
コード領域モジュールを方向性に、かつ連続的に組み立てる別の方法は、環状DNAに加えて直鎖状DNAを使用する。例えば、上記で説明された連続的クローニング過程のように、コード領域に隣接する制限酵素部位は、制限酵素NgoM IVおよびCla I;またはXma IおよびCla Iによって認識される配列である。図3を参照すると、3' Xma I突出部および5' Cla I突出部を有する直鎖状DNAを生じるために、Xma IおよびCla Iで第一の環状DNA試薬を切断する。第一のDNA試薬は、アクセプターである。第二の試薬は、相補的オリゴヌクレオチドを合成し、かつアニールすることによって、または、PCR増幅に続いて突出部を生成する適切な制限酵素での消化によって生成した直鎖状二本鎖DNA断片である。第二の直鎖状DNAは、第一の直線化されたDNAアクセプターの付着末端と適合する3' Cla I突出部および5' NgoM IV突出部を有する。
本発明の方法はまた、開始試薬として二つの直鎖状DNAを用いて実施され得る。本実施例において、NgoM IVおよびXma I部位は、本明細書における他の実施例と比較して交換されている。図4を参照すると、二つの開始試薬は、1)3' NgoM IV突出部および5' Cla I突出部を有するベクターバックボーンであって、関心対象のオープンリーディングフレームであるORF ZがNgoM IV部位の上流に位置し、かつ、Xma I部位がORF Zとインフレームで、かつその上流に位置するベクターバックボーン、および2)5' Xma I突出部および3' Cla I突出部を含む二本鎖DNAオープンリーディングフレームであって、Xma I突出部が関心対象のオープンリーディングフレームであるORF Xのすぐ上流で、かつインフレームであり、かつ、ORF Xの下流にORF XとまたインフレームであるNgoM IV部位が続く二本鎖DNAオープンリーディングフレームである。
上記の実施例において議論された開始試薬は、試薬の収集物またはライブラリーの要素であってもよい。収集物の要素は、多数の特性を共有する。共有される最小限の特性は、関心対象のオープンリーディングフレームに隣接する同一の所定の制限酵素部位(例えば、NgoM IV、Xma I、Cla I)を含む。また、関心対象のオープンリーディングフレームは、内部にこれらの同一の制限酵素部位が欠如するように操作される。さらに、少なくとも第一の開始試薬(アクセプターDNA)は環状DNAであり、かつ、環状DNA全体において1箇所のみ、すなわち関心対象のオープンリーディングフレームに隣接する部位に、これらの同一の制限酵素部位を含むように操作される。換言すれば、アクセプターDNAベクターは、他の場所ではこれらの所定の制限酵素部位が欠如するように操作される。
ホモ多量体融合タンパク質の例は、例えばORF 8の二量体または多量体を含むポリペプチドである(図5参照)。関心対象のオープンリーディングフレームであるORF 8は、任意の長さまたは組成であり得る。例えば、ORF 8は、プロテインキナーゼDのペプチド阻害物質であり得る。本発明による環状DNAよりホモ多量体を構築する場合、二つの核酸開始試薬は同一である。しかしながら、二つの別々の制限酵素消化が行われる。実施例1における同様の例示的制限酵素部位の位置を利用して、Xma IおよびCla Iを用いた一つの消化が一つの容器中で実施され、NgoM IVおよびCla Iを用いたもう一つの消化がもう一つの容器中で実施される。直線化されたベクターDNAを遊離した挿入断片と混合し、アニールし、かつライゲーションすることによってORF 8の融合二量体が生じる。これらの段階は、所望の数のモジュールが達成されるまで、多くの回数繰り返され得る。また、図5に示されるように、二つの直鎖状DNA分子から開始してもよい。当業者は、本方法に利用可能である変形物を認識すると考えられる。
ヘテロ多量体融合タンパク質の例は、少なくとも二つの同一ではないモジュールを含むポリペプチドである。図6A〜6Cは、関心対象のエクソン由来ORFを含むモジュールから構築されたいくつかのヘテロ多量体融合タンパク質を示す。図7A〜7Cは、一つのモジュールが局在化シグナルを含む融合ポリペプチドの例を示す。図8A〜8Cは、一つのモジュールがエピトープタグを含むキメラポリペプチドの例を示す。図9A〜9Cは、異なる機能ドメインを含むモジュールから構築されたキメラタンパク質の例を示す。本明細書において説明される方法の任意が、ヘテロ多量体融合タンパク質を調製するために使用されてもよい。さらに、融合タンパク質はその後、哺乳動物細胞において発現されてもよい。図10は、本発明によって作製されたキメラタンパク質コード配列を含む例示的な発現カセットを示す。融合タンパク質は、研究ツールまたは治療学として有用である。
本発明の方法は、いくつかのヘテロ多量体の同時のコンビナトリアル合成に有用である。消化、精製、混合、およびライゲーションの段階は上記で説明した通りであるが、反応容器が以下のように組み合わされる。本実施例は、モジュールA、モジュールB、モジュールC、およびモジュールDとして示される異なるモジュールコード配列を各々含む4種類の環状DNA開始試薬から組み立てられる、あらゆる可能なヘテロ多量体融合タンパク質の合成である。例えば、結果として生じるヘテロ多量体は理想的には44の総数であると考えられ、モジュールA‐モジュールB‐モジュールC‐モジュールD;モジュールB‐モジュールC‐モジュールD‐モジュールA;モジュールC‐モジュールA‐モジュールD‐モジュールBなど、ならびに各モジュールのホモ多量体を含む。

上記の方法に加えて、本発明のモジュールライブラリーは、同様に多数のキメラを同時に生成すると考えられる動的コンビナトリアル合成において利用されてもよい。動的コンビナトリアル合成は、同一の反応混合物に多くの適合する突出部が存在し、かつ、互いにアニールおよびライゲーションすることが可能である場合に起こる。融合タンパク質がN末端からC末端へ(または逆に)構築される上記で説明された循環的な方法と比較して、動的方法は、後方および前方の配向において、ならびに特に順序無く連結される挿入断片をもたらす。融合タンパク質構築物の二つの末端は、選択されたモジュール(上記で使用されるような第一および第二の試薬と類似したもの)によって固定されているが、融合タンパク質の「中間」部分は、可変的である。
(a)少なくとも2個のアミノ酸を含む第一の群のアミノ酸をコードする第一の制限酵素部位;
(b)関心対象のポリペプチドのコード配列が、第一の制限酵素部位の少なくとも2個のアミノ酸のコード配列とインフレームである、関心対象のポリペプチドをコードする第一のオープンリーディングフレーム;
(c)第二の制限酵素部位の第二の群のアミノ酸のコード配列が、第一のオープンリーディングフレームの関心対象のポリペプチドのコード配列とインフレームであり、第二の制限酵素部位のポリヌクレオチド配列が、第一の制限酵素部位のポリヌクレオチド配列と同一ではない、少なくとも2個のアミノ酸を含む第二の群のアミノ酸をコードする第二の制限酵素部位;

a)追加的でありかつ再作製された第三の制限酵素部位を認識する制限酵素で上記由来のライゲーションされた発現ベクターを消化して、第三および第四の一本鎖突出部を有する直鎖状ベクターを作製する段階であって、該第三および第四の突出部が互いに適合しない、段階;
b)第二の挿入断片を提供する段階であって、該第二の挿入断片が、少なくとも一つの追加的なオープンリーディングフレームを含み、該少なくとも一つの追加的なオープンリーディングフレームが、第三および第四の突出部と適合する5'および3'突出部を含み、該少なくとも一つの追加的なオープンリーディングフレームの各々が、少なくとも一つの追加的な関心対象のポリペプチドをコードする、段階;
c)第二の挿入断片を直鎖状ベクター中にライゲーションして、新たなライゲーションされた発現ベクターを形成させる段階であって、少なくとも一つのオープンリーディングフレームの5'突出部のライゲーションが、直鎖状ベクターの第三の一本鎖突出部とアニールし、少なくとも一つのオープンリーディングフレームの3'突出部のライゲーションが、直鎖状ベクターの第四の一本鎖突出部とアニールし、該3'突出部のライゲーションが、第三の制限酵素部位を再作製する、段階。
a)第二および第三の制限酵素部位を認識する制限酵素で上記のベクターを消化して、第一および第二の一本鎖突出部を有する直鎖状ベクターを作製する段階であって、該第一および第二の突出部が互いに適合しない、段階;
b)第一の挿入断片を提供する段階であって、該第一の挿入断片が、少なくとも一つの追加的なオープンリーディングフレームを含み、該少なくとも一つの追加的なオープンリーディングフレームが、第一および第二の突出部と適合する5'および3'突出部を含み、該少なくとも一つの追加的なオープンリーディングフレームの各々が、少なくとも一つの追加的な関心対象のポリペプチドをコードする、段階;
c)第一の挿入断片を直鎖状ベクター中にライゲーションして、ライゲーションされた発現ベクターを形成させる段階であって、少なくとも一つのオープンリーディングフレームの5'突出部のライゲーションが、直鎖状ベクターの第一の一本鎖突出部とアニールし、少なくとも一つのオープンリーディングフレームの3'突出部のライゲーションが、直鎖状ベクターの第二の一本鎖突出部とアニールし、該3'突出部のライゲーションが、第三の制限酵素部位を再作製する、段階。
a)第一および第二のポリヌクレオチド試薬を提供する段階であって、各試薬が、関心対象のオープンリーディングフレームを含み、各関心対象のオープンリーディングフレームには、少なくとも3箇所の所定の制限エンドヌクレアーゼ部位が隣接しており、所定の部位のうちの2箇所が、適合する突出部を有する、段階、
b)所定の制限エンドヌクレアーゼ部位のうちの2箇所で切断する2種類の異なる制限エンドヌクレアーゼにより第一のポリヌクレオチド試薬を消化して、2個の異なる突出部を有するポリヌクレオチドを生成する段階、
c)所定の制限エンドヌクレアーゼ部位のうちの2箇所で切断する2種類の異なる制限エンドヌクレアーゼにより第二のポリヌクレオチド試薬を消化する段階であって、関心対象のオープンリーディングフレームを含むポリヌクレオチドが遊離し、該遊離したポリヌクレオチドが2個の異なる突出部を有し、該突出部のうちの1個が段階b)において消化されたポリヌクレオチドの突出部と適合する、段階、
d)段階b)において生成されたポリヌクレオチドと、段階c)の関心対象のオープンリーディングフレームを含む遊離したポリヌクレオチドとを一緒に混合し、アニールし、かつライゲーションする段階であって、オープンリーディングフレームの融合物を含む第三のポリヌクレオチドが生成され、関心対象のオープンリーディングフレーム間の接合部が、前記3箇所の所定の制限酵素部位で切断するいかなるエンドヌクレアーゼによる消化にももはや感受性ではなく、該関心対象のオープンリーディングフレームの融合物に隣接する配列が、前記3箇所の所定の制限酵素部位を含む、段階。
a)第一および第二のポリヌクレオチド試薬を提供する段階であって、各試薬が、関心対象のオープンリーディングフレームを含み、各関心対象のオープンリーディングフレームには、少なくとも3箇所の所定の制限エンドヌクレアーゼ部位が隣接しており、所定の部位のうちの2箇所が、適合する突出部を有する、段階、
b)所定の制限エンドヌクレアーゼ部位のうちの2箇所で切断する2種類の異なる制限エンドヌクレアーゼにより第一のポリヌクレオチド試薬を消化して、2個の適合しない突出部を有するポリヌクレオチドを生成する段階、
c)所定の制限エンドヌクレアーゼ部位のうちの2箇所で切断する2種類の異なる制限エンドヌクレアーゼにより第二のポリヌクレオチド試薬を消化する段階であって、制限エンドヌクレアーゼのうちの1種類が段階b)の制限エンドヌクレアーゼと同一であり、関心対象のオープンリーディングフレームを含むポリヌクレオチドが遊離し、該遊離したポリヌクレオチドが2個の適合しない突出部を有する、段階、
d)段階b)において生成されたポリヌクレオチドと、段階c)の関心対象のオープンリーディングフレームを含む遊離したポリヌクレオチドとを一緒に混合し、アニールし、かつライゲーションする段階であって、オープンリーディングフレームの融合物を含む第三のポリヌクレオチドが生成され、該関心対象のオープンリーディングフレームの融合物には、前記所定の制限酵素部位が隣接しており、該関心対象のオープンリーディングフレーム間の接合部には、前記3箇所の所定の制限エンドヌクレアーゼ部位が欠如している、段階。
Claims (18)
- モジュールオープンリーディングフレームを形成するように連続的に配置された下記の要素の各々を含むベクターであって、
(a)少なくとも2個のアミノ酸を含む第一の群のアミノ酸をコードする第一の制限酵素部位;
(b)関心対象のポリペプチドのコード配列が、第一の制限酵素部位の少なくとも2個のアミノ酸のコード配列とインフレームである、関心対象のポリペプチドをコードする第一のオープンリーディングフレーム;
(c)第二の制限酵素部位の第二の群のアミノ酸のコード配列が、第一のオープンリーディングフレームの関心対象のポリペプチドのコード配列とインフレームであり、第二の制限酵素部位のポリヌクレオチド配列が、第一の制限酵素部位のポリヌクレオチド配列と同一ではない、少なくとも2個のアミノ酸を含む第二の群のアミノ酸をコードする第二の制限酵素部位;
(d)スペーサー配列のコード配列が、第二の制限酵素部位のポリヌクレオチド配列とインフレームである、少なくとも3個のスペーサーアミノ酸をコードするスペーサーポリヌクレオチド配列;および
(e)第三の制限酵素部位の第三の群のアミノ酸のコード配列が、少なくとも3個のスペーサーアミノ酸のコード配列とインフレームであり、第三の制限酵素部位のポリヌクレオチド配列が、第一の制限酵素部位のポリヌクレオチド配列と同一ではなく、第三の制限酵素部位が、第二の制限酵素部位のポリヌクレオチド配列と同一ではない、少なくとも2個のアミノ酸を含む第三の群のアミノ酸をコードする第三の制限酵素部位
を含み、
要素(a)〜(e)が、モジュールオープンリーディングフレームを形成するように互いにインフレームで連結されており、
前記第一の制限酵素部位が、少なくとも2個のヌクレオチドを含む第一の一本鎖突出部をもたらす制限酵素に感受性であり、
第二の制限酵素部位が、少なくとも2個のヌクレオチドを含む第二の一本鎖突出部をもたらす制限酵素に感受性であり、
前記第一および第二の突出部が互いに相補的であり、
前記第一および第二の突出部がアニーリングすると前記第一の制限酵素部位および第二の制限酵素部位が破壊される、
ベクター。 - 関心対象のポリペプチドが、全長タンパク質もしくはポリペプチド、タンパク質機能ドメイン、タンパク質構造ドメイン、タンパク質酵素ドメイン、タンパク質阻害ドメイン、タンパク質結合ドメイン、タンパク質局在化ドメイン、またはエピトープである、請求項1記載のベクター。
- 第一の制限酵素部位が、NgoM IV制限酵素によって認識されるポリヌクレオチド配列を含む、請求項1〜2のいずれか一項記載のベクター。
- 第二の制限酵素部位が、Xma I制限酵素によって認識されるポリヌクレオチド配列を含む、請求項1〜3のいずれか一項記載のベクター。
- 第三の制限酵素部位が、Cla I制限酵素によって認識されるポリヌクレオチド配列を含む、請求項1〜4のいずれか一項記載のベクター。
- スペーサーポリヌクレオチド配列が、3個のグリシンアミノ酸をコードする、請求項1〜5のいずれか一項記載のベクター。
- 少なくとも一つの追加的なオープンリーディングフレームが、第一のオープンリーディングフレームと第二の制限酵素部位との間に挿入されており、該少なくとも一つの追加的なオープンリーディングフレームの各々が、該第一のオープンリーディングフレームおよび第二の制限酵素群の双方とインフレームであり、該追加的なオープンリーディングフレームの各々が、少なくとも一つの追加的な関心対象のポリペプチドをコードする、請求項1〜6のいずれか一項記載のベクター。
- 第一または第二の制限酵素部位のいずれかにおいて生じる制限酵素部位が、第一と第二の制限酵素群の間のいかなるオープンリーディングフレーム中にも存在しない、請求項7記載のベクター。
- 第一のオープンリーディングフレームと、少なくとも一つの追加的なオープンリーディングフレームとが互いにインフレームでライゲーションされる、以下の段階を含む融合ポリペプチドの作製方法:
a)第二および第三の制限酵素部位を認識する制限酵素で請求項4〜8のいずれか一項記載のベクターを消化して、第一および第二の一本鎖突出部を有する直鎖状ベクターを作製する段階であって、該第一および第二の突出部が互いに適合しない、段階;
b)第一の挿入断片を提供する段階であって、該第一の挿入断片が、少なくとも一つの追加的なオープンリーディングフレームを含み、該少なくとも一つの追加的なオープンリーディングフレームが、第一および第二の突出部と適合する5’および3’突出部を含み、該少なくとも一つの追加的なオープンリーディングフレームの各々が、少なくとも一つの追加的な関心対象のポリペプチドをコードする、段階;
c)第一の挿入断片を直鎖状ベクター中にライゲーションして、ライゲーションされた発現ベクターを形成させる段階であって、少なくとも一つのオープンリーディングフレームの5’突出部のライゲーションが、直鎖状ベクターの第一の一本鎖突出部とアニールし、少なくとも一つのオープンリーディングフレームの3’突出部のライゲーションが、直鎖状ベクターの第二の一本鎖突出部とアニールし、該3’突出部のライゲーションが、第三の制限酵素部位を再作製する、段階。 - 以下の段階を含む、融合タンパク質DNA構築物の作製方法:
a)第一および第二のポリヌクレオチド試薬を提供する段階であって、各試薬が、関心対象のオープンリーディングフレームを含み、各関心対象のオープンリーディングフレームには、少なくとも3箇所の所定の制限エンドヌクレアーゼ部位が隣接しており、所定の部位のうちの2箇所が、適合する突出部を有する、段階、
b)所定の制限エンドヌクレアーゼ部位のうちの2箇所で切断する2種類の異なる制限エンドヌクレアーゼにより第一のポリヌクレオチド試薬を消化して、2個の異なる突出部を有するポリヌクレオチドを生成する段階、
c)所定の制限エンドヌクレアーゼ部位のうちの2箇所で切断する2種類の異なる制限エンドヌクレアーゼにより第二のポリヌクレオチド試薬を消化する段階であって、関心対象のオープンリーディングフレームを含むポリヌクレオチドが遊離し、該遊離したポリヌクレオチドが2個の異なる突出部を有し、該突出部のうちの1個が段階b)において消化されたポリヌクレオチドの突出部と適合する、段階、
d)段階b)において生成されたポリヌクレオチドと、段階c)の関心対象のオープンリーディングフレームを含む遊離したポリヌクレオチドとを一緒に混合し、アニールし、かつライゲーションする段階であって、オープンリーディングフレームの融合物を含む第三のポリヌクレオチドが生成され、関心対象のオープンリーディングフレーム間の接合部が、前記3箇所の所定の制限酵素部位で切断するいかなるエンドヌクレアーゼによる消化にももはや感受性ではなく、該関心対象のオープンリーディングフレームの融合物に隣接する配列が、前記3箇所の所定の制限酵素部位を含む、段階。 - 以下の段階を含む、融合タンパク質DNA構築物の作製方法:
a)第一および第二のポリヌクレオチド試薬を提供する段階であって、各試薬が、関心対象のオープンリーディングフレームを含み、各関心対象のオープンリーディングフレームには、少なくとも3箇所の所定の制限エンドヌクレアーゼ部位が隣接しており、所定の部位のうちの2箇所が、適合する突出部を有する、段階、
b)所定の制限エンドヌクレアーゼ部位のうちの2箇所で切断する2種類の異なる制限エンドヌクレアーゼにより第一のポリヌクレオチド試薬を消化して、2個の適合しない突出部を有するポリヌクレオチドを生成する段階、
c)所定の制限エンドヌクレアーゼ部位のうちの2箇所で切断する2種類の異なる制限エンドヌクレアーゼにより第二のポリヌクレオチド試薬を消化する段階であって、制限エンドヌクレアーゼのうちの1種類が段階b)の制限エンドヌクレアーゼと同一であり、関心対象のオープンリーディングフレームを含むポリヌクレオチドが遊離し、該遊離したポリヌクレオチドが、2個の適合しない突出部を有する、段階、
d)段階b)において生成されたポリヌクレオチドと、段階c)の関心対象のオープンリーディングフレームを含む遊離したポリヌクレオチドとを一緒に混合し、アニールし、かつライゲーションする段階であって、オープンリーディングフレームの融合物を含む第三のポリヌクレオチドが生成され、該関心対象のオープンリーディングフレームの融合物には、前記所定の制限酵素部位が隣接しており、該関心対象のオープンリーディングフレーム間の接合部には、前記3箇所の所定の制限エンドヌクレアーゼ部位が欠如している、段階。 - 所定の制限酵素部位が、NgoM IV、Xma I、およびCla Iによって切断可能な配列である、請求項10または11記載の方法。
- 単離されたポリヌクレオチドのライブラリーであって、各ポリヌクレオチドが、少なくとも3箇所の所定の制限エンドヌクレアーゼ部位が隣接する関心対象のオープンリーディングフレームを含み、3箇所の所定の制限酵素部位のうちの2箇所が互いに適合せず、3箇所の所定の制限酵素部位のうちの2箇所が互いに適合し、各関心対象のオープンリーディングフレームには、該所定の制限酵素部位が欠如している、ライブラリー。
- 関心対象のオープンリーディングフレームを含む単離されたポリヌクレオチドであって、該関心対象のオープンリーディングフレームの一方の末端には、NgoM IVによって切断可能な配列が隣接しており、該関心対象のオープンリーディングフレームの他方の末端には、Xma IおよびCla Iによって切断可能な配列が隣接している、単離されたポリヌクレオチド。
- 単離されたポリヌクレオチドのライブラリーであって、各ポリヌクレオチドが、関心対象のオープンリーディングフレームを含み、各関心対象のオープンリーディングフレームの一方の末端には、NgoM IVによって切断可能な配列が隣接しており、各関心対象のオープンリーディングフレームの他方の末端には、Xma IおよびCla Iによって切断可能な配列が隣接しており、該関心対象のオープンリーディングフレームには、NgoM IV、Xma I、およびCla Iによって切断可能な配列が欠如している、ライブラリー。
- 請求項15記載のライブラリーの単離されたポリヌクレオチドを含む、ベクター。
- 請求項15記載のライブラリーを含む、組換え宿主細胞。
- 前記請求項1〜8のいずれか1項に記載のベクターを含む宿主細胞。
