Description
本出願は、2002年8月4日に提出された米国仮出願第60/401,191号からの優先権を請求する。本出願は、参照としてその全体が本明細書に組み入れられている。
本発明での使用のための適切な開始物質は、これに限定されるわけではないが、アミド基を含有するC13側鎖を備えたいずれかのタキサン分子を含む。N−アシル化フェニルイソセリン側鎖をC13位置に含有するタキサンは、本発明の開始物質として作用する。加えて、開始物質は、これらのタキサンの2つ以上の混合物でもよい。これらの化合物は、変化する置換パターンを有することがある;たとえばC10位置はアセテートまたはヒドロキシルを含有し、C7位置はヒドロキシルまたはキシロシル基を含有する。これらの化合物の例は図16および図17に見出され、さらなる例は、J.Natural Products,1999,62,1448−1472,およびPhytochemistry,1999,50,1267−1304に見出される。各刊行物は、その全体が参照として本明細書に組み入れられている。潜在的な開始物質は、これらの参考文献に見出される開始物質に限定されない。開始物質は、タキサン分子を還元的に脱酸素化する前に精製または単離しても、しなくてもよい。
本発明で使用する開始タキサンは、OH保護されていてもよいし、されている必要もない。一部の場合ではOH保護が望ましく、これに限定されるわけではないがC7、C10、およびC2’におけるOH基を含む、活性基の1つ以上がヒドロキシル保護基によって保護されるように実施できる。C2’ヒドロキシル基での保護は、次の還元的脱酸素化ステップで観察される側鎖開裂の量を劇的に減少させるため、特に重要であることが示されている。
本発明において、ヒドロキシル保護または非保護タキサンアミドは、保護または非保護タキサンイミンあるいはイミニウム化合物を生成するために、還元的に脱酸素化される。I開始タキサンアミドにおいて、RNが水素である場合は、タキサンイミンが形成される。1つの別の実施形態において、イミン化合物は一般に、図9に示す式(式中、R1、R2、R4、R7、R10、R2’、およびRACは、上で定義された通りである)を有する。開始タキサンアミドにおいて、RNがアルキル基である場合は、タキサンイミニウム化合物が形成される。1つの別の実施形態において、イミニウム化合物は一般に、図10に示す式(R1、R2、R4、R7、R10、R2’、およびRACは、上で定義された通りである)を有する。
還元的脱酸素化反応の完了に続いて、遷移金属および/または遷移金属副生成物の大半は、次の反応を進める前に除去できる。たとえば1つの別の実施形態において、すべてのまたは実質的にすべての遷移金属または金属副生成物は、得られた混合物が10,000ppm未満、好ましくは5,000ppm未満、さらに好ましくは1,000ppm未満の遷移金属または遷移金属副生成物を含むように除去できる。本発明では、遷移金属またはいずれかの金属副生成物の除去は任意である。
イミン化合物への変換後、イミン官能基の加水分解は、アミンまたはアミン塩をC−3’位置に生成する。イミン化合物の加水分解を実施するためには、どの一般的な酸でも使用できる。そのような酸は、これに限定されるわけではないが:(i)フッ化水素酸、塩酸、臭化水素酸、またはヨウ化水素酸;(ii)硝酸および他の窒素含有酸;(iii)硫酸または他の硫黄含有酸;(iv)トリフルオロ酢酸を除くカルボン酸;(v)リン酸または他のホスフェート含有酸;(vi)酒石酸;(vii)過塩素酸;(viii) p−トルエン硫酸;(ix)ピクリン酸を含む。好ましくは、酸は硫酸である。酸は、水性および/またはプロトン性溶媒、たとえばエタノールおよび/またはメタノールによる溶液である。本発明の別の実施形態において、アミン塩は、図11に示す式(式中、R1、R2、R4、R7、R10、R2’、およびRNは、上で定義した通りである)を有する。ここでXは、ハロゲンを含む脱プロトン化無機酸を含む。そのような酸は、これに限定されるわけではないが、フッ化水素酸、塩酸、臭化水素酸、またはヨウ化水素産を含む。またXは:(i)脱プロトン化硝酸または他の窒素含有酸;(ii)脱プロトン化硫酸または他の硫黄含有酸;(iii)トリフルオロ酢酸を除く、脱プロトン化カルボン酸;(iv)脱プロトン化リン酸または他のホスフェート含有酸;(v)脱プロトン化酒石酸;(vi)脱プロトン化過塩素酸;(vii)脱プロトン化p−トルエン硫酸;(viii)脱プロトン化ピクリン酸を含む。別の実施形態において、イミン1モル当たり、約2モル当量以上の酸を使用すべきである。
タキサンアミン塩は、適切な量のより極性の低い溶媒、好ましくはメチルtert−ブチルエーテルを添加することによって凝固させることができる。他の溶媒、たとえばジクロロメタン、ヘプタン、ヘキサン、トルエン、またはトリフルオロトルエンも使用できる。タキサンアミン塩はいったん凝固すると、容易に濾過できるため、精製を提供する。本質的にすべての非アミン副生成物または未反応開始物質は濾液に残るため、除去される。
タキサンアミン塩は、アミンを各種の方法で反応させることによって、パクリタキセルまたは他の有用なタキサンに変換できる。たとえば適切なアミン塩は、それに塩化ベンゾイルまたは他のベンゾイル化剤を反応させることによって、パクリタキセルに変換できる。これは、最初にアミン塩を溶媒、好ましくはTHFに溶解させ、次にアミン塩を過剰の塩基、好ましくは無機塩基、最も好ましくは炭酸ナトリウムによって中和し、続いてやや過剰のベンゾイル化剤、好ましくは塩化ベンゾイルを添加することによって実施できる。そのような反応の非制限的な例を図15に示す。他のタキサンアミドは、アシル化剤を変えることによって生成される。
本発明では、以下の物質を以下に述べるように使用する:
・HPLC(XTerra Column;0.05%ギ酸を含む、水−アセトニトリル勾配);UV検出
・タキサンアミド(500g);
・無水THF 3リットル;
・4−(ジメチルアミノ)ピリジン(DMAP)[1122−58−3]7.2g;
・(Hunig塩基)[7175−49−7]N−エチルジイソプロピルアミン 338mL;
・クロロトリメチルシラン(TMSC1)[75−77−4]165mL;
・ビス(シクロペンタジエニル)ジルコニウムクロライドヒドリド(Schwartz試薬)469g;
・ビシン溶液(1M)[150−25−4];
・エチルアセテート 1リットル;
・10%硫酸溶液 350mL;
・メチルtert−ブチルエーテル(MTBE) 10リットル;
・4:1 MTBE:THF 4リットル;
・重炭酸ナトリウム 317g;
・塩化ベンゾイル[ベンゼン環置換不純物を含まない]68mL;
・ヘプタン15リットル;および
・塩水溶液 3リットル。
本実施例は、主にタキソールCおよびタキソールEより成るタキサンアミド混合物の、タキソールA(パクリタキセル)への変換を説明する。
本実施例は、エトキシエチルエーテル保護を用いた、主にタキソールAおよびタキソールBより成るタキサンアミド混合物の、タキソールA(パクリタキセル)への変換を説明する。
本実施例は、エトキシエチルエーテル保護を用いた、主にタキソールCおよびタキソールEより成るタキサンアミド混合物の、タキソールA(パクリタキセル)への変換を説明する。
本実施例は、トリメチルシリルエーテル保護を用いた、主にタキソールBより成るタキサンアミド混合物の、タキソールA(パクリタキセル)への変換を説明する。
本実施例は、トリメチルシリルエーテル保護を用いた、主にタキソールAより成るタキサンアミド混合物の、タキソールA(パクリタキセル)への変換を説明する。
タキソールA 79.2%、タキソールB 17.4%、タキソールC 1.6%、およびタキソールE 0.3%より成るタキサンアミド混合物(2.5g.,〜2.9mmol)を無水THF 7.5mLに溶解させ、続いたN,N−ジメチルアミノピリジン(DMAP)18mg(0.13mmol)、ジイソプロピルエチルアミン(DIPEA)1.54mL(8.8mmol)、およびクロロトリメチルシラン(TMS−C1)1.1mL(8.7mmol)を添加した。反応物を0℃にて攪拌し、1時間後に完了させた。反応混合物を次に濾過した(アミン塩酸塩を除去するため)。濾液を、無水THF 3.3mL中のSchwartz試薬2.26グラム(8.8mmol)のスラリーに5℃にて、窒素雰囲気下で添加した。4時間後、反応は、1Mビシン13mLと共に30分間攪拌することによって停止させた。有機層は、さらなる1Mビシン13mLと共に30分間攪拌した。合わせた水層は、エチルアセテート2.5mLと共に20分間攪拌した。THFおよびエチルアセテート層を合わせ、10%H2SO4 1.75mLによって1時間処理して、続いてメチルtert−ブチルエーテル(MTBE)65mLをゆっくり添加し、さらに30分間攪拌した。アミン塩を濾過して、THF 10mLおよび塩化ベンゾイル0.41mL(3.5mmol)に溶解させ、飽和NaCl溶液2.5mL、およびpH7リン酸緩衝液12.5mLを添加し、反応物を室温にて15分間攪拌した。反応溶液を次に濃水酸化アンモニウム0.17mLによって処理し、15分間攪拌した。次に溶液を飽和NaCl溶液5mLによって洗浄した。分離した有機層をMgSO4 12.5グラム上で乾燥させ、濾過し、ヘプタン73mL中で沈殿させた。濾過および真空乾燥は、粗パクリタキセル(2.3グラム、純度80.9%、収率76.5%)を生じた。
本実施例は、N−メチルタキソールAをN−メチルタキソールCへの変換を説明する。 N−メチルタキソールA(3.5グラム、4.0mmol)のサンプルを無水THF 10mLに溶解させ、続いてN,N−ジメチルアミノピリジン(DMAP)30mg(0.24mmol)、ジイソプロピルエチルアミン(DIPEA)2.0mL(11.5mmol)、およびクロロトリメチルシラン(TMS−C1)1.4mL(11.4mmol)を添加した。反応物を0℃にて攪拌し、1時間後に完了させた。反応混合物を次に濾過した(アミン塩酸塩を除去するため)。濾液を、無水THF 5mL中のSchwartz試薬3.0グラム(11.8mmol)のスラリーに0℃にて、窒素雰囲気下で添加した。4時間後、反応は、1Mビシン18mLと共に30分間攪拌することによって停止させた。有機層は、さらなる1Mビシン18mLと共に30分間攪拌した。合わせた水層は、エチルアセテート7mLと共に20分間攪拌した。THFおよびエチルアセテート層を合わせ、10%H2SO4 2.2mLによって1時間処理して、続いてメチルtert−ブチルエーテル(MTBE)83mLをゆっくり添加し、さらに30分間攪拌した。アミン塩を濾過して、THF 12mLおよびヘキサノイルクロライド0.6mL(4.0mmol)に溶解させ、飽和NaCl溶液3.1mL、およびpH7リン酸緩衝液17mLを添加し、反応物を室温にて15分間攪拌した。反応溶液を次に濃水酸化アンモニウム0.22mLによって処理し、15分間攪拌した。次に溶液を飽和NaCl溶液6mLによって洗浄した。分離した有機層をMgSO4 1.7グラム上で乾燥させ、濾過し、ヘプタン95mL中で沈殿させた。濾過および真空乾燥は、粗パクリタキセル(2.83グラム、収率75%)を生じた。
Claims (5)
- タキサンアミン塩を生成する方法であって:
(i)タキサンイミンに第一の溶媒を接触させるステップと;
(ii)前記タキサンイミンに、イミン官能基の加水分解のためにサルフェート含有酸に接触させるステップと;
(iii)前記第一の溶媒よりも極性の低い第二の溶媒を添加することによって、凝固したタキサンアミン塩を分離するステップと;
を含み、
前記タキサンイミンが次式:
(式中、R1=アルキル、アリール、カルボニルまたはエーテル基;
R2=H、アルキル、アリール、エステル、エーテル、アセテート、ベンゾイル、トリメチルシリル、トリエチルシリル、2,2,2−トリクロロエトキシカルボニルまたはエトキシルエチルエーテル基;
R3=H、アルキル、アリール、エーテル、エステル、キシロシル、アセテート、ベンゾイル、トリメチルシリル、トリエチルシリル、2,2,2−トリクロロエトキシカルボニルまたはエトキシルエチルエーテル基;および
R4=H、アセテート、ベンゾイル、トリメチルシリル、トリエチルシリル、2,2,2−トリクロロエトキシカルボニルまたはエトキシルエチルエーテル基)
を有する、
タキサンアミン塩を生成する方法。 - タキサンアミン塩を生成する方法であって:
(i)タキサンイミンにプロトン性溶媒を接触させるステップと;
(ii)前記タキサンイミンに、イミン官能基の加水分解のために、脱プロトン化窒素含有酸、脱プロトン化リン含有酸、酒石酸、過塩素酸、脱プロトン化リン含有酸、脱プロトン化ピクリン酸、p−脱プロトン化p−トルエンスルホン酸、または塩酸を除く脱プロトン化ハロゲン含有酸を接触させるステップと;
(iii)前記プロトン性溶媒よりも極性の低い溶媒を添加することによって、凝固したタキサンアミン塩を分離するステップと;
を含み、
前記タキサンイミンが次式を有するタキサンアミン塩を生成する方法。
(式中、R1=アルキル、アリール、カルボニルまたはエーテル基;
R2=H、アルキル、アリール、エステル、エーテル、アセテート、ベンゾイル、トリメチルシリル、トリエチルシリル、2,2,2−トリクロロエトキシカルボニルまたはエトキシルエチルエーテル基;
R3=H、アルキル、アリール、エーテル、エステル、キシロシル、アセテート、ベンゾイル、トリメチルシリル、トリエチルシリル、2,2,2−トリクロロエトキシカルボニルまたはエトキシルエチルエーテル基;および
R4=H、アセテート、ベンゾイル、トリメチルシリル、トリエチルシリル、2,2,2−トリクロロエトキシカルボニルまたはエトキシルエチルエーテル基) - 請求項1に記載のタキサンアミン塩を生成する方法であって、
前記ステップ(ii)で用いるサルフェート含有酸が、硫酸であることを特徴とするタキサンアミン塩を生成する方法。 - 請求項2に記載のタキサンアミン塩を生成する方法であって、
前記ステップ(ii)で用いる酸が、硝酸、リン酸、酒石酸、p−トルエン硫酸、ピクリン酸、または、これらの組み合わせから選択されるタキサンアミン塩を生成する方法。 - 請求項1〜4のいずれか1つに記載のタキサンアミン塩を生成する方法であって、
前記ステップ(iii)で用いる溶媒が、メチルtert−ブチルエーテル、ジクロロメタン、ヘプタン、ヘキサン、トルエン、トリフルオロトルエン、または、これらの組み合わせから選択されるタキサンアミン塩を生成する方法。
