Description
(実験操作:鉄フタロシアニン錯体と酸化剤との反応による化学発光強度測定)
化学発光用ガラスセルに蒸留水(株式会社島津製作所製SWAC−500の蒸留水製造装置により作製、以下同じ)340μlをマイクロシリンジにより注入し、pH緩衝溶液(和光純薬工業株式会社製四ホウ酸ナトリウム(Borax)を蒸留水に溶解させ、0.1molL−1に希釈したものを使用、以下同じ)を100μl加え、pHを調整した後(株式会社堀場製作所pHメーターF−22を使用、以下同じ)、0.08molL−1過酸化水素水溶液(和光純薬製工業株式会社製30%過酸化水素を蒸留水により希釈したものを使用、以下同じ)を40μl加え混合した。そして、化学発光装置のセルホルダーにセルをセットし、Fe−PTS水溶液(Aldrich社製鉄フタロシアニンスルホン酸ナトリウムを蒸留水に希釈して作製した)を20μlマイクロシリンジにより注入した。その後、注入直後に現れる化学発光シグナルを化学発光測定装置(株式会社日音医理科機器製作所製LUMICOUNTER NU−600)により測定した。
水溶液中における他の金属フタロシアニン錯体(銅、鉄、ニッケル、コバルト、マグネシウム、鉛、白金、亜鉛)および遊離のフタロシアニンの化学発光現象について検討するため、Fe−PTS水溶液のかわりに上記の金属フタロシアニン錯体および遊離のフタロシアニン錯体を添加した(銅フタロシアニン錯体はAldrich社製銅フタロシアニンスルホン酸ナトリウムを蒸留水に希釈して作製;ニッケルフタロシアニン錯体はAldrich社製ニッケルフタロシアニンスルホン酸ナトリウムを蒸留水に希釈して作製;その他の合成フタロシアニンはスルホン化したフタロシアニンと金属塩をDMSO中において150℃、1時間の条件で合成し、再結晶したものを使用)。それぞれの化学発光強度を表1に示す。Fe−PTSの化学発光強度(ICL)が1270で最大となり、遊離のフタロシアニンスルホン酸とその他の金属フタロシアニンスルホン酸ではほとんど化学発光現象が観測されなかった。
Fe−PTSと酸化剤との化学発光反応における酸化剤の種類について検討を行った。酸化剤としてH2O2、および2KHSO5・KHSO4・K2SO4を用いて、pH2.3、pH7.0、またはpH10.0における場合のFe−PTSの化学発光強度について検討した。その結果を、表2および表3に示す。過酸化水素では、pH10.0において最大の化学発光強度が得られた。これは、代表的な化学発光であるルミノール化学発光の挙動と同様である。また、2KHSO5・KHSO4・K2SO4では、酸性〜アルカリ性条件下において微弱ではあるが、ほぼ同様な発光強度が得られた。
(pHの影響)
Fe−PTSと過酸化水素との化学発光反応系に及ぼすpHの影響を検討した。Fe−PTS濃度2.0×10−5M、過酸化水素濃度6.4×10−3Mの条件で行った。その結果を図3に示す。pH変化に伴う最大化学発光強度はpH10.0において観測された。これは他の化学発光系(ルミノール化学発光、鉄−クロロフィリン錯体化学発光など)とほぼ同様の値となった。pH10.0前後における化学発光強度のピークは、過酸化水素からスーパーオキシドラジカルの生成される最適なpHが10前後であるためではないかと考えられる。
Fe−PTSと過酸化水素との測定方法に使用する化学発光反応系における過酸化水素濃度の影響を検討した。Fe−PTS濃度2.0×10−5M、pH10.0の条件でおこなった。その結果を図4に示す。添加する過酸化水素濃度の変化に伴う最大化学発光強度は過酸化水素濃度6.4×10−3Mにおいて観測された。極大値以上の濃度では発光強度の減少がみられた。これは一般の化学発光系と同様の挙動であった。
Fe−PTSと過酸化水素との化学発光反応系に及ぼすFe−PTS濃度の影響を検討した。過酸化水素濃度6.4×10−3M、pH10.0の条件で行った。その結果を図5に示す。添加するFe−PTSの濃度の増加に伴い化学発光強度も増加した。また、Fe−PTSの濃度に伴う化学発光強度の増加率低下は、Fe−PTSの濃度が過剰となり、過酸化水素から生成されるスーパーオキシドラジカルにより酸化分解される割合が低下してしまうためであると考えられる。よって、本実施例においては、Fe−PTSの濃度は2.0×10−5Mとすることとした。
Fe−PTSと過酸化水素との化学発光反応系に及ぼす温度変化の影響を検討した。Fe−PTS濃度2.0×10−5M、過酸化水素濃度6.4×10−3M、pH10.0の条件下で温度を15℃〜40℃の間で変化させた。その結果を図6に示す。図6に示すように20℃〜40℃の間において化学発光強度の変化はほとんど見られなかった。
Fe−PTSの化学発光反応系における共存物質の影響について検討した。
アミノ酸の化学発光強度に与える影響を表4に示す。アミノ酸のうち、L−システイン、L−ヒスチジン、L−チロシン、L−トリプトファンにおいて消光現象がみられた。その他のアミノ酸については特に影響を及ぼさなかった。L−システインは還元性の物質であり、酸化反応に影響を及ぼしていると推測される。L−ヒスチジンはヘモグロビンのタンパク中に含まれ、ヘム鉄と結合している。本化学発光系においても、Fe−PTSの軸配位部位に配位し、触媒活性の低下が起こり、発光強度の現象がみられたと考えられる。L−チロシン、L−トリプトファンは一重項酸素の消去剤として用いられることから(H. Goto, A. Masuda, M. Yamada, BUNSEKI KAGAKU, 46, 711-717 (1997))、活性酸素種に影響を与えているものと考えられる。
(実験操作)
化学発光用セルに蒸留水100μlをマイクロシリンジにより注入し、pH緩衝液100μlを加えてpHを調整した後、L−チロシンを含む水溶液200μlとFe−PTS水溶液50μlを加え混合した。化学発光装置のセルホルダーにセルをセットし、過酸化水素水溶液50μlをマイクロシリンジにより注入し、化学発光シグナルを化学発光測定装置(マイクロテック・ニチオン社製GENELIGHT GL−200S)で測定した。
pHを変化させたときの化学発光強度の影響を検討した。過酸化水素濃度6.4×10−3M、Fe−PTS濃度2.0×10−5M、L−チロシン濃度2.0×10−7Mの条件でおこなった。その結果を図7に示す。pH変化に伴う最大化学発光強度はpH10において観測された。pH10前後における化学発光強度のピークは、過酸化水素からスーパーオキシドラジカルの生成される最適なpHが10前後であるためではないかと考えられる。この結果より、L−チロシンの濃度2.0×10−7Mにおける発光強度の差が大きいことから、pH10を最適pHとした。
反応系における過酸化水素濃度の影響を検討した。pH10、Fe−PTS濃度2.0×10−5M、L−チロシン濃度2.0×10−7Mの条件でおこなった。その結果を図8に示す。過酸化水素濃度の変化に伴う最大化学発光強度は過酸化水素濃度6.4×10−3Mにおいて観測され、極大値以上の濃度では発光強度の減少が見られた。これは一般の化学発光系と同様の挙動であった。しかし、L−チロシンの濃度2.0×10−7Mにおける発光強度の差が大きい3.2×10−3Mを最適濃度とした。
Fe−PTS濃度の影響を検討した。pH10、過酸化水素濃度6.4×10−3M、L−チロシン濃度2.0×10−7Mの条件でおこなった。その結果を図9に示す。Fe−PTSの濃度の増加に伴い化学発光強度は増加した。L−チロシンの濃度2.0×10−7Mにおける発光強度の差が大きいことから、4.0×10−5Mを最適濃度とした。
pH10、過酸化水素濃度3.2×10−3M、Fe−PTS濃度4.0×10−5Mの条件下でL−チロシンの濃度を変化させ、L−チロシンの検量線を作成した。GENELIGHT GL−200SによるL−チロシン(Tyr)の検量線を図10に示す。その結果、2.0×10−8M〜2.0×10−5Mにおいて良好な直線が得られた。検量線の相関係数は0.9842であった。また、L−チロシンの濃度2.0×10−7Mにおいて、5回測定での変動係数は0.68%であり、検出限界(3σ)は1.81×10−8M(3.27ppm)であった。

Claims (6)
- 鉄フタロシアニンスルホン酸と過酸化水素又は2KHSO 5 ・KHSO 4 ・K 2 SO 4 とを含む化学発光試薬であって、前記鉄フタロシアニンスルホン酸と前記過酸化水素又は前記2KHSO 5 ・KHSO 4 ・K 2 SO 4 とを反応させることにより化学発光を生じる化学発光試薬。
- 鉄フタロシアニンスルホン酸と過酸化水素又は2KHSO 5 ・KHSO 4 ・K 2 SO 4 とを反応させて、化学発光を生じさせる化学発光方法。
- 試料液中に含まれる被検出物質の濃度を測定する方法であって、
前記試料液と、鉄フタロシアニンスルホン酸水溶液と、過酸化水素又は2KHSO 5 ・KHSO 4 ・K 2 SO 4 とを混合する工程と、
前記混合溶液中に生じる発光の発光強度を測定する工程と、
前記測定した発光強度と、予め求めた検量線とから前記被検出物質の濃度を求める工程と
を含む測定方法。 - 前記試料液のpHがpH8〜12に調整されている請求項3に記載の測定方法。
- 前記被検出物質が、L−システイン、L−フェニルアラニン、L−ヒスチジン、L−トリプトファン、およびL−チロシンからなる群より選択されるアミノ酸と、チアミン塩酸塩、リボフラミン、およびL−アスコルビン酸からなる群より選択されるビタミンと、グルコースと、過酸化水素と、銅イオン、ニッケルイオン、亜鉛イオン、マンガンイオン、およびコバルトイオンからなる群より選択される金属イオンとからなる群のうちの少なくとも一つである請求項3または4に記載の測定方法。
- 試料液中に含まれる被検出物質の濃度を測定するためのキットであって、前記キットは、鉄フタロシアニンスルホン酸と過酸化水素又は2KHSO 5 ・KHSO 4 ・K 2 SO 4 とを含み、前記被検出物質の存在下で、前記試料と前記鉄フタロシアニンスルホン酸と前記過酸化水素又は前記2KHSO 5 ・KHSO 4 ・K 2 SO 4 とを混合させることにより前記被検出物質の濃度に応じて化学発光を生じるキット。
