Description
(1)LMWおよびMMWを分解する酵素による前処理を行い、酵素分解されずに残ったアディポネクチンを測定することによって、HMWだけを検出する方法(特許文献1)。
(2)アディポネクチン単量体には反応しないが、3量体及び/又はそれが凝集した構造を有する高分子アディポネクチンを認識する抗体を用い、当該高分子アディポネクチン抗原だけを検出する方法(特許文献2)。
本明細書において、「高分子アディポネクチン」とは、HMWアディポネクチン、または単にHMWとも称され、6個より多くのアディポネクチン単量体が結合したアディポネクチン多量体を指す。また本明細書において、「高分子アディポネクチン」は、6量体以下のアディポネクチンとそれより大きなアディポネクチンとを区別するための用語としても使用され得る。
本発明に用いるポリクローナル抗体としては、上記と同じくアディポネクチンヒンジ部を含むか又は当該部位に絞り込んだペプチドを作製し、これを抗原としてポリクローナル抗体を作製すれば、HMWに特異的な抗体を作製することが可能である。
1)ANOC9121
エピトープ:ヒトアディポネクチンアミノ酸配列の36番システイン残基の近傍領域(Bio Clinica 19(13):52−57,2004)。
2)Clone 5A(大塚製薬株式会社より取得)
エピトープ:不明(但し、単量体等の低分子量帯アディポネクチンには反応しない(後述)ことから、高次構造を認識していると考えられる)。
上記抗体の認識するアディポネクチン分子を、Western Blottingにより解析した。サンプルとして、非還元非加熱条件下(レーンNR)および2メルカプトエタノールによる還元条件下(レーンR)でヒト血液を電気泳動し、これに上記2種類の抗体を反応させ、染色した。結果を図1に示す。ヒト血中アディポネクチンは、非還元非加熱条件下では、HMWやMMW、LMW等の多様な形態で存在する一方、還元条件下では、3量体以上の形態が消失し、単量体である約28,000Daの形態若しくは2量体として存在した。モノクローナル抗体5Aは、約9万Da(LMW)以上の分子量のアディポネクチンのみを検出した一方、2量体以下の低分子アディポネクチンは認識しなかった。これに対し、モノクローナル抗体ANOC9121は、全ての分子量体を認識した。
ヒト血清を、Superdex200カラム(GEヘルスケア)を用いてHPLC分画した。各画分のアディポネクチン濃度は、以下の3種類の方法で測定を試みた。
1)Totalアディポネクチン(図2中、白四角)
各画分の抗原を、前処理してアディポネクチン基本骨格に変換した後に、サンドイッチELISA法にて定量した(ヒトアディポネクチンELISAキット(大塚製薬(株))使用)。測定値を、各アディポネクチン画分のコントロール値とした。
2)ANOC9121(図2中、黒丸)
各画分の抗原を、固相化抗体(一次抗体)及び二次抗体にANOC9121を使用したサンドイッチELISA法にて定量した:各画分は緩衝液で適宜希釈後、前処理を行わずに測定した。
3)Clone 5A(図2中、白丸)
各画分の抗原を、固相化抗体及び二次抗体に5Aを使用したサンドイッチELISA法にて定量した:各画分は緩衝液で適宜希釈後、前処理を行わずに測定した。
3,3’,5,5’−テトラメチルベンジジン−HRP結合ストレプトアビジンを添加し、インキュベートした。プレートに結合したHRP結合ストレプトアビジンより得られる発色の強度を測定することにより、各画分のアディポネクチン量を決定した。
Claims (6)
- 寄託番号FERM BP−10940であるハイブリドーマにより生産されるモノクローナル抗体ANOC9121を一次抗体及び二次抗体として使用するサンドイッチ免疫測定系を用いた、高分子アディポネクチンを選択的に測定する方法。
- 高分子アディポネクチンが、6量体より大きな多量体アディポネクチンである、請求項1記載の方法。
- ヒト高分子アディポネクチンを選択的に測定する、請求項1又は2記載の方法。
- 寄託番号FERM BP−10940であるハイブリドーマにより生産されるモノクローナル抗体ANOC9121を一次抗体及び二次抗体として使用するサンドイッチ免疫測定系を用いた、高分子アディポネクチンを選択的に測定するためのキット。
- 高分子アディポネクチンが、6量体より大きな多量体アディポネクチンである、請求項4記載のキット。
- ヒト高分子アディポネクチンを選択的に測定するための、請求項4又は5のいずれかに記載のキット。
