培養終了後、マイクロプレートの各穴へ、1%(v/v) FBS含有DMEM培地に安息香酸ベンジルを溶解した試料溶液120μLを添加して更に24時間培養した。培養後、上清100μLを、VEGF捕捉抗体を吸着させたELISAプレートに移し換えた。その後、上清をELISAプレートから除去し、0.05%(v/v) Tween-20を含むリン酸生理緩衝液(PBS-T)にて洗浄を行った。ELISAプレートへビオチン化抗ヒト抗体を添加し、捕捉されたVEGFと反応させた後にPBS-Tにて洗浄を行い、その後、ストレプトアビジン-HRP希釈溶液を添加した。PBS-Tにて洗浄後、HRP基質溶液を添加してHRPとの酵素反応を起こし、反応によって生じた光を450nmの吸光度として測定した。
