環状構造を形成し得るポリペプチド領域(即ち、両端のシステインを除くアミノ酸配列)において、(1)全てのポリペプチドには塩基性アミノ酸のKまたはRが含まれ、(2)残りのアミノ酸残基は、10種のアミノ酸〔G、A、V、L、S、T、P、Q、H、N〕のいずれかから構成される。

本発明の好ましい態様としては、上記のポリペプチドの両末端のシステイン残基がリジンへ置換されたポリペプチドにおいて、両末端の該リジンがアミド結合によって連結した構造の環状化ポリペプチドを例示することができる。
本発明の好ましい態様において「脳移行モチーフ配列を含む環状化ポリペプチド」とは、例えば、配列番号:13〜24のいずれかに記載の脳移行モチーフ配列を含むアミノ酸配列であって、該配列の両末端にそれぞれリジンが付加され、該リジン同士がアミド結合によって連結した構造の環状化ポリペプチド(その鎖長は例えば、100アミノ酸以内であり、好ましくは50アミノ酸以内であり、より好ましくは30アミノ酸以内であり、さらに好ましくは15アミノ酸以内であり、さらに好ましくは12乃至9アミノ酸であり、最も好ましくは9アミノ酸である。)を例示することができる。
さらに好ましくは、「脳移行モチーフ配列を含む環状化ポリペプチド」は、例えば、配列番号:13〜24のいずれかに記載の脳移行モチーフ配列の両末端にそれぞれリジンが付加され、該リジン同士がアミド結合によって連結した構造の環状化ポリペプチドを例示することができる。
(a)任意の分子と本発明のポリペプチドとが結合した構造の脳移行性の環状化ポリペプチドを製造する工程
(b)前記環状化ポリペプチドを動物体内へ投与する工程
なお本明細書において引用された全ての先行技術文献は、参照として本明細書に組み入れられる。
〔実験手法〕
・N-succinimidyl-4-[18F]fluorobenzoate ([18F]SFB)の合成
t-butyl 4-N,N,N-trimethylammoniumbenzoate(5 mg)を出発物質として、Acetonitrile(0.5 mL)中[18F]KF / Kryptofix222との反応(90℃, 10 min)、引き続く塩酸加水分解(100℃, 5 min)により得られる4-[18F]fluorobenzoic acidをC-18 Sep-Pakにて精製した。精製した中間体4-[18F]fluorobenzoic acidにAcetonitrile(1 mL)中25% tetramethylammnoium hydroxide(20μL)存在下、O-(N-succinimidyl)-N,N,N’,N’-tetramethyluronium tetrafluoroborate (TSTU、15 mg)を加え加熱(90℃, 5 min)、冷却後反応液を5% 酢酸溶液(10 mL)に捕集した。[18F]SFBの精製はC18 Sep-Pakを用いて10% 酢酸で洗浄後、3 mLのDichloromethaneで溶出させた。溶出液を窒素気流下で乾燥させた。
FE-21(22.8 mg, 60μmol)を含むN,N-dimethylformamide (DMF)(1 mL)溶液に、60 % NaH(12.07 mg, 303μmol)存在下6-bromohexanoic acid(11.77 mg, 60μmol)を加え、60℃にて2時間加熱した。脱イオン水、0.05 N HClを添加後、反応混合物を酢酸エチルにて抽出した。酢酸エチルを乾燥、濃縮後得られた残渣をシリカゲルカラムクロマトグラフィー(CHCl3 / 酢酸エチル = 2 / 1)にて精製し、C6-FE-21を得た。Acetonitrile(2 mL)中 diisopropylethylamine(11μL)存在下、C6-FE-21(10 mg)とTSTU(11.5 mg)を80℃にて2時間反応させた。反応液を留去して得られる残渣を酢酸エチルに溶かし、水洗、乾燥後シリカゲルカラムクロマトグラフィー(CHCl3 / 酢酸エチル = 2 / 1)にて精製すると、FE-21-C6-スクシンイミド誘導体が得られた。本発明ではこれをL-703,717スクシンイミド誘導体として使用した。
K004Kペプチド1mgを100μLのDMFに溶解させ、Triethylamine (TEA)添加によりpHを7-8に調整した。調整したペプチド溶液全量を乾燥[18F]SFBに添加し、溶解させた後室温で20-30分反応させた。精製には高速液体クロマトグラフィー(HPLC)を使用した。YMC J’sphereカラム(10 x 250 mm)、23% Acetonitrile, 0.1% Trifluofoacetic acid (TFA)イソクラティック溶出、5 mL/minで約8.5 minに溶出されるピークフラクションを分取した。分取したピークフラクションにTween80を20μL添加後ロータリーエバポレーターで300-400μLに濃縮し、インジェクションサンプルとした。
K004Kペプチド0.6 mgを100μLのDMFに溶解させ、TEA添加によりpHを7-8に調整した。
調整したペプチド溶液全量を乾燥[18F]SFBに添加し、溶解させた後室温で20-30分反応させた。次にL-703,717スクシンイミド誘導体0.3 mg/25μLのDMF溶液を全量添加し、室温で20-30分間反応させた。精製にはHPLC)を使用し、YMC J’sphereカラム(10x250mm)、38% Acetonitrile, 0.1% TFAイソクラティック溶出、5 mL/minで約19分に溶出されるピークフラクションを分取した。分取したピークフラクションにTween80を30μL添加後ロータリーエバポレーターで300-400μLに濃縮し、インジェクションサンプルとした。

Sprague-Dawleyラットをネンブタール腹腔内投与することにより麻酔し、右総頚動脈より[18F]Fluorobenzoate(FB)標識ペプチドを投与した。投与後30分間Micro PETにより脳のイメージング画像を取得した。
Sprague-Dawleyラットをネンブタール腹腔内投与することにより麻酔し、右総頚動脈より標識ペプチドを投与した。投与後30分間のMicro PETスキャン終了後脳を摘出しドライアイスにより凍結させた。凍結脳は厚さ0.2μmの水平断の凍結切片を作製し、20-30分間イメージングプレートとコンタクトさせた後、BAS5000を用いてイメージングプレートから信号の取り込みを行い、ARG画像を取得した。
ヨウ素化反応にはChloramine T法によりヨウ素化を行った。
0.2 M Phosphate buffer 20μLに10 mM [C]C004CY 15μL、[131I]NaI 5μL (100-200μCi)を混和後、0.1M Phosphate bufferで溶解させた15 mM Chloramine T 20μL加え撹拌後、30秒間室温で反応させた。反応溶液は直ちにHPLC精製を行った。Inertsil ODS-2カラム(4.6 x 150 mm)、20% Acetonitrile, 0.1% Trifluofoacetic acid (TFA)イソクラティック溶出、1 mL/minで約16分に溶出されるピークフラクションを分取した。分取したピークフラクションにTween80を5μL添加後凍結乾燥させ、生理食塩水に再溶解させてインジェクションサンプルとした。
標識ペプチド([131I]標識ペプチド;約10μCi, [18F]FB標識ペプチド;約100μCi)をFVB/NJclマウス尾静脈より投与後1分もしくは10分後に血液、脳を採取した。
血液は200μLとりMethanol 1 mLを添加し激しく撹拌した。脳にはMethanolを2 mL添加し、ポッター型ホモジェナイザーでホモジェナイズした。血液、脳のホモジェネートを遠心し、上清を組織抽出液とし、ペプチド成分を回収した。抽出液を薄層クロマトグラフィー(TLC)により展開し(溶媒:Buthanol: Acetic acid: Water=4:1:2)、代謝物スポットの有無を調べた。

脳移行性ペプチドがその活性を発揮するためには脳移行性アミノ酸モチーフが環状構造を保つことが必要である。環状化にはシステインのSH基間のS-S架橋によるものと、N末端のαアミノ基とC末端のカルボシキル基の間でアミド結合(ペプチド結合)を形成することによるものが考えられた。[C]C004CYペプチドでは、配列番号:4の脳移行性アミノ酸配列が2つのシステイン(C)で挟まれた配列を持ち、システインのSH基間のS-S架橋により環状化しており、C末端にヨウ素標識に利用できるチロシン(Y)が追加されている。[C]は環状であることを表す。一方、[C]K004Kペプチドは同じ脳移行性アミノ酸配列の両端にリジン(K)が存在し、N末端のαアミノ基とC末端のカルボシキル基の間でアミド結合(ペプチド結合)を形成することにより環状化したペプチドである。これらを模式図で示す(図1)。
In vivo代謝試験の結果から[C]K004Kのペプチドは[C]C004CYに比べin vivo で安定であることが示された。この[C]K004Kがどの程度安定性が向上したのか比較するために、アイソトープ標識の代わりに蛍光色素をトレーサーとしマウス肝ホモジネートを用いてin vitroで評価する実験系を作成して比較を行った。

Co-senseを用いてHPLC分析
[C](Flu)-C004C: 004の配列の両端にCを加えてC同士をジスルフィド結合により環状化したペプチドのN末にFluoresceinで標識した。
[C](Flu)-K004C: 004の配列のN末側にK、C末側にCを加えてN末とC末をペプチド結合させることにより環状化したペプチドのN末にFluoresceinで標識した。
[C](Flu)-K004K: 004の配列の両端にKを加えてN末とC末をペプチド結合させることにより環状化したペプチドのN末にFluoresceinで標識した。
[C](Flu)-C004CYK: 004の配列のN末側にC、C末側にCYKを加えてC同士をジスルフィド結合により環状化したペプチドのN末にFluoresceinで標識した。
[C]C004CYK(Flu): 004の配列のN末側にC、C末側にCYKを加えてC同士をジスルフィド結合により環状化したペプチドのC末K側鎖にFluoresceinで標識した。
次に[C]K004Kの脳実質への移行を検証するために、[C]K004Kのアミノ基に18F標識したSFBを反応させることによってK004Kペプチドをポジトロン標識し[C] ([18F]FB)-K004K を合成してin vivo脳移行性を検討した。

その結果、PETでは脳にもある程度のシグナルが見られたが、顔のシグナルが強すぎるため脳が抜けているように見えてしまった。Ex vivo ARGでは脳実質にある程度のシグナルが見られることから尾静注でもある程度は脳に入っていると考えられた(図7)。
[C]K004Kの脳実質への移行が確認されたことから、次に[C]K004KがPETリガンドを脳内に運搬できるかどうかを検証するために、[C]([18F]FB)-K004K-(L-703,717)のPETイメージングを行った(図8)。[C]K004Kと[18F]SFBを反応させ、L-703,717スクシンイミド誘導体を引き続き反応させることにより[C]([18F]FB)-K004K-(L-703,717)を合成した。合成した[C]([18F]FB)-K004K約0.33 μCi/300μLをSprague-Dawleyラット右総頸動脈より投与し、マイクロPETにより30分間スキャンを行い脳の生体イメージング画像を取得した。
その結果、投与側で強いシグナルが検出され、反対側、小脳でもシグナルが検出された。スキャン後の脳を摘出しEx vivo ARGを行った(図8D、図9)。

これらのことから[C]K004Kは頚動脈投与によりリガンドを脳実質へ移行させることができることがわかった。
Claims (13)
- 配列番号:13〜24のいずれかに記載された脳移行モチーフ配列の両末端部にリジンを有する配列からなるポリペプチドであって、該リジン間のペプチド結合によって環状化され、代謝安定性が向上していることを特徴とする、多価結合手を有する環状化ポリペプチド。
- リジンのεアミノ基にPET核種で標識された分子が結合し、および/または、他のリジンのεアミノ基に脳内受容体のリガンド分子が結合している、請求項1に記載の環状化ポリペプチド。
- 請求項1に記載の環状化ポリペプチドを有効成分とする、脳内移行用キャリア分子。
- 請求項1に記載の環状化ポリペプチドのリジン残基のεアミノ基に、薬剤が結合していることを特徴とする、脳移行性薬剤。
- 請求項2に記載の環状化ポリペプチドを有効成分とする、PET検査用試薬。
- 以下の物質を少なくとも構成要素として含む、脳移行性薬剤製造用キット。
(a)請求項1に記載の環状化ポリペプチド
(b)薬剤 - 以下の物質を少なくとも構成要素として含む、PET検査用キット。

(b)PET核種で標識された分子 - 配列番号:13〜24のいずれかに記載された脳移行モチーフ配列の両末端部にリジンを有する配列からなるポリペプチドについて、両末端部をペプチド結合させる工程を含む、多価結合手を有し環状化され代謝安定性が向上した脳移行性ポリペプチドの製造方法。
- 請求項1に記載の環状化ポリペプチドのリジンのεアミノ基と、薬剤とを結合する工程を含む、代謝安定性が向上した脳移行性薬剤の製造方法。
- 請求項1に記載の環状化ポリペプチドのリジンのεアミノ基と、PET核種で標識された分子とを結合する工程を含む、代謝安定性が向上したPET検査用試薬の製造方法。
- 請求項1に記載の環状化ポリペプチドのリジンのεアミノ基と、脳内受容体リガンド分子とを結合する工程を含む、代謝安定性が向上した脳移行性リガンド分子の製造方法。
- 配列番号:13〜24のいずれかに記載された脳移行モチーフ配列の両末端部にリジンを有する配列からなるポリペプチドについて、両末端部をペプチド結合させる工程を含む、多価結合手を有する状態で脳移行性ポリペプチドを環状化する方法。
- 配列番号:13〜24のいずれかに記載された脳移行モチーフ配列の両末端部にリジンを有する配列からなるポリペプチドについて、両末端部をペプチド結合させる工程を含む、脳移行性ポリペプチドの代謝安定性を向上させる方法。
