Description
青色タンパク質
いくつかの形態のリナブルータンパク質着色剤を評価した。Health and Herbs製の原料粉末藻を用いて、乳鉢及び乳棒で圧潰し、遠心分離して細胞残屑をペレット状にし、青色溶液を得た。リナブルーAE及びリナブルーHGEをDainipponから、精製アロフィコシアニン及びフィコシアニンをSigmaから入手した。リナブルー着色剤をさらなる検討に用いたが、それはリナブルー着色剤がBlue Visionレンズケア製品(CIBA Vision)に用いられているからである。
スブチリシン、トリプシン、及びパパインの3種のプロテアーゼを試験した。3種のプロテアーゼは全て、リナブルーAE、並びにタンパク質の他の形態を脱色することができた。眼用製品での使用が許可されているので、スブチリシン、例えばUnizyme錠剤(CIBA Vision)を選択した。スブチリシンは、Clear Lens Pro(CLP)と呼ばれる製品として、Novozymeから入手できる。NovozymeからのCLPの液体サンプルを、直接に試験に用いた。
Clear Lens Pro(CLP)及びUnizyme錠剤の存在下での、リナブルーの脱色速度を検討するために、実験を行った。この結果から、リナブルーAE溶液は4〜6時間の時間枠で脱色されうることが分かった。
Sigma製スブチリシンを、種々の樹脂:Amberlite XAD7HP、並びにビーズEC-HA及びEC-EP(違いは表面上の官能基であり、それぞれアミン及びエポキシ)の上に固定化した。Amberliteに結合後、スブチリシンは失活した。ビーズ上に固定後、スブチリシンはまだ活性であり、結合の後9日まで活性を留めた。非固定化酵素より活性がいくらか失われた。ビーズ上に固定化されたスブチリシンは、7回までうまく再使用することができたが、その後の脱色反応について活性の損失がいくらか見られた。
Claims (15)
- コンタクトレンズの洗浄及び消毒用レンズケアキットであって:
水溶性有色タンパク質及び殺菌剤を含む有色レンズケア溶液;並びに
タンパク質分解酵素、ここで、該タンパク質分解酵素は、固体支持材上に固定化されており、そして、該タンパク質分解酵素は、スブチリシン、トリプシン、パパイン又はこれらの組み合わせである、
を含み、
有色レンズケア溶液と接触すると、有色レンズケア溶液を実質的に脱色させるのに十分な期間にわたって、タンパク質分解酵素が有色タンパク質を分解し、これにより有色レンズケア溶液により消毒及び洗浄されているレンズが使える状態にあることを示す、コンタクトレンズの洗浄及び消毒用レンズケアキット。 - レンズケアキットが、処理するコンタクトレンズ及び一定量の有色レンズケア溶液を収容するためのウェルを有するレンズケース、ウェルと流体連通にある区画を含み、固体支持材上のタンパク質分解酵素がレンズケースの区画に入れられた、請求項1のレンズケアキット。
- レンズを処理する直前に、レンズケース中で、一方の着色剤を含まず過酸化水素を含有する溶液と、もう一方の過酸化水素を含まず着色剤を含有する溶液の、二つの溶液を混合することにより有色レンズケア溶液を得る、請求項2のレンズケアキット。
- 有色レンズケア溶液が過酸化水素を含まない多目的溶液である、請求項2のレンズケアキット。
- 殺菌剤が、ヘキサメチレンビグアニドポリマー(PHMB)である、請求項4のレンズケアキット。
- PHMBが0.01〜10ppmの量で存在する、請求項5のレンズケアキット。
- 有色レンズケア溶液が、最初に青又は緑又は紫の色を有する、請求項6のレンズケアキット。
- 有色レンズケア溶液がタンパク質分解酵素と接触後、有色溶液の色が、制御された期間にわたって徐々に退色する、請求項7のレンズケアキット。
- 制御された期間の終わりに、有色レンズケア溶液の色が実質的に脱色され、実質的に透明かつ無色になる、請求項8のレンズケアキット。
- 制御された期間がコンタクトレンズの消毒に十分な長さである、請求項9のレンズケアキット。
- 制御された期間が少なくとも約2時間である、請求項10のレンズケアキット。
- 有色タンパク質が、フィコビリタンパク質、フィコシアニン、アロフィコシアニン、フィコエリトリン、緑色蛍光タンパク質(GFP)、GFP様タンパク質、バクテリオロドプシン、又はこれらの組み合わせである、請求項2のレンズケアキット。
- 有色タンパク質が、フィコシアニン、アロフィコシアニン、又はこれらの組み合わせである、請求項2のレンズケアキット。
- 有色タンパク質が、リナブルーAE、リナブルーHGE、リナブルーA、リナブルーHK、リナブルーHG、及びこれらの組み合わせよりなる群から選択される、藻の抽出物である、請求項2のレンズケアキット。
- 有色タンパク質が、精製アロフィコシアニン、精製フィコシアニン、又はこれらの組み合わせである、請求項2のレンズケアキット。
