Description
Claims (14)
- クロマトグラフ用カラムであって、入口、出口、分析要素、および1つまたは複数の保持要素を備え、前記1つまたは複数の保持要素が、前記入口の端部または/および前記出口の端部に縦軸に沿って互いに隣接して配置され、前記分析要素が、前記分析要素内に画定される管腔域内に配設される固定相を有し、前記保持要素が、保持要素内に画定される管腔域を有し、前記保持要素が、前記分析要素の前記管腔域と位置合わせされ、被分析物が、前記保持要素と分析要素との間を移動し、前記保持要素および分析要素を通り抜けることができ、各前記保持要素が、残る前記分析要素から取り外されることができるカラム。
- 前記分析要素が、前記入口に隣接して配設される保持要素を有する請求項1に記載のカラム。
- 前記分析要素が、前記入口の端部で縦軸に沿って互いに隣接して配設される複数の保持要素を有する請求項2に記載のカラム。
- 前記分析要素が、前記入口の端部で縦軸に沿って互いに隣接して配設される1つまたは複数の保持要素と、前記出口の端部で縦軸に沿って互いに隣接して配設される1つまたは複数の保持要素とを有する請求項1に記載のカラム。
- 前記分析要素が、約1cmから約20cmの範囲の長さの縦軸を有する請求項1に記載のカラム。
- 前記分析要素が、100μmの内側直径および360μmの外側直径の管腔寸法を有する請求項1に記載のカラム。
- 前記1つまたは複数の保持要素の長さが、前記縦軸に沿って約0.5cmから約25cmの範囲であることができる請求項1に記載のカラム。
- 前記1つまたは複数の保持要素が、前記分析要素の管腔域よりも直径が小さい管腔域を有する請求項1に記載のカラム。
- 前記1つまたは複数の保持要素を、前記分析要素に固定する手段をさらに備える請求項1に記載のカラム。
- 前記固定する手段が接着剤からなる請求項9に記載のカラム。
- 前記接着剤がPDMSエラストマである請求項10に記載のカラム。
- 1つ以上の被分析物を含む試料を分離する方法であって、
前記試料を適切な移動相と混合することと、
前記試料を、縦軸に沿って構成される、入口、出口、分析要素、および1つ以上の保持要素を有するクロマトグラフ用カラムに導入し、前記分析要素が、前記分析要素内に画定される管腔域内に配設される固定相を有し、被分析物が、前記保持要素と分析要素との間を移動し、前記保持要素および分析要素を通り抜けることができるように、前記保持要素が、前記保持要素内に画定される管腔域を有し、前記保持要素が、前記分析要素の前記管腔域と位置合わせされることと、
前記被分析物を、前記分析要素内に配設された前記固定相によって分離することと、および、
適切な移動相を使用して、前記被分析物を前記固定相から溶離することとを含む方法。 - クロマトグラフ用カラムを作る方法であって、
入口端部、出口端部、および内部に配設された固定相を有する第1キャピラリを獲得することと、
前記第1キャピラリよりも小さな直径の保持要素を、前記第1キャピラリの前記出口端部に装着し、かつ前記第1キャピラリよりも小さな直径の1つ以上の保持要素を、前記第1キャピラリの前記入口端部に装着することとを含み、前記1つ以上の保持要素が、前記入口端部で縦軸に沿って互いに隣接して配設され、前記保持要素のそれぞれの管腔が、連続する管腔域を形成するために前記入口端部で位置合わせされ、前記保持要素それぞれが、前記第1キャピラリから取り外されることができ、方法がさらに、
前記保持要素それぞれを、接着剤を使用することによって定位置に固定することを含む方法。 - 前記1つ以上の保持要素が、前記第1キャピラリに沿ってまたは前記第1キャピラリ内で隣接して配置される請求項13に記載の方法。
