Description
また、ここで言う「厳密に」とは生体や細胞の状態のより詳細な解析を指すばかりではなく診断等において試料間、測定装置間、施設間の差を超えて定量的な比較検討をするには試料中に設定した内部標準反応等を同時に測定しなければならず、例えある一つの酵素活性を病態診断に用いる場合でも内部標準反応として別種の反応の同時測定が行われなければならない事も指す。
また、酵素活性測定用人工基質などが多く開発されているものの、それらを並列してハイスループット的に使用することは困難である。なぜなら、従来技術では、測定対象が全く異なる場合でも用いる試薬が類似のものが殆どで同時に別種の活性は測定不可能な場合が殆どだからである。
また、量子ドットと蛍光分子を標識した生体分子により構成されるバイオプローブにおけるFRETを利用した解析方法もこれまでに報告されているが、効率的で測定対象に依存しないFRETの観測が困難性あるなどの理由から、測定精度を担保した利用は実現されていない(特許文献2、非特許文献3、非特許文献4及び非特許文献5)。
また、量子ドットなどを用いる点においても、広範な用途のための一般化された解析システムの安定性等における問題が存在していた。
さらに、GFPに結合させる蛍光色素でラベルした生体分子の分子量が大きい場合、GFPとの複合体を得る事は可能であるが、結合効率が極めて悪く実用化には適さないことから、種々の生体反応、酵素反応を測定するのに必要な測定分子(例えば、GFPと他の蛍光標識タンパク質との複合体、又はGFPと蛍光標識DNAとの複合体)を大量に効率よく取得するのが困難であった。
よって、本発明は測定環境に左右されず、かつ、複数の生体反応又は生体内状態変化を同時検出するための活性測定分子の安定した提供を目的とする。
また、本発明は該活性測定分子を使用した生体反応複数同時解析法の提供を目的とする。
すなわち、本発明は以下の(1)〜(19)に関する。
(1)本発明の第1の態様は、量子ドット上に、生体反応又は生体内状態変化の対象となる、蛍光分子で標識した生体分子及び/又は標識しない生体分子を1以上結合していることを特徴とする生体反応及び/又は生体内状態変化の複数同時解析用活性測定分子である。
(2)本発明の第2の態様は、前記生体分子がペプチドであり、該ペプチドが前記蛍光分子と共有結合で結合していることを特徴とする上記(1)に記載の活性測定分子である。
(3)本発明の第3の態様は、前記共有結合がペプチド結合であることを特徴とする上記(2)に記載の活性測定分子である。
(4)本発明の第4の態様は、前記量子ドットをアビジンでコートし、前記生体分子をビオチン化して、ビオチン−アビジン結合を介して、前記生体分子が量子ドットに結合することを特徴とする上記(1)乃至(3)のいずれかに記載の活性測定分子である。
(5)本発明の第5の態様は、前記蛍光標識した生体分子の数が0〜40であり、及び/又は前記蛍光標識していない生体分子の数が0〜40であることを特徴とする上記(1)乃至(4)のいずれかに記載の活性測定分子である。
(6)本発明の第6の態様は、前記生体分子が、核酸、タンパク質、ペプチド、アミノ酸、補酵素、糖、糖鎖、脂質、それらの誘導体、及びそれらの複合体からなるグループより選択されることを特徴とする上記(1)乃至(5)のいずれかに記載の活性測定分子である。
(7)本発明の第7の態様は、前記生体反応が酵素反応であることを特徴とする上記(1)乃至(6)のいずれかに記載の活性測定分子である。
(8)本発明の第8の態様は、前記酵素が、核酸分解酵素、核酸修飾酵素、タンパク質分解酵素、タンパク質修飾酵素、糖鎖分解酵素、糖ヌクレオチド合成酵素及び糖転移酵素からなるグループより選択されることを特徴とする上記(7)に記載の活性測定分子である。
(9)本発明の第9の態様は、前記酵素が、核酸分解酵素であり、前記生体分子が核酸であることを特徴とする上記(8)に記載の活性測定分子である。
(10)本発明の第10の態様は、前記酵素が、タンパク質分解酵素であり、前記生体分子がペプチドであることを特徴とする上記(8)に記載の活性測定分子である。
(11)本発明の第11の態様は、前記蛍光分子がBFP、CFP、GFP、YFP、RFPを代表とする蛍光蛋白質とその変異体から選択されることを特徴とする上記(1)乃至(10)のいずれかに記載の活性測定分子である。
(12)本発明の第12の態様は、上記(1)乃至(6)のいずれかに記載の第1の生体分子を結合した1以上の活性測定分子と蛍光分子で標識した又は標識しない1以上の第2の生体分子からなる試験サンプルを混合し、量子ドット上の第1の生体分子と第2の生体分子が相互作用するために適した条件下でインキュベートし、インキュベートの間又はインキュベート後、各量子ドットに適した波長で各量子ドットを励起し、各量子ドット上に結合している第1の生体分子及び/又は各第2の生体分子に結合した蛍光標識など、系に存在するすべての蛍光分子から得られる蛍光強度の変化を検出することを含んでなる、生体分子の相互作用複数同時解析方法である。
(13)本発明の第13の態様は、前記量子ドット上の第1の生体分子が、核酸、タンパク質、ペプチド、糖、糖鎖、それらの誘導体、及びそれらの複合体からなるグループより選択されることを特徴とする上記(12)に記載の方法である。
(14)本発明の第14の態様は、前記第2の生体分子が、核酸、タンパク質、ペプチド、糖、糖鎖、それらの誘導体、及びそれらの複合体からなるグループより選択されることを特徴とする上記(12)又は(13)に記載の方法である。
(15)本発明の第15の態様は、前記試験サンプルが細胞抽出液、細胞培養上清、血液、体液であることを特徴とする上記(12)乃至(14)のいずれかに記載の方法である。
(16)本発明の第16の態様は、上記(7)乃至(11)のいずれかに記載の1以上の活性測定分子の混合物と、活性検出の対象である1以上の酵素を混合し、該混合物を該酵素の活性に至適な条件下でインキュベートし、インキュベートの間又はインキュベート後、各量子ドットに適した波長で各量子ドットを励起し、各量子ドット及び各基質に結合した蛍光標識など系に存在するすべての蛍光分子から得られる蛍光強度の変化を検出することを含んでなる、酵素活性複数同時解析方法。
(17)本発明の第17の態様は、前記酵素の少なくとも1つがタンパク質分解酵素であることを特徴とする上記(16)に記載の方法である。
(18)本発明の第18の態様は、活性測定分子の混合物に、前記酵素の蛍光標識した第2の基質をさらに混合することを特徴とする上記(16)又は(17)に記載の方法である。
(19)本発明の第19の態様は、前記活性測定分子の混合物中に含まれる量子ドットの蛍光特性が相互に異なるものであることを特徴とする上記(12)乃至(18)のいずれかに記載の方法である。
ここで本発明の「活性測定分子」は、生体反応又は生体内状態変化等の対象となる生体分子、又は、該生体分子(例えば、タンパク質、核酸、脂質、糖など)の結合パートナー(例えば、タンパク質、核酸、脂質、糖など)などに蛍光分子標識したもの又は蛍光分子標識していないものを量子ドット上に結合させたものである。ここで、蛍光標識をしていないものを量子ドット上に結合させたものは、該蛍光標識をしていない生体分子と結合する第2の生体分子との結合様式を検出するために使用することができる。「生体分子」とは、生体内において、機能的又は構造的な役割を担って存在する全ての分子のことであり、例えば、核酸、タンパク質、ペプチド、アミノ酸、補酵素、糖、糖鎖、脂質、又はそれらの誘導体及び複合体(例えば、糖タンパク質、糖脂質など)、各種酵素(例えば、核酸分解酵素、核酸修飾酵素、核酸合成酵素、タンパク質分解酵素、タンパク質修飾酵素、糖鎖分解酵素、糖ヌクレオチド合成酵素又は糖転移酵素)、酵素反応の基質(例えば、タンパク質、核酸、脂質、糖など)などが含まれる。
ここでいう「生体反応」には、生体分子同士の反応の全てが含まれ、例えば、酵素反応などがその典型的な例である。また、「生体内状態変化」とは、化学反応は生じない場合であっても、生体分子同士の相互作用などの結果、1の生体分子が分解したり、2の生体分子が結合して一体化するような状態変化の全てが含まれる。さらに、生体分子同士の相互作用のみならず、測定対象である生体分子の濃度(反応中に起きる関連分子の濃度変化など、例えば、カルシウムイオン濃度変化、プロトンの濃度変化であるpH変化など)や膜電位、酸化還元状態なども本発明の生体内状態変化に含まれる。
本発明の活性測定分子に用いられる生体分子に蛍光標識色素(蛍光分子)を導入する場合には、量子ドットとの間で蛍光共鳴エネルギー移動(FRET)または蛍光2分子のゆらぎを検出する(FCCS)ことができるような蛍光標識色素であれば、いかなるものでも利用可能である。限定はしないが、例えば、そのような蛍光標識色素として、Alexa dye、BODIPY、Cy dye、quencherなどが好適に利用可能である。
また、上記抗体やアビジンでコートされた量子ドットを用いる方法以外では、例えば、量子ドット上に存在する官能基(例えば、−COOH、−NH2など)を介して結合させる方法なども利用可能である。これらの方法に適した量子ドットが市販されており、これらの市販品を使用することもできる。
蛍光タンパク質とペプチドを共有結合させる場合、両者の位置関係は、反応の前後で大きなFRET変化、FCCSなどが観測可能な位置関係にある必要がある。例えば、蛍光タンパク質として、GFP、RFPやその色変異体などを使用する場合、GFP、RFPのN末端、C末端、11本のβ鎖をつなぐ11本のループ領域のいずれかに挿入することができる。この場合、当該ペプチドと蛍光タンパク質又は量子ドットとを直接結合させてもよいが、場合によっては、スペーサーとなるいくつかアミノ酸(1〜20程度)を挿入してもよい。
量子ドット上に標識された生体分子が結合している状態において、最も高い効率のFRETが検出される。また、FCCS法を適用する場合、量子ドットと標識された生体分子が同一分子としてのゆらぎのパターンが検出される。ここで活性測定分子に含まれる生体分子がタンパク質、核酸、糖鎖などの場合に、タンパク質分解酵素、核酸分解酵素、糖鎖分解酵素で処理すると、生体分子が切断されることで、生体分子に標識された蛍光分子が量子ドットから解離するため、FRETが解消され、また、2つの蛍光分子が別の分子としての挙動を示しゆらぎのパターンが変化する。この過程を蛍光分光光度計によるFRET、蛍光寿命、時間分解蛍光等の測定、蛍光相互相関分光計によるFCCSの測定及びそれらの測定対応蛍光マイクロプレートリーダーを用いて解析することで酵素活性のモニターが可能である。
さらに、タンパク質同士、タンパク質と核酸、核酸同士、タンパク質と糖鎖などの結合反応に関しては、一方の生体分子を蛍光標識せずに量子ドット上に結合させた活性測定分子を調製し、該活性測定分子と結合パートナーとなる蛍光標識した生体分子を接触させた結果、FRETの最も低い状態から高い状態への変化が検出されるか、または、独立した蛍光分子としてのゆらぎパターンが1つの大きな分子上にある2つの蛍光分子としてのゆらぎパターンに変化した場合には、両生体分子が結合または相互作用し得ると判断することができる。
従って、異なる生体反応又は生体内状態変化を測定するための生体分子を結合させた蛍光特性の異なるドットを共存させた場合においても、同一の波長で異なる蛍光特性を有する量子ドットの全てを励起することができることから、複数の生体反応を同時にモニターすることができる。例えば、1の量子ドットには、蛍光標識したDNAを結合させ、2の量子ドットには別の蛍光標識を行ったタンパク質を結合させ、3の量子ドットには蛍光標識しない糖鎖を結合させて活性測定分子を調製した場合、DNA分解酵素、タンパク質分解酵素、糖転移酵素及び蛍光標識した糖を含んだ試験サンプル(精製した酵素などの混合物、または細胞抽出物、培養上清など)を該活性測定分子と混合し、適切な条件下でインキュベートしている間に励起波長を照射することによって得られる蛍光波長の特性変化を、FRET変化検出法又はFCCS法によって解析することにより、上記3種の酵素活性を確認することが可能となる。さらに、蛍光標識しないタンパク質を結合させた量子ドットと蛍光標識したタンパク質の混合ではタンパク質間相互作用を検出することができ、生体分子との結合(金属イオンをキレートするなども含め)によりその蛍光特性が変化する環境依存型蛍光色素を適切なスペーサー分子等を用いて量子ドットに結合させた場合には、該生体分子の濃度変化をFRET変化検出法に適応する事も可能である。
以上のように、本発明の方法は、多くの生体反応又は生体内状態変化の複数同時解析に適用することができる。
キット中に含まれる活性測定分子は、構成成分が活性を長期間有効に持続し、容器の材質によって吸着されず、変質を受けないような何れかの種類の容器中に供給される。例えば、封着されたガラスアンプルは、窒素ガスのような中性で不反応性ガスの下において包装されたバッファーを含む。アンプルは、ガラス、ポリカーボネート、ポリスチレンなどの有機ポリマー、セラミック、金属、又は試薬を保持するために通常用いられる他の何れかの適切な材料などから構成される。他の適切な容器の例には、アンプルなどの類似物質から作られる簡単なボトル、及び内部がアルミニウム又は合金などのホイルで裏打ちされた包装材が含まれる。
DNA分解酵素の活性測定
1.活性測定分子の調製
DNA分解酵素の活性を測定するための実施例を示す。
基質となる二重鎖DNAは、5‘AGAACCACTCCCAGCAGCAGTTACAAACTC3‘(配列番号1)及び5‘ACTCCAATTGGCGATGGCCCTG3‘(配列番号2)のプライマーセットを使用し、UV5casS22tagをコードしているプラスミド( 非特許文献2を参照のこと)をテンプレートにしてPCR法により一部を(およそ150mer)増幅し、増幅された産物は、電気泳動により塩基長の確認を行った。具体的には、dUTPAlexa(Alexa Fluor 568-5dUTP又はAlexa Fluor 532-5dUTP, Moleclar Probes社)を、PCR反応液に加え(dATP、dCTP、dGTPを各0.2mM、0.14mM dTTPを含み、Alexa-dUTPは、0.05mM濃度になるように添加した)、94℃2分後、94℃30秒、63℃30秒、72℃30秒を30サイクル、最後に72℃7分の反応条件にてPCR反応を行わせ、Alexa dyeで標識したDNaseの基質となるDNAを調製した。また、配列番号1又は2で示される配列からなるプライマーの末端をビオチンラベルしておくことで、アビジンコートした量子ドット(住商バイオサイエンス株式会社)と容易に結合することができた。この結合反応は、PCR後精製した二本鎖DNAと量子ドットを添付バッファー中にて(組成2% BSA in 50mM borate, pH8.3 with 0.05% sodium azide)室温で2時間インキュベートの条件で行った。量子ドットはアビジンコートにより多価となっているため、複数の基質DNAの結合が可能である(図1)。
1分子の量子ドット(FRETのドナー)に対し複数個Alexa dyeでラベルされた多数の基質DNA(FRETのアクセプター)が結合しても量子ドットの量子収率は極めて高く効率の良いFRETが観察された(図2、図3)。このようにして調製した活性測定分子にDNase(Promega社)を混合し、37℃にて60分インキュベートし、基質DNAの分解反応を行わせたところ、FRETの解消が観察された(図2、図3)。
本測定を量子ドットの代わりにGFPなどを用いると、GFPとDNAを一対一で結合させた場合、DNAに取り込まれた複数の蛍光ラベルが、GFPの蛍光量子収率以上(ラベルされた蛍光分子の蛍光のみとなる)になると高効率のFRETが観察されず、基質として使用することができなくなる欠点を呈することとなる。
さらに、GFPとDNAを有機化学的に共有結合させる場合、タンパク質と核酸の効率の良い結合方法はこれまでに殆ど知られていない。最近、150merまでのdsDNAをタンパク質に効率よく結合させる方法が報告された。しかしながら、この方法論では煩雑なステップを経なければならない事やモニターしたいDNaseの種類によっては活性測定プローブとして成立するためには150mer程度では不十分であると考えられることから従来の方法では限界がある。それに対し、本実施例は簡潔なステップであるばかりではなく、より長鎖にすぐさま応用可能であると思われる。
DNAポリメラーゼ活性の測定
1.活性測定分子の調製
活性測定分子は、量子ドット(QD525)上に、予めプライマーをハイブリダイズさせたssDNA(一本鎖DNA)を、上述の方法により固定化して調製した。
2.DNA分解活性の測定
調製した活性分子に対し、DNAポリメラーゼとして、Klenow fragment(クレノウフラグメント)を用いて、DNA鎖伸長反応を行わせた。その結果、DNAポリメラーゼで処理する前は、FRETを観察することができなかったが(最低の状態)、DNAポリメラーゼ処理により、FRETが観察されるようになった(最高の状態)(図4)。
トリプシンの活性測定
1.トリプシン感受性部位のGFP標識及び活性測定分子の調製
本実施例、生体分子としてトリプシン感受性部位を含むペプチドを用い、トリプシンの活性測定を行ったものである。
トリプシン感受性部位を含むペプチド(配列番号3)が、図5Aに示すようにGFPのC末端領域に挿入され、さらにそのC末端側にHisタグが付加されるように、GEP−トリプシン認識ペプチド挿入改変タンパク質の発現ベクターを構築した。本発現ベクターは、非特許文献1に示したがpET21a(NOVAGEN社)にPCRによりGFPUV5配列を組み込みさらにそのC末端領域に、配列番号4で示されるアミノ酸配列をコードするDNA断片を挿入することで構築した。構築した発現ベクターを用い、GEP−トリプシン認識組換えタンパク質を発現させ、Hisタグを利用して、認識ペプチド挿入組換えタンパク質精製を行った。
アミン(Amine)処理された量子ドット(Amine Evitag Lake Placid Blue, Evident Technologie社)とアミン基と反応可能なホモ二価性架橋試薬をインンキュベートし(0.1μM 炭酸バッファー中)、未反応のホモ二価性架橋試薬を限外濾過スピンカラムにて除去する。得られた量子ドットとNi−NTAのアミン誘導体をさらにインキュベートする(0.1μM 炭酸バッファー中)。Ni−NTAの結合した量子ドットより限外濾過スピンカラムにて未反応のNi−NTAのアミン誘導体の除去及びバッファー置換を行い(リン酸バッファー)精製した認識ペプチド挿入組換えタンパク質とインキュベートすると、Hisタグ部位を介して、認識ペプチド挿入組換えタンパク質を量子ドット上に固定化することができる。
上述のように調製した活性測定分子にトリプシンを添加し、37℃120分のインキュベートを行ったところ、FRETの解消が観察された(図6)。つまり、蛍光強度のピーク波長が、トリプシン処理によって513nm(トリプシン未処理)から508nm(トリプシン処理)に変化した。
従来、量子ドットを用いたタンパク質分解酵素活性の測定においては、酵素と基質とのインキュベーションに極めて長時間(〜47時間)必要であったため、真の酵素活性を反映しているか疑わしい点が存在していた。これに対し、本実施例において、本発明の構成をとることにより、合理的なインキュベーション時間により、明確な測定結果が得ることができた。
キャスパーゼ−3の活性測定
1.キャスパーゼ−3感受性部位のGFP標識及び活性測定分子の調製
本実施例、生体分子としてキャスパーゼ−3を含むペプチドを用い、トリプシンの活性測定を行ったものである。
キャスパーゼ−3感受性部位を含むペプチド(配列番号5)が、図5Bに示すようにGFPのC末端領域に挿入され、さらにそのC末端側にHisタグが付加されるように、GEP−キャスパーゼ−3認識ペプチド挿入組換えタンパク質の発現ベクターを構築した。本発現ベクターは、上述のトリプシン活性を測定する場合と同様にして調製した。
2.キャスパーゼ−3の活性測定
上述のように調製した活性測定分子にキャスパーゼ−3を(Medical & Biological Laboratories社)添加し、30℃120分のインキュベートを行ったところ、FRETの解消が観察された(図7)。
従来、量子ドットを用いたタンパク質分解酵素活性の測定においては、酵素と基質とのインキュベーションに極めて長時間(〜47時間)必要であったため、真の酵素活性を反映しているか疑わしい点が存在していた。これに対し、本実施例において、本発明の構成をとることにより、合理的なインキュベーション時間により、明確な測定結果が得ることができた。つまり、蛍光強度のピーク波長が、キャスパーゼ−3処理によって510nm(トリプシン未処理)から506nm(トリプシン処理)に変化した。
なお、キャスパーゼ−9(図5Cに、実験に使用したGEP−キャスパーゼ−9認識ペプチド挿入改変タンパク質のアミノ酸配列を示す)、カテプシン−E(図5Dに、実験に使用したGEP−カテプシン−E認識ペプチド挿入改変タンパク質のアミノ酸配列を示す)に関しても同様に使用可能である。
また、上記トリプシン及びキャスパーゼ−3の実施例はいずれもGFPを用いているがRFPを用いた場合も概ね同様の結果が得られている。さらに、GFP又はRFPのN末端側に酵素認識ペプチドを配置した場合も、同様の結果に使用可能である。
pH変化の測定
アミン処理された量子ドット(Amine Evitag Lake Placid Blue , Evident Technologies社)にTraut試薬を反応させアミン基をチオール基に変換する。この量子ドットとpH感受性蛍光色素(慶応大学小松広和博士より分譲)のマレイミド誘導体(チオール基反応性)をリン酸バッファー中でインキュベートし量子ドット上にpH感受性蛍光色素を結合させる。未反応のpH感受性蛍光色素をゲル濾過カラムにて除去する。得られたpH感受性蛍光色素結合量子ドットをそれぞれpHモニター用バッファーに分散させ、量子ドットを励起し蛍光スペクトルを測定した。ここで、量子ドット単体、pH感受性蛍光色素単体も同様のpHモニター用バッファー中にてそれぞれ蛍光スペクトルを測定した。この結果量子ドット上に固定化した場合のみpHに依存した大きな蛍光スペクトル変化が得られ(図8、9、10)、pH変化測定用分子を作製出来た事が示された。
酵素活性の複数同時検出
本実施例は、本発明の構成を用いて、核酸分解酵素とタンパク質分解酵素の各々の活性を同時に検出した結果を示す。
DNA分解酵素活性の活性測定分子は、量子ドットとしてQD525を、蛍光分子としてAlexa532を用い、〔実施例1〕と同様な方法で調製を行った。一方、タンパク質分解酵素(ここでは、トリプシンを用いた)の活性測定分子は、量子ドットとしてQD565を、蛍光分子としてGFP(図5A)を用い、〔実施例2〕と同様な方法で調製を行った。調製した活性測定分子を4つのチューブに分注して、DNase未処理及びトリプシン未処理(図11、−DNase−Trypsin)のチューブをコントロールとして、DNase処理及びトリプシン処理(図11、+DNase+Trypsin))、DNase処理及びトリプシン未処理(図11、+DNase−Trypsin))、DNase未処理及びトリプシン処理(図11、−DNase+Trypsin))を行ったチューブに対しFRETの測定を行った。その結果、図11に示す通り、DNase未処理及びトリプシン未処理のチューブのピーク波長から、各処理によって検出される蛍光強度のピーク波長のずれが観察された。詳細には、本実施例の核酸分解酵素及びタンパク質分解酵素の同時測定においては、−DNase−Trypsinの波形パターンを基本とし、各酵素処理による波形パターンの変化を観察し、各酵素の活性状況をモニターする。510、560、600nm付近に3つのピークが存在する。+DNase−Trpsinでは、核酸分解酵素のみの添加であるが、600nm付近のピークが減少し560nm付近のピークが増加した。−DNase+Trpsinではトリプシンのみの添加であるが、510nm付近のピークがずれているが観察された。+DNase+Trpsinは、両酵素を添加した場合であるが、上記2点の変化の両方が観察された。なお、+Trpsinでトリプシンを添加した場合、 560nm付近のピークの増加は、DNAを固定化している方の量子ドットの蛍光波長が観察されたものと思われるが、本実施例の観察方法(コントロールに対して波形パターンの変化を観察する方法)においては、特に、支障は出なかった。
以上の結果から、本発明の構成を用いることで複数の生体内反応を同時に検出できることが分かった。
Claims (9)
- 量子ドット上に蛍光分子を標識した酵素反応の基質となる生体分子が1以上結合していることを特徴とする酵素活性測定分子の2種類以上の混合物であって、該酵素活性測定分子の少なくとも1種類は、該蛍光分子が蛍光タンパク質である混合物と、測定対象となる1種類以上の酵素を含む試験サンプルを混合し、活性測定分子に含まれる生体分子と試験サンプルに含まれる酵素が反応する条件下でインキュベートし、インキュベートの間又はインキュベート後、各活性測定分子に含まれる量子ドットの励起波長で各量子ドットを励起し、各活性測定分子に含まれる量子ドット及び蛍光分子から得られる蛍光強度の変化を検出することを含んでなる、複数の酵素活性の同時解析方法。
- 前記活性測定分子に含まれる生体分子が、核酸、蛋白質、ペプチド、糖、糖鎖、それらの誘導体、及びそれらの複合体からなるグループより選択されることを特徴とする請求項1に記載の方法。
- 前記試験サンプルが細胞抽出液、細胞培養上清、血液、体液であることを特徴とする請求項1又は2のいずれかに記載の方法。
- 前記酵素反応の少なくとも1つが蛋白質分解酵素反応であることを特徴とする請求項1乃至3のいずれかに記載の方法。
- 前記酵素反応の少なくとも1つが核酸分解酵素反応であることを特徴とする請求項1乃至3のいずれかに記載の方法。
- 前記活性測定分子の混合物中に、相互に異なる蛍光特性をもつ量子ドットが含まれることを特徴とする請求項1乃至5のいずれかに記載の方法。
- 前記活性測定分子の混合物中に、相互に異なる蛍光特性をもつ蛍光蛋白質又は蛍光分子が含まれることを特徴とする請求項1乃至6のいずれかに記載の方法。
- 前記蛍光蛋白質が、BFP、CFP、GFP、YFP、RFPからなる蛍光蛋白質又はその変異体から選択されることを特徴とする請求項1乃至9のいずれかに記載の方法。
- 量子ドット上に、蛍光タンパク質で標識した酵素反応の基質となる生体分子が1以上結合している酵素活性測定分子の2種類以上を含んでなる、複数の酵素活性の同時解析用キット。
