カロチノイド及びルテイン含有量の分析方法
本発明に従って製造したマイクロカプセル類は、カロチノイド含有量を分析する。この分析方法は、わずか約5〜7mgカロチノイド類試料の使用を必要とする。従って、かかる方法の最初に添加する試料の量は、期待量がこの範囲となるように選択する。10重量%のカロチノイド類を含有するとされるマイクロカプセルを使用する場合、75mgのマイクロカプセルの試料を計量し、止め具を備えた50mlのガラスの遠心分離管に添加する;25%のカロチノイド類を含有するとされるマイクロカプセルを使用する場合、30mgの試料を計量し、止め具を備えた50mlのガラスの遠心分離管に添加する;以下同様である。約20mlの0.1 M EDTA/2NaH2O(37.224g/L)を添加する。止め具を管に取り付け、管を60℃の水槽で20分間加熱する。管を水槽から取り除き、時々管を回しながら15分間超音波処理する。管を室温まで冷却させ、次に30mlのジクロロメタンをピペットで添加する。止め具を付けて内容物を繰り返し反転させて混合し、通気のために止め具をたびたび取り外す。管を暗所に置き、層を約30分間分離させる。ピペットを使用して最上層の水を吸い上げ廃棄する。2〜3gの硫酸ナトリウムをジクロロメタン層に添加し、取り付けた止め具及び管を穏やかに振る。硫酸ナトリウムは沈殿させ、1mlのジクロロメタン層をピペットで取り除いて100mlの容量フラスコに添加し、印までエタノールで希釈する。エタノール混合物を、あらかじめエタノールに対してゼロに合わせた紫外可視分光光度計(Agilent, Palo Alto, CA)に入れる。446nmでの吸光度を読み取る。全カロチノイド類の割合は、以下の式に従って決定する:
本プロセスは、Coating Place, Inc., Verona, Wisconsinからの4"/6"移動式流動床コーターを使用して実施した。コーターは、バッチあたり0.60〜1.7Lの容量を有する。カロチノイド類の供給源は、Kemin Industries, Inc., Des Moines, IowaからのFloraGLO(登録商標)ブランド結晶性ルテインであった(ここでは「ドライケーキ」と称する)。このカロチノイド類の供給源を分析すると、典型的な結果として製品が85%のカロチノイド類(78%のルテイン、5%のゼアキサンチン、及び2%の他のカロチノイド類)、8%のパラフィン蝋類、及び1%のパルミチン酸塩のような脂肪酸を含むことがわかった。
Claims (5)
- マイクロカプセルの摂取時にカロチノイドを放出する保護コーティングを有する、一つ以上のカロチノイドの結晶性形状のマイクロカプセルを形成する方法であって、以下のステップ:
(a) 保護コーティングを有さない最初は保護されていない結晶性カロチノイドを含有する出発原料の循環流を生成するコーティング装置で、カロチノイドを含有する出発原料を循環させること;
(b) 糖、デンプン又はマルトデキストリン、及び場合によりコーティング蛋白質を含む液体コーティング材を調製すること;並びに
(c) 出発原料に保護コーティングを形成するため、循環するカロチノイドを含有する出発原料に液体コーティング材を噴霧すること
を含む、前記方法。 - マイクロカプセルに存在するカロチノイドの量が、出発原料中のカロチノイドの量の80%より大きい、請求項1に記載の方法。
- 糖がグルコース、フルクトース、ガラクトース、スクロース、ソルビトール、及びラクトースからなる群から選択され、デンプンがタピオカデンプン、コーンスターチ、じゃがいもデンプン、及びアロールートデンプンからなる群から選択され、蛋白質が牛、豚、及び魚のゼラチンからなる群から選択される、請求項1又は請求項2に記載の方法。
- 一つ以上のカロチノイドが、アクチニオエリスロール、アスタキサンチン、ビキシン、カンタキサンチン、カプサンチン、カプソルビン、β−8’−アポ−カロテナール、β−12’−アポ−カロテナール、α−カロチン、β−カロチン、α−及びβ−カロチンの混合物、γ−カロチン、β−クリプトキサンチン、ルテイン、リコピン、ビオレリトリン、ゼアキサンチン、及びそれらのうちヒドロキシル又はカルボキシルを含有するもののエステルからなる群の一つ以上を含む、請求項1乃至請求項3のいずれか1項に記載の方法。
- マイクロカプセルのカロチノイド含有量が5%〜20重量%である、請求項1乃至請求項4のいずれか1項に記載の方法。
