Description
a)E1B55k、E3ADPならびにE4orf6及びE4orf4を含む群から選択される、YB−1核陽性細胞内の少なくとも1つのウイルス性遺伝子がトランス活性化される;そして
b)細胞核内、特にウイルス性癌遺伝子タンパク質が存在する細胞の細胞核内で、YB−1が誘導されない。
B群:核にYB−1を有さず、特に細胞周期とは無関係ではないが、調節解除されたYB−1を含む細胞;及び
C群:核にYB−1を有さず、特に細胞周期とは無関係ではなく、調節解除されたYB−1を含まない細胞
−YB−1が細胞周期とは無関係に核に局在するかどうか又は細胞が調節解除されたYB−1を含有するかどうかを決定すること。
図1はAdE1/E3マイナス、即ちE1/E3欠失アデノウイルスと野生型アデノウイルスおよびアデノウイルスdl520の構造デザインを示している。
図2はp300,p107およびp105の結合に関係するE1Aタンパク質の結合ドメインを示している。P300およびP107は、細胞質結合タンパク質である。腫瘍抑制タンパク質である網膜芽細胞腫タンパク質(pRb)の結合は、CR1およびCR2を通じて媒介される。研究からpRbおよびp107/p300は細胞転写因子E2Fと組み合わさり効率的に転写制御することが示されている。野生型E1Aタンパク質はE2FのRbへの結合を妨害する。即ち放出されたE2FはE2初期プロモーターに結合して、アデノウイルスの複製を誘導する。
100,000個のU2OS細胞をウエルに播いた。翌日細胞に図3に示すような各種アデノウイルスを感染させた。感染は無血清DMEM培地500μl中で、37℃で1時間行った。続いて感染培地を取り除き、完全培地(10%FCS/DMEM)2mlと交換した。3日後にクリスタルバイオレット染色で分析を行った。
100,000個の257RDB細胞をウエルに播いた。翌日細胞に図4に示すような各種アデノウイルスを感染させた。感染は無血清DMEM培地500μl中で、37℃で1時間行った。続いて感染培地を取り除き、完全培地(10%FCS/DMEM)2mlと交換した。3日後にクリスタルバイオレット染色で分析を行った。
図5に示すように、E1Aタンパク質のアミノ酸4〜138およびそれをコードする核酸を欠失しており、さらにアミノ酸218の後に停止コドンを含むために発現する断頭型(truncate)E1Aタンパク質が完全なE1Aタンパク質のCR3領域を含んでいるアデノウイルスdl1119/1131をYB−1核陰性U2OS細胞に感染した場合、20pfu/細胞のMOIでは溶解は起きなかった。陰性コントロールには非感染細胞層を用いた。
本実施例は核内YB−1が転写因子としてmdr1プロモーター(多剤耐性プロモーター)内にあるY−ボックス(CAAT配列)に結合しなければならないという考察に基づいている。これを検出するために、いわゆるEMSA分析(電気泳動移動度シフトアッセイ)を行った。これに関連して、核タンパク質を単離し、続いてタンパク質1〜10μgを短いDNA鎖(オリゴ)と共の37℃でインキュベーションした。核内YB−1を決定するために以下のオリゴヌクレオチドを使用した;U2O3と反対のmdr1プロモーター(位置−86〜−67):TGAGGCTGATTGGCTGGGCA(Xボックスには下線を付した)。
本実施例は初期ウイルス遺伝子E1B−55KおよびE4orf6をプラスミドpE4orf6によるトランスフェクションおよびE1/E3欠失アデノウイルスAd−55Kによる感染で置換えられることを示す。Ad−55KはE1B−55KをE1にクローニングし、CMVの制御下に置いたE1/E3欠失ウイルスである。この置換は、AdYB−1、即ちYB−1を発現するアデノウイルスがこれら初期遺伝子を発現していないという事実、および本発明者が核内にYB−1を含む複製系でのこれら初期遺伝子を置換することで複製効率および粒子形成能率をそれぞれ野生型アデノウイルスであるAd5型のそれと同程度まで上げることができることを認識していたという事実より必要であった。
リポフェクタミンを用いた各105個のU2OS細胞へのプラスミドpE4orf6のトランスフェクション。プラスミドpEorf6はCMV制御下にある初期ウイルス遺伝子E4orf6をコードするDNA配列を保持している。プラスミドpE4orf6をトランスフェクションした24時間後に、細胞にYB−1発現E1/E3欠失アデノウイルスAdYB−1(50pfu/細胞)およびE1/E3欠失E1B−55KアデノウイルスAd−55K(50pfu/細胞)を感染させた。Ad−55KはCMV制御下にあるウイルス遺伝子E1B−55Kをトランス遺伝子として保持するE1/E3欠失ウイルスである。
各種細胞増殖抑制薬の添加がヒト転写因子YB−1の核内局在化を誘導することは、先行技術において知られていた。本発明者が見出したように、核内に局在したYB−1は、アデノウイルスのE2−後期プロモーターの活性化によりアデノウイルスの複製をコントロールする。両効果の組み合わせを利用すれば特異的な腫瘍溶解を得ることができる。
本インビボ試験で使用したHeLa(YB−1核陰性)および257RDB(YB−1核陽性)細胞は、無菌細胞培養条件にて拡大した。細胞をマウス(系統CD1NuNu)に注射して皮下腫瘍を形成させる前に、細胞をトリプシン処理して集め、DMEM培地(10%FCS)に加え、細胞数を測定してからPBSで1回洗浄した。続いて細胞を遠心分離してPBSを除き、細胞を希望数する細胞数になるよう新鮮PBS中に分配した。本研究で皮下注射した細胞の数は両細胞系統とも5×106細胞であった。注射は動物の一方の側腹部に行い、より区別し易いようにHeLa細胞は右側に、そして257RDB細胞は左側に注射した。腫瘍の増殖を週2回調べ、ノギスを使って腫瘍の長さおよび幅を測定した。その結果を用い、次式より腫瘍の容積を計算した:

実施例9で発育させた腫瘍の中央部から取り出した腫瘍試料からDNAを抽出した。単離のためにQiagenのDneasy Tissue Kitを用いた。製造元の指示書に従ってDNA抽出を行った。それに従って、アルカリ溶解を介して細胞からDNAを取り出した。続いて、単離したDNAをカラムで精製した。続いて、260nmの光度測定により単離したDNAの濃度を決定した。制限酵素KpnI、10単位で消化したDNA試料2μgを用いて解析を行った。続いて、0.8%アガロースゲルで試料を電気泳動的に分離した。続いて、DNAをナイロン膜にブロットした(Schleicher & Schuellの系に従って)。膜にブロットさせたDNAを特異的な1501bpのDNAプローブとハイブリッド形成させた。1501bpのプローブは、E2AをコードするAd5の配列の中で3369bpのKpnI断片に特異的に結合する。プローブはそれに先立ってPCRで調製し(プライマー:5’−GTCGGAGATCAGATCCGCGT(配列番号2)、5’−GATCCTCGTCGTCTTCGCTT(配列番号3))、32Pを用いて放射性標識を行った。続いて、膜を洗浄し、フィルムに暴露した。
100、000〜200、000個の各細胞を6個のウエルを持つプレート(6ウエルプレート)、FCSを10%含むL15培地(耐性細胞)およびDMEM(非耐性細胞)に接種した。24時間後にdl520および野生型アデノウイルスを感染させた(10pfu/細胞)。感染3日後(インフェクション後)、凍結と融解を3回繰り返して細胞浮遊液よりウイルス粒子を放出させた。次に293細胞を使ってプラークアッセイを行い、形成した感染粒子(プラーク形成単位/ml(pfu/ml))を決定した。結果を図15に示す。プラークアッセイの結果は、dl520がYB−1核陽性細胞(257RDBおよび181RDB)では野生型アデノウイルスの場合と同様に複製することを示した。この限りにおいて、本発明に従ってここに記載のアデノウイルスを使用する場合には、野生型アデノウイルスと同様の複製効率が観察できた。
図16はアデノウイルスベクターXvir03の構造デザインを示す。アデノウイルスXvir03は、いわゆるE1/E3欠失アデノウイルスである。このことは、アデノウイルス複製に機能するE1A、E1BおよびE3タンパク質が作られないことを意味する。E1領域の欠失は342〜3528の範囲である;アミノ酸位置27865〜30995のE3領域の欠失。本明細書で使用する場合、用語「E1欠失ウイルス」とは、E1がもはや機能的に作用しないウイルスを意味する。これは、その他の核酸配列およびアミノ酸配列は未変性な状態で不活性化することにより達成できるが、しかしこのことは欠失したE1領域がコードするタンパク質が様々なサイズを持つことを意味することにもなる。E1AおよびE1Bタンパク質およびそれらをコードする核酸を欠くことにより、E4orf6の様なE4領域は弱くにしか発現されないか(野生型アデノウイルスの1〜5%)、または全く発現されない。ウイルス遺伝子E1B55kおよびE4orf6はXvir03内に導入された異種のCMVプロモーター(Clontech: Plasmid pShuttle)によりE1領域内で発現する。上記CMVプロモーターに代わって、E1Aの発現に関連して明細書中に開示したその他プロモーターを用いることができる。両遺伝子のオープンリーディングフレームは、いわゆるIRES配列(内部リボソーム進入部位)(Plletier, J.およびSonenberg, N.Nature、1988年、334、320〜325)により互いに連結している。このエレメント(Novagen:pCITE)は1種類のmRNAから2種類のタンパク質を発現させる。
プラスミドE1B55k−pShuttleを、M.Dobelstein(マールブルグ大学)のpCGNE1Bより、XbaIおよびBfrIを用いてE1B55kのオープンリーディングフレームをClontech社製のpShuttleベクター内にクローニングして作製した。続いてpShuttle内のE1B55kをApaIを使って直線化し、末端を平滑末端とし、NheIを使って切断した。
図17から分かるように、Xvir03/01はXvir03を更に発展させたものである。例えばここに記した遺伝子の様な治療目的の遺伝子やトランス遺伝子をE3領域内にクローニングできる。さらに、E4領域内に欠失を導入してXvir03の発現カセットに由来するE4orf6との間で相同的組換えが起こるのを回避した。これによってより大型のトランス遺伝子(導入遺伝子とするか??)をこの構築体内にクローニングすることが可能になった。欠失型のE3領域にはカセット導入に適したSacI、NdeIおよびNheI制限部位があり、ここに例えば治療用トランス遺伝子をクローニングすることができる。
Clontech社製pAdenoX−Plasmidには3’ITR領域の後に野生型アデノウイルスには無いSfuI向けの制限部位がある。E3〜E4領域をSpeI(位置23644)およびSfuIを使ってpAdenoX(Clontech)から取り出し、pcDNA3.1(+)(Invitrogen)内にトランスフェクションした=pcDNA3.1−E3Δ27865−30995−E4。E4ORF6の大部分、即ち33241〜33875をPstIを用いて取り出した=pcDNA3.1−E3Δ27865−30995、E4Δ33241−33875。Xvir03を更に発展させるために、pcDNA3.1−E3Δ27865−30995、E4Δ33241−33875由来の欠失E3/E4領域をSfuIおよびSpeIを使ってプラスミドpAdenoX内にクローニングした=pAdnoXE3Δ27865−30995、E4Δ33241−33875。
100、000個の細胞(257RDBおよび181RDB)を6個のウエルを持つプレート(6ウエルプレート)の各ウエルに接種した。翌日細胞に、図18に示すようにAd312(20pfu/ml)およびXvir03(5pfu/ml)を感染させた。感染は無血清DMEM培地500μl中で、37℃、1時間実施した。次に感染培地を取り除き、完全培地(10%FCS/DMEM)2mlと交換した。5日後にクリスタルバイオレット染色による分析を行った。結果を図18Aおよび18Bに示す。
各場合において、100万個のA549細胞及びU20S細胞を10cmのペトリ皿に入れた。翌日、Ad312(50pfu/細胞)及びAdwt(対照として働く、5pfu/細胞)を細胞に感染させた。使用した高いウイルス力価のAd312は結果として、腫瘍細胞でE1非依存性の複製を生じた。感染は、1〜2mLの無血清DMEM培地にて37℃で1時間行った。続いて、感染培地を取り除き、10mLの完全培地(10%FCS/DMEM)に取り替えた。3日後、RNAを単離した。続いて、260nmでの光度測定により単離したRNAの濃度を測定した。次いで、0.8%ホルムアルデヒドアガロースゲルにてRNA試料を電気泳動的に分離した。続いて、RNAをナイロン膜にブロットした(Schleicher & Schuellの系に従って)。膜にブロットしたRNAを「早期プローブ」E2及び「後期プローブ」E2に対してブロットした。1501bpの「後期プローブ」はE2後期プロモーターの後に特異的に結合する。それに先立って、プローブをPCRで調製し、
32Pを用いて放射性標識を行った。対照的に、早期プローブはE2早期プロモーターとE2後期プロモーターの間(226791〜227002位)に結合し、PCR
によって生成した。続いて膜を洗浄し、フィルムに暴露した。
それぞれ100万個のU20S細胞を10cmのペトリ皿に入れた。翌日、アデノウイルスデルタ24(Adデルタ24)(10pfu/細胞)及び野生型アデノウイルス(Adwt)(対照として働く、10pfu/細胞)を細胞に感染させた。使用した組換えアデノウイルスAdデルタ24(Fueyo J. et al., Oncogene 19:2-12, 2000)はE1Aタンパク質のCR2領域に特異的な欠失を有するので、Rb陰性の腫瘍でのみ複製可能である。さらに、ウイルスは、野生型アデノウイルスに匹敵する遺伝子E1B55K及びE4orf6を発現している。感染は、1〜2mLの無血清培地にて37℃で1時間行った。続いて、感染培地を取り除き、10mLの完全培地(10%FCS/DMEM)に取り替えた。12時間後及び24時間後、RNAを単離した。続いて、260nmでの光度測定により単離したRNAの濃度を測定した。次いで、0.8%ホルムアルデヒドアガロースゲルにてRNA試料を電気泳動的に分離した。続いて、RNAをナイロン膜にブロットした(Schleicher & Schuellの系に従って)。膜にブロットしたRNAを「早期プローブ」及び「後期プローブ」に対してハイブリッド形成させた。1501bpを含む「後期プローブ」はE2後期プロモーターの後に特異的に結合する。それに先立って、プローブをPCRで調製し、
32Pを用いて放射性標識を行った。対照的に、早期プローブはE2早期プロモーターとE2後期プロモーターの間に結合し、PCR
によって生成した。続いて膜を洗浄し、フィルムに暴露した。
ベクターの説明:本明細書でグループIのアデノウイルスと呼ばれるウイルスの実施態様であり、アデノウイルスのベクター、XvirPSJL1及びXvirPSJL2で例示されるXvirPSJL群のベクターは、アデノウイルスdl520と同様に、YB−1核陽性の細胞、特に腫瘍細胞で複製可能であるばかりでなく、YB−1が過剰発現された及び調節解除された腫瘍細胞でも複製可能である。ウイルス遺伝子E1B55K及びE4orf6は、それぞれE1Bプロモーター及びE4プロモーターの影響下dl520を感染させたYB−1核陽性細胞で発現されるが、XvirPSJLにおけるE1B55K及びE4orf6の発現はサイトメガロウイルス(cmv)プロモーターによって生じる。しかしながら、cmvプロモーターの代わりに、他のプロモーター、特に腫瘍特異的プロモーター、組織特異的プロモーター及び器官特異的プロモーターも使用してもよい。E1B55K及びE4orf6の発現のために、過剰発現されたYB−1及び調節解除されたYB−1が核に輸送され、アデノウイルスの複製が開始される。本明細書で開示されるXvirPSJL群のアデノウイルスベクターは、種々の要素を組み合わせ、アデノウイルスベクターdl520、Xvir03及びAdYB−1の機能を単一のベクターで組み合わせる。ベクターdl520と同様に、XvirPSJLウイルスはE1A12S遺伝子を含有する。この遺伝子及び相当する遺伝子産物は、感染細胞のS期誘導に関与し、ウイルスの複製並びに化学療法及び照射の効果を促進する。Xvir03と同様に、XvirPSJLウイルスは、発現カセットCMV−E1B55K/IRES/E4orf6を含有し、それは、効率的な複製及び調節解除されたYB−1の、好ましくは腫瘍細胞に含有される核への直接的又は間接的な輸送に必要とされる。従って、複製は、YB−1が過剰発現されている、又は調節解除されている細胞、特に腫瘍細胞においてのみ可能である。さらに、E1B55K/E4orf6複合体によりp53を分解の対象とする。ヒトの転写因子YB−1をコードする配列をウイルスAdYB−1から選ぶ。内因性の、すなわち、細胞にすでに存在するYB−1はウイルス複製を増幅する。E1A12S及びYB−1双方の発現は、YB−1に依存したアデノウイルスE2後期プロモーターによって制御される。それに関連して、特異的プロモーター、特に腫瘍特異的プロモーター、組織特異的プロモーター又は器官特異的プロモーターを使用してもよい。これらウイルスのさらなる特徴はE4領域を欠失しているということである。ベクターはそこに制限部位を含有し、それによって、アデノウイルスベクターXvirPSJL1及びXvirPSJL2の場合、明細書で開示されるように、たとえば、リボザイム、アンチセンス分子、siRNA、アポトーシス誘導遺伝子、サイトカイン及びプロドラッグの遺伝子のような種々の導入遺伝子を発現してもよい。明細書で開示されるように、その発現は、腫瘍特異的プロモーター、組織特異的プロモーター又は器官特異的プロモーターに制御されてもよい。発現カセットの局在化は、特にE1、E3及びE4に関して又はその範囲内で固定されず、しかしいかなる方法でも配置することができる。ベクターは、腫瘍細胞のp53又はRbの状況に無関係に複製する。
本明細書で出発物質として使用されたClontech/BD BiosciencesのpAdenoXプラスミドは、アデノウイルスAd5のゲノム核酸を含み、野生型アデノウイルスには存在しない3’ITR領域の後にSfuI制限部位を有する。SpeI(23644位)及びpAdenoX(Clontech)のSfuIによって、E3−E4領域をpcDNA3.1(+)(Invitrogen)に転移させ、pcDNA3.1−E3Δ27865−30995−E4と呼んだ。E4orf6の大半、すなわち、塩基33241〜33875をPstIにより除いた。そのように得た断片をpcDNA3.1−E3Δ27865−30995、E4Δ33241−33875と呼んだ。
ディッシュ当たり100,000個のHela細胞を入れた。翌日、種々の力価のアデノウイルスdl520を細胞に感染させた。感染は、500μLの無血清DMEM培地にて37℃で1時間行った。続いて、感染培地を除き、完全培地(10%FCS/DMEM)2mLに取り替えた。クリスタルバイオレット染色を用いて、3〜5日後、分析を行った。
YB−1が核においてアデノウイルスE2後期タンパク質に結合し(Holm et al., JBC 277:10427-20434, 2002)、このプロモーターが核酸の発現によく適していることは既知である。アデノウイルスのE2後期プロモーターの使用は、それがYB−1によって調節されることができ、その際、YB−1は陽性のエフェクターとして作用する、すなわち、該プロモーターは核におけるYB−1の存在下でのみ活性があるという事実によって特に動機付けられる。その程度において、前記アデノウイルスE2後期プロモーターは、高度に選択的な様式で調節されることができるので、核にYB−1が存在する系で使用され、実際、YB−1がエフェクター又はレギュレータとして核に存在しない場合、アデノウイルスE2後期プロモーターの制御下にある核酸のいかなる発現も回避する。E2後期プロモーターは、E2遺伝子の活性化に関連する3つのYボックス(CCAAT)を含む。様々なE2後期プロモーターの構築物が調製され、その特異性及び活性について調べられている。分析は以下のように行った。
1.塩基25932〜26179に相当するYボックスI、II、及びIIIを含む(野生型アデノウイルス配列をいう。後で提供されるアデノウイルスE2領域の部分も参照のこと)
2.塩基25932〜26127に相当するYボックスII、及びIIIを含む(野生型アデノウイルス配列をいう。後で提供されるアデノウイルスE2領域の部分も参照のこと)
3.塩基25932〜26004に相当するYボックスIIIを含む(野生型アデノウイルス配列をいう。後で提供されるアデノウイルスE2領域の部分も参照のこと)
4.ブランクの読み取りとして作用するようにYボックスを含まない。
E1/E3を欠失したアデノウイルスベクターAdYB−1及びE1Aのみの欠失を有するAd312を、50pfu/細胞のMOIにてヒト骨肉種細胞(U20S)に感染させた。AdYB−1は、ゲノムに、細胞性転写因子YB−1をコードする配列を含有するので、Y−ボックス結合タンパク質1(YB−1)を発現する。感染後、「プラーク形成単位(pfu)」としてのウイルス粒子の放出を評価するために、培養培地の上清及び残りの細胞層をそれぞれ、感染後2日目及び5日目に単離した。細胞内の粒子は3回の凍結乾燥により放出させた。293細胞におけるプラークアッセイによって粒子数を分析した。

- E1Aタンパク質からなる群から選択される第2のタンパク質に先立って、E1B55KDタンパク質及びE4orf6タンパク質の組み合わせである第1のタンパク質を発現する組換えアデノウイルスであって、アデノウイルスがE1A12タンパク質をコードする少なくとも1つの核酸を含み、E1Aタンパク質の発現がE1Aプロモーターとは異なるプロモーターの制御下にあり、アデノウイルスは、核内に調節解除されたYB−1を含むか又はYB−1を有する細胞内で複製することができる、組換えアデノウイルス。
- E1Aタンパク質がE1A12Sタンパク質であることを特徴とする、請求項1に記載のアデノウイルス。

- 腫瘍が、固形腫瘍、非固形腫瘍、悪性腫瘍及び良性腫瘍を含む群から選択されることを特徴とする、請求項37に記載の使用。
- 腫瘍を形成する細胞の少なくとも一部が核内にYB−1を有し、好ましくは、細胞周期とは無関係に核内にYB−1を有することを特徴とする、請求項36〜38のいずれか1項に記載の使用。
- 腫瘍を形成する細胞の少なくとも一部が、調節解除されたYB−1を含むことを特徴とする、請求項36〜39のいずれか1項に記載の使用。
- 腫瘍を形成する細胞の少なくとも一部が、Rb陽性又はRb陰性であることを特徴とする、請求項36〜40のいずれか1項に記載の使用。
- 腫瘍を形成する細胞の少なくとも一部が、薬剤的に活性な薬剤に対して耐性、好ましくは多剤耐性を有することを特徴とする、請求項36〜40のいずれか1項に記載の使用。
- 耐性が多剤耐性であることを特徴とする、請求項42に記載の使用。
- 耐性が抗腫瘍剤、好ましくは細胞増殖抑制剤に対するものであり、及び/又は耐性は照射により誘発されることを特徴とする、請求項42又は43に記載の使用。
- 薬物を意図する患者が複数の細胞を含み、その際、細胞が請求項39〜43のいずれか1項に記載したような細胞であることを特徴とする、請求項36〜44のいずれか1項に記載の使用。
- 薬物が少なくとも1つのさらなる薬剤的に活性な薬剤を含むことを特徴とする、請求項33〜45のいずれか1項に記載の使用。
- 薬物が、さらなる薬剤的に活性な薬剤とともに投与される、又はそのために意図されることを特徴とする、請求項33〜45のいずれか1項に記載の使用。
- さらなる薬剤的に活性な薬剤が、サイトカイン、メタロプロテイナーゼ阻害剤、血管形成阻害剤、細胞増殖抑制剤、チロシンキナーゼ阻害剤及び細胞周期阻害剤を含む群から選択されることを特徴とする、請求項45又は47に記載の使用。
- 照射の前、その間、その後に薬物が投与されることを特徴とする、請求項36〜45のいずれか1項に記載の使用。
- 腫瘍を治療する目的で、放射線が投与されることを特徴とする、請求項49に記載の使用。
- 処置されるべき細胞又は生物が手段の対象とされ、その際、手段が、照射、細胞増殖抑制剤の投与及び温熱療法を含む群から選択されることを特徴とする、請求項36〜50のいずれか1項に記載の使用。
- 手段が局所的に、又は全身性に適用されることを特徴とする、請求項36〜51のいずれか1項に記載の使用。
- 照射が、高エネルギー放射線を使用する、好ましくは、腫瘍性疾患の治療で使用されるいかなる照射も使用することを特徴とする、請求項49〜52のいずれか1項に記載の使用。
- 腫瘍性疾患が、乳腫瘍、骨腫瘍、胃腫瘍、腸腫瘍、胆嚢腫瘍、膵臓腫瘍、肝腫瘍、腎臓腫瘍、脳腫瘍、卵巣腫瘍、皮膚腫瘍、皮膚付属器の腫瘍、頭頚部腫瘍、子宮腫瘍、滑膜腫瘍、咽頭腫瘍、食道腫瘍、舌腫瘍、前立腺腫瘍を含む群から選択される、腫瘍性疾患を治療するための薬物を製造するための、請求項1〜28のいずれか1項に記載のアデノウイルス又は請求項29に記載の核酸の使用。
- 腫瘍を形成する細胞の少なくとも一部が核内にYB−1を有し、好ましくは、細胞周期とは無関係に核内にYB−1を有する、
腫瘍を形成する細胞の少なくとも一部が、調節解除されたYB−1を含む、
腫瘍を形成する細胞の少なくとも一部が、Rb陽性又はRb陰性である、および/または
腫瘍を形成する細胞の少なくとも一部が、薬剤的に活性な薬剤に対して耐性、好ましくは多剤耐性を有する、
請求項54に記載の使用。 - 腫瘍特異的プロモーターが、薬物が使用される腫瘍に特異的であるプロモーターである、腫瘍性疾患を治療するための薬物を製造するための、請求項1〜28のいずれか1項に記載のアデノウイルス又は請求項29に記載の核酸の使用。
- 請求項1〜28のいずれか1項に記載のアデノウイルス又は請求項29に記載の核酸、及び場合により薬学上許容可能な担体を含む医薬組成物。
