Description
現在、色素沈着状態や度合いを客観的に評価する方法として、皮膚表面の「しみ」「そばかす」を形成するメラニン顆粒の分布や深さを近紫外線、可視、近赤外線の組み合わせにより評価する方法(特許文献1)、皮膚上の参照領域と判定領域における光の広がりの変化に基づいて色素沈着の深さを判定する方法(特許文献2)、ビデオマイクロスコープなど画像データーとコンピューターとの組み合わせにより色素沈着の評価を測定する方法(特許文献3)など、肌の色を光による反射や吸収をパラメーターとして定量する評価系が一般的に用いられている(非特許文献1、2、3)。また、皮膚の色素沈着の度合いを複数の色見本と対比照合することによって評価する方法などもある(特許文献4)。
しかし、これらの方法は肌の色合いを数値化したものであって色素沈着の種類やメカニズム(生成機構)などを特定するにはいたらない。
一方、色素沈着ができる初期段階においては皮膚表面に顕在化する前に様々な刺激により情報伝達物質のα-MSH, bFGF, CGRP, エンドセリンなどが多量に作られ、それらの発現によりメラノサイトが刺激され、メラニン色素が大量に作られるといったような細胞レベルにおける様々なタンパク質の変化が行われていると考えられる。さらに、in vitroの系を用いた実験においてもメラノサイトにUVを照射した場合、細胞の増殖因子マーカーである、p73, Nup88, p27, Id1,PCNAが、 またアポトーシス関連タンパク質であるbcl-2の発現が増減するという報告もある(非特許文献4)。また色素沈着の種類によっても皮膚表面での形、色合いが異なることから皮膚内部で変化している色素沈着部位のタンパク質の種類は異なると考えられる。これまでに、シミの状況をバイオプシーによる皮膚組織を用いて侵襲的にシミ部と正常部を比較し、シミ部位においてNT−3、ADAM9、HB−EGFといったタンパク質の発現が顕著に増加しているという報告はある(特許文献5)。また、非侵襲的な方法としてテープストリッピングもしくは擦過を介して採取した皮膚に由来する角層を試料として、インターロイキン1(IL−1)とインターロイキン1レセプターアンタゴニスト(IL−1ra)の存在量分析をし、肌質の評価をする方法が特許文献6に開示されている。
しかしながら、非侵襲的なテープストリッピングなどの方法を用いて肌表面の角質層から色素沈着に特異的なタンパク質の検索を行ったり、それらのタンパク質を用いて色素沈着の度合いを判定するといった方法は今までに報告がない。
その結果、9種類のタンパク質が、非色素沈着部位に比べて色素沈着部位において特異的に発現量が増加していることがわかった(図1、表1)。これらのタンパク質を質量分析(MS)で同定を行ったところ8種類が同定され、これまで色素沈着との関係が知られていなかった、Bleomycin hydrolase, Annexin II, Cathepsin D, Arginase 1, β-actin, Keratin 14, Keratin 1, SCCA2のタンパク質が含まれていた。
さらに、これらのタンパク質の特異的な抗体を用いてウェスタンブロッティング法で発現量を確認したところ、Bleomycin hydrolase, Annexin II, Cathepsin D, Arginase 1の4種類は後述する全ての色素沈着部位(図2及び表2参照)において発現が増加している一方、β-actin, Keratin 14, Keratin 1, SCCA2は後述する3つ若しくは4つの色素沈着部位(図3及び表2参照)のみで発現が増加していることがわかった。
さらに、これら色素沈着マーカーと色彩色差計による肌の明るさとの相関を調べたところこれらマーカータンパク質の発現量が肌の明るさと逆相関することがわかった。つまり、色素沈着が重度ほどこれらのタンパク質の発現量が増加し、色素沈着が軽度ほどこれらのタンパク質の発現量が少なくなることを見出した。

(2)非侵襲的な手段を用いて入手された被験者由来の皮膚細胞試料及び/又は皮膚組織試料を測定対象とすることを特徴とする(1)記載の色素沈着の度合いを分析する方法。
(3)下記4種類のタンパク質の内1種類又は複数種類からなる皮膚の色素沈着測定マーカーもしくは色素沈着リスク予測マーカー。

(4)皮膚細胞及び/又は皮膚組織における下記4種類のタンパク質であって、色素沈着の度合いに伴って発現が変化する前記タンパク質の内1種類又は複数種類の発現を測定することができる試薬を含む、色素沈着の度合いを判定するためのキット。

本発明は、非侵襲的なテープストリッピング手段を用いた皮膚サンプルを用いて分析することができるので、安全かつ簡便である。
本発明により、色素沈着の度合いを判定できる客観的なマーカーを指標とする評価系が確立できる。
この評価系を用いて、従来法に比べてより詳細に、色素沈着の度合いを判定することができる。
また、この色素沈着マーカーを利用して色素沈着の予防または改善に有用な成分をスクリーニングする方法や色素沈着の度合いを判定できるキットも提供される。
本発明により、シミ対策、シミ予防の美容が容易となる。即ち、シミ等色素沈着部位に対する化粧料、医薬部外品、医薬品又は美容用健康食品の選択、シミ等の危険性のある部位に対する化粧料、医薬部外品、医薬品又は美容用健康食品の選択及びそのために有効な情報を提供することができる。シミ対策等の美容カウンセリングを的確に行うことができる。
発現が変化する態様としては、タンパク質発現の有無が変わること、発現量が増減することなどが挙げられる。
特定のタンパク質は、Bleomycin hydrolase, Annexin II, Cathepsin D, Arginase 1, β-actin, Keratin 14, Keratin 1, SCCA2の8種類からなる群より選択することが好ましい。選択するタンパク質は1種でも複数種であってもよい。
(メラノサイト)、悪性黒色腫細胞(メラノーマ)などを挙げることができる。皮膚細胞は国内ではヒューマンサイエンス研究資源バンク、理化学研究所細胞銀行、海外ではthe American Type Culture Collectionから供給される。またClontech社、クラボウ社、PromoCell社から入手できる。また、公知の方法により皮膚から採取することができる(分子生物学研究のための新細胞培養実験法,p57−71,羊土社,1999)。
特定のタンパク質は、Bleomycin hydrolase, Annexin II, Cathepsin D, Arginase 1, β-actin, Keratin 14, Keratin 1, SCCA2の8種類からなる群より選択することが好ましい。
キットには、さらに、皮膚組織を採取するための粘着テープ、粘着テープ上のタンパク質を免疫化学的に検出するための試薬一式、取扱説明書などが含まれてもよい。取扱説明書には、キットの使用方法の他、色素沈着の度合いの判定基準なども記載しておくとよい。
(a)被験物質と皮膚細胞及び/又は皮膚組織を接触させる工程、
(b)工程(a)で被験物質と接触させた皮膚細胞及び/又は皮膚組織を所定時間培養する工程、
(c)工程(b)で培養した皮膚細胞及び/又は皮膚組織における特定のタンパク質の発現及び/又はその遺伝子発現を測定する工程であって、前記特定のタンパク質は色素沈着の度合いに伴って発現が変化するものである前記工程、及び
(d)工程(c)で測定した特定のタンパク質の発現及び/又はその遺伝子発現を対照の皮膚細胞及び/又は皮膚組織における特定のタンパク質の発現及び/又はその遺伝子発現と比較することにより、皮膚細胞及び/又は皮膚組織における特定のタンパク質の発現及び/又はその遺伝子発現に対する被験物質の効果を評価する工程を含む。
被験物質と皮膚細胞及び/又は皮膚組織との接触は、いかなる方法によってもよい。例えば、被験物質を皮膚細胞及び/又は皮膚組織の培養液に添加する方法、被験物質を塗布又は固定した培養容器又は培養シート上で皮膚細胞及び/又は皮膚組織を培養する方法などを挙げることができる。また、ヒト又はヒト以外の哺乳動物(例えば、マウス、ラット、モルモット、ウサギ、ブタなど)などの生体を用いて、被験物質を直接皮膚に塗布する方法、被験物質を経口投与する方法でもよい。ヒトについては、採取された皮膚組織、細胞あるいは培養皮膚細胞を用いることもできる。
例えば、皮膚細胞としてヒト正常表皮角化細胞を用いた場合には、12〜48時間が適当であり、12〜24時間が好ましい。ここで、「皮膚細胞及び/又は皮膚組織を培養する」とは、皮膚細胞及び/又は皮膚組織を生育・増殖させることを言う。これには、単離した皮膚細胞及び/又は皮膚組織を生育・増殖させることの他、皮膚細胞や皮膚組織を有する生体自体を生存させること、飼育すること、養うことなども含まれる。
<材料と実験方法>
1.色素沈着部位の判別法
今回サンプルとして採取した色素沈着部位とは顔面に存在する直径0.5mm以上の周囲の肌の色とは明らかに異なる茶褐色等の色彩を帯びた箇所、非色素沈着部位とはUVが比較的あたっていないとされる背中、顎下部位の箇所とした。今回は色素沈着部位を顔面から採取したのでより顔に近い部分の顎下部分を非色素沈着部位としてサンプルを採取した。
皮膚顔面に色素沈着を有するボランティア3名からそれぞれ色素沈着部位2箇所、比較対照として非色素沈着部位2箇所、また色素沈着部位の近傍部位2箇所に角質チェッカー(アサヒバイオメッド社)を貼り、角質層を採取した。角質チェッカーは一つの部位に対して同じ箇所で5枚を用いて回収した。1枚のシール当たり50μlの抽出buffer (PBS、1% SDS、10% glycerin) でホモジナイゼーション用ペッスルを用いてシールからサンプルを掻き取った(特願2007-089737「タンパク質抽出方法」)。10,000×g、4℃、10分で遠心後上清を回収し、DC protein assay kit (BIO-RAD社)を用いてタンパク定量を行った。
3−1 SDS-PAGE
Tris-glycin系のバッファー(0.02M Tris-HCl, 0.2M Glycine, 0.1% SDS)を用いて7.5%アクリルアミドゲルにおいてSDS-PAGEを行った。
3−2 電気泳動ブロット法
SDS-PAGE終了後のゲルをセミドライタイプ転写装置(BIO-RAD社)を用いて、PVDF膜 (ProBlott Membranes (Applied Biosystemus社)) に200mAの定電流で2時間転写した。転写バッファーは48mM Tris-HCl (和光純薬株式会社)、39mM Glycine(MP Biomedicals社)、20% Methanol(和光純薬株式会社)を用いた。転写後はTTBSバッファー(20mM Tris-HCl (pH7.5) (BIO-RAD社)、500mM NaCl、0.3% Tween-20 (BIO-RAD社)で20分間、3回洗浄した後MQWで2分間、3回洗浄した。次に膜をシーリングして50mlの金コロイド溶液(Colloidal Gold Total Protein Stain (BIO-RAD社))で満たし、1〜2時間振盪させてタンパク質を染色した。膜を染色後、金コロイド溶液を除き、純水で1分間、5回洗浄した後、乾燥させた。
4−1 PVDF膜上タンパク質の還元S-アルキル化とプロテアーゼ消化
PVDF膜に転写されたタンパク質のスポット部分を切り取り、チューブに入れて還元用バッファー(8M guanidine-HCl (pH8.5)、0.5M Trisbase、0.3% EDTA-2Na (w/v)、5% アセトニトリルを100〜300μl加えた。還元用バッファーに溶解したDTT (dithiothreitol) 1mg相当分を加えてチューブ内を窒素ガスに置換して室温、1時間静置してタンパク質の還元を行った。反応終了前に1M NaClに溶解したモノヨード酢酸3mg相当分を加えて遮光状態で15〜20分間撹拌を続けてS-カルボキシメチル化を行った。終了後PVDF膜を取り出し純水で5分間撹拌洗浄し、その後2%アセトニトリル中で同様に撹拌した。PVDF膜を取り出しLys-C消化用バッファー(70%アセトニトリル/ 20mM Tris-HCl (pH9.0))を入れたチューブに移し2〜3回すすいだ後完全に浸る程度にLys-C消化用バッファーを加えて1時間消化した。
4−2 質量分析とペプチドマスフィンガープリンティング
プロテアーゼ消化した溶液を7倍希釈してアセトニトリル濃度を10%にした。前処理としてZipTipc18ピペットチップ(MILLIPORE社)の充填剤部分を活性化するために50%アセトニトリル/ 0.1%TFA (trifluoro-acetic acid)で数回、2%アセトニトリル/ 0.1% TFA (trifluoro-acetic acid)で数回吸引、排出を繰り返した。次に活性化したZipTipc18ピペットチップでプロテアーゼ消化液を数回吸引、排出して断片化ペプチドを充填部分に吸着させた。さらに2%アセトニトリル/ 0.1% TFA (trifluoro-acetic acid)を数回吸引、排出して塩類を除いた。50%アセトニトリル/ 0.1%TFA (trifluoro-acetic acid)に溶解した飽和マトリックス溶液0.5〜1μlを吸引して10秒間程置いた後、質量分析計の付属のターゲットプローブに滴下した。サンプルを乾固させ、質量分析計(MALDI-TOF MS)で測定した。得られた質量値をもとにデータベース(MS-Fit, Mascot Search)に登録されているタンパク質の同定を行った。
SDS-PAGE終了後のゲルをセミドライタイプ転写装置(BIO-RAD社)を用いて、PVDF膜(ProBlott Membranes (Applied Biosystemus社)) に200mAの定電流で2時間転写した。転写後、5% スキムミルク/ PBS(−)を用いて4℃、終夜でブロッキングした。膜を0.1% Tween 20/ PBS(−)で3回洗浄した後、1次抗体を室温1時間で反応させた。次に2次抗体としてHRP labeled anti-mouse IgG (Amersham Bioscience社) (1:10000 dilution), HRP labeled anti-rabbit IgG (Amersham Bioscience社) (1:10000 dilution)で1時間振盪し、Enhanced chemiluminescence plus (ECL) (Amersham Bioscience社)キットを用いて蛍光発色により検出した。使用した1次抗体の種類と希釈度は次の通りである:mouse anti-Bleomycin hydrolase monoclonal antibody(Abnova社)(1:1000 dilution)、rabbit anti-annexin II polyclonal antibody (Santa Cruz Biotechnology社)(1:1000 dilution)、mouse anti-Cathepsin D monoclonal antibody (R&D systems社)(1:1000 dilution)、rabbit anti-Arginase 1 polyclonal antibody (Santa Cruz Biotechnology社) (1:1000 dilution)、mouse anti-beta actin monoclonal antibody (Cytoskelton社) (1:1000 dilution)、mouse anti-Keratin 14 monoclonal antibody (Chemicon社) (1:1000 dilution)、mouse anti-Keratin 1 monoclonal antibody (Zymed Laboratories社) (1:1000 dilution)、mouse anti-SCCA2 monoclonal antibody (Santa Cruz Biotechnology社) (1:1000 dilution)。
市販の分光側色計(CM-500、MINOLTA社製)を用いて測定した。その測定原理は分光センサーで皮膚表面から反射された光を各波長ごとに反射率を測定し、皮膚色を数値化する方法である。
変量Xと変量Yの間に相関関係があるかどうかを計算するには以下の計算式を使用した。
0〜0.2: ほとんど相関無し、

1.色素沈着部位の角層抽出物を用いたタンパク質発現変化の解析
皮膚顔面に色素沈着を有するボランティア3名からそれぞれ色素沈着部位2箇所、非色素沈着部位2箇所から角質チェッカーを用いて同じ箇所から5枚ずつ角層部位を採取した。
次に、角質チェッカーに付着した角層タンパク質に抽出バッファーを加えて、ホモジナイゼーション用のペッスルでタンパク質を抽出した。
その後、色素沈着部位と非色素沈着部位のサンプルを用いてSDS-PAGEし、金コロイド染色をしたところ色素沈着部位特異的に発現が増加しているタンパク質が分子量37〜118KDaあたりに9個検出された(図1)。
それらのバンドを質量分析装置(MALDI TOF-MS)を用いて同定した結果、Bleomycin hydrolase, Annexin II, Cathepsin D, Arginase 1, β-actin, Keratin 14, Keratin 1, SCCA2の8個タンパク質が同定された(表1)。なお、表1に示されるタンパク質の番号は図1中に示す○付き数字を示している。
上記で同定された色素沈着部位特異的に変化するタンパク質の発現を確認するために種々の抗体を用いてウェスタンブロッティング法による解析を行った。
ボランティア3名の色素沈着部位2箇所、非色素沈着部位2箇所から採取した角層抽出サンプルを用いてBleomycin hydrolase, Annexin II, Cathepsin D, Arginase 1, β-actin, Keratin 14, Keratin 1, SCCA2の各抗体でウェスタンブロッティングを行った。
その結果、Bleomycin hydrolase, Annexin II, Cathepsin D, Arginase 1,の4種類のタンパク質は被験者3人のすべての色素沈着部位(+)(6箇所)において非色素沈着部位(−)に比べて有意に発現量が増加していることがわかった(図2(a)〜(d))。
さらに、β-actin, Keratin 14, Keratin 1, SCCA2の4種類のタンパク質は被験者3人の色素沈着部位(+)の6箇所のうち3箇所若しくは4箇所において非色素沈着部位(−)に比べて有意に発現量が増加していることがわかった(図3(a)〜(d))。
図2、図3を表2にまとめて示す。
これまで色素沈着の状態を測定する方法として肌の色を光による反射や吸収によって定量されるのが一般的な方法として広く知られている。色彩色差計で肌色を測定した場合、数値が小さくなるにつれて肌の色が暗い、すなわち色素沈着が濃くなることを意味し、数値が大きくなるにつれて肌の色が明るい、すなわち色素沈着が薄い、もしくは無いことを意味する。そこで、この色彩色差計で測定した肌色数値と色素沈着マーカーの発現量とを比較し、候補マーカーの有用性を検証した。
肌の明るさと4種の候補マーカー(Bleomycin hydrolase, Annexin II, Cathepsin D, Arginase 1)の発現度との相関を調べた結果を図4(a)〜(d)に示す。グラフのX軸は肌の明るさを、またY軸は候補マーカータンパク質の発現量を示している。
その結果、Bleomycin hydrolase, Annexin II, Cathepsin D, Arginase 1の4種類のマーカーに対して、肌の明るさが暗くなるに従ってこれらのタンパク質の発現量が増加していることがわかった。
さらに、この候補マーカーの中でも、Bleomycin hydrolase(相関係数R2=0.74), Annexin II(相関係数R2=0.33), Cathepsin D(相関係数R2=0.44)の3種類に関しては相関係数が0.3以上であることから有意に肌の明るさと候補マーカーの発現量との相関があると考えられる。このことから、この方法を用いることによって色素沈着の度合いを測定できるマーカーとして使用できることが明らかとなった。
一方、色素沈着部位は皮膚表面に顕在化する前に皮膚内においては刺激により情報伝達物質であるα-MSH, bFGF, CGRP, エンドセリン等の発現が上昇し、メラノサイトの増加、メラニンの増加、メラニンの増加に関与するチロシナーゼ酵素の増加によって様々なステップにおいて種々のタンパク質の変化が起こることが知られている。また、色素沈着部位近傍は非色素沈着部位に比べて色素沈着が顕在化しやすいということが報告されている。
そこで、上記とはまた別の実験系において色素沈着部位2箇所、非色素沈着部位2箇所、色素沈着近傍部位2箇所から角質チェッカーを用いて同じ箇所から5枚ずつ角層部位を採取し、同時に同じ箇所で色彩色差計を用いて肌色を測定した。これらのサンプルを用いて候補マーカータンパク質の発現と色彩色差計による肌の明るさとの相関を調べた。その結果を図5(a)〜(d)に示す。
その結果、色素沈着近傍部位の肌の明るさは非色素沈着部位とほぼ近い値であるにもかかわらず5種類のマーカー(Bleomycin hydrolase, Annexin II, Cathepsin D, Arginase 1,)に対する発現量は色素沈着部位での発現量の半分程度まで増加している(非色素沈着部位<色素沈着近傍部位<色素沈着部位 の順に発現量が増加)ことがわかった。このことから、この方法を用いることによって色素沈着の危険度を予測できるマーカーとしても使用できる。
Claims (4)
- 皮膚細胞及び/又は皮膚組織に含まれる下記タンパク質4種類の内1種類又は複数種類の発現を測定し、色素沈着の度合いを分析する方法。
タンパク質:Bleomycin hydrolase(ブレオマイシンハイドロラーゼ), Annexin(アネキシン)II, Cathepsin(カテプシン) D 、Arginase (アルギナーゼ)1
- 非侵襲的な手段を用いて入手された被験者由来の皮膚細胞試料及び/又は皮膚組織試料を測定対象とすることを特徴とする請求項1記載の色素沈着の度合いを分析する方法。
- 下記4種類のタンパク質の内1種類又は複数種類からなる皮膚の色素沈着測定マーカーもしくは色素沈着リスク予測マーカー。
タンパク質:Bleomycin hydrolase(ブレオマイシンハイドロラーゼ), Annexin(アネキシン)II, Cathepsin(カテプシン) D 、Arginase (アルギナーゼ)1
- 皮膚細胞及び/又は皮膚組織における下記4種類のタンパク質であって、色素沈着の度合いに伴って発現が変化する前記タンパク質の内1種類又は複数種類の発現を測定することができる試薬を含む、色素沈着の度合いを判定するためのキット。
