Description

The present invention improves the compatibility of the denture with the living body contact surface and the living body, and various symptoms such as dryness, discomfort, pain, caries, periodontal disease, tongue coating, and bad breath in the oral cavity when the denture is worn. The present invention relates to a denture surface modifier that can prevent, ameliorate, or alleviate the above.

Normally, saliva is constantly supplied into the oral cavity. Saliva contains proteins such as albumin and inorganic components such as salt, and is important for maintaining an optimal oral environment, such as maintaining a moist condition in the oral cavity, pH buffering action, antibacterial effect, and oral cleaning effect. Has many functionalities.

However, it is said that the secretion amount of saliva decreases with aging, and dryness in the oral cavity due to a decrease in the secretion amount of saliva has become a problem in elderly people. Regardless of age, salivary secretion is significantly reduced due to the effects of many diseases and drugs used for the treatment, stress on complex social life conditions, etc., causing dry mouth There is.

Especially in recent years, dryness in the oral cavity when using dentures has become a problem. There are many denture users who feel a great deal of stress when living with their dentures in their mouths in various everyday situations, and it is known that saliva secretion is extremely reduced by these stresses.

Furthermore, when using a denture, a denture base stabilizing paste such as a paste or sheet may be used to strongly bond and fix the denture and the oral tissue so that the denture does not come off. However, forcibly adhering a denture, which is a foreign body to a living body, in the oral cavity gives great stress to the body and spirit of the denture user. Therefore, simultaneously with the start of use of dentures, there is a problem that saliva secretion is significantly reduced or depleted and the oral cavity is dried.

These xerostomia are generally called dry mouth, burning in the mouth, pain, tongue pain, caries, taste disorder, inflammation of the oral mucosa, erosion, ulceration, tongue coating, halitosis, periodontal disease, difficulty in chewing It is known to cause dysphagia, difficulty in conversation, and the like. In addition, the denture is not fixed due to dryness in the oral cavity, and it rubs against the periodontal tissue, causing pain and making it impossible to eat and talk, which is a very serious problem. In such dryness in the oral cavity, even if the mouth is swollen, the oral cavity is only temporarily moisturized and the dryness in the oral cavity is not improved, and causes a great deal of pain in daily life. For these reasons, there is a strong demand for appropriate measures against dryness in the oral cavity.

Thus, in order to prevent, improve or alleviate dryness in the oral cavity, development of a composition that moderately stimulates the salivary glands and moistens the oral cavity has been promoted. It has been proposed to use salivary secretion promoters. However, the acidity is strongly irritating, and in addition to the difference in personal preference, the use is limited because of lack of sustainability of the effect. In addition, for example, in the case where the salivary secretion function is remarkably reduced by surgery or radiation therapy for disease treatment, the above-mentioned method by stimulation of the salivary gland does not provide a sufficient salivary secretion promoting effect, Inside drying is not improved.

In addition, artificial saliva and gargles are being developed for the purpose of improving dryness in the oral cavity. So far, artificial saliva and gargles to which glycerin, hyaluronic acid and the like have been added have been proposed (Patent Document 2, Patent Document 3), but these components are not originally present in large quantities in saliva, A feeling of use or the like may not be preferable for the user, or a sufficient effect may not be obtained. In Patent Document 4, artificial saliva containing salivary proteins such as mucin and albumin has been developed. However, proteins derived from mammals such as pigs are sometimes used, and safety to humans is a problem. It has become. Furthermore, these artificial saliva and gargles have problems in sustaining effects, taste, and feeling of use.

On the other hand, Patent Document 5 proposes a method in which a biocompatible material is contained in the denture to increase the hydrophilicity of the living body contact surface of the denture so that the denture is easily adapted to the oral cavity. However, in these methods, since it is necessary to make a new denture, the burden on the user may be large. Therefore, there has been a demand for a simple method for modifying the surface of existing dentures so that it can be easily adapted to the oral cavity.

For these reasons, there is still a demand for an improvement measure for preventing, improving or alleviating dryness in the oral cavity when a denture is mounted.

特開平10−182392号公報Japanese Patent Laid-Open No. 10-182392 特開2004−136102号公報JP 2004-136102 A WO00−056344号WO00-056344 特表平9−508898号公報Japanese National Patent Publication No. 9-508898 WO00−05049号WO00-05049

The present invention has been made in view of the above circumstances, and by enhancing the ease of compatibility between the living body contact surface of the denture and the tissue in the oral cavity, the dry oral cavity is moistened, giving a smooth sensation in the oral cavity, An object of the present invention is to provide a denture surface modifier that can prevent, ameliorate, or alleviate various symptoms and conditions caused by drying.

As a result of repeated studies to achieve the above object, the present inventors have found that sericin modifies the surface of the denture and exhibits an excellent effect as a salivary protein. That is, sericin increases the compatibility of the denture's biological contact surface with oral tissues, moisturizes the dry oral cavity, gives a coating feeling and a smooth sensation in the oral cavity, and various symptoms caused by dry mouth I found that I can prevent, improve, and alleviate the condition.

The application of sericin to skin as a cosmetic has been disclosed so far, but there is no idea of using sericin as a denaturing agent for a living body contact surface of a denture as in the present invention. In addition, as shown in the examples described later, there is no disclosure or suggestion about the remarkable effect recognized in the present invention, and sericin can prevent, ameliorate, or alleviate various symptoms and conditions caused by dry mouth. Was previously unknown.

The present inventors have found that sericin is highly safe and excellent in biocompatibility, and is a material that has been conventionally used for foods, surgical sutures, etc. Therefore, the knowledge that it was useful as a component of the denture surface modifier was obtained.

Furthermore, the present inventors have found that the surface modification effect of the denture can be dramatically enhanced by the synergistic effect of the plant extract, polyhydric alcohol and sericin.
The present invention has been completed based on these findings.

The denture surface modifier of the present invention increases the ease of compatibility between the denture bio-contact surface and the oral tissue, moisturizes the dry oral cavity, gives a smooth sensation in the oral cavity, and is caused by dry mouth. It is possible to prevent, ameliorate, or alleviate various symptoms and conditions such as oral discomfort, dental caries, periodontal disease, oral pain, tongue coating, and bad breath. Further, the denture surface modifier of the present invention has excellent biocompatibility, exerts a buffering action between the artificial denture and the oral tissue, and is used in the oral cavity during denture use. Eliminate problems such as discomfort, discomfort, and pain in the oral cavity.

Furthermore, the denture surface modifier of the present invention uses sericin that is used in foods, cosmetics, surgical sutures, etc., so it has high safety and has an unpleasant taste, smell, and color. In some cases, sufficient effects may not be obtained.

The origin of sericin used in the denture surface modifier of the present invention is not particularly limited, and silk glands such as rabbits and wild silkworms, produced silkworms, raw silk, silk fabrics, and the like can be used. In the extraction of sericin, a known extraction method can be applied, for example, a method in which silkworms, raw silk, or silk fabric is used as a raw material and partially hydrolyzed and extracted with hot water, acid, alkali, enzyme, etc. Can be adopted. The obtained sericin is purified by a known method such as filtration, dialysis, ultrafiltration, etc., and then prepared in the form of powder or the like dissolved or suspended in water, buffer solution, physiological saline, alcohol, etc. Can be used as a denture surface modifier.

Moreover, the surface modification effect of denture can be heightened by adding a polyhydric alcohol and / or a plant extract to said denture surface modifier. Examples of the polyhydric alcohol used in the present invention include glycerin, propylene glycol, polyethylene glycol and sorbitol. Examples of the plant extract include grapefruit extract, olive extract, tea extract, bear extract, sesame extract, safflower extract, ginkgo biloba extract and the like.

The dosage form of the denture surface modifier of the present invention is not particularly limited as long as it is applied to a denture. Can improve and alleviate discomfort and discomfort when touching the tissue in the oral cavity, and pain in the oral cavity. Furthermore, from these, mastication and conversation of the denture user are more activated than before, and as a result, the salivary secretion function is improved and symptoms resulting from dry mouth are improved.
Further, the denture surface modifier of the present invention can be widely applied not only to humans but also to mammals.

EXAMPLES Hereinafter, although an Example is shown and this invention is demonstrated concretely, this invention is not restrict | limited to the following Example.

In this example, sericin obtained by alkaline hydrolysis of silkworms or raw silk was used. The average molecular weight of this sericin was measured by gel filtration chromatography and found to be about 20,000.

(Preparation of Denture Storage Solution) Sericin, grapefruit extract, and glycerin were added to pure water at the concentrations shown in Table 1, respectively, to prepare denture storage solutions having a function as a denture surface modifier.

Figure 0004942944
Figure 0004942944

(Hydrophilicity evaluation of denture base material surface) The chip | tip of the denture base material was produced using Physioresin (made by Nissin Co., Ltd.), and the hydrophilization effect of the denture base material surface by the said denture base storage liquid was evaluated. The denture base material was immersed in the denture storage solution for 30 minutes, washed with running water for 0 seconds, 30 seconds, 1 minute, 5 minutes, 15 minutes, and 30 minutes, and then the moisture on the surface of the denture base material was wiped off with a filter paper. 2 μL of water was dropped on the surface of the denture base material, and the contact angle of the water droplet was measured to evaluate hydrophilicity. A goniometer type contact angle measuring device (manufactured by Elma Engineering Co., Ltd.) was used for measuring the contact angle. As a comparative example, the same test was performed using pure water. The results are shown in Table 2.

Figure 0004942944
Figure 0004942944

From the results shown in Table 2 above, it has been clarified that the contact angle of water droplets on the denture base material surface is reduced by sericin. Further, even after washing with running water, a state of higher hydrophilicity than pure water was maintained. That is, the denture base material surface was modified by sericin, and the wettability of the denture base material surface, which is inherently hydrophobic, was greatly improved. Furthermore, it was shown that by adding the grapefruit extract to a concentration of 0.2%, the hydrophilicity of the denture base material is higher than when using sericin alone, and a synergistic effect with sericin can be obtained. . Further, by adding glycerin to a concentration of 20.0%, the wettability of the denture base material is improved more than the combination of sericin alone and sericin and grapefruit extract, and sericin and grapefruit extract It was shown that a synergistic effect can be obtained.

(Monitor test of denture surface modification effect) The denture surface modification effect was evaluated using the above denture storage solution. The denture was immersed in the above-mentioned denture storage solution for 30 minutes and then taken out. The denture was attached to the oral tissue, and the sense of discomfort, pain, taste, and oral wetness at the time of attachment were evaluated. As a comparative example, the same test was performed using pure water.
