Description
Wanner法(PNAS June 6,2000.97(12):6640−45)の変法を使用してeda欠失を実施した。PstI部位を含むS.enteritidis SE5609の5’及び3’edaフランキング配列のプライマー(PstI eda 5’:cta gct gca ggt gct aag cgg taa tct ggg及びPstI eda 3’:cta gct gca gaa gag att gct cgt cat gtg g)を設計し、PCR産物をpBluescript SKII+(pBSeda)にクローニングした。BglIIを含むedaプロモーター領域(ctag aga tct ctcgcctgattacta gtgtg)と3’末端(ctag agatct aag ccg ttaaatgcccgatgg)のプライマーを使用してpBSedaからのフランキング配列とベクター配列をリバースPCR増幅した。BglII消化とそれに続く連結によりeda欠失クローンを作製し、pBSedaΔと命名した。BamHIで消化した1.2kbクロラムフェニコール耐性遺伝子をpBSedaΔのBglII部位に挿入し、pBSedaΔcamを作製した。pBSedaΔcamをPstIで消化し、インサートをベクター配列から分離し、PCRの鋳型として使用した。反応液を100μlずつ8本貯留し、5μlをゲル上にて調べ、直線性PCR産物をエタノール沈殿させ、水2〜4μlに再懸濁した。
安全性と効力の両者を試験するために、SE5609−eda(−)1.1×108CFUと4.1×108CFUをニワトリに夫々6週齢と14週齢で経口接種した。
サルモネラ菌に汚染されていない集団から商業用産卵鶏を入手した。
どちらの経口ワクチン接種後も臨床異常は観察されなかった。
Claims (9)
- eda遺伝子の変異の結果として機能的Eda蛋白質を発現することができないサルモネラ・エンテリカ(Salmonella enterica)種の弱毒生菌を含む、サルモネラ・エンテリカ種感染又はその病原作用に対する動物又はヒトの防御のための経口投与用弱毒生ワクチン。
- 前記弱毒生菌がS.エンテリカ(S.enterica)血清型チフィムリウム(typhimurium)、S.エンテリカ血清型エンテリチジス(enteritidis)、S.エンテリカ血清型コレラエスイス(choleraesuis)、S.エンテリカ血清型デュブリン(dublin)、S.エンテリカ血清型チフィ(typhi)、S.エンテリカ血清型ガリナルム(gallinarum)、S.エンテリカ血清型アボルツソビ(abortusovi)、S.エンテリカ血清型アボルツス−エクイ(abortus−equi)、S.エンテリカ血清型プロルム(pullorum)から構成される群から選択される請求項1に記載のワクチン。
- 前記変異が挿入及び/又は欠失を含むことを特徴とする請求項1又は2に記載の弱毒生菌を含むワクチン。
- 異種遺伝子がeda遺伝子に挿入されていることを特徴とする請求項3に記載の弱毒生菌を含むワクチン。
- 医薬的に許容可能なキャリヤーをさらに含有することを特徴とする、請求項1から4のいずれか一項に記載のワクチン。
- アジュバントをさらに含有することを特徴とする請求項1から5のいずれか一項に記載の弱毒生ワクチン。
- 凍結乾燥形態であることを特徴とする請求項1から6のいずれか一項に記載の弱毒生ワクチン。
- 請求項1から7のいずれか一項に記載のワクチンの製造における、eda遺伝子の変異の結果として機能的Eda蛋白質を発現することができないサルモネラ・エンテリカ(Salmonella enterica)種の弱毒生菌の使用。
- eda遺伝子の変異の結果として機能的Eda蛋白質を発現することができないサルモネラ・エンテリカ(Salmonella enterica)種の弱毒生菌と医薬的に許容可能なキャリヤーを混合することを特徴とする請求項1から7のいずれか一項に記載のワクチンの製造方法。
