Description
1.猫から得た大便試料をスクリーニングするためのプロトコール:
大便試料を正常で健康な猫及びIBDに関して診断された猫から集めた。正常な猫をサイエンスダイエット(登録商標)フィーラインメンテナンス(登録商標)ドライ(Science Diet (登録商標) Feline maintenance(登録商標) dry)で維持し、一方、IBDを有する猫を治療のための胃腸用の食餌(therapeutic gastrointestinal diet)で維持した。大便試料を分析の前に−70℃に凍結した。分析のために、試料をリン酸緩衝液と1:10(w/w)の比に混合し、ガラスビーズと共に撹拌し、遠心分離して、粒子状物質を除去した。375μlの試料のアリコートを、1.125mlの4%パラホルムアルデヒドを含む管に加え、4℃で4〜5時間放置した。試料を遠心分離し、PBS中で2回洗浄し、次に150μlのろ過済みエタノールと混合し、蛍光インシトゥハイブリダイゼーション分析(微生物計数のためのFISH)の前に−20℃で貯蔵した。属特異的16SrRNA標的プローブを合成し、5’末端で蛍光染料を用いて単一標識して(monolabelled)、発酵媒質中の考察の対象となっている細菌を検出した。総核酸を染色して、総細胞計数を得た。データをlog10細胞/gの大便として表す。FISHは貯蔵した試料の細菌の定量化を可能にし、培養可能な及び非培養可能な多様性の両方を含む。
結果
実施例2:IBD及び正常な猫においてAGがデサルホビブリオに及ぼすインビボ効果
2.供給の研究のためのプロトコール
IBDを有する11匹の猫及び10匹の正常で健康な猫に、ウェスタンラーチから得た0.6%のアラビノガラクタン抽出物と共に1.0%のビートパルプを含むフードを供給した。抽出物は、約90重量%のアラビノガラクタン及び約4重量%のポリフェノールであり、主なポリフェノールはタキシフォリンであり、残りは水分であり、全て2週間の乾物ベースだった。これに続いて、猫を、1.5%のビートパルプを単独で含むフードに切り換えた。大便試料を、0日、14日及び28日に集めた。試料をFISH分析のために次の通り作製した:各大便試料を凍結するために、5.0gの大便を、無菌の袋またはプラスチック容器中、無酸素性のリン酸緩衝食塩水(PBS)中でpH7.3で懸濁して、最終濃度10%(5gの場合45ml)を与えた。スラリーを袋中で均質化/混合して汚染を避けた。異なる容器を各試料につき使用した。5mlのスラリーを等しい量のグリセロールと混合して、50:50混合物を与え、これをFISHによる分析のために凍結した。
結果:
13の完全な組の大便試料を得た。猫が0.6%のAG抽出物を含むフードを摂取した場合、13匹の猫につき4匹は、0.3log単位以上の減少したデサルホビビロspp.(Desulfovibiro spp.)を有した。13匹の猫につき8匹は、デサルホビブリオspp.のレベルの小さな低下を有するかまたは変化がなく、一方、13匹の猫につき1匹のみがデサルホビブリオspp.の増大を有した。猫を、AGを有しないフードに切り換えた場合、13匹の猫につき10匹は、0.3log単位以上のデサルホビブリオspp.の増大したレベルを有し、13匹の猫につき2匹は変化がなく、13匹の猫につき1匹のみがデサルホビブリオspp.の低下したレベルを有した。
実施例3−様々な繊維がデサルホビブリオspp.のレベルを低下したことを示すインビトロ実験
無酸素性のリン酸緩衝媒質を含む発酵容器を用意し、1mlの犬の大便接種材料(10%w/vの大便試料を緩衝させた)を加えた。媒質の組成は、Sunvold GD, Hussein HS, Fahey GC, Merchen NR, and Reinhart GA (1995), In vitro fermentation of selected fiber sources by dogs fecal inoculum and in vivo digestion and metabolism of fiber supplemental diets. J. Animal Sci. 73: 1099-1109 (1995)において説明されている。発酵を39℃で実行した。実験を、様々な繊維を用いてブラインドコード化方法(blind-coded manner)で行った。8時間のインキュベーションの後、1mlの培養流体を除去した。これのアリコートをFISHのために作製した。8時間後に、1mlの培養流体を除去し、PBS中の4%パラホルムアルデヒドと混合し、FISHのために固定した。属特異的16SrRNA標的プローブを合成し、5’末端で蛍光染料を用いて単一標識して、発酵媒質中の考察の対象となっている細菌を検出した。総核酸を染色して、総微生物計数を得た。結果は、幾つかの異なるタイプの繊維は、8時間の発酵の最中デサルホビブリオspp.の増殖を0.5〜1.0log単位減少できることを示した(表2を参照されたい)。
Claims (8)
- 犬または猫のGI管におけるデサルホビブリオ及び/またはヘリコバクターspp.の量を低減する方法であって、前記犬または猫にフードを経口投与することを含み、前記フードは、オリゴ糖、ガラクタン、ベータグルカン及びこれらの混合物からなる群から選択される繊維を乾物ベースで0.1から20重量%含む方法。
- 前記犬または猫は前記投与を必要とする、請求項1に記載の方法。
- 前記犬または猫は、GI管炎症が主な構成要素である疾患を有する、請求項2に記載の方法。
- 前記繊維は、アラビノガラクタン、キシロオリゴ糖、ガラクトオリゴ糖、フルクトオリゴ糖、イヌリン、発芽した大麦及びこれらの混合物からなる群から選択される、請求項3に記載の方法。
- フード中に繊維と共にポリフェノールも存在する、請求項1に記載の方法。
- GI管におけるデサルホビブリオ及び/またはヘリコバクターspp.の高いレベルを有する犬または猫におけるGI管炎症を治療する方法であって、前記犬または猫にフードを経口投与することを含み、前記フードは、オリゴ糖、ガラクタン、ベータグルカン及びこれらの混合物からなる群から選択される繊維を乾物ベースで0.1から20重量%含む方法。
- フード中に繊維と共にポリフェノールも存在する、請求項6に記載の方法。
- デサルホビブリオ及び/またはヘリコバクターspp.の高いレベルを有する犬または猫における腸ガス臭気、大便臭気及び任意のこれらの混合物からなる群から選択される臭気を低減する方法であって、前記犬または猫にフードを経口投与することを含み、前記フードは、オリゴ糖、ガラクタン、ベータグルカン及びこれらの混合物からなる群から選択される繊維を乾物ベースで0.1から20重量%含む方法。
