本発明は、分注良否判定装置、特に、親液体を複数に小分けして子液体を作成するときの作業の失敗を迅速に検出することのできる分注良否判定装置に関する。

【従来の技術】
人の健康状態やアレルギー体質などの検査を行う検体検査の分野において、最も頻繁に行われるものの一つに、血液中の血清を調べる方法がある。この検査を行う時の一般的な処理方法は、採血後の全血を遠心分離装置にかけ、血清を分離し、その血清を分析項目ごとの必要量に小分けし、所定の分析装置に提供して分析処理を行うものである。この時の小分けを自動で迅速かつ正確に行う装置として、自動分注装置がある。なお、前記自動分注装置は、試薬等の小分け作業にも使用される。従って、自動分注装置は液体を扱う検査においては、必要不可欠な装置となっている。特に検査項目が多く、所定量の液体を複数に小分けする必要のある検体検査においては重要な装置である。

従来、自動分注装置は、個々に異なる分注元の検体試験管(親検体)に収容された液体(例えば、血清)の液面高さを、前記液体を吸引する分注ノズル等に設けられたセンサにより検出し、その液面高さ位置に基づき分注ノズルの液への挿入量を算出し、液体の吸引を行っている。そして、所定の液量を吸引した後、吸引した液を各分注先の試験管(子検体)に所定量ずつ吐出し分配して分注処理を完了する。なお、親検体の液面高さは、例えば、静電容量センサや光学センサを用いて検知している。

【発明が解決しようとする課題】
しかし、前述のように親検体の液体(検体:例えば血清)の液面高さを検出するとき、静電気や外来ノイズ、液面の泡などの存在が原因となり、誤検知が発生する場合がある。例えば、泡の存在により実際の高さより高い位置(空中位置)に液面が存在すると検知してしまった場合、分注ノズルは検体の吸引ができず、結果的に子検体に検体が吐出されないことになってしまう。また、分注ノズルによる吸引量が所定量より少ない場合には、所定数の分注が行われなかったり、行われても必要十分量でなかったりしてしまう。このように、子検体を収納する容器(例えば試験管)に実際にあるべき検体が存在しない場合、または不足している場合、分注処理の後に行われる分析処理において、その分析結果は正常な範囲を逸脱した値として出力されてしまう。この時、分析結果を異常値を見ただけでは、所定量の検体に対し、正常な検査が実施されたが検体自体の異常により所定範囲を逸脱した値が出力されたのか、あるいは、検体が存在せず、異常値が出力されたのかの判断を行うことが困難であるという問題がある。このような場合、分析処理終了後に、再検査が必要になり、再度親検体からの子検体を作成したり、再分析を行う等の無駄な作業が発生する。そして、検査結果の出力迅速性も損なわれる。試薬等の分注を行う場合にも、試薬の有無、量の適否により結果が変化し、同様な問題が発生する。

なお、小規模施設等では、手動による分注も行われるが、この場合も分注漏れ(分注し忘れ)の可能性は存在し、前述と同様な問題が発生する。

本発明は、上記従来の課題に鑑みなされたものであり、その目的は、分注処理における分注ミスの発生を迅速に検出し、後段の処理に影響を与えることを確実に防止することのできる分注良否判定装置を提供することを目的とする。

【課題を解決するための手段】
上記目的を達成するために、本発明は、第1容器に収納された親検体である血清を吸引し、それを透明性を有する第2容器に小分け吐出して子検体を作成する分注処理について、分注処理の良否判定を行う分注良否判定装置であって、分注処理の後に前記第2容器に対して、光を照射し、第2容器を透過した光の吸光度測定を行う吸光度測定手段と、測定した吸光度に基づいて、第2容器内の分注処理の分注良否判定を行う判定手段と、を含むことを特徴とする。

上記目的を達成するために、本発明は、上記構成において、前記吸光度測定手段は、血清中のビリルビンの吸収波長での吸光度測定を行うことを特徴とする。

この構成によれば、第2容器内に血清が存在することにより、所定の吸光度が得られる。特に、血清中には、黄色の色素を有するビリルビンが必ず存在するので、第2容器に血清が正しく分注されたか否かを、ビリルビンの吸収波長での吸光度の値により判定することが可能になり、分注処理後、直ちに分注処理の良否判定が可能になる。

【発明の実施の形態】
以下、本発明の好適な実施の形態(以下、実施形態という)を図面に基づいて説明する。

図1は、本実施形態の分注良否判定装置10の処理工程におけるレイアウトを説明する説明図である。図1において、分注元である親検体は、第1容器12に収納されている。本実施形態において、親検体は、例えば採血後の全血を遠心分離させて得た血清14である。そして、自動分注装置16は、吸引ノズルにより所定量の血清14を吸引し、複数の第2容器18に小分け分配し複数の子検体を作成する。なお、第2容器18は、例えば、ラック(不図示)に収納され整列された透明性(後述する光源の光が透過可能)を有する容器である。そして、各第2容器18または、ラックは、個々にIDを持ち、どの第2容器18にどの親検体がどれだけの量で分注されるかが個々に管理されている。そして、本実施形態において、自動分注装置16、分注良否判定装置10、分析装置20はそれぞれ搬送装置(例えばコンベア)等で接続され、第2容器18が順次所定の分析装置20に搬送されるようになっている。つまり、自動分注装置16により分注され第2容器18に小分けされた子検体は、分注良否判定装置10により、その分注処理が正常に行われたか否かが判定され、正常な分注処理が行われたことが確認された第2容器18のみが、各種分析を行う分析装置20に投入され、そこで所定の分析が実施されるようになっている。

本実施形態の特徴的事項は、分注良否判定装置10において第2容器18に光を照射し、そのときの吸光度を測定することにより、第2容器18内に所定の液体(例えば、血清14)が分注されているか否かを判定するところである。

図2には、本実施形態の分注良否判定装置10の構成を示す概略構成ブロック図が示されている。本実施形態において、分注良否判定装置10は、光源22から照射された光が第2容器18を透過することによって生じる特定物質の吸収波長での吸光度を検出器(受光器)24により測定することにより行う(前記光源22と検出器24で吸光度測定手段の一部を構成している)。この時、検出器24からは電流値変化として測定値が出力されるため、測定データは、電流−電圧変換回路(I−V変換)26、AD変換器(ADC)28を介して、判定処理を行う判定手段としてのCPU30に提供される。このCPU30は、各種データの提供を受けると共に、そのデータに基づいて、第2容器18に関する吸光度演算を行う。本実施形態の場合、CPU30は吸光度により血清14の有無検出を行い、その結果を出力部32を介して出力する。出力部32では、例えば、分注が正常に行われた状態を示す『分注OK』や、分注が失敗した状態を示す『分注NG』等を表示したり、『分注NG』の場合のアラームを出力したり、『分注NG』の第2容器18を搬送ラインから排出したりする。この他、CPU30には、前記第2容器18を収納したラックを間欠的駆動することにより、第2容器18の子検体に対し所定位置で光源22の光を照射し、透過光を検出器24で受光し、分注良否判定を最適なタイミングで行うための搬送駆動回路34や、どの親検体をどの第2容器18に対して、どのくらいの分注量で分注を行ったかという情報を提供する情報提供部36、所定検体の吸光度と分注量と関係をあらかじめ測定し基準値として記憶した基準テーブル38等が接続されている。

図3(a)、(b)には、第2容器18と、光源22及び検出器24との配置関係の一例が示されている。検出器24は、血清(検体)14による吸光度を測定するのみであるので、光源22及び検出器24は対向配置されれば、第2容器18との関係は任意であり、例えば、図3(a)に示すように、第2容器18の上方に光源22を配置して、第2容器18の下面に検出器24を配置してもよいし、図3(b)に示すように、第2容器18の側方の一方側に光源22を配置し、対向側面側に検出器24を配置するようにしてもよい。

本実施形態において検体として使用する血清は、その成分として黄色の色素を持つビリルビンを含む。このビリルビン濃度は、ビリルビンの代謝障害者で例えば30〜40mg/dlであり、健常者でも0.4〜1.2mg/dl程度含まれる。従って、吸光度測定の結果、ビリルビン濃度が所定値以上、例えば、0.4mg/dl以上であることが確認できれば、子検体に血清が分注されていると見なすことができる。つまり、第2容器18の収納された液体(血清)の色の濃度により分注良否判定を行うことができる。

ところで、子検体(血清14)の量(分注量)によって、光の透過する長さ、すなわち光路長が変化し、同じ血清14が存在していても吸光度が変化する。従って、前述したように、基準テーブル38を使用し、分注量に応じた補正を行う必要がある。基準テーブル38の一例を図4に示す。図4は、図3(a)の状態で検出を行った場合の分注量と吸光度(ビリルビン濃度0.4mg/dlの血清)の対応関係を示すものである。従って、CPU30は、情報提供部36から取得できる子検体(サンプル)ごとの分注量情報に基づき、実際に検出器24で、検出したその子検体(サンプル)の吸光度を補正することにより、子検体(サンプル)毎の吸光度を正確に算出することができる。なお、本実施形態における血清14に対する吸光度測定は、他の物質(溶血や乳び等)の影響を抑制するためや空の第2容器18との区別を行うために2波長測光を行うことが好ましい。そして、2波長によるビリルビンの吸光度計算式は、(式1)のようになる。

このように、分注処理が完了した第2容器18に対して吸光度測定を行った結果、ビリルビンの吸収波長での吸光度が所定値以上確認された場合、分注処理が正常に行われたと判断することができる。逆に、ビリルビンの吸収波長での吸光度が所定値以上確認されない場合、分注処理に失敗したと判断され、分析処理前に、分注ミスの子検体を排除し、分析処理が実施されることを未然に防止することができるので、一連の分析をスムーズかつ効率的に行うことができる。また、分析装置で、異常な値が検出されてもその原因が、分注処理ミスによる異常でないことが保証されるので、分析結果の信頼性向上にも寄与することができる。

なお、図3(a)のような第2容器18と光源22と検出器24の配置を行った場合、前述したように、分注量によって、透過光の光路長が変化するので、図4の基準テーブルによる補正が必要になるが、図3(b)のような配置を行った場合、第2容器18の直径により光路長が決まり、一定であるので図4の基準テーブルによる補正を省略することができる。

また、本実施形態では、第2容器18が個別の試験管形状である例を示しているが、子検体は、板状のプレートに複数の凹状の窪みが形成され、その窪みに子検体を順次分注するマイクロプレートを使用する場合もある。この場合、各窪みに対して図3(b)のような光源22と検出器24の配置は困難であるので、図3(a)のような光源22と検出器24の配置が適する。

図5には、本実施形態の分注良否判定装置を用いた分注良否判定の他の利用例が示され、図6には、そのときに処理フローチャートが示されている。図5の例では、検体が第2容器18に実際に分注されたか否かを判断することに加えて、分注前後で検体が同一であるか否か、すなわち、検体の一致性も含めた分注良否判定を行う例を示している。

まず、第1測定工程40では、上述した方法と同様に、親検体が収納された第1容器12に対して分注良否判定装置42の第1吸光度測定部(第1吸光度測定手段)42aを用いて、光源から光を照射し、その透過光に基づいて、吸光度を測定し、親検体に関するデータ1の取得を行い(S100)、データ1を分注良否判定装置42の判定手段としてのCPU42bに提供する。この時のデータ1は、親検体の量に基づき補正された単位量あたりの吸光度である。続いて、分注工程44で、親検体は、所定の量に小分けされ第2容器18に分配され子検体が形成される(S101)。さらに、第2測定工程46では、図3(a)等で示したように各第2容器18に対して、分注良否判定装置42の第2吸光度測定部(第2吸光度測定手段)42cを用いて光を照射し吸光度を測定し、子検体に関するデータ2の取得を行い(S102)、データ2を分注良否判定装置42のCPU42bに提供する。この時のデータ2も子検体の量に基づき補正された単位量あたりの吸光度である。

CPU42bでは、取得したデータ2に基づき、検体が第2容器18に実際に分注されたか否かを判断すると共に、データ1とデータ2の比較を行い、第2容器18に分注された検体が、親検体と同一のものであるか否かの確認を行う(S103)。すなわち、第2容器18に親検体が正常に分注され、子検体が作成されていれば、データ1とデータ2は一致する。従って、その第2容器18は、正常な分注処理が行われたと判断される。その結果に基づき、搬送・振り分け工程48では、該当する子検体を分析工程(分析装置)50へ搬送する。すなわち、検体の正常処理を行う(S104)。その後、第2容器18の子検体は、分析工程50において所定の分析処理が行われ、一連の処理が終了する。

一方、分注に失敗し、親検体が分注されなかったり、他の検体が分注された場合には、CPU42bにおいてデータ1とデータ2は一致しない。従って、その第2容器18は、異常な分注処理が行われたと判断される。その結果に基き、搬送・振り分け工程48では、該当する子検体を異常処理工程52へ搬送する。すなわち、検体の異常時処理を行う(S105)。前記異常時処理工程52では、例えば、該当する子検体である第2容器18が正規ラインから排出されたり、警報出力が行われたりし、利用者に対し、分注処理の失敗を分析処理前に通知する。その結果、分注に失敗した第2容器18に対する分析処理が実施されることがなく、効率的に分注処理及びそれに続く分析処理を行うことができる。

図7には、本実施形態の分注良否判定装置を用いた分注良否判定のさらに他の利用例が示され、図8には、そのときに処理フローチャートが示されている。図7の例では、検体が第2容器18に実際に分注されたか否かを判断することに加えて、分注後に搬送、その他、所定時間のストック等が行われ、第2容器18の流れが不連続になった場合に、不連続の前後で検体が同一であるか否か、すなわち、検体の一致性も含めた分注良否判定を行う例を示している。

まず、分注工程54で第1容器12に収納された親検体を所定数の第2容器18に小分けして子検体を作成する(S200)。続いて、第1測定工程56で、図3(a)等で示したように各第2容器18に対して、分注良否判定装置58の第1吸光度測定部(第1吸光度測定手段)58aを用いて光を照射し、吸光度を測定し、分注直後の子検体に関するデータ1の取得を行い(S201)、データ1を分注良否判定装置58のCPU58bに提供する。この後、子検体を収納した第2容器18は、別工程における処理や所定の時間のストックのために自動または手動で搬送される搬送工程60に移行する(S202)。つまり、この時点で、第2容器18の処理が中断される。その後、第2容器18の処理が再開された場合、再び第2測定工程62で、図3(a)等で示したように各第2容器18に対して、分注良否判定装置58の第2吸光度測定部(第2吸光度測定手段)58cを用いて光を照射し、吸光度を測定し、分注直後の子検体に関するデータ2の取得を行い(S203)、データ2を分注良否判定装置58のCPU58bに提供する。CPU58bでは、搬送工程60に投入される前に測定されたデータ1と、搬送後に測定されたデータ2との比較を行い、検体が第2容器18に実際に分注されたか否かを判断すると共に、データ1とデータ2の比較に基づき、処理しようとしている第2容器18の子検体が、搬送前の子検体と同一であるか否かの確認を行う(S204)。すなわち、第2容器18が搬送中やストック中に他のものと入れ替わったりしていない場合には、データ1とデータ2は一致する。従って、その第2容器18は、分注処理で所定の親検体が所定の分量で分注されたものであることが確認され、分注装置で取得している情報(親検体の種類や分注量等)が正確に分析装置に伝えられることが保証される。その結果に基づき、搬送・振り分け工程64では、該当する子検体を分析工程(分析装置)66へ搬送する。すなわち、検体の正常処理を行う(S205)。その後、分析工程66において第2容器18の子検体は、所定の分析処理が行われ、一連の処理が終了する。

一方、分注に失敗し、親検体が分注されなかったり、搬送時やストック時に他の検体と入れ替わる等した場合には、CPU58bにおいてデータ1とデータ2は一致しない。従って、その第2容器18は、異常な分注処理が行われたと判断される。その結果に基づき、搬送・振り分け工程64では、該当する子検体を異常処理工程68へ搬送する。すなわち、検体の異常時処理を行う(S206)。この異常時処理工程68においても、例えば、第2容器18が正規ラインから排出されたり、警報出力が行われたりして、利用者に対し、分注処理の失敗を分析処理前に通知する。その結果、分注に失敗した第2容器18に対する分析処理が実施されることがなく、効率的に分注処理及びそれに続く分析処理を行うことができる。なお、分注自体の失敗(未分注)は、第1測定工程56で確認することができるので、この時点で、失敗した第2容器18に対する異常処理を行うようにしてもよい。

このように、分注後、子検体の搬送が不連続的に行われても、その前後で検体の一致性を確認することにより、未分注の早期発見と共に子検体の取り違いミスの防止を確実に行うことができる。

また、上述した各実施形態では、分注の対象となる液体として、検体(血清)を例に取り、吸光度の測定は、ビリルビンに関して行う例を示したが、分注の対象は任意であり、例えば、試薬等でもよい。この場合、吸光度の測定は、試薬に含まれる物質に関して行うことになる。また、本実施形態において、親検体は、一度分注された後、試薬等が混合された二次液体でもよく、本実施形態と同様な効果を得ることができる。

なお、本実施形態においては、分注良否判定装置を独立した構成で説明したが、分注良否判定を子検体の分注後、分析装置投入前に行えれば、その配置は任意である。また、自動分注装置に分注良否判定装置の機能を付加してもよい。

【発明の効果】
以上説明したように、本発明によれば、分注処理が完了した第2容器に対して吸光度測定を行い、所定物質の関する吸光度が所定値以上と確認された場合、その物質が第2容器に分注され、分注処理が正常に行われたと判断することができる。逆に、吸光度が所定値以上と確認されない場合、分注処理に失敗したと判断され、分析処理前に、分注ミスの第2容器を排除する。必要に応じて再分注することが可能である。その結果、分注に失敗した第2容器に対する分析処理が実施されることを未然に防止することができ、一連の分析をスムーズかつ効率的に行うことができる。また、分析装置で、異常な値が検出されてもその原因が、分注処理ミスによる異常でないことが保証されるので、分析結果の信頼性向上にも寄与することができる。
