Description
しかしながら、前記特許文献1及び2の方法では、絶縁油中に含まれている有機塩素化合物もPCBとして定量されてしまうこと、塩素量を基にしたPCB濃度と実際のPCB濃度とが一致しにくいことなどの問題がある。
<1> n種(ただし、nは2以上の自然数)の組成物からなる被測定物質を含む試料から、
前記被測定物質を分離する工程と、
前記被測定物質をクロマトグラフィーにより解析し、クロマトグラムを得る工程と、
得られた前記被測定物質のクロマトグラムのm個(ただし、mは2以上の自然数)のピークを抽出し、該m個のピークに基づくピーク要素からなるピークパターンPを特定し、該ピークパターンPと近似するように、予め求めた前記n種の各組成物が示すn種のピークパターンP1、P2、…Pnを組合せて合成するピークパターン解析工程と、

本発明の被測定物質分析方法は、n種(ただし、nは2以上の自然数)の組成物からなる被測定物質を含む試料中の前記被測定物質を分析する方法であって、(1)被測定物質分離工程と、(2)クロマトグラム取得工程と、(3)ピークパターン解析工程と、(4)組成解析工程と、を含み、必要に応じて適宜選択したその他の工程を含む。
前記分離の方法としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、固相カラム処理、バッチ処理、液液抽出、電気泳動、各種カラム処理(イオン交換、疎水、アフィニティー、サイズ除去、分配、吸着、塩析、逆相、超臨界)等が挙げられる。
前記クロマトグラフィーとしては、前記クロマトグラムが得られる限り、特に制限はなく、目的に応じて適宜選択することができ、例えば、ガスクロマトグラフィー、液体クロマトグラフィー、薄相クロマトグラフィー、電気泳動、免疫クロマトグラフィー、質量分析、電子スピン、X線等が挙げられる。
前記ピーク要素が、前記ピーク面積である場合、前記ピークパターンとしては、前記ピーク面積の比からなるものであれば、特に制限はなく、前記ピークの面積の比は、例えば、チャートやグラフ等で表したものであってもよく、数値や数式であってもよい。
また、前記ピーク要素が、前記ピーク高さである場合、前記ピークパターンとしては、前記ピーク高さの比からなるものであれば、特に制限はなく、前記ピークの高さの比は、例えば、チャートやグラフ等で表したものであってもよく、数値や数式であってもよい。あるいは、ピーク面積と高さの組み合わせであっても良い。
前記被測定物質の総濃度は、前記各組成物の含有比率に基づき求めた各組成物の絶対濃度の総和から求めることができる。
本発明の被測定物質分析方法における前記被測定物質としては、n種の同族体及び異性体の混合物からなる物質であることが好ましく、特に、PCBが好ましい。
以下、本発明のPCB分析方法の説明を通じて、本発明の被測定物質分析方法を明らかにする。
前記PCB分析方法は、前記被測定物質分析方法により、n種(ただし、nは2以上の自然数)の複数同族体混合物からなるPCBを含む試料中の該PCBを分析する方法であって、(1)PCB分離工程と、(2)クロマトグラム取得工程と、(3)ピークパターン解析工程と、(4)組成解析工程と、を含み、必要に応じて適宜選択したその他の工程からなる。
前記PCB分離工程は、前記試料からPCBを分離する工程である。
前記分離の方法としては、特に制限なく、目的に応じて適宜選択することができるが、例えば、前記試料中のPCBを有機溶媒で液液抽出する方法、及び前記試料中のPCBを有機溶媒で液液抽出し、抽出された前記PCBを含む前記有機溶媒を吸着剤で固相抽出し、前記吸着剤に吸着した成分を所望の溶媒に転溶させる方法などが挙げられる。
前記絶縁油としては、脱塩素化処理等のPCB分解処理を行った後の絶縁油も好適に挙げられる。
前記クロマトグラム取得工程は、前記試料から分離したPCBをクロマトグラフィーにより解析し、クロマトグラムを得る工程であり、前記クロマトグラフィーとしては、例えば、ガスクロマトグラフィーが好ましい。
前記ピークパターン解析工程は、得られた前記PCBのクロマトグラムの26個のピークを抽出し、該26個のピークに基づくピーク要素からなるピークパターンPを特定し、該ピークパターンPと近似するように、予め求めた前記n種の同族体及び異性体の混合物が示すn種のピークパターンP1、P2、…Pnを組合せて合成する工程である。
前記PCB(カネクロールKC−300、KC−400、KC−500、KC−600の等量混合物、総濃度0.5mg/Kg)をGC−ECD法により解析して得たクロマトグラムの一例を図1に示す。
前記n種の同族体及び異性体の混合物が示すn種のピークパターンP1、P2、…Pnは、予め求められたものが好ましい。
n種の同族体及び異性体の混合物X1、X2、…XnのピークパターンP1、P2、…Pnの前記p(a)に相当するn種のピークp1(a)、p2(a)、…pn(a)によって、下記式(1)を満たす組合せとなるn種の係数x1、x2、…xnを求めることにより行われる。
また、前記ピークパターンは、複数の要素について組合せ(例えば、p(b)に相当するn種のピークp1(b)、p2(b)、…pn(b)の組合せ、p(c)に相当するn種のピークp1(c)、p2(c)、…pn(c)の組合せ)を求め、各要素間の誤差を最小となる係数x1、x2、…xnを求めることが好ましい。
前記組成解析工程(4)は、前記ピークパターンP1、P2、…Pnの組合せから、前記n種の同族体及び異性体の混合物の含有比率を求めて前記PCBの組成を解析する工程である。
具体的には、前記式(1)で求めた前記係数x1、x2、…xnを、前記n種の同族体及び異性体X1、X2、…Xnの含有比率(質量%)として、前記PCBの組成を規定することができ、該含有比率の合計から、PCB総量を求めることができる。
算出された同族体ごと、異性体ごと、カネクロールの種類ごとの濃度をそれぞれ合計することにより、試料中のPCBの総濃度を求めることができる。
カネクロールKC−300、KC−400、KC−500、及びKC−600をそれぞれ等濃度で混合し、総濃度として0.5mg/Kg含む試料を調製し、これをGC−ECD法により解析し、図3に示すクロマトグラムを得た。また、カネクロールKC−300、KC−400、KC−500、及びKC−600をそれぞれ0.5mg/Kgずつ含む試料を調製・測定し、予め得た各カネクロールのクロマトグラムを図4〜7に示す。
JIS K0093(用水・排水中のポリクロロビフェニル(PCB)の試験方法)に基づいてカネクロール混合物を検量線として、各カネクロールを定量した。具体的には、カネクロールKC−300、KC−400、KC−500、及びKC−600をそれぞれ等濃度で混合し、総濃度として0.5mg/Kg含む試料を調製し、これをGC−ECD法により解析し、クロマトグラムを得た。このクロマトグラム上のピーク1から26番に番号を付け、全ピーク高さの積算から、各ピークの割合を求めた。次にカネクロールKC−300、KC−400、KC−500、及びKC−600をそれぞれ0.5mg/Kgずつ含む試料を調製し、GC−ECD法にて測定した。この4種のクロマトグラムをもとに、検量線から定量値を求めた。具体的には、表5に例を示す。KC−600を含む試料を例にすると、まずクロマトグラム上のピーク1から26番までの高さを求めた。この後、検量線におけるピーク高さとKC−600におけるピーク高さから、カネクロール混合物としての換算値を算出した。算出方法は、それぞれのピーク番号について、KC−600におけるピークの割合=(KC−600のピーク高さ/カネクロール混合物のピーク高さ)×カネクロール混合物におけるピーク割合とした。求めたKC−600におけるピークの割合の積算値は、カネクロール混合物の存在比の積算値と同意であるので、0.5mg/kg×(KC−600の積算値/カネクロール混合物の積算値)×(希釈倍率や試料量などその他の因子)から、KC−600として実濃度0.5mg/kgの試料は、カネクロール混合物の換算値として総PCB濃度0.37mg/kgと定量された。
カネクロールKC−300、KC−400、KC−500、及びKC−600をそれぞれ0.5mg/Kgずつ含む試料を調製し、GC−ECD法にて測定した。この4種のクロマトグラムから、それぞれピーク1から26番の高さを求めた。この後、PCB濃度不明の実試料を調製し、GC−ECD法にて測定し、各ピークの高さを求めた。実試料のピーク1から26番の高さのパターンが、予め測定した前記のカネクロールKC−300、KC−400、KC−500、及びKC−600のピークパターンの組み合わせとして一致するように、上記と同様に計算機にて解析し、実試料における各カネクロールの組成比と、PCB濃度値を求めた。
同時に前記参考例と同様にしてJIS K0093法で前記試料中のPCB濃度値を算出した。このようにして本発明の方法及びJIS K0093法により得られたそれぞれのPCB濃度値と、特別管理一般廃棄物及び特定管理産業廃棄物に係る基準の検定方法別表第二(平成4年厚告192号、平成15年環告14号改正)により測定した質量分析値との相関を図17に示す。
また、図17図において、前記質量分析値と前記JIS K0093法の分析値、前記質量分析値と本発明の方法による分析値との絶対誤差を図18に示す。

前記被測定物質を分離する工程と、
前記被測定物質をクロマトグラフィーにより解析し、クロマトグラムを得る工程と、
得られた前記被測定物質のクロマトグラムのm個(ただし、mは2以上の自然数)のピークを抽出し、該m個のピークに基づくピーク要素からなるピークパターンPを特定し、該ピークパターンPと近似するように、予め求めた前記n種の各組成物が示すn種のピークパターンP1、P2、…Pnを組合せて合成するピークパターン解析工程と、
前記ピークパターンP1、P2、…Pnの組合せから、前記n種の各組成物の含有比率を求めて前記被測定物質の組成を解析する組成解析工程と、
