oolas RPら、Gynecol Oncol, 1995; 59(1):111-6; Zhang Zら、Gynecol Oncol, 1999; 73(1): 56-61; Zhang Zら、Use of Multiple Markers to Detect Stage I Epithelial Ovarian Cancers: Neural Network Analysis Improves Performance. American Society of Clinical Oncology 2001; Annual Meeting, Abstract)、全ての試験の特異度をあげるという有望な結果を示したが、これは、比較的罹患率の低い卵巣がんのような疾患にとっては重要である。
本発明は、マーカーを測定することにより、卵巣がんの状態を決定するために有用な、感度が高く迅速な方法及びキットを提供する。患者のサンプルにおいてこれらのマーカーを測定することで、診断者がヒトのガンの予想される診断、又は陰性(たとえば正常又は疾患ではない)と関連付けられる情報が提供される。マーカーは分子量、および/または、それらの蛋白質の独自性で特徴付けられる。マーカーは、たとえばマススペクトロメトリーを伴うクロマトグラフィーによる分離、固定化された抗体を使用する、又は伝統的なイムノアッセイによる蛋白質の捕獲等の、様々な分画技術を使用して、サンプル内のほかの蛋白質から分析されることができる。好ましい態様では、分析法は、表面活性化レーザー脱離/イオン化(SELDI)マススペクトロメトリーを含み、その方法では、マススペクトロメトリープローブの表面は、マーカーに結合する吸収剤を含んでいる。

A.アポリポプロテインA1
本発明の方法において有用なマーカーの一例は、アポリポプロテインA1であり、これは本明細書では「ApoA1」と表される。ApoA1はm/z28043のピークとして、マススペクトロメトリーで検出される。本明細書に開示されるマーカーの分子量は、開示されるSELDIマススペクトロスコピー手順により決定される特定された値と0.15%以内の正確性であると考えられている。ApoA1は手順に従って血液を分画化し、IMACチップへ適用後のSELDIによる検出で、見出された。精製された蛋白質はトリプシンで消化され、アポリポプロテインA1と同定された。ApoA1の単離及び同定のための手順は、以下の実施例に開示されている。ApoA1はある段階の卵巣がん患者においてダウンレギュレートされている。よってApoA1の欠如または正常な対照と比較したApoA1量における統計学的に顕著な減少は、卵巣がん状態と関連があるだろう。統計学的に顕著な減少とは、たとえばp値が0.05より低いものであり、当該技術分野で公知のものである。
本発明の方法において有用はマーカーのほかの例には、プレアルブミンの形態であり、本明細書で「トランスサイレチンΔN10」と表されるものが含まれる。トランスサイレチンΔN10はm/z12870.9のピークとして、マススペクトロメトリーで検出される。トランスサイレチンΔN10は手順に従って血液を分画化し、IMACチップへ適用後のSELDIによる検出で、見出された。免疫沈降およびタンデムマススペクトロメトリーにより、精製された蛋白質はN末端のアミノ酸を欠如するプレアルブミンの切断された形態(本明細書では「トランスサイレチンΔN10」と表される)であることが見出された。トランスサイレチンΔN10の単離及び同定のための手順は、以下の実施例に開示されている。トランスサイレチンΔN10もまたある段階の卵巣がん患者においてダウンレギュレートされている。よってトランスサイレチンΔN10の欠如または正常な対照と比較したトランスサイレチンΔN10量における統計学的に顕著な減少は、卵巣がん状態と関連があるだろう。
本発明の方法において有用なマーカーのほかの例は、インターαトリプシン阻害剤重鎖H4の解裂した断片であり、本明細書では「IAIH4」と表されるものである。IAIH4断片はm/z3272ピークとして、マススペクトロメトリーで検出される。IAIH4断片は手順に従って血液を分画化し、IMACチップへ適用後のSELDIによる検出で、見出された。ピークは一連のクロマトグラフィー分離技術を使用して、卵巣がんの集められた血清から精製された。その配列はMNFRPGVLSSRQLGLPGPPDVPDHAAYHPF(配列番号1)と決定され、ヒトインターαトリプシン阻害剤、重鎖H4(ITIH4、PK−120)の660−689アミノ酸断片であった。この結果はマーカーのペプシン消化産物の分析により確認された。ITIH4断片はある段階の卵巣がん患者においてアップレギュレートされている。よってITIH4断片の存在または正常な対照と比較したITIH4断片量における統計学的に顕著な増加は、卵巣がん状態と関連があるだろう。
さらなるバイオマーカーが、卵巣がん疾患状態と関連のある、pH4及びpH9で溶出された分画において同定された。pH4では、これらのバイオマーカーに対応する蛋白質またはその断片が、以下の値を中心とする分子量の強度ピークとして、SELDI(表面活性化レーザー脱離/イオン化)プロテインチップ/マススペクトルにおいて表れた。
本発明のある態様はまた、ApoA1、トランスサイレチンΔN10,及び、IAIH4断片から選択される一つ又はそれ以上のマーカーと組み合わせて、既知の卵巣がんバイオマーカーを使用するものである。「マーカー4」という用語は、本明細書では既知の卵巣がんマーカーを表すのに使用される、マーカー4として有用なマーカーの例には、CA125、CA125II、CA15−3、CA19−9、CA72−4、CA195、TATI、CEA、PLAP、シアリルTN、ガラクトシルトランスフェラーゼ、M−CSF、CSF−1、LPA、p110EGFR、組織カリクレイン、プロスタシン、HE4、CKB、LASA、HER−2/neu、尿ゴナドトロピンペプチド、ダイアノンNB70/K、TPA、オステオポンチン、ハプトグロビン、及び蛋白質変異体(例えば開裂体やアイソフォーム等)が含まれるが、これらに限定されるものではない。
A)生体型
サンプルは、例えば卵巣がんの状態を定めることを望む女性などの生体から集められる。生体はその家系に基づいて卵巣がんの高度の危険性があることが決定されている女性でもあろう。他の患者には、卵巣がんである女性が含まれ、試験は患者が受ける治療または処置の効果を決定するために使用される。また、患者には、お決まりの試験の一部としてまたはバイオマーカーのベースラインレベルを確立するために、テストを受ける健常人が含まれるだろう。サンプルは卵巣がんと診断され、ガンを軽減するための処置を受け、またはおそらく回復した女性から集められることもできるだろう。
マーカーは生理学的サンプルの異なる型において測定されることができる。サンプルは好ましくは生理学的液体サンプルである。有用な生理学的液体サンプルの例には、血液、血清、血漿、膣分泌、尿、涙、精液等が含まれる。マーカーは全て血清内で見出されることから、本発明の態様において血清が好ましいサンプル源である。
マーカーは好ましくは、個々に開示されるいかなるバイオチップ、マルチウエルマイクロタイタープレートまたは樹脂のような、固体支持体に固定化された捕獲試薬により捕獲される。特に、本発明のバイオマーカーは、好ましくはSELDI蛋白質バイオチップ上に捕獲される。捕獲はクロマトグラフィーの表面または生体特異的表面において行われる。反応性表面を含むSELDI蛋白質バイオチップのいかなるものでも、本発明のバイオマーカーを捕獲及び検出するために使用されることができる。しかしながら、本発明のバイオマーカーは固定化された金属キレートによく結合する。IMAC−3及びIMAC30バイオチップは、ニトリル酢酸がキレートによりCu++やNi++のような遷移金属イオンを吸着するもので、本発明のバイオマーカーを捕獲するための好ましいSELDIバイオチップである。反応性表面を含むSELDI蛋白質バイオチップであれば、どれでも本発明のバイオマーカーの捕獲及び検出に使用できる。これらのバイオチップは生体分子を特異的に捕獲する抗体を運搬することができ、または、イムノグロブリンに結合するプロテインA又はプロテインGのような捕獲試薬とともに運搬することができる。それから生体分子は特異的な抗体を使用して溶液内で捕獲することができ、捕獲された生体分子は捕獲試薬を通じてチップ上で単離される。
たとえばバイオチップ又は抗体のような基質に一旦捕獲されれば、適切な方法であればどれでも、サンプル内の一つ又はそれ以上のマーカーを測定するために使用できる。たとえば、マーカーは、ガス相イオンスペクトロメトリー法、光学的手法、電気化学的手法、原子力マイクロスコーピー、放射頻度方法を含む、様々な検出方法により、検出及び/又は測定することができる。これらの方法を使用することで、一つ又はそれ以上のマーカーが検出できる。
バイオマーカーの検出及び/又は測定のひとつの好ましい方法は、マススペクトロメトリー、特に、「表面活性化レーザー脱離/イオン化」または「SELDI」を使用する。SELDIとは、分析物がプローブインターフェースを司るSELDIプローブの表面に捕獲される、脱離/イオン化ガス相イオンスペクトロメトリー(たとえばマススペクトロメトリー)の方法のことである。「SELDI MS」では、ガス相イオンスペクトロメーターはマススペクトロメーターである。SELDI技術は詳しく上述されている。ApoA1、トランスサイレチンΔN10、IAIH4断片は、それぞれm/z28043、m/z約12870.9、及びm/z3272として検出される。
他の態様では、イムノアッセイがサンプル内のマーカーの検出及び分析に使用される。この方法は、(a)マーカーに特異的に結合する抗体を提供し、(b)サンプルを抗体と接触させ、(c)サンプル内でマーカーに結合した抗体の複合体の存在を検出する、ことを含む。
たとえばマススペクトロメトリーにより、サンプルが測定され、データが産生されると、データはそれからコンピュータソフトウェアプログラムにより分析される。一般的にソフトウェアはマス由来のシグナルをコンピュータで読み取り可能な形に変換するコードを含んでいる。ソフトウェアはまた、シグナルが、本発明のマーカーまたは他の有用なマーカーに対応する「ピーク」を表しているか否かを決定するために、シグナル分析のためのアルゴリズムを適用するコードを含むこともできる。ソフトウェアはまた、試験サンプル由来のシグナルを「正常」及びヒトガンの典型的なシグナルの特徴と比較して、両シグナル間の適合の親密さを決定するアルゴリズムを実行するコードを含むこともできる。ソフトウェアはまた、試験サンプルが最も近く、それゆえ予想される診断を提供するコードを表示することを含むことができる。
好ましい態様では、血清サンプルは患者から集められ、上述したように陰イオン交換樹脂を使用して分画化される。サンプル内のバイオマーカーはIMAC銅プロテインチップアレイを使用して捕獲される。マーカーはそれからSELDIを使用して検出される。そのような試験においては、ApoA1、トランスサイレチンΔN10、IAIH4断片が検出できる。結果はコンピュータシステムへ挿入され、そのコンピュータシステムは、バイオマーカーをもともと決定するための学習アルゴリズムと分類アルゴリズムにおいて使用されたものと同じパラメータを使用するようにデザインされたアルゴリズムを含む。アルゴリズムは個々のバイオマーカーに関する受容されたデータに基づいて診断する。
個別的にいかなるバイオマーカーもが、卵巣がん状態を決定する上での助けとして有用である。まず、選択されたバイオマーカーが、例えばマススペクトロメトリーによる検出に続くSELDIバイオチップでの捕獲のような、ここに開示される方法を用いて、生体において測定される。それから、測定結果は卵巣がんの状態を、ガンでない状態から区別する対照の診断量と比較される。診断量はガンでない状態に比較して、ガン状態において特有のバイオマーカーがアップレギュレートまたはダウンレギュレートされているという情報を反映するだろう。当業界においてよく理解されているように、使用される特有の診断量は、診断する人の意向に基づいて、診断アッセイの感度や特異度を増強させるために調整されることができる。診断量と比較される試験量はよって、卵巣がんの状態を示す。
VIII.キット
原料と方法
サンプル
プロテオミックプロファイリングデータはグローニンゲン大学病院(グローニンゲン、オランダ)、デューク大学メディカルセンター(ダーハム、NC)、ロイヤルホスピタルフォーウイメン(シドニー、オーストラリア)、MDアンダーソン癌センター(ヒューストン、TX)で集められた全部で503例の血清試料から過去にさかのぼって得たものである。卵巣がんの群は、65人のステージI/IIの浸潤性上皮卵巣がん患者、88人のステージIII/IVの浸潤性上皮卵巣がん患者、28人の境界型腫瘍の患者及び14人の再発性患者からなる。ガンのケースはFIGOクライテリアに基づく病理学者により最適なステージに分けられる。ステージI/IIの浸潤性卵巣がん患者のうち、20人は重症、17人は粘液性、15人は子宮内、8人は明細胞、1人はがん肉腫、及び4人は混合上皮がん腫であった。サンプルはまた、良性骨盤塊と診断された166人の患者及び142人の健常人を対照として含むものであった。年齢及びCA125レベルを含む研究集団(群)の特徴と基本的な記述統計を、表1に記す。
血清分画:血清サンプルは氷上で溶かされ、それから沈殿を除去するために20000gで10分間遠心された。血清の20μlは30μlの変性緩衝液(U9:9Mのウレア、2%のCHAPS、50mMのトリスpH9.0)と混合され、4℃で20分間撹拌された。個々のサンプルのために、180μlのハイパーQDFアニオン交換樹脂が、200μlのU1緩衝液(50mMのトリスpH9.0で1:9に希釈されたU9)で三回平衡化された。変性した血清は樹脂に添加され、30分間結合させられた。結合しない物質は集められ、それから100μlの0.1%OGPを含む50mMトリス9.0が樹脂へと添加された。この洗浄物は収集され、結合しない物質(フロースルー、分画1)とあわせられた。分画はそれからpH7,5,4,3の洗浄緩衡液と有機溶媒それぞれ100μlを二回使用する段階的pH勾配で集められた。これで、全部で6つの分画が集められた。分画はバイオメック2000自動化液体ハンドラー(ベックマン)及びマイクロミックスシェイカー(DPC)で処理された。対照の収集されたヒト血清(インタージェン)のサンプルはアッセイをモニターするために、同様に処理された。

血清変性
20μlの血清を96穴V底プレートにピペットする。血清を含む個々のウエルに30μlのU9を添加する。96穴プレートをシーリングフィルムで覆う。樹脂が平衡化される間、少なくとも20分間4℃で撹拌する。
50mMのトリス塩酸 pH9.0の3ベッド(bed)容量で樹脂を5回洗浄する。これは50mLの遠心チューブで行われる。樹脂に50mMのトリス塩酸 pH9.0の当量を添加することで樹脂の50/50スラリーを調製する。180μlの50/50スラリーを96ウェルフィルタープレートの個々のウェルに添加する。樹脂の緩衝液に対する比率を一貫したものに保つために、スラリーを含む(2又は3分取毎に)チューブを定期的に撹拌する。それから緩衝液をろ過し、200μlのU1を添加し、再度ろ過する。これを同様にさらに2回行う。
次に血清を樹脂に結合させる。本工程の最初の段階は、フィルタープレート内の対応するウェルに50μlの個々のサンプルをピペットする。次にサンプルプレートの個々のウェルに50μlのU1を添加し、5分間混合する。それからサンプルプレートの個々のウェルから50μlをフィルタープレートの対応するウェルへピペットする。30分間4℃で撹拌する。
チップ結合手順までの間、分画は凍結させておく。
バイオマーカー検索: M/Z2kD−50kDのマスの幅で限定されたマスピークが(S/N>5、0.3%のクラスターマスウインドウ)、SELDIスペクトルから選択された。興味あるスペクトルの幅でのデータのばらつきのより一貫したレベルを得るために、さらなる分析の前に対数変換がピーク強度に適用された。初期の上皮卵巣がん患者、及び、デューク大学医療センター(Ca n=36、HC n=47)並びにグロニンゲン大学病院(Ca n=20、HC n=30)由来の健常コントロールのデータ強度が、まずマイクロアレイデータ分析に、次いで蛋白質発現データ分析(プロピーク、3Zインフォマティックス)に用いられた、ユニファイドマキシマムセパラビリティー分析(UMSA)アルゴリズムを使用して分析された。(Li Jら、Clin Chem 2002, 48: 1296-304、Rai AJら、Zhang Zら、Arch Pathol Lab Med 2002, 126: 1518-26、Zhang Zら、Applying classification separability analysis: papers from CAMDA ’00 Boston: Kluwer Academic Publishers, 2001: 125-136; Zhang Zら、Fishing Expression- a Supervised Approach to Extract Patterns from a Compendium Of Expression Profiles. 全てのマーカーにおいて、血清は蛋白質発現プロファイリングに使用されたアニオン交換プロトコールを使用してはじめに分画化された。個々の精製工程では、分画はNP20又はIMAC−銅プロテインチップアレイにおいてモニターされた。
精製された蛋白質はトリプシンで消化され、トリプシンで消化された断片はプロテインチップリーダーで分析された。個々のスペクトルは平均少なくとも250レーザーショットであり、既知のペプチドの混合物に対して外見的に校正され、又はトリプシン自動溶解およびマトリックスピークを使用して内部的に校正された。ピークマスはプロファウンド検索ペプチドマッピングサイト(http://129.85.19.192/profound_bin/WebProFound.exe)に供された。蛋白質配列はNCBIデータベースを使用して得られた。これらのデータベース合致は、プロテインチップアレイインターフェース(サイファーゲン)を装着したPE Sciex QStar(コンコルド、カナダ)により確かめられた。MS/MS実験では、スペクトルはサイファーゲンPCI1000プロテインチップ(登録商標)アレイインターフェースを装着したSciex QStar(コンコルド、オンタリオ、カナダ)タンデム四極飛行時間マススペクトロメーターで得られた。イオンは、平均電圧流が130μJ/mm2の秒辺り30パルスで作動する、電圧をかけられた窒素レーザー(レーザーサイエンスVSL337NDS、フランクリン、MA、USA)を使用してつくられた。窒素ガスは、全ての低エネルギー衝突誘起解離(CID)実験と同様に、形成されたイオンの衝突冷却のために、圧力10mトールで使用された。適用された衝突エネルギーは一般に50eV/kDaの支配へ続く。MS及びMS/MSモードでは、システムは既知のペプチドの混合物を使用して外見的に校正された。蛋白質の同定はUCSFプロテインプロスペクターMS−タグプログラム(http://prospector.ucsf.edu)を使用して行われた。MS−タグによるデータベース検索は、以下の値を使用して行われた。ヒト、トリプシン消化(許容される解裂の二つがかけた)、カルバミドメチル化により修飾されたシステイン、親及び断片イオンマス許容性50ppm、NCBI又はSwiss−Protデータベース。
これらの同定は、EIA又はプロテインチップアレイに基づくイムノアッセイにより確かめられた。
発見セット内で、初期の卵巣がん患者と健常人の対照との間における、3つのバイオマーカーの発現レベルにおける違いは統計学的に顕著であった(m/z12828及び28043のマーカーではP<0.000001、m/z3272のマーカーではP<0.003)(表1)。
非線形UMSA分類を使用して、二つの多変量予測モデルが構築された。最初のものは、その入力として3つのバイオマーカーのみを使用し、二番目のものはCA125レベルとともに、3つのバイオマーカーを使用した。図2のパネルE−Hは、二つのモデルの全体的な診断性能を、ROC分析を使用してCA125の全体的な診断性能と比較している。パネルE−Hでは、○:CA125、□:3つのバイオマーカーを使用する多変量モデル、△:3つのバイオマーカーとCA125を使用する多変量モデル。トレーニングデータでは、0.5のカットオフ値が感度及び特異度の合計をおおよそ最大化した。このカットオフを使用して、モデルは試験データと個々の検証データに適用された(表2)。独立した検証セットにおいて健常対照とステージI/II浸潤性卵巣がんとを差別するために、82.6%の感度(95% Cl 61.2−95.1%)で三つのバイオマーカーとCA125を使用する多変量モデルが、93.7%(84.5−98.2%)の特異度を有していた。表2はまた、独立した検証セットで、良性状態、後期浸潤がん、または境界の腫瘍である患者に関する結果を含んでいる。
マイクロタイタープレート形式(和光化学、USA)で行われる濁度免疫アッセイを使用して、アポリポプロテインA1について、そして、ジメンションRxL器具(デイド−ベーリング)での、粒子増強濁度免疫アッセイを使用して、トランスサイレチンΔN10について、142の試料が分析された(表3)。
図4において図式的に描かれる分類アルゴリズムに関して、モジュール1−3はUMSA学習アルゴリズムでトレーニングされた。しかしながら、最終分類モジュールは、通常の支持ベクター機械分類と同じ数学的形態を有していた。

Claims (32)
- (a)生体由来のサンプルにおいて、ApoA1、トランスサイレチンΔN10、及びIAIH4断片からなる群より選択される複数のバイオマーカーを測定し、(b)その測定結果を正常な対照と比較して、生体における卵巣がんの状態を認定する方法。
- 前記バイオマーカーが、ApoA1及びIAIH4断片である請求項1に記載の方法。

- さらなるバイオマーカーが、CA125、CA125II、CA15−3、CA19−9、CA72−4、CA195、腫瘍関連トリプシン阻害剤(TATI)、CEA、胎盤アルカリンフォスファターゼ(PLAP)、シアリルTN、ガラクトシルトランスフェラーゼ、マクロファージコロニー刺激因子(M−CSF、CSF−1)、リゾフォスファチジック酸(LPA)、上皮成長因子受容体(p110GFR)の細胞外ドメインの110kD構成体、例えばカリクレイン6及びカリクレイン10(NES−1)等の組織カリクレイン、プロスタシン、HE4、クレアチンキナーゼB(CKB)、LASA、HER−2/neu、尿ゴナドトロピンペプチド、ダイアノンNB70/K、組織ペプチド抗体(TPA)、オステオポンチン及びハプトグロビンからなる群から選択されるものである、請求項6に記載の方法。
- さらなるバイオマーカーが、CA125である、請求項6又は7に記載の方法。
- 測定が、(a)血液又は血液由来の生体サンプルを提供し、(b)陰イオン交換樹脂でサンプル内の蛋白質を分け、ApoA1、トランスサイレチンΔN10、及びIAIH4断片からなる群より選択される複数のバイオマーカーを含む分画を集め、(c)当該バイオマーカーに結合する捕獲試薬を含む基質の表面で、分画由来の当該バイオマーカーを捕獲し、測定することを含む、請求項1〜8のいずれか1項に記載の方法。
- 基質が、IMAC銅表面を含むSELDIプローブであり、当該バイオマーカーがSELDIで検出されるものである、請求項9に記載の方法。
- 基質が、ApoA1、トランスサイレチンΔN10、及びIAIH4断片からなる群より選択されるバイオマーカーに結合する生体特異的親和性試薬を含むSELDIプローブであり、当該バイオマーカーがSELDIで検出されるものである、請求項9又は10に記載の方法。
- 基質が、ApoA1、トランスサイレチンΔN10、及びIAIH4断片からなる群より選択されるバイオマーカーに結合する生体特異的親和性試薬を含むマイクロタイタープレートであり、当該バイオマーカーがイムノアッセイで検出されるものである、請求項9〜11のいずれか1項に記載の方法。
- 測定が、前記バイオマーカーの存否を検出すること、当該バイオマーカーを定量すること、及び当該バイオマーカーのタイプを認定することから選択される、請求項1〜12のいずれか1項に記載の方法。
- 少なくとも一つの前記バイオマーカーが、バイオチップアレイを用いて測定されるものである、請求項1〜13のいずれか1項に記載の方法。
- バイオチップアレイが、蛋白質チップアレイである、請求項14に記載の方法。
- バイオチップアレイが、核酸アレイである、請求項14に記載の方法。
- 少なくとも一つの前記バイオマーカーが、バイオチップアレイに固定化されるものである、請求項14〜16のいずれか1項に記載の方法。
- 前記バイオマーカーが、SELDIで測定されるものである、請求項1〜17のいずれか1項に記載の方法。
- 前記バイオマーカーが、イムノアッセイで測定されるものである、請求項1〜15のいずれか1項に記載の方法。
- 正常な対照との比較が、ソフトウエア分類アルゴリズムにより行われるものである、請求項1〜19のいずれか1項に記載の方法。
- サンプルが、血液、血清及び血漿から選択されるものである、請求項1〜20のいずれか1項に記載の方法。
- ApoA1、トランスサイレチンΔN10、及びIAIH4断片からなる群より選択される複数のバイオマーカーに結合する捕獲試薬を含んでなる生体における卵巣がんの状態を認定するための診断剤。
- 前記バイオマーカーが、ApoA1及びIAIH4断片である請求項22に記載の診断剤。
- 前記バイオマーカーが、ApoA1、トランスサイレチンΔN10、及びIAIH4断片である請求項22に記載の診断剤。
- 捕獲試薬が、複数のバイオマーカーに結合するものである、請求項22〜24のいずれか1項に記載の診断剤。
- 捕獲試薬が、SELDIプローブである、請求項22〜25のいずれか1項に記載の診断剤。
- さらに、CA125に結合する捕獲試薬を含む、請求項22〜26のいずれか1項に記載の診断剤。
- さらに、第一の捕獲試薬が結合しないバイオマーカーの一つと結合する第二の捕獲試薬を含むものである、請求項22〜27のいずれか1項に記載の診断剤。
- (a)ApoA1、トランスサイレチンΔN10及びIAIH4断片から選択される少なくともひとつのバイオマーカーを結合する第一の捕獲試薬、及び(b)第一の捕獲試薬とは結合しないバイオマーカーの少なくとも一つと結合する第二の捕獲試薬、を含む請求項28に記載の診断剤。
- 少なくとも一つの捕獲試薬が抗体である、請求項22〜29のいずれか1項に記載の診断剤。
- 少なくとも一つの捕獲試薬が結合している、又は結合できるMSプローブをさらに含む、請求項22〜30のいずれか1項に記載の診断剤。
- 捕獲試薬が、固定化された金属キレートである、請求項22〜31のいずれか1項に記載の診断剤。
