Description
【0001】

BACKGROUND OF THE INVENTION
The present invention relates to an analgesic composition comprising a phenylpropionic acid antipyretic anti-inflammatory analgesic and a phenothiazine antihistamine.
[0002]
[Prior art]
Phenylpropionic antipyretic anti-inflammatory analgesics are widely used as analgesics and anti-inflammatory agents. Among these, for example, in Japan, for example, loxoprofen, ibuprofen, aluminoprofen, thiaprofen, fenoprofen or pranoprofen is also used for analgesia of acute upper respiratory inflammation such as cold.
[0003]
On the other hand, examples of the phenothiazine-based antihistamine include mequitazine, promethazine, alimemazine, or methodirazine, which are used to treat sneezing, nasal discharge, and cough associated with upper respiratory tract inflammation such as the common cold. Furthermore, in Japan, mequitazine or promethazine is also included in general cold medicine.
[0004]
As a known combination example of a phenylpropionic acid antipyretic anti-inflammatory analgesic and an antihistamine,
1) It was found that a combination of loxoprofen and mequitazine, which is a phenothiazine-based antihistamine, was effective in improving olfactory sensation in patients with nasal congestion (see, for example, Patent Document 1).
2) The analgesic action is enhanced by the combined use of loxoprofen and the aminoalkyl ether antihistamine carbinoxamine maleate (for example, see Patent Document 2).
3) The analgesic effect is enhanced by the combined use of ibuprofen and epinastine (for example, see Patent Document 3).
Is mentioned.
[0005]
However, phenylpropionic acid antipyretic anti-inflammatory analgesics cannot be said to enhance the analgesic action when used in combination with any antihistamine. For example, according to Patent Document 2, even if the analgesic action is enhanced, the degree is the kind of antihistamine. It varies greatly depending on.
[0006]
Furthermore, at present, there is no report that the analgesic action is synergistically enhanced by the combined use of a phenylpropionic acid antipyretic analgesic and a phenothiazine antihistamine.

[Table 4]
(2) Manufacturing method Take the above components and quantities, and make a syrup according to the Japanese general formulation “Syrup” section, then fill it into a brown glass bottle to make a syrup.
(Test Example 1 ) Analgesic effect (1) Test substance The test substance was used as a suspension in 0.5% tragacanth solution on the test day. The administration volume of the test substance was 2.5 mL per kg body weight, and the same amount of 0.5% tragacanth was administered to the control group.
(2) Animals
Wister-Imamichi Male rats 3 to 4 weeks old (purchased from the Institute for Animal Breeding) controlled at 20-26 ° C, humidity 30-70%, lighting time 7: 00-19: 00 (Japan) 6-8 animals in a stainless steel rat breeding gauge within Clair), and feed water (mouse / rat breeding F-2, Funabashi Farm) and tap water freely passed through a water filter. After 9 days of pre-breeding, fast from the day before the test (approximately 20 hours, water ad libitum). On the day of the test, the health condition was visually observed and a good animal was selected and used for the test.
(3) Method (1) Induction of pain by yeast The yeast solution is adjusted to a concentration of 20% by adding physiological saline (Otsuka Pharmaceutical) to brewer's yeast (yeast, Sigma Chemical) on the day before the test. Stored. Inflammation is induced by administering 0.1 mL of the yeast solution subcutaneously to the right hind footpad of rats.
(2) Pain threshold and grouped inflammation When 2, 4, and 4.5 hours later, the inflamed foot was pressurized using a balance-type pressure device (manufactured by Ugo Basile), and the rat showed hoarseness and struggling symptoms The pressure value (g) is measured and used as a pain threshold value. Rats having a pain threshold of 75 to 150 g 4.5 hours after yeast administration are selected and divided into groups (5 animals per group) so that the pain threshold is averaged.
(3) Analgesic action of test substance The test substance is administered using an oral sonde 5 hours after administration of yeast. Pain threshold is measured at 0.5, 1 and 2 hours after dosing. The pain threshold is measured in the same manner for the control group and the non-yeast control group.
[0032]
Rats whose test substance-administered rats had a pain threshold that was at least twice the average pain threshold of the control group were determined to be effective (Blane evaluation method), and the efficacy rate was calculated.
[0033]
ED 50 (50% effective dose) was calculated from the relationship between the effective rate and the dose (Probit method).
(4) Test result 1
