BACKGROUND OF THE INVENTION
The present invention relates to a medicament (especially used for the suppression of rhinitis, sneezing or runny nose) containing pseudoephedrine and an antihistamine (especially chlorpheniramine), the amount of either or both of pseudoephedrine and antihistamine And a drug containing pseudoephedrine and epinastine or ketotifen (especially those used for the suppression of rhinitis or sneezing or runny nose).
[0002]
[Prior art]
The internal medicine for rhinitis used for allergic rhinitis such as hay fever and nasal inflammation such as the common cold contains a sympathomimetic drug for removing nasal mucosal hyperemia and an antihistamine for suppressing allergic reactions. Sometimes. The reason why these drugs are used in combination is that nasal mucosal decongestants are effective for nasal congestion, while antihistamines are effective for nasal itchiness (sneezing), sneezing, and runny nose. This is because a complementary effect is expected (see, for example, Non-Patent Document 1). The following is well-known about the combination of such pseudoephedrine and an antihistamine. That is,
1) Oral medicine for rhinitis containing pseudoephedrine and chlorpheniramine or triprolidine (for example, see Non-patent Document 2),
2) Pseudoephedrine and chlorpheniramine for night use, divided into prescriptions that do not contain antihistamines for morning use, and filled in the same package to avoid the side effects of antihistamines such as drowsiness or sedation during the day Rhinitis drugs (see, for example, Patent Document 1),
3) A combination of pseudoephedrine and cetirizine (see, for example, Non-Patent Document 3),
4) A preparation of pseudoephedrine and diphenhydramine (for example, see Patent Document 2),
5) Combined use of pseudoephedrine and terfenadine (for example, see Non-Patent Document 4),
6) A combination of pseudoephedrine and acribastine (for example, see Non-Patent Document 5),
7) Combined use of pseudoephedrine and fexofenadine (see, for example, Non-Patent Document 6),
8) A combination of pseudoephedrine and astemizole (see, for example, Non-Patent Document 7), and
9) A combination of pseudoephedrine and loratadine (for example, see Non-Patent Document 8).
However, until now, there has not been known a medicine combining pseudoephedrine and epinastine or ketotifen.
In addition, in these combination examples, the compounding amount is set based on the dose of each single agent, and no combination agent that reduces the amount of either one or both of pseudoephedrine and antihistamine has been known.

(2) Induction of sneezing induced by antigens After checking and checking the health condition of the animals, the animals are distributed so that the weights of five animals per group are averaged. Next, 0.1 mL of the antigen solution (50 mg / mL) is dropped into each nasal cavity of the guinea pig. Thereafter, the symptom is observed for 15 minutes, and the number of sneezing is counted to obtain the intensity of the antigen-induced nasal symptom.
(3) Inhibitory action of test substance The test substance is administered using an oral sonde one hour before the antigen nasal drop in (2) above. The sneezing suppression rate (%) is calculated from the following formula.
[0025]
[Expression 1]
Inhibition rate (%) = [1-average number of sneezing in test substance administration group /
Average number of sneezing in control group] × 100
(4) Test results Table 5 shows the results of sneezing inhibition rates for each single agent and combination of pseudoephedrine hydrochloride [P (mg / Kg)] and chlorpheniramine maleate [C (mg / Kg)].
[0026]
[Table 5]
[0027]
Pseudoephedrine hydrochloride or chlorpheniramine maleate alone showed an excellent sneezing-inhibiting effect by combining both even at low doses where no sneezing-inhibiting effect was exhibited.
Table 6 shows the results of the sneezing inhibition rate for each single agent and combination of pseudoephedrine hydrochloride [P (mg / Kg)] and epinastine hydrochloride [E (mg / Kg)].
[0028]
[Table 6]
[0029]
Pseudoephedrine hydrochloride or epinastine hydrochloride alone showed an excellent sneezing inhibitory effect by combining both even at low doses that did not exhibit a sneezing inhibitory effect.
Table 7 shows the results of sneezing inhibition rates for each single agent and combination of pseudoephedrine hydrochloride [P (mg / Kg)] and ketotifen fumarate [K (mg / Kg)].
[0030]
[Table 7]
[0031]
The combination of pseudoephedrine hydrochloride and ketotifen fumarate improved sneezing suppression.
[0032]
