Description
。
1) 主にラット乳仔を対象としており、より小さなマウス乳仔を対象とした場合にはその成長に追随できない。
2) この装置では乳仔に対して必要に応じて手持ちでミルクを与えるような形態で簡便に試験液等を与えることはできない。従ってこの装置ではラット乳仔の自発的な意志に従った給餌は可能となったが必要に応じて試験液等を与える等、実験の運用上より細かく制御された形で定期的、定量的に試験液等を与えることは難しい。
3) マウス、ラット等の小動物用の人工乳首としてはこの装置の乳首は構造が複雑すぎる為、乳首間で性能差が出やすく給餌量等が安定しにくい。
試験方法; BALB/Cマウスを自家繁殖させ自然出産後、1日令から本発明のマウス用サイズ小の乳首と哺乳瓶を用いて人工哺育を開始した。人工哺育用の哺育ケージは市販のホットカーペット上で保温した。保温ケージ内に竹籠を入れ、中にチップを敷いて仔マウスを入れた。竹籠の外側のチップは常に湿り気を保つよう時々注水した。人工哺育開始の前には仔マウスを親から3−4時間離しておいた。人工乳は市販のイヌ用ミルクを温湯10 mlに3.8 g 溶かして調製した。該ミルクをポンプで人工乳首をセットした哺乳瓶の中に導入後、温水を還流しながら保温中の銅管に哺乳瓶をさしこみ約3―5分間加温した。加温は各乳仔に哺乳させる前には必ず、また必要に応じて随時行った。ミルクは朝8時頃から夜10時頃までの間に5−6回、10日令以後は4−5回授乳させた。ミルクの摂取量は、授乳の前後で哺乳瓶中の筒内のミルクの減少量(筒の長さ:cm)を測定することおよび哺乳前後で仔の体重を量ることにより行った。人工乳哺育仔では、夜10時頃から翌朝8時ころまでの授乳を行わなかった。人工哺育を開始した乳仔は群飼いし、以後一度も母獣哺育には戻さなかった。排尿および排便は必要に応じて日に数回膀胱の上腹部と背部を同時に軽く圧迫することにより行った。母乳哺育群の乳仔は母獣と同一の通常ケージ内で通常通りに飼育した。
実験結果:結果を図5に示す。
人工哺育仔は成長率においては母乳仔に劣っているものの本発明品を用いた結果、人工乳を給餌時に自発的に摂取し続け、1匹の脱落もなく成長することが確認された。
試験方法;日本SLCから妊娠12日目のBALB/C マウスを3腹購入し、クリーンキャップをかぶせたステンレスケージ内で自然出産させた。生後24時間以内に仔を親毎に2群に分け、それぞれの親からの仔が均等に含まれるように組み合わせて、2匹の親にそれぞれ仔の総数が8匹ずつになるように哺育させた。今実験で用いた試験液は以下のようである。即ち0.5%-フラクトオリゴ糖-0.2M-スクロース水溶液を試験液として用いた。この試験液を滅菌した先細のチップを接続したディスポーザブル注射器を用いて本発明のマウス用哺乳瓶に充填し、50℃のインキュベーターに4分間保温した。マウス乳仔は、生後1日目と2日目に親から離し、43℃の水槽に浮かべたプラスチックケージに2時間保温しておいた。マウス乳仔へ試験液を本発明のマウス用小サイズ人工乳首を用いマウス用哺乳瓶の目盛りで0.7 mm(約50 μl)飲ませたところで、直ちに乳首を口から離した。その後0.2M-スクロース水溶液を同上量飲ませた。こうすることにより、飲用試験液量を正確に決めることが出来た。対照群には、0.2M-スクロース水溶液を同様の方法で飲ませた。試験液投与後、マウス乳仔はただちに母獣(里親)のケージに戻した。
実験結果 乳仔の生育曲線を図6に示す。本発明品を用いて試験液を経口投与したマウス乳仔は順調に生育した。
経口的に薬剤等を投与してその影響を検討する実験系として胃ゾンデ法やカテーテル法の実施が難しいマウスやラット乳仔等の実験動物を対象とした場合でも本発明品が該実験系に有効に用いられることが確認できた。

Claims (7)
- 乳首内に交換可能で内径を維持できるステンレス管からなる細管部を有し、且つ乳頭部と細管部以外の乳首部分が弾力性のある部材で充填することで成されている実験動物用人工乳首を哺乳瓶に装着した実験動物用液体供給装置であって、哺乳瓶中に交換可能な筒状部分を有し、該筒状部分に計量用の目盛りをつけたことを特徴とする、前記実験動物用液体供給装置。
- 筒状部分に可動式の目印をつけたことを特徴とする、請求項1に記載の実験動物用液体供給装置。
- 実験動物用人工乳首の乳頭部に、切れ込みまたは孔が設けられている、請求項1または2に記載の実験動物用液体供給装置。
- 哺乳瓶が逆止弁を有する、請求項1〜3のいずれかに記載の実験動物用液体供給装置。
- 哺乳瓶が、実験動物が所定量又は任意量の液体を飲む事で哺乳瓶中の内圧が陰圧となって液体の流れが止まる機構を有する、請求項1〜4のいずれかに記載の実験動物用液体供給装置。
- 哺乳瓶が、実験動物が所定量又は任意量の液体を飲む事で哺乳瓶中の内圧が陰圧となって液体の流れが止まり、その時点で哺乳瓶外部から圧を加えることによってさらに実験動物が液体を自発的に飲むことが可能となる機構を有する、請求項5に記載の実験動物用液体供給装置。
- 請求項1〜6のいずれかに記載の実験動物用液体供給装置において、実験動物(人間を除く)の大きさや種類に応じて人工乳首の大きさ及び/又は形状を変化させることを特徴とする実験動物用液体供給方法。
