【発明の属する技術分野】
本発明は、植物に含有される抗糖尿病成分の抽出分離方法に関するものであり、さらに詳しくは、エゾウコギ((学名:Acanthopanax senticosusharms)(別名シベリア人参 Eleutherococcus))を原料とする植物性抗糖尿病成分の抽出分離方法および該抽出方法により得られる抗糖尿病成分含有画分ならびにその利用に関するものである。

【従来の技術】
生活習慣病としての糖尿病は世界的にも増加の傾向にある。我が国においても、糖尿病患者数は、人口の約5%にも達する状況にあり、疫学的に予測される肥満者等のいわゆる糖尿病潜在者はこの数倍にのぼると推定されている、また、近年には15才以下の子供にも糖尿病を発症する事例が増加している。かかる糖尿病の発症は、運動不足の生活環境における高カロリー食物の摂取等の現代社会の生活様式によるところが極めて大きいものと考えられる。そして糖尿病にはインスリン依存型とインスリン非依存型の二種があるが、現在、インスリン非依存型糖尿病が大部分を占めている。

かかる状況下において、多種の糖尿病治療薬が提案されているが、これらの糖尿病治療薬は、すべてインスリンの分泌を促進するための薬剤である。
しかしながら、従来提案されている糖尿病治療薬には副作用の強いものが多く、効果が疑問視されるものも多い。また、ほとんどのものにインスリン抵抗性成分が含まれていることが判明し、そのためにかかるインスリン抵抗性を抑制する成分が必要であることが新たに認識されるに至っている。

従って、インスリン抵抗性解除剤として、例えば、抗酸化性ビタミンE、Cおよび天然カロテノイドを強化した食用人参ジュースにシベリア人参エキス(エゾウコギエキス)を配合した飲料が提案されている(特許文献1;特開2000−247898号公報)。しかし、提案されたシベリア人参エキス配合物は、インスリン抵抗性解除剤としての効果は未だ十分でなく、また、シベリア人参エキスの抽出にその植物体の全体または根部分を、そのまま用いることは多くの狭雑物の混入を許し、その効力を希薄にするばかりでなく、阻害物の混入により目的とする効力を著しく低減させるという問題点があった。また、エゾウコギ類の有機成分の解析などにより、種々の成分が知られているが(非引用文献1;p.1101〜1106)、これらの成分のうち、糖尿病に有効な成分は、植物体のどの部位に存在するのか、またはどの画分に存在するかなどについては全く知られていなかった。

さらに、エゾウコギ等に含有される抗糖尿病成分の科学的構造が具体的には開示されていないので、該有効成分の含有される画分を選択して用いることはできなかった。

また、従来から提案されているエゾウコギエキスの抽出方法としては、例えば、熱水を用いる方法、エタノールまたはエタノールと蒸留水を用いる方法等が提案されている。熱水を用いる抽出方法により得られる抽出エキスは、ストレス性胃潰瘍、ストレス性急性腸炎、ストレスによる不眠症、ストレスによるイライラ、ストレスによる円形脱毛症、小人症、パーキンソン病、注意力不足活動過多症等の治療または予防に用いることが開示されている(特許文献2;特開2002−3391号公報)。また、特許文献3(特開平10−136938号公報)によれば、シベリア人参の根を40%エチルアルコールにより還流抽出して得られるエキスを生理活性賦活飲料の成分として用いることが提案されている。

しかしながら、従来のいずれの方法によっても抗糖尿病成分を十分効率よく選択して抽出することができず、前記特許文献等においてもその開示についてはもちろん示唆する点もなかった。

【発明が解決しようとする課題】
従って、本発明の課題は、前記の如き開発状況に鑑み、エゾウコギ植物体に含有されている成分を化学的に把握し、該植物体の含有成分のうち、糖尿病に有効な抗糖尿病成分のみを特定し効率よく分離すると共に、植物体全体に含まれる種々の狭雑物および糖尿病への有効性に寄与しない成分を排除することにより、高純度の抗糖尿病成分を含有する画分を収得することが可能な新規抽出分離方法を提供することにある。

【課題を解決するための手段】
そこで、本発明者らは、前記課題の解決を図り、植物体全体または部分に含有される抗糖尿病成分を効率よく抽出し、高濃度で分画可能な新規抽出分離方法を提供することを目的に鋭意検討を重ねた結果、従来提案されているアルコール類からなる抽出溶媒に特定の塩類を配合した混合溶媒を用いることにより抗糖尿病成分の抽出効果が向上し、かつ、クロマトグラフィーにより抗糖尿病成分の選択的な分離が可能となることを見い出し、これらの知見に基づいて本発明の完成に到達したものである。

かくして、本発明によれば、
エゾウコギ(学名:Acanthopanax senticosusharms)の植物体を
塩類およびアルコールからなる抽出溶媒と抽出条件下において接触させ;
前記抽出工程にて得られた抽出物を含有する抽出溶液からクロマトグラフィーにより抗糖尿病成分を分画し;
前記分画工程にて得られた抗糖尿病成分含有画分を収得する;
ことを特徴とする植物性抗糖尿病成分の抽出分離方法
が提供される。

本発明は、前記の如き、エゾウコギに含有される抗糖尿病成分をアルコール/塩類からなる特定の抽出溶媒を用いて抽出する方法および該抽出方法により得られる抽出物から抗糖尿病成分を分画する方法ならびに該抽出分離方法により得られる抗糖尿病成分含有画分に関するものであるが、さらに、好ましい実施の態様として次に示す1)〜8)に掲げるものを包含する。

1)前記エゾウコギの植物体が、根、根茎、茎または葉である前記抗糖尿病成分の抽出分離方法。
2)前記塩類が、塩化カリウム、塩化ナトリウムおよび塩化リチウムからなる群より選択される少なくとも一種の塩である前記抗糖尿病成分の抽出分離方法。
3)前記塩類の配合量が、抽出溶媒全重量基準で1〜8重量%である前記抗糖尿病成分の抽出分離方法。
4)前記抽出工程が、前記エゾウコギの植物体と前記アルコール水溶液と接触させる第一の抽出工程と、該第一の抽出工程にて得られた抽出物を前記塩類と前記アルコールとの混合水溶液と接触させる第二の抽出工程からなる前記抗糖尿病成分の抽出分離方法。
5)前記第一および第二の抽出工程において、前記エゾウコギの植物体に対する前記抽出溶媒の添加量がそれぞれ重量基準で1〜5倍である前記抗糖尿病成分の抽出分離方法。
6)前記エゾウコギの前記抽出物の分画工程において、液体クロマトグラフィー用の疎水性充填剤が、粒状スチレン系重合体である前記抗糖尿病成分の抽出分離方法。
7)前記抗糖尿病成分含有画分が、前記分画工程の溶出物をさらに濃縮処理された液体または粉状体である前記抗糖尿病成分の抽出分離方法。
8)エゾウコギの植物体と、抽出溶媒全重量基準でアルコールを2.5〜25重量%含有する水溶液に塩化カリウムを2〜5重量%添加してなる抽出溶媒とを接触させることにより抽出物を含有する抽出溶液を調製し、該抽出溶液を、疎水性担体によるカラムクロマトグラフィーにおいて、アルコールの濃度勾配によりインスリン抵抗性解除活性成分を分画することからなる前記抗糖尿病成分の抽出分離方法。

【発明の実施の形態】
以下、本発明について詳細に説明する。
本発明の植物性抗糖尿病成分の抽出分離方法に用いられるエゾウコギ(学名:Acanthopanax senticosusharms)は、別名シベリア人参(Eleutherococcus)と称され、朝鮮人参と同様にウコギ科(Araliaceae)に属する植物であり、北海道東部、ロシアおよび韓国等には自生しているが、栽培・増殖方法も検討または実施されている。前記抗糖尿病成分の抽出にはエゾウコギの植物体、特に、茎、根または葉等の部位が主として用いられる。また、かかる植物体は、採取した生の状態のもの、または乾燥したもの、さらには蒸焼に供したもの等のいずれの処理によるものでもよいが、乾燥処理したものが好ましい。

(抽出工程)
本発明の抗糖尿病成分の抽出分離方法において、抽出工程は、エゾウコギ植物体と抽出溶媒とを抽出条件下において接触させ、該エゾウコギから前記抗糖尿病成分を含有する抽出物を含有する抽出溶液を得る工程を包含する。

抽出工程においては、抽出原料としてのエゾウコギの前記の茎、根および葉等の植物体を所定量の抽出溶媒に浸漬し、ブレンダー、ミキサーで攪拌することにより該植物体を細かく粉砕すると同時に含有成分を抽出溶媒中に抽出させた抽出物を粉砕物および溶媒等の除去処理に供される。

前記エゾウコギの植物体と抽出溶媒との接触による抽出工程は、次の二種のいずれかの方式を採用することができる。
すなわち、
(1)抽出容器内にエゾウコギ植物体を採取し、これに、塩類とアルコールの混合溶媒を添加し、抽出条件下においてミキサーを攪拌し該植物体の粉砕処理を行なうと同時に含有成分の抽出を行ない、得られた粉砕物含有抽出溶液を粉砕物等の除去処理に供する方式。

または、
(2)抽出容器内にエゾウコギ植物体を採取し、これにアルコール溶媒を添加し、ミキサーにて該植物体の粉砕処理を行なった後、粉砕物含有溶液に塩類を添加し、抽出条件下にて抽出処理に供し、得られた粉砕物含有抽出溶液を粉砕物等の除去処理に供する方式。
のいずれかの方法が挙げられる。

抽出溶媒は、溶媒全重量基準でアルコールを1〜30重量%および塩類を1〜10重量%の割合で含有させた水溶液である。特に、アルコール含有量5〜25重量%、塩類含有量2〜8重量%を含有させた抽出溶媒が好ましい。アルコール含有量が1重量%に達しないと抗糖尿病成分がほとんど抽出されず、一方、30重量%を超えても増量に見合う効果は得られない。また、塩類含有量が1重量%に満たないと抗糖尿病成分の抽出が十分でなく、一方、10重量%を超えても増量に見合う効果が得られないばかりでなく、後続の分離除去操作が煩雑になるという難点が生ずる。

前記抽出工程の前記方式(2)においては、抽出溶媒として第一段ではアルコール1〜30重量%の水溶液を用い、第二段では塩類1〜10重量%を添加したものを用いることができる。

抽出溶媒のアルコールとしては脂肪族低級アルコールが好ましく、例えば、メタノール、エタノール、プロパノール、イソプロパノール、ブタノールおよびイソブタノール等からなる群より選択される少なくとも一種のアルコールを挙げることができるが、実用上エタノールが好ましい。

また、塩類としては、アルカリ金属ハロゲン化物およびアルカリ土類金属ハロゲン化物からなる群より選択される少なくとも一種の金属ハロゲン化物を用いることができる。例えば、塩化カリウム、塩化ナトリウム、塩化リチウム、塩化カルシウム、塩化マグネシウム、塩化バリウム、塩化ストロンチウム等を挙げることができるが、これらのなかで、アルカリ金属ハロゲン化物、特に塩化カリウムが好ましい。該塩類の前記抽出処理における作用については、まだ十分に解明されてはいないが、エゾウコギ植物体内からの抗糖尿病成分の抽出促進に有用であり、本発明者らの検討により、かかる塩類をアルコールと共存させることによりエゾウコギから前記抗糖尿病成分、特にインスリン抵抗性解除活性成分の効率的な抽出をはじめて達成したものである。

エゾウコギ植物体に対する抽出溶媒の添加量は、特に限定されるものではなく、植物体が十分浸漬される量であれば、任意に決定することができるが、通常、植物体に対し、重量基準で1〜5倍の割合が好適である。

前記抽出工程における抽出条件は、エゾウコギから抗糖尿病成分が可及的完全に抽出されるように適宜調整することができるが、温度120〜200℃、好ましくは120℃以上であり、圧力としては、1〜5気圧、好ましくは1.5〜3気圧の範囲で抗糖尿病成分が完全に抽出される抽出時間(浸漬時間)(例えば、30分以上)を採用することができる。

かかる抽出処理により、得られる抽出物は、必要に応じ精製処理に供し、植物体その他の破砕物、塩類および溶媒等が分離除去される。精製処理方法としては、濾過静置または遠心分離等の分離方法を採用することができる。
精製された抽出物を含有する抽出溶液は、抗糖尿病成分を含有するアルコール水溶液であり、次の分画工程に供される。

(分画工程)
本発明に係る抗糖尿病成分の抽出分離方法における分画工程は、前記の抽出工程にて得られた抽出物からクロマトグラフィーにより抗糖尿病成分を分画する工程を包含する。すなわち、該分画工程は、尿中のグルコース量、クレアチニン量およびインスリン前駆体であるC−ペプチド量の減少効果を奏する画分を他の成分画分から分離するものである。具体的には、前記抽出物を含有する抽出溶液を移動相とし、疎水性充填剤を固定相とするカラムクロマトグラフィー(液−固クロマトグラフィー)により、前記抽出物をカラム充填剤の充填層に通過させ、抗糖尿病成分を充填剤上に吸着させた後、該充填剤層に展開溶媒を通過させ、前記充填剤に吸着された前記抗糖尿病成分を溶出させ、カラムの下端より流下させて、該抗糖尿病成分含有画分を収得するものである。

前記カラムクロマトグラフィーにおいて用いられるカラム充填剤としては、抽出物中の抗糖尿病成分の性質上、疎水性担体が好適である。疎水性担体としては、芳香族系、置換芳香族系、またはアクリル系の化学構造を有する多孔性合成吸着剤から選択し使用することができる。

を有する架橋スチレン系多孔質重合体を、また、置換芳香族系吸着剤としては該芳香族重合体の芳香核に臭素原子を結合させたものを挙げることができる。市販品としては、例えば三菱化学(株)製DIAION HPシリーズ、SEPABEDAS SP シリーズ等を選択して使用することができる。カラム充填剤の形状は、特に限定されるものではないが、通常、粒状体のものが好ましく、粒度分布として粒径>250μmの粒子が90%以上含有するものを採用することができる。また、細孔容積1.3ml/g以上、比表面積600m2/g以上の多孔性吸着剤が好ましい。

本発明に係る抗糖尿病成分の分画に用いられるクロマトグラフィーとしては、操作上の形式は任意に選択することができるが、カラムクロマトグラフィーが好適であり、具体的には、液体クロマトグラフィーまたは高圧下での高速液体クロマトグラフィー(HPLC)を採用することができる。

また、前記抗糖尿病成分の抽出物を含有する抽出溶液を固定相とする逆相クロマトグラフィーを利用してもよい。
高速液体クロマトグラフィー(HPLC)を用いる場合の分画条件としては、温度20〜25℃、圧力10〜15kg/cm2 および流通速度0.2〜0.5ml/分の範囲が好ましく、これらの条件は任意に選択することができる。

前記カラムクロマトグラフィーにおいて、カラム内の充填剤に保持された抗糖尿病成分の溶出に用いられる展開溶媒としては、通常、脂肪族低級アルコールが好ましく、例えば、メタノール、エタノール、イソプロパノール等が挙げられるが、エタノールが好ましい。

(濃縮工程)
前記抽出・分画処理を経て得られる前記抗糖尿病成分含有画分は、必要に応じ濃縮処理に供され、アルコール溶媒と共に水分が除去される。濃縮処理には、例えば、エバポレーター等を任意に選択し使用することができる。また、濃縮処理により抗糖尿病成分画分は、溶液または粉末のいずれかの形態で得ることもできる。

前記抽出工程および分画工程を経て得られた抽出物の画分成分は、極性を有する低分子量水溶性成分であり、抗糖尿病成分を含有する。かかる抗糖尿病成分としては、疎水性および親水性の両方の性質を有するものであり、分子量200〜10000の糖鎖ペプチドおよび蛋白質を含有するものである。

該抗糖尿病成分は、インスリン抵抗性解除活性成分を少なくとも25〜50%以上含有するものであり、第II型糖尿病モデルマウスに投与することにより、図1、図2および図3に示すように、尿中に放出されるグルコース排泄量、クレアチニン排泄量およびインスリン前駆物質C−ペプチド排泄量を著しく低減させることができる。かかる成分の排泄量が低減することは糖尿病が改善されていることを示すものである。

前記抗糖尿病成分含有画分には、ビタミン類、甘味料および栄養補助剤を配合することによりインスリン抵抗性解除に効果を有する飲料を提供することができる。ビタミン類としてはビタミンA、CおよびビタミンE等を、また、甘味料および栄養補助剤としては通常使用されているものを選択して用いることができる。
また、さらに、賦形剤を配合し、例えば錠剤状の成形物を提供することもできる。

【実施例】
以下、本発明について実施例および比較例により具体的に説明する。もっとも、本発明は、これらの実施例等により限定されるものではない。

実施例1
エゾウコギの根茎および根の部分1kgに2.5%のエチルアルコールを含有する蒸留水を2.5kg加え、大型ミキサーで5分間粉砕した。
さらに、このようにして得られた粉砕物を含有する溶液に塩化カリウムを4.0重量%加え、120℃に加熱し、2気圧で55分間抽出を行なった。抽出液をガーゼにより濾過した後、2000回転で15分間遠心分離し1kg以上の上澄液を取得した。

次に、この上澄液を直径10cm、高さ20cmの疎水性担体(三菱化学(株)製合成吸着剤ダイヤイオンHP20)充填カラムに通過させ、カラムクロマトグラフィーにより分画化した。得られた画分は、極性を有する低分子量の水溶性成分であり、植物体に比較して5〜10倍の高い抗インスリン抵抗性活性を示すものであった。

抗インスリン抵抗性活性は、第II型糖尿病モデルマウスKK-AY/Ta Jcl(日本クレア(株))を用い、64日間の投与実験により、尿中に排泄されたC−ペプチド量を定量することにより判定した。

投与実験においては、前記第II型糖尿病モデルマウス KK-AY/Ta Jcl に体重1gに対し、抽出液17.3mgまたは分画液16.2mgを水溶液として1日1回経口投与した。尿中のC−ペプチドの定量はHPLCによる定量曲線から算出した(N=5)。

また、グルコース排泄量、クレアチニン排泄量についても分画液についてそれぞれ定量し、図2および図3に示す。いずれも投与開始後15日経過時には著しく減少したことが示されている。

さらに、投与実験に供した前記モデルマウスの投与期間における体重の変化を観察したところ、図4に示す通り増加の傾向にあり、投与成分による体調への著しい阻害効果はないものと推定される。

図1に前記抽出液および分画液をそれぞれ用いた場合の尿中C−ペプチド量を示した。同図により、投与開始には1時間当たりの尿中C−ペプチドの排泄量が5μgを超えていたものが2週間後には1μgに激減し、しかも開始時には抽出液と分画液との間にC−ペプチド量の差がなかったが、投与日数の経過と共に分画液を用いた場合の尿中C−ペプチド量が抽出液の投与の場合に比較して著しく減少していることが確認された。

この結果、分画液の抽出液に対するC−ペプチド排泄量の抑制効果が極めて大きく、従って、分画液が顕著なインスリン抵抗性解除効果を奏することが把握された。

(安全性評価)
前記抗糖尿病成分について以下の如く安全性を評価し、その結果、本発明により得られる抗糖尿病成分が有用であることが示された。
(1)急性
10週令のマウスに、1gの体重あたり1mlの画分水溶液(固形物10〜30重量%含有)を11週間経口で投与し、31gから46gの体重の増加を得ており体調に変化はみられなかった。
(2)催奇性
予想されない。
(3)突然変異性
UME試験((株)日本抗体研究所)により陰性であり、ほとんど影響しない。
(4)精子変異性
予想されない。
(5)悪急性
急性と同様に体調に変化はみられなかった。

比較例1
塩化カリウムを使用しなかったこと以外すべて実施例1と同一の原材料および同一の操作により分画液を調製した。投与実験の結果によると、抽出液および分画液による尿中C−ペプチド排泄量は、投与後30日経過時においてそれぞれ3μg/時間および2μg/時間であり、抽出効率の低いものであり、また、インスリン抵抗性解除効果も十分得られなかった。

【発明の効果】
以上述べた如く、本発明に係る抽出工程および分画工程によりエゾウコギの植物体から抗糖尿病成分、例えば、インスリン抵抗性解除効果を示す画分を高効率で取り出すことができる。第II型糖尿病モデルマウスによる投与実験でも尿中に排泄されるグルコース量、クレアチニン量、インスリンの前駆体であるC−ペプチド量が投与日数14日程度で激減するという著しく顕著な効果を奏するものである。
